  • Purpose : Continuous renal replacement therapy(CRRT) has been the first choice for the treatment of acute renal failure in critically ill children not only in western countries but also in Korea. However, there are very few studies that have analyzed the outcome and prognosis of this modality in Korean children. We performed this study to evaluate the factors associated with the outcome and prognosis of patients treated with CRRT. Methods : We retrospectively reviewed the medical records of 32 children who had received CRRT at Severance hospital from 2003 to 2006. The mean age was 7.5 years(range 4 days-16 years) and the mean body weight was 25.8 kg (range 3.2-63 kg). Results : Eleven(34.4%) of the 32 patients survived. Bone marrow transplantation and malignancy were the most common causes of death and underlying disease leading to the need for CRRT Mean patient weight, age, duration of CRRT, number of organ failures, urine output, estimated glomerular filtration rate(eGFR), C-reactive protein, and blood urea level did not differ significantly between survivors and nonsurvivors. (1) Pediatric risk of mortality(PRISM) III score at CRRT initiation($9.8{\pm}5.3$ vs. $26.7{\pm}7.6$, P<0.0001), (2) maximum pressor number ($2.1{\pm}1.2$ vs. $3.0{\pm}1.0$, P=0.038), and (3) the degree of fluid overload($5.2{\pm}6.0$ vs. $15.0{\pm}8.9$, P=0.002) were significantly lower in survivers than in nonsurvivors. Multivariate analysis revealed that fluid overload was the only independent factor reducing survival rate. Conclusion : CRRT was successfully applied to the treatment of acute renal failure in a wide range of critically ill children. To improve survival, we suggest the early initiation of CRRT to prevent the systemic worsening and progression of fluid overload in critically ill children with acute renal failure. • Naringin, major citrus flavonoids, has been identified to exert antioxidative, antidiabetic, and lipid lowering effects. In this study, we examined the effect of 0.2 g/kg, 0.5 g/kg naringin supplementation for 3 times/week for 5 weeks on lipid metabolism and antithrombotic capacity in rat. Eighteen five week-old Sprague Dawley(SD) female rats, which had initial body weights of $246{\pm}9g$, were randomly divided into three groups: Control (non naringin group); Low (0.2 g/kg naringin-supplemented group); High (0.5 g/kg naringin-supplemented group). Three groups of rats were supplemented with three experimental diets for 5 weeks and we investigated antithrombotic capacity before sacrifice. Naringin did not significantly alter the body weight gain, relative organ weight. However, the level of serum triglyceride, serum free fatty acid, serum total lipid and serum glucose levels were significantly lowered compared to those of control. The high group (0.5 g/kg naringin-supplemented group) was showed significantly increased bleeding time compared to control group. These results suggest that naringin supplemental diets reduces the level of hypertension, glycosuria and fatness on the female SD rats, when orally administered below the dosage 0.5 g/kg for 5 weeks.

  • This study was performed to investigate the effect of pork on the cadmium detoxification in rats. Ninety male Sprague-Dawley rats weighing $125.3{\pm}1.4g$ were divided into five groups based on cadmium treatment(0, 25, 50, 100, 250ppm) and five levels of Cd in AIN-76 purified diet had been fed for 8 weeks. Cadmium was supplemented with a form of $CdCl_2$.. During following 8 weeks of intoxication, casein was replaced by pork and the effect of pork on cadmium- detoxification was compared with casein. After 8 weeks of Cd intake had resulted in apparent cadmium intoxication; reduced growth rate, enlarged kidney and testis, decreased hematocrit value and hemoglobin content in response to the supplemented Cd levels in the diets. Discontinuing cadmium feeding, the body weights were relieved. Pork-fed groups seemed to have higher body weight than casein-fed groups. Hemoglobin content and hematocrit value became normal range at detoxification stage. The weights of liver, kidney, and testis were decreased along with cadmium intake. However, organ weight ratio was not affected by cadmium. Cadmium accumulation in liver and kidney showed a tendency to increase in the cadmium-exposed groups. The levels of metallothionein were also significantly elevated in the tissues of liver in response to the levels of Cd supplemented(p<0.05). Cadmium concentration in kidney was two times higher than that in liver. Cadmium removal rate of liver was higher than that of kidney. Cadmium accumulation of the pork-fed group was lower than that of casein. Especially, the factors which affected the cadmium contents in kidney were $Cd^{***}$ and $Cd{\times}pork^{***}$. Metallothionein(MT) was increased with cadmium, and MT was not likely to be affected by pork. Based on the findings from gross lesion, rats fed 250ppm of Cd were externally emaciated, had exposed penis and observed atrophies of kidney and testis. Histopathological observation seemed that the liver of groups feeding Cd supplemented diets showed cellular degeneration and accumulation of eosinophilic materials in the capillaries. In kidney, rats fed Cd diets had shown tubular epithelium degeneration and lesions of basophilic materials, while testis were weakened in numbers of spermatid and sporadically enlarged of giant cells. But the rats administered cadmium-detoxified diet supplemented pork for 7 weeks were shown individually decreased lesions compared with the rats supplied with casein diet.

  • Effects of various combinations of corn oil (CO) and perilla oil (PO) as respective dietary sources of ${\omega}$-6 and ${\omega}$-3 polyunsaturated fatty acids on fatty acid profiles of immune organs were studied in young chicks. Seventy-five 1-day-old male (ISA Brown) chicks were assigned to five treatments with three replications. Semi-purified-type diets containing glucose and soybean meal as major ingredients were added with 8% CO, 6% CO+2% PO, 4% CO+4% PO, 2% CO+6% PO and 8% PO and fed for 7 weeks. There were no significant differences in body weight gain, feed intake and relative weights of liver and immune organs (g/100g weight) among dietary groups. Dietary fatty acid patterns were generally reflected in the fatty acid compositions of all immune organs such as spleen, thymus and bursa of Fabricius. The levels of a-linolenic acid(LNA), eicosapentaenoic acid (EPA) and docosahexaenoic acid in various immune organs increased with increasing levels of perilla oil in the diets, whilet the levels of linoleic acid (LA) and arachidonic acid (AA) decreased. Thymus appeared to have capacity to retain remarkably higher (P<0.05) levels of LA and LNA up to 37 and 22%, respectively, compared to the other organs. Thymic tissue contained ${\omega}$-3 fatty acid and ${\omega}$-6 fatty acid 10~36 times and 3~5 times higher than the other organs, respectively. Spleen tissue was specifically higher (P<0.05) in the levels of AA and EPA and the ratios of AA/LA and EPA/LNA, compared to the other organs, suggesting that the tissue might have high desaturase activity to convert LA or LNA to AA or EPA, respectively. BSA antibody production tended to increase by 18 ~ 32% with higher levels of perilla oil in diet, although the increase was not statistically significant. In conclusion, fatty acid compositions of immune organs very depending on the lipid composition of the diets and each organ appears to respond differently for its fatty acid profile to dietary lipids. Considering AA and EPA are precursors of many important eicosanoids, further studies are required to clarify the responses of the immune organs to the dietary fatty acids.

  • Effects on yields, yield components and nutrient content of potassium depression for two or three weeks at various growth stages were investigated in rice (var. Jinheung) under sand culture system.(K 40 ppm 1973) 1. Analysis of variance showed significant difference among treatments of both two-week (at p=0.01) and three-week depression (at p=0.05) in yield. 2. Most sensitive stage to potassium depression on yield appeared two weeks until heading (42% yield decrease) and sensitivity decreased the growth stage is apart from heading either before or after. During 30 days after transplanting two-week potassium depression increased yield, but three-week depression decreased yield. Until about 30 days after heading depression caused poor yield. 3. Root potassium involves in harvest index, filled grain ratio and grain weight with significant correlation and considerably in spikelet per panicle while potassium in leaf sheath+culm involves considerably in spikelet per panicle and panicle per hill. Relative total dry matter weight was significantly correlated with panicle per hill, spikelet per panicle and K or K/Ca+Mg only in leaf sheath+culm. The indications are that root potassium contributes for building sink and efficiency of structure while potassium in leaf sheat+culm primarily for building source, productive structure. 4. Relative yield was significantly correlated with potassium content in root and leaf sheath+culm and with K/Ca+Mg and its ratio before and after depression in root indicating that potassium depression occurs greatly in root and that K/Ca+Mg might have more important role than K content alone under depression. 5. Optimum level of $K_2O$ appears around 3% in leaf blade. 4% in leaf sheath+culm and 1% in root under the assumption that below these level the same content has the same role in relation to yield during growth. The K/Ca+Mg appeares to be 2.5 in root and should not decrease throughout the growth stages. 6. The increase of sodium content in plant by K depression was highest, especially in leaf sheath during the most insensitive period to K depression suggesting that insensitivity may be attributed to partial replacement of Na for K. Partial replacement seems very little in sensitive stage (later stage) and sensitive organ (root).

  • This study investigated the effect of improved liver function in rats administered with ethanol by kimchi lactic acid bacteria with high GABA producing capacity. Sprague-Dawley male rats were divided into four groups; normal diet control (NC), ethanol control (EC), ethanol+Lactobacillus sp. OPK2-59 normal powder (EL1), ethanol+Lactobacillus sp. OPK2-59 GABA powder (EL2) and fed for 6 weeks. Analysis showed that there were no significant differences in body weight and feed consumption among the groups during the experimental period. Also, there were no significant differences in organ weight among the groups. The test results showed total cholesterol and triglyceride in the blood concentration that were increased by ethanol administration were significantly lowered in EL2 group. Liver triglyceride was also significantly lowered in the EL2 group compared with the EC group. Serum GOT and GPT, and liver GOT levels were significantly lower in the EL2 group compared with the EC group. Serum ethanol concentration was lower in the EL1 and EL2 groups compared with the EC group. SOD activities in liver were significantly increased in the EL1 and EL2 groups compared with the EC group. These results suggest that Lactobacillus sp. OPK2-59 GABA powder improves lipid and enzyme profiles of rats administered with ethanol.

  • We investigated the effects of fermented Sargassum thunbergii (FST) on platelet aggregation and serum lipid levels in rats made obese by a high fat diet. Six-week-old male SD-rats were randomly assigned to four groups as CON, HF-CON induced by high fat diet (HF), ST supplemented with HF (HFST100), and the fermented ST supplemented with HF group (HF-FST100). After 6 weeks, the results showed that the final weight and weight gain had decreased in the HF- FST100 group compared to the HF-CON group. Also, the food efficiency ratio was significantly reduced in the HF-FST100 group compared to HF-CON. The organ weights other than heart and spleen were significantly lower in the HF-FST100 group than in the HF-CON group. The levels of serum GOT and GPT significantly decreased in the HF-FST100 group over the HF-CON group. In addition, the total cholesterol, triglyceride and LDL-cholesterol levels were lower in the HF-FST100 group than in HF-CON, while the HDL-cholesterol level was higher in the HF-FST100. The ability of platelet aggregation of groups supplemented with FST was lower than the HF-CON group. These results suggest that FST may be beneficial in improving lipid profile and platelet aggregation in obesity.

  • Herbal medicines are widely used as therapeutic products in many countries. Corni fructus (CF), the dried ripe sarcocarp of Cornus officinalis Sieb. et Zucc (Cornaceae), has been used for thousands of years in traditional medicine and has been reported to be effective for the prevention and treatment of various diseases, such as kidney diseases and diabetes. Recent research on CF has documented a wide spectrum of therapeutic properties, which include anti-inflammatory, ant-oxidative, immunomodulatory, and anti-cancer effects. However, there is no information on its safety. Therefore, in this study, the toxicity of water extract of CF to ICR mice was investigated. The mice received a single dose of water extract of CF (1,000, 2,000, and 5,000 mg/kg of body weight) via the oral route. Mortality, clinical signs, body weight changes, gross findings, and weights of the principal organs after 14 d were then assessed. The results revealed no adverse effects of CF as determined by clinical signs, body weights, or organ weights and no gross pathological findings in any of the treatment groups. These results suggest that the 50% lethal dose and approximated lethal dose of CF extract is over 5,000 mg/kg. The findings provide scientific evidence for the safety of CFs.

  • To identify the physiological phenomena on the sudden wilting of japonica/indica crossed varieties, Pot experiment was carried out under the heavy N application with various levels of potassium in Japan. The results obtained are as follows. 1. Sudden wilting was occurred in both varieties used, Yushin and Milyang 23. The former showed a higher degree than the latter. 2. Sudden wilting was occurred into two types, one at early ripening stage and the other at late ripening stage. The former type was found in the field with low potassium supply and the latter was seemed to be related to varietal wilting tolerence. 3. By the investigation of concerning the effective tillering rate and the change of dry weight of each organ at the heading stage, it was inferred that the growth status from young panicle formation stage to heading stage were related to sudden wilting tolerence. 4. Manganese content at heading stage, ratio of Fe/Mn and Fe. Fe/Mn in stern at late ripening stage and $K_2$ O/N ratio of stem at harvesting stage were recognized as the specific factors in connection with sudden wilting. Mn content in the sudden wilting rice plant was already in creased remarkably at heading stage. In relation to root age and absoption characteristics of Mn, the senility of root before heading stage was inferred as the cause of increase the value of Fe/Mn or Fe. Fe/Mn. 5. The $K_2$ O/N ratio of culm at harvesting stage was lower in upper node than lower node in relation to sudden wilting. And it was well accordance with the fact that the symptoms of sudden wilting proceeded from upper leaf to lower leaf. These phenomenon was different from the usual one that the effect of potassium deficiency was more remarkable in lower node than upper node. 6. All varieties which have a condition of potassium deficiency have a high degree of nitrogen content of leaves at heading stage and the $K_2$ O/N ratio of each organ was low, Especialy, $K_2$ O/N ratio is much lower in sheath and culm than leaves.

  • Two experiments were conducted to determine whether water extracts from Artemisia capillaries (A. capillaries) and Camellia sinensis (C. sinensis) could be used as alternatives to antibiotic growth promoters in broiler feed. The experiment 1 was verified their chemical composition, extracts yields, total phenolic compounds concentration, antioxidant activity, antimicrobial activity, and chicken splenocytes proliferation through in vitro test. The extract yields of A. capillaries and C. sinensis were 26.5 and 16.8%, respectively. Total phenolic compounds concentrations of them expressed as gallic acid equivalent were 15.28 and 26.74 mg/mL, respectively. Electron donating abilities of them expressed as $SC_{50}$ showing 50% DPPH radical scavenging were 0.30 and 0.06 mg, respectively. Bacterial inhibitory rates of them against Escherichia coli, Staphylococcus aureus, and Salmonella Typhimurium were ranged from 42.1 to 52.3% and from 21.6 to 33.7%, respectively. And, these extracts increased proliferation of chicken splenocytes. Especially, A. capillaris was more excellent than Echinacea and Concanavalin A known as T-cell stimulator. The experiment 2 was investigated their effects on growth performance, relative organ weight, cecal microflora, blood biochemical parameters, and splenic cytokines mRNA expression in broiler chicks. Four hundred eighty 1-day-old male broiler chicks (Ross 308) were divided in to 4 treatment groups with 4 replicates of 30 birds in each group: NC (control, no antibiotics), PC (avilamycin, 10 ppm; salinomycin, 60 ppm), AC (A. capillaries, 100 ppm), and CS (C. sinensis, 100 ppm); treatments were administered through water supplementation. Final body weight was significantly higher in all treated groups than in NC (p<0.05). Cecal Salmonella numbers were significantly or somewhat decreased in all treated groups than in NC (p<0.05). The relative weights and lengths of the small intestine were more significantly decreased in the PC and AC groups than in the other groups. Cecal Salmonella numbers were significantly or somewhat decreased in all treated groups than in the NC group (p<0.05). The contents of total cholesterol, aspatate aminotransferase, and alanine aminotransferase in blood serum were more significantly decreased in all treated groups than in NC (p<0.05). In conclusion, these results suggested the possibility that these extracts could serve as alternatives for antibiotic growth promoters.
