This study was performed to investigate the effects of peking-duck extracts added with medicinal herbs(DJ) on the intoxication of heavy metals (Hg, Pb, Cd, As) in rats. Sprague-Dawley rats weighing 150$\pm$15g, were randomly assigned to groups: basal diet only in normal control group(NCG): basal diet and heavy metals without DJ injection in heavy metal control group(HMC); basal diet, heavy metals and DJ(3mg/ml) injection in heavy metal low duck-juice group(HMLD), basal diet, heavy metals and DJ(30mg/ml) injection in heavy metal middle duck-juice group(HMMD): basal diet, heavy metal and DJ(300mg/ml) injection in heavy metal high duck-juice group(HMHD). Hg and As was injected by 50ppm and Cd and Pb by 25ppm for 17days. Also DJ oral feeding was conducted for 28days. The result of this study were as follows: Food intake and body weight gain in heavy metal administered groups were lower than those of NCG. Liver, kidney and testis weights were not significantly different among 5 groups. GOT, GPT and BUN activities were significantly reduced in DJ treated groups as compared to HMC. DJ showed the suppressing effect on the accumulation of Hg, Pb and Cd in serum, liver and kidney. Fecal Hg and Cd excretions increased with DJ feeding. The results suggested that DJ may have some protective effects on Hg, Cd and Pb intoxication by reducing the accumulation in tissues and increasing excretion. This study also showed the effective way of using duck-extract and its application to the oriental medicine.
Objectives : For the screening of neuroprotective effects of medicinal herbs, the complex system of animal models suffer some disadvantages in controlling critical parameters such as blood pressure and body temperature. Additionally, application of drugs to the appropriate brain area sometimes is difficult, due to poor permeability though the blood brain barrier, and so potential protective effects might be masked. Methods : Organotypic hippocampal slice culture (OHSC) method has the advantages of being relatively easy to prepare and of maintaining the general structure, including tissue integrity and the connections between cells. Drugs can easily be applied and neuronal damage can easily be quantified by using tissues and culture media. This study demonstrates neuroprotective effects of Puerariae radix (葛根, PR), Salviae miltiorrhizae radix (丹蔘, SR), Rhei rhizoma (大黃, RR), and Bupleuri radix (柴胡, BR). These were screenedand compared to MK-801, antagonist of NMDA receptors, by using OHSC of 1 week-old Sprague-Dawley rats. Oxygen/glucose deprivation (OGD) were conducted in an anaerobic chamber $(85%\;N_2,\;10%\;CO_2\;and\;5%\;H_2)$ in a deoxygenated glucose-free medium for 60 minutes. Water extracts of each herbs were treated to culture media with $5\;{\mu}g/ml$ for 48 hours. Results : Neuronal cell death in the cultures was monitored by densitometric measurements of the cellular uptake of propidium iodide (PI). PI fluorescence images were obtained at 48 hours after the OGD and medicinal herb treatment. Also TUNEL-positive cells in the CAI and DG regions and LDH concentrations in culture media were measured at 48 hours after the OGD. According to measured data, MK-801, PR, SR and BR demonstrated significant neuroprotective effect against excessive neuronal cell death and apoptosis induced by the OGD insult. Especially, PR revealed similar neuroprotective effect to MK-801 and RR demonstrated weak neuroprotective effect. Conclusions : These results suggest that OHSC can be a suitable method for screening of neuroprotective effects of medicinal herbs. Obesity can be defined as a metabolic disease due to an increased fat accumulation in the body caused by an imbalance of calorie intake and output The prevalence of obesity has increased substantially over the past 2-3 decades in developed and developing countries. The health impact of weight gain is so marked that obesity has now been classified as a major global public health problem In order to investigate the effect of diet conversion and oral administration of Platycodon grandiflorum extracts on the treatment of obesity, male Spraque-Dawley rats were divided into four groups: a group converted to normal diet (Control group), a group maintained high fat (30%) diet (H), and two groups with Platycodon grandiflorum extract added to the previously mentioned two groups. All animals were fed high fat diet for 7 weeks to induce the obesity. Then they were divided as mentioned above. Animals were fed experimental diet and Platycodon grandiflorum extract (150 mg/ml/rat/day) for 7 weeks. Body weight, adipose tissue weight (subcutaneous, epididymal, peritoneal fat pads) and serum lipids (total cholesterol and triglyceride) showed some differences among groups. The Platycodon grandiflorum feeding markedly decreased both body weight and adipose tissue weight in control group compared to H, high fat diet maintaining, group. Platycodon grandiflorum extracts significantly decreased the concentrations of serum lipids compared to H group. Fat cell numbers and sizes were significantly reduced in the oriental medicinal herb extract administrated group. Increased fatty acid binding protein (FABP) expression in high fat diet group was decreased by the dietary conversion to normal diet and the oral administration of Platycodon glandiflorum extracts. In contrast, there was no significant effect on FABP expression in the high fat maintenance group. In this study, the conversion from high fat diet to low fat or normal diet had a beneficial effect on body weight loss and serum lipid profiles. Dietary Platycodon glandiflorum extracts had an additive beneficial effect on the prevention and treatment of obesity.
Objectives: The purpose of this study was to investigate the effects of herbal medicine treatment for patients with migraine and to suggest research methods for herbal medicine treatment on migraine. Methods: In this study, a search was conducted through several academic sites using a combination of terms '편두통', 'Migraine', '한약', '한약치료', 'Herbal medicine', 'Herbal medicines', 'Herb', 'Traditional Chinese Medicine', and 'TCM'. Randomized controlled trials using herbal medicine treatments for adult patients with migraine were selected. Results: Ultimately, 46 papers were selected and analyzed. A statistically significant improvement was noted in the treatment group in terms of clinical migraine symptoms and other migraine evaluation tools before and after the herbal medicine treatment. Herbal medicines were administered in decoction, pill, and granule formulations. Many kinds of medicinal herbs, such as 解表藥類, 補益藥類, 淸熱藥類, 活血祛瘀藥類, and 平肝藥類, have been used for migraine. Among them, 川芎, belonging to 活血祛瘀藥類, is mentioned 36 times and is the most frequently used medicine. Herbal medicine was used safely for migraine treatment, without major adverse reactions, and the recurrence rate was significantly lower in the treatment group than in the control group. Conclusions: In conclusion, the herbal medicine treatment for patients with migraine showed a statistically significant improvement in 46 papers. Future studies should utilize standardized and objective evaluation tools, along with appropriate experimental design. The relevant articles should be increased to a significant level to verify the effect of herbal medicine treatments on migraine.
We evaluated the hepatoprotective activity of crude hot-water extracts of the traditional Korean medicinal herb, Phellodendri Cortex (PC; Bark of Phellodendron amurense RUPR.), in an experimental model of hepatic damage induced by carbon tetrachloride ($CCl_4$). The serum marker of liver damage, sGOT, sGPT and sALP, were significantly decreased in the liver of the PC treated rats, compared with that of $CCl_4$ treated group. The histological observation of liver section of rats, showed the same protective effect of PC treatment. And the protective activity of PC was more significant in the post-treated group than pre-treated group. The significant decrease of malodialdehyde and increase of glutathion, catalase activity were observed in the liver homogenate of PC treated rats. Based on these findings, it is suggested that PC has potent hepatoprotective effects and the mechanism of the protection may be related to antioxidation pathways.
Clematis chinensis is the root of Clematis chinensis OSBECK and is classified in Ranunculaceae. Clematis chinensis is a traditional medicinal herb possesses analgesic, diuretic, anti-tumorigenic, and anti-inflammatory effects. In this study, the effect of aqueous extract of Clematis chinensis against lipopolysaccharide-induced inflammation was investigated in mouse BV2 microglial cells. The aqueous extract of Clematis chinensis at the respective concentration was treated one hour before the lipopolysaccharide treatment in mouse BV2 microglial cells. From the present results, pre-treatment with the aqueous extract of Clematis chinensis suppressed prostaglandin E2 synthesis and nitric oxide production by inhibiting on the lipopolysaccharide-stimulated cyclooxygenase-2 and inducible nitric oxide synthase expressions in mouse BV2 microglial cells. The present study suggests that Clematis chinensis may offer a valuable mean of therapy for brain inflammatory diseases.
