부숙과정중(腐熟過程中) 낙엽류(落葉類) 가수분해물(加水分解物)의 유기산함량(有機酸含量) 변화(變化) (Changes of Some Organic Acids in the Hydrolysates of Decomposing Litters of Wild Grasses and Tree Leaves)

  • 산야초(山野草), 활엽수(闊葉樹)와 침엽수낙엽(針葉樹落葉)을 실험실조건하(實驗室條件下)에서 90일간(日間) 부숙(腐熟)시키면서 경시적(經時的)으로 시료(試料)를 채취(採取)하여 산가수분해(酸加水分解)시킨후 6개(個)의 저분자량(低分子量)의 유기산(有機酸)을 분석(分析)한 결과(結果)는 다음과 같다. 1. Formic, acetic, succinic, malic, tartaric와 citric acid가 검출(檢出)되었으며 이중(中) malic acid는 전시료중(全試料中)에서 극(極)히 미량(微量)으로 함유(含有)되어 있었다. 2. 산야초(山野草)의 경우 저분자량(低分子量) 유기산(有機酸)의 농도(濃度)는 부숙기간(腐熟期間)이 경과(經過)함에 따라 감소(減少)했으며 formic acid와 acetic acid등(等) monocarboxylic acid는 dicarboxylic acid와 tricarboxylic acid에 비(比)해 훨씬 높은 농도(濃度)를 보였고 formic acid와 acetic acid는 상호보충(相互補充) 관계(關係)가 있었다. Citric acid는 부숙기간(腐熟期間)이 경과(經過)함에 따라 농도(濃度)가 증가(增加)하였다. 3. 활엽수낙엽(闊葉樹落葉)의 경우 부숙기간(腐熟期間)이 경과(經過)함에 따라 monocarboxylic acid농도(濃度)가 상대적(相對的)으로 높았다. Formic acid와 acetic acid사이는 상호보충(相互補充) 관계(關係)가 있음이 구명(究明)되었다. 4. 침엽수낙엽(針葉樹落葉)의 경우 부숙기간(腐熟期間)이 경과(經過)함에 따라 유기함유량(有機含有量)의 변화(變化)가 별(別)로 나타나지 않았다. 산야초(山野草)와 활엽수낙엽(闊葉樹落葉)과는 대조적(對照的)으로 dicarboxylic acid인 succinic acid농도(濃度)가 가장 높았다.

Synergistic Effects of Bacteriocin-Producing Pediococcus acidilactici K10 and Organic Acids on Inhibiting Escherichia coli O157:H7 and Applications in Ground Beef

  • When used in combination with organic acids, Pediococcus acidilactici K10 or its bacteriocin was effective in inhibiting Escherichia coli O157:H7 in vitro and in situ. P. acidilactici K10, a strain of bacteriocin-producing lactic acid bacteria (LAB), was previously isolated from kimchi in our laboratory, and the molecular weight of its bacteriocin was estimated to be around 4,500 Da by SDS-PAGE. Initially, P. acidilactici K10 and its bacteriocin could not inhibit E. coli O157:H7, when used alone. However, when they were used together with organic acids such as acetic, lactic, and succinic acids, they greatly inhibited E. coli O157:H7 in vitro. Based on these in vitro results, a real sample test with ground beef was conducted at $4^{\circ}C$ with acetic acid (0.25%) or lactic acid (0.35%) alone, and then in combination with P. acidilactici K10 (10^5 CFU/g of sample). Combined treatment of P. acidilactici K10 with lactic acid showed the most inhibitory effect: a 2.8-$log_{10}$-unit reduction of E. coli O157:H7 in ground beef during storage at $4^{\circ}C$. This result suggests that the combination of bacteriocin-producing P. acidilactici K10 and organic acids has great potential as a food biopreservative by inhibiting the growth of E. coli O157:H7.

  • Changes in taste components of Backilju, an traditional alcoholic beverage of Korea, were investigated. Ethanol(17%) was the most abundant, and then isoamylalcohol(23mg%) and methanol(8mg%) were also detected in a small amount in Backilju. Major non-volatile organic acids were lactic and malic acid, followed by citric, fumaric and succinic acid. Arginine, phenylalanine and glutamic acid were major free amino acids and essential amino acid content was 230∼560 mg%, which was 45∼48% of total free amino acids. The major fatty acid of total lipid was palmitic acid(37∼43%). It has been found that the Backilju contained more free amino acids and alcohols than other Korean Yokjus.

잎담배 엽육과 주맥의 휘발성 정유성분 및 비휘발성 유기산의 비교 (Comparison of Volatile Aroma Components and Non-volatile Organic Acids in Tobacco Lamina and Stems.)

  • Volatile aroma components, non-volatile organic acids in lamina and stems of flue-cured(NC 2326) and burley ( Burley 21) were analyzed by gas chromatography and mass spectrometry, respectively. Then compositional differences of these components between lamina and stems were discussed. The contents of volatile components were higher in flue-cured than in burley tobacco, and it was also higher in lamina then in stem. The major aroma components in lamina were neophytadiene , nicotine, solanone and benzyl alcohol but those in stems were palmitic acid, neophytadiene, nicotine, solanone and phenyl ethyl acetate. On the other hand, the contents of non-volatile organic acids were higher in burley than in flue-cured tobacco, and these values of burley tobacco were higher in lamina than in stem but flue-cured tobacco were higher in stem than in lamina. The major acids in all the above four tabacco samples were malic, citric, oxalic and linolenic acid.

산화물 표면의 U(VI) 흡착에 미치는 살리실산과 피콜린산의 영향 (Effect of Salicylic and Picolinic Acids Acids on the Adsorption of U(VI) onto Oxides)

  • 세 종류의 산화물($TiO_2$(아나타제), $SiO_2$(비결정성) 및 $Al_2O_3$(비결정성)) 표면에 U(VI)이 흡착될 때 유기산이 미치는 영향을 연구하였다. 유기산으로는 살리실산과 피콜린산을 사용하였다. 유기산의 존재 여부에 따라 달라지는 U(VI)의 흡착률 변화를 pH 함수로 측정하였다. 또한 U(VI)의 존재 여부에 따라 달라지는 유기산의 흡착량을 pH 함수로 측정하였다. $TiO_2$의 경우, 살리실산과 피콜린산이 U(VI)과 수용성 착물을 형성함으로써 U(VI)의 흡착률을 저하시킨다. $SiO_2$의 경우, 살리실산은 U(VI) 흡착에 영향을 주지 않지만, 피콜린산은 오히려 U(VI) 흡착을 증가시킨다. 이 현상을 삼성분 표면착물(ternary surface complex) 생성으로 해석하였으며 U(VI) 흡착에 의존하는 피콜린산의 흡착량 변화, 그리고 흡착된 U(VI)의 형광 특성 변화로 이를 확인하였다. $Al_2O_3$의 경우, 살리실산과 피콜린산 모두 U(VI) 흡착과 무관하게 높은 흡착량을 보였으나 U(VI) 흡착을 감소시키지는 않았다. 따라서 삼성분 표면착물 생성을 배제할 수 없으나 이를 확인하기 위해서는 분광 분석과 같은 추가 연구가 필요하다.

Comparative Chemical Composition among the Varieties of Korean Chili Pepper

  • From the point of breeding view for our future, 20 Korean varieties of red pepper for the contents of capsaicinoids, free amino acids, free fatty acids and organic acids with powder product and eating qualities were chosen, and the sensory properties of their water and ethanol extract were compared in order to investigate the influence of the composition of test components on sensory acceptability of Korean red pepper. The composition of taste components in red pepper powder varied wildly depending on the varieties; total capsaicinoids content variety from 0.029 to $0.913\%$, free sugar 8.45 to $20.2\%$, and organic acid 4.58 to $19.54\%$. Capsaicinoid contents especially dihydrocapsaicin content, were highly correlated with pungent taste of the methanol extract of red pepper power, but did not show significant relationship to the overall sensory acceptability. However, the components analyzed here and eating and processing qualities include high variations and future breeding sources.

저분자량 유기산 세척을 이용한 오염토양으로부터의 Cu제거에 관한 연구 (Remediation of Copper-Contaminated Soil using Low Molecular Weight Organic Acid Flushing Technique)

  • 저분자량 유기산을 이용한 in-situ flushing 토양세척기법의 실제 현장 적용성 검토를 위해 실험실에서 회분식 실험을 통하여 적당한 세척제를 선정하고 이에 따른 여러 가지 운전변수를 찾아보았다. 대상 중금속으로는 Cu를 택했고, 세척제로는 실제 토양환경에 영향이 비교적 적은 저분자량 유기산인 acetic, citric, oxalic, succinic acid를 이용했다. 유기산을 이용하여 처리한 결과 실험에 사용된 유기산들 모두 1 mM의 저농도에서는 효과를 나타내지 못했고 50 mM 이상의 고농도에서 보다 좋은 제거율을 보였는데, 50 mM과 100mM의 경우 제거율이 큰 차이를 보이지 못했다. citric arid와 oxalic acid의 경우 중성 및 약산성 상태에서 최대의 제거율이 나타났고 특히 citric arid의 경우 87.1%의 높은 제거율을 보였다. 유기산이 없을때는 강산성 상태에서 Cu의 최고제거율이 70% 정도였다. 그러나 각 경우에서의 speciation을 살펴보면 유기산이 없을 경우에는 추출된 Cu의 대부분이 유해한 free ion상태로 존재하게 되고 유기산이 있을 경우에는 추출된 Cu의 대부분이 유기산과 complex를 이룬다는 것을 알 수 있었다.

Adsorption of Organic Compounds onto Mineral Substrate Prepared from Oyster Shell Waste

  • Humic acids react with chlorine to produce Trihalomethanes(THMs), known as carcinogens, during disinfection, the last stage in water purification. Currently, the removal of organic humic acids is considered the best approach to solve the problem of THM formation. Accordingly, the current study examined the adsorption of organic compounds of humic acids onto an inorganic carrier prepared from oyster shell waste. The adsorbent used was activated oyster shell powder(HAP) and silver ion-exchanged oyster shell powder(HAP-Ag), with CaCO$_3$ as the control. The adsorbates were phthalic acid, chelidamic acid, catechol, dodecylpyridinium chloride(DP), and 2-ethyl phenol(2-EP). The adsorption experiments were carried out in a batch shaker at $25^{\circ}C$ for 15 hours. The equilibrium concentration of the adsorbate solution was analyzed using a UV spectrophotometer and the data fitted to the Langmuir isotherm model. Since the solution pH values were found to be greater than the pKa values of the organic compounds used as adsorbates, the compounds apparently existed in ionic form. The adsorptive affinities of the organic acid and phenolic compounds varied depending on the interaction of electrostatic forces, ion exchange, and chelation. More carboxylic acids and catechol, rather than DP and 2-EP, were adsorbed onto HAP and HAP-Ag. HAP and HAP-Ag exhibited a greater adsorptive affinity for the organic compounds than CaCO$_3$, used as the control.

Effects of Organic Acids on Availability of Phosphate and Growth of Corn in Phosphate and Salts Accumulated Soil

  • Accumulated Phosphate can be released by ligand exchange reaction of organic acids. The objective of this study was to evaluate effects of the organic acids on the availability of phosphate and the growth of crop in phosphate and salts accumulated soil. Soil samples were collected from farmer's plastic film house. Available phosphate and electrical conductivity of soil were $3,005mg\;kg^{-1}$ and $16.63mg\;kg^{-1}$ which were 6 and 8 times higher than the optimum range of soil for crop growth, respectively. Corns were cultivated in pots for 2 months. Treatments were no treatment (control), phosphate fertilizer (P), citric acid (CA) 1, 5, 10 mM, and oxalic acid (OA) 1, 5, 10 mM. Water soluble phosphorus, available phosphate, corn growth and uptake were determined after cultivation. Results showed that organic acids increased water soluble phosphorus and available phosphate. For the level of 10 mM, the order of effectiveness of organic acids for water soluble P was citric acid (44%) > oxalic acid (32%). Height and dry weight of corns were increased significantly by the treatment of citric acid 1 and 5 mM. Also, corn absorbed more phosphorus, nitrogen, potassium, calcium and magnesium in the treatment of citric acid 1 mM than these of other treatments. Even though phosphate availability of soil was enhanced by addition of citric acid 10 mM, the growth of corns decreased because high concentration of citric acid caused salt damage by increasement of electrical conductivity. Thus, the citric acid of 1 mM has the potential to improve the availability of phosphate and the healthy growth of corns.
