Production of Recombinant GG1234-DsRed Fusion Protein and Its Effect on in vitro CaCO3 Crystallization (재조합 GG1234-DsRed 융합 단백질의 생산 및 In vitro 탄산칼슘 결정화에 미치는 영향에 대한 연구)

  • Eggshell-based biocomposites have become attractive due to their exquisite nanostructure and biological properties, which are mainly composed of highly organized calcium carbonate crystals controlled by organic macromolecules such as proteins and polysaccharides. Here, we designed the recombinant fusion protein of a putative eggshell matrix protein named as GG1234 and a fluorescent reporter protein of DsRed. The protein was successfully over-expressed in E. coli and purified by Ni-NTA affinity chromatography. In vitro calcium carbonate crystallization was conducted in the presence of the fusion protein, and morphological change was investigated. The protein inhibited the calcite growth in vitro, and spherical calcium carbonate micro-particles with the diameter of about $20-30{\mu}m$ were obtained. We expect that this study would be helpful for better understanding of eggshell-based biomineralization.

  • This review paper evaluates different kinds of synthesis methods for calcite precipitated calcium carbonates by using different materials. The various processing routes of calcite with different compositions are reported and the possible optimum conditions required to synthesize a desired particle sizes of calcite are predicted. This paper mainly focuses on that the calcite morphology and size of the particles by carbonation process using loop reactors. In this regard, we have investigated various parameters such as $CO_2$ flow rate, Ca $(OH)_2$ concentration, temperature, pH effect, reaction time and loop reactor mechanism with orifice diameter. The research results illustrate the formation of well-defined and pure calcite crystals with controlled crystal growth and particle size, without additives or organic solvents. The crystal growth and particle size can be controlled, and smaller sizes are obtained by decreasing the Ca $(OH)_2$ concentration and increasing the $CO_2$ flow rate at lower temperatures with suitable pH. The crystal structure of obtained calcite was characterized by using X-ray diffraction method and the morphology by scanning electron microscope (SEM). The result of x-ray diffraction recognized that the calcite phase of calcium carbonate was the dominating crystalline structure.

Solidification/Stabilization of Heavy Metals in Sewage Sludge Prior to Use as a Landfill Cover Material (매립지 복토재로의 활용을 위한 하수슬러지 내 중금속의 고형화/안정화)

  • The effects of chemical binders (ladle slag, ordinary portland cement (OPC), hydroxyapatite and calcium hydroxide) on the solidification/stabilization of heavy metals (Cd, Cu, Ni, Pb, Zn) in sewage sludge were evaluated by chemical leaching tests such as EDTA extraction, TCLP and sequential extraction. The results of EDTA extraction showed that heavy metal concentrations in sewage sludge were highly reduced after solidification/stabilization with slag, cement or calcium hydroxide. However, EDTA interrupted solidification/stabilization of heavy metals by hydroxyapatite. The TCLP-extracted heavy metal concentrations in sewage sludge after solidification/stabilization with chemical amendments were highly reduced. However, Cu concentration in the sewage sludge solidified/stabilized with slag, cement or calcium hydroxide increased because the pH of TCLP solution was higher than 7. Mixtures of sludge 1 : slag 0.2 : calcium hydroxide 0.1 (wt ratio) showed the least leachability in batch TCLP and EDTA extraction. The results of sequential extraction (SM&T, formaly BCR) indicated that the distribution of heavy metals changed from exchangable and carbonate fractions to strongly bound organic fraction. It was found that maximum leachate concentrations of Ba, Cd, Cr and Pb from sewage sludge amended with slag and calcium hydroxide were far below US EPA TCLP regulations.

Effects of compound organic acid calcium on growth performance, hepatic antioxidation and intestinal barrier of male broilers under heat stress

  • Objective: The aim of this study was to evaluate the effects of compound organic acid calcium (COAC) on growth performance, hepatic antioxidant status and intestinal barrier of male broilers under high ambient temperature (32.7℃). Methods: Nine hundred healthy one-d-old Cobb-500 male broiler chicks were randomly assigned into three groups with six replicates of 50 birds each. A basal diet supplemented with 0% (control), 0.4% and 0.8% COAC, respectively were fed to birds for 6 weeks. All treatments were under high ambient indoor temperature of 32.7℃, and had a constant calcium and available phosphorus ratio. Results: The results showed that, compared with control, the average daily gain of broilers in 0.4% and 0.8% was significantly increased and the ratio of feed to gain in in 0.4% and 0.8% was significantly decreased at 1 to 21, 22 to 42 and 1 to 42 days of age (p<0.05). Compared with control, 0.8% COAC slightly decreased (p = 0.093) the content of malondialdehyde in liver at 42 days of age while 0.4% COAC significantly decreased (p<0.05) the activity of alkaline phosphatase. Furthermore, 0.4% COAC significantly enhanced the intestinal barrier function via increasing jejunal and ileal ocln transcription, promoting jejunal mucin 2 transcription at 42 days of age (p<0.05), and decreasing jejunal toll-like receptor 2 (TLR-2) and ileal TLR-15, inducible nitric oxide synthase compared with control group (p<0.05). Whereas, no significant differences on the transcription of interleukin-1β in jejunum and ileum were observed among three treatments (p>0.05). Overall, heat stress caused by high natural environment temperature may induce the damage to hepatic antioxidation and intestinal barrier. Conclusion: Dietary inclusion of COAC can improve the tolerance of broilers to thermal environment through the modification of antioxidative parameters in liver and the mRNA expression of genes in intestinal barrier, resulting in an optimal inclusion level of 0.4%.

Fundamental Study on Developing Lignocellulosic Fillers for Papermaking(I) (목질계 제지용 충전제 개발을 위한 기초연구(I))

  • Wastewoods (logging residues) generated in Korea were used to make lignocellulosic fillers for papermaking. Lignocellulosic fillers could play great roles to increase retention efficiency and thus decrease turbidity of white water in papermaking process. In addition, lignocellulosic fillers could be used to improve physical properties of paper through their high affinity to cellulosic fibers, leading to the less use of chemical additives like retention aids. Wastewoods including Pinus densiflora and Quercus variabilis were chemically and mechanically treated for making fine particles passing through 100 mesh wire. The newly generated fillers showed larger particle size distribution than ground calcium carbonates but similar distribution to talc. In particular, pretreatment by hot water was more effective to generate smaller particle size than by alkali treatment. Lignocellulosic particles mixed with ground calcium carbonates under intense hybridizing condition greatly contributed to surface coverage of organic fillers in addition to filling to lumen and pits.

Community Structure of Ectomycorrhizal Fungal communities Colonizing Quercus spp. in Limestone Areas of Korea (석회암 지대 참나무속 식물에 공생하는 외생균근균의 군집구조)

  • In this study, we analyzed the diversity of ectomycorrhizal (ECM) fungal communities of Quercus spp. roots in the limestone area. We identified 45 generd of ECM using next generation sequencing (NGS) analysis. Soil chemical composition analysis confirmed soil pH, substitution calcium concentration, total nitrogen content, organic phosphate, and organic matter content. Shannon's Index was calculated according to the changes in soil chemical composition. The results of cluster analysis showed that Sebacina, Tomentella, Tuber, Densospora, Inocybe, Suillus, and Piloderma were the main genera of symbiotic ECM fungi that thrived in soil with high pH and calcium content.

Preparation of Natural Polymer-CaP Composite Films (천연 고분자-칼슘 포스페이트 복합 박막 제조)

  • We investigated the surface modification method for the preparation of organic-inorganic hybrid composite thin film. Gelatin obtained from the decomposition of collagen was allowed to adsorb in a polystyrene tissue culture dish for 2 h to from layers of gelatin. Supersaturated ionic solution of calcium and phosphorus was injected on the gelatin adsorbed layer to form calcium phosphate thin film. During the initial period of incubation, nucleates were formed. With increase of the incubation time, CaP (calcium phosphate) thin film grew on the surface of the culture dish. The gelatin/CaP thin film displayed the highly porous three-dimensional surface structure. Attenuated, total reflectance Fourier transform, infra-red spectroscopy (ATR-FTIR) was used to analyze the chemical properties of CaP film. The analysis demonstrated that the CaP film formed at initial period of treatment appeared to be amorphous. With increase of incubation time, the crystallinity of the film was slightly increased, but the presence of the peaks for the low crystalline CaP confirmed that the CaP thin film prepared in this study was poorly crystallized.

  • This study examined the effect of calcium (AC) extracted from salt fermented anchovy Engraulis japonicus on osteoporosis in ovariectomized (OVX) SD female rats. After AC treatment for 8 weeks, the body weight of the OVX-AC group decreased, while the weight of the OVX group (control) increased 43.9%. The blood osteocalcin, calcium, and ALP levels were examined as physiological markers for osteoporosis. The concentrations of all markers were elevated in the OVX-AC group compared to the OVX group. Thus, anchovy calcium is useful as an organic calcium supplement.

Characteristics of the Shells and Calcined Powders from the Butter Clam Saxidomus purpuratus and Littleneck Clam Ruditapes philippinarum as a Natural Calcium Resource (천연칼슘소재로서 개조개(Saxidomus purpuratus)와 바지락(Ruditapes philippinarum) 패각 및 소성분말의 특성)

  • Shell waste from the butter clam Saxidomus purpuratus and littleneck clam Ruditapes philippinarum is a large by-product of shellfish aquaculture, and it is desirable to convert it into value-added products for industrial applications. In this study, calcium carbonate (CaC) polymorphs from butter clam (BCSP) and littleneck clam (LCSP) shell powders and commercial CaC were characterized using Fourier transmission infrared spectroscopy (FT-IR), X-ray diffraction (XRD), and scanning electron microscopy (SEM). The results revealed that the optimal calcination conditions to eliminate organic substances and improve solubility for both BCSP and LCSP were $800^{\circ}C$ for 8 h in an electrical furnace. Calcination improved the white index of the butter clam (BCCP) and littleneck clam (LCCP) calcined powder compared with shell powders. The calcium content in BCCP (51.1%) was higher than that of LCCP (44.9%) or commercial calcium oxide (CaO, 44.7%). The XRD patterns of BCCP and LCCP were similar to that of CaO. Cubic-like crystals of CaC and irregular crystals of BCCP and LCCP were observed by SEM. The FT-IR and XRD analyses revealed the presence of calcite and aragonite in the BCSP and aragonite in the LCSP, whereas the CaC contained calcite. These results indicate that butter and littleneck clam shells are potential biomass resources for calcium carbonate and calcium oxide.

Effects of Mild Heat and Organic Acid Treatments on the Quality of 'Daebo' Peeled Chestnut during Storage (열수 및 유기산 처리가 '대보' 박피밤의 저장 시 품질에 미치는 영향)

  • The effects of dip treatments of chemicals (ascorbic acid, citric acid, calcium chloride: 1% solution) and physicals (vacuum packing, $50^{\circ}C$ distilled water) on the browning and quality of 'Daebo' peeled chestnut were studied. During the storage, the surface color of the samples showed higher ${\Delta}E$ values and lower L values than that of the initial sample. The color with the normal packing treatment hardly changed, unlike in the chemical and physical treatments. The calcium chloride treatment showed less color change than other treatments. The marketable quality was maintained for 10 days with the normal packing and for 35 days with the chemical and physical treatments. Thus, the chemical and physical treatments, especially with vacuum packing after 1% calcium chloride treatment, extended the shelf-life of the 'Daebo' peeled chestnut by inhibiting the browning.
