The composition of dissolved gases and nutrients in a liquid medium were determined for establishment of the optimum conditions for in vitro culture of Helicobacter pylori. A microaerobic condition facored by the organism was prepared by adjusting the partial pressure of the gas, agitation speed, and viscosity of the medium. The gaseous concentrations were controlled by utilizing CampyPak Plus that reduced oxygen while augmenting carbon dioxide. Agitation of the broth facilitated the oxygen transfer to the cells, yet inhibited the growth at high rates. An increase of viscosity in the medium repressed the culture although this variable was relatively insignificant. The chemical constituents of the liquid broth were examined to establish an economic model for H. pylori cultivation. The microbe required a neutral pH for optimum growth, and yet was also able to proliferate in an acidic condition, presumably by releasing the acidity-modulating enzyme, urease. Cyclodextrin and casamino acid were investigated as growth enhancers in place of serum, while yeast extract unexpectedly inhibited the cells. A low concentration of glucose, the unique carbon source for the organism, increased the cell density, yet high concentrations resulted in an adverse effect. Under optimally dissolved gas conditions, the cell concentration in brucella broth supplemented with serum substitutes and glucose reached $1.6{\times}10^8$ viable cells/ml which was approximately 50% higher than that obtained in the liquid medium added with only cyclodextrin or serum.
To exploit a suitable vector and recipient strain for molecular cloning in Bacillus subtilis, oxytetracycline-resistant plasmic DNA has been prepared from Streptomyces rimosus by phenol-buffer extraction of lysozyme-lysed cells and introduced into B. subtilis KPM 60 [St $r^{R}$-mutant of RM 125 (leu A8, arg 15, hsm $M^{-10}$ , hsr $M^{-10}$ )] by transformation. Oxitetracycline-resistant plasmid was well transferred into B. subtilis KPM 60 with average frequency of 10$^{-4}$ per $\mu\textrm{g}$ of DNA. The highest frequency of plasmid transformation was obtained after 3 hours incubation of recipient cells in the growth medium and 30 to 60 minutes incubation in the competence medium, and then 20 minutes contact of DNA and host cells. Optimum pH for competence was 7.5, and optimum temperature for transformation was 2$0^{\circ}C$.>.
Mycological characteristics of Phytophthora nicotianae var. nicotianae SPC10 (A1 type) causing Phytophthora rot of strawberry and the resistances of 11 strawberry cultivars against the pathogen were examined. Optimum temperature for the mycelial growth of the pathogen was obtained in the range of 30~35$^{\circ}C$, and the growth was completely stopped under 13$^{\circ}C$ or over 42$^{\circ}C$. Aerial mycelia were abundant on oatmeal agar (OMA), V-8 juice agar (V8A) and lima bean agar (LBA) medium, although there were slight differences, however, on cornmeal agar (CMA) medium, it was a shape of stellate without aerial mycelia. The colony shape on potato dextrose agar (PDA) medium was rough and irregular whereas the mycelial growth was slow, and some aerial mycelia were only produced in the middle of PDA medium. Optimum temperature for sporangial formation was 3$0^{\circ}C$, and zoospores were mostly released at $25^{\circ}C$ from the sporangia. Sporangia were more produced in C/Z solution with pH 5. 0~6.$0^{\circ}C$ than sterilized distilled water (DSW) and distilled water (DW), and zoospores were also released much more than other solutions. Eleven strawberry cultivars such as Reiko, Hokowase, Eyeberry, Akaneko, Sistakara, Toyonoka, Nyoho, Sulhong, Suhong, Myhong and Wonkyo #3104 revealed the disease incidence up to 88.9~100% by the leaf inoculation with mycelial disk. However, Nyoho and Suhong showed higher level of resistance against the pathogen by root inoculation.
Effects of various kinds of sawdusts, supplements and culture conditions on activities of several enzymes such as protease, phenoloxidase and cellulase produced from mycelium of P. ostreatus grown on sawdust medium were studied and the results are as follows; Higher specific activity of these enzymes was observed when oak tree sawdust and poplar tree sawdust were supplemented with rice bran or wheat bran at rate of 30%, 20% and 10% in total volume respectively. Higher total activities of protease, phenoloxidase and cellulase were observed at 70% of the moisture contents of culture media, while lower activity of these enzymes was observed with 40% moisture contents of sawdust culture medium. The pH 4 and 9 of the sawdust media appeared to be optimum pH for the. production of protease while pH 5 and 7 were optimal for the production of phenoloxidase. The pH 6 of the sawdust medium was optimal for the production of cellulase. The optimum incubating temperature for the production of protease, phenoloxidase and cellulase was $25^{\circ}C$. Higher total activities of protease and phenoloxidase were observed when culture medium was added with wood vinegar at the control, and 0.5% for cellulase.

So this study has compared with characteristics of resistance depending on the shape of the lower part of fin keel in the same surface of water submersion, we have attached three different types of models of lower part of fin keels to the model ships using circulating water channel and analyzed resistance characteristics per shape to arrive at the optimum shape of reduction of resistance.
In the present study, the optimization of poly(${\gamma}$-glutamic acid) (${\gamma}$-PGA) production by Bacillus sp. FBL-2 was studied using a statistical approach. One-factor-at-a-time method was used to investigate the effect of carbon sources and nitrogen sources on ${\gamma}$-PGA production and was utilized to select the most significant nutrients affecting the yield of ${\gamma}$-PGA. After identifying effective nutrients, response surface methodology with central composite design (CCD) was used to obtain a mathematical model to identify the optimum concentrations of the key nutrients (sucrose, $\text\tiny{L}$-glutamic acid, yeast extract, and citric acid) for improvement of ${\gamma}$-PGA production. The optimum amount of significant medium components appeared to be sucrose 51.73 g/l, $\text\tiny{L}$-glutamic acid 105.30 g/l, yeast extract 13.25 g/l, and citric acid 10.04 g/l. The optimized medium was validated experimentally, and ${\gamma}$-PGA production increased significantly from 3.59 g/l (0.33 g/l/h) to 44.04 g/l (3.67 g/l/h) when strain FBL-2 was cultivated under the optimal medium developed by the statistical approach, as compared to non-optimized medium.
Small scale mushroom growers take a lot of the costs in the bottle cultivation than the large scale growers. Therefore, they are not competitive in the market. Mushroom cultivation in small scale mushroom farm is labor-intensive and this experiment was carried out to establish the method for the cultivation of various kinds of mushrooms together under the similar conditions in the production system. As a result, the common medium were selected based on the growing conditions of Pleurotus eryngii and Agrocybe cylindracea, and the level of medium moisture content were studied. The results were shown below. When the medium in the input stage for bottle cultivation was filled by using automatic machine, the range of the three state rates in the bottle is different depending on ingredients and the mixing ratio. The optimum moisture content of the medium for some mushroom production was a different trend based on the medium cluster of the raw materials. The optimum moisture content for P. eryngii in the medium was 70% (douglas fir sawdust, rice bran or sawdust, dried bean curd refuse). In the medium containing douglas fir sawdust, wheat bran was 65%, and the medium including douglas fir sawdust, wheat bran, dried bean curd refuse was 67%. The optimum moisture content of the above three media for A. cylindracea was 70%. The suitable medium for the bottle cultivation of P. eryngii was selected as douglas fir sawdust 75%, wheat bran 20%, dried bean curd refuse 5%, and moisture content 67%. The medium of A. cylindracea was selected as douglas fir sawdust 75%, wheat bran 25%, and moisture content 70%. The suitable medium for bottle cultivation of P. eryngii and A. cylindracea was shown as douglas fir sawdust 75%, rice bran20%, dried bean curd refuse 5%, and 70% moisture content to be used as a common medium of the growing. The incubation period, primordial formation, and growth environment conditions of P. eryngii and A. cylindracea were a similar trend. Therefore, the small scale farmers of P. eryngii can cultivate A. cylindracea together with P. eryngii.

In order to find the optimum culture condition for marine free-living nematoda in the laboratory, various agar media were developed and experiments were carried on nematodes. Nematodes have collected from the bottom of the sandy sediments' surface layer (about 5cm sediment) of Taean Mallipo beach's intertidal zone. Especially, with regard to agar medium, Killian medium was transformed slightly, density of agar had made a difference and this agar medium(height 2.0 mm on diameter 60 mm Petri dish) was divided. It was mixed with 5 different species of microorganism as nematodes' live food and added each culture medium. Five individuals of Enoplolaimus sp. on each culture medium were grown in a culture medium which was set on $20^{\circ}C$ and light blocked. Moreover, as a result of the optimum culture condition, 1.0% density of agar showed the highest survival rates (the average time of survival is 246.8 hours). On the other hand, the 0.4% density of agar adding Killian medium, bacto peptone and beef extract showed the lowest survival rates, which indicates the average time of survival is 27.6 hours. About Bathylaimus sp., on Killian medium's 1.0% density of agar(no feeding amount) showed the highest survival rates, which connects that the hatching rate 94.7% after 99.5 hours and it laid 7 eggs averagely on the spawning amount and the hatching rate experiment.
The optimum culture conditions for an antibiotics from Actinomyces sp. were investigated. The optimum composition of medium for antibiotics production was 1% glucose, 1% soybean meal, 0.5% NaCl, 0.1% $CaCO_2$, and the optimum initial pH was 7.0. And the antibiotics showed highest activity when the strain isolated from soil was aerobically cultivated at $28^{\circ}C$ for 72hours under the optimum conditions. A production of the antibiotics from Actinomyces sp. begins at the 36th hours and then reached the maximum at the stationary phase developed at the 72th hours under the optimum conditions.
Celluloytic microorgnasims were isolated form the various sources and four of them were identified as Trichoderma roningi, Aspergillus niger, Penicillium chrysogenum, and Streptomyces sp. The induction of extracellular5 cellulase of these species in the liquid culture media containing carboxymethylcellulose (CMC) or Avicel as inducer showed that CMC was a better effective inducer for the production of CMCase(Cx cellulase component) as well as Avicelase(C$_{1}$ cellulase component) than Avicel. It is believed that certain hydrolysis products of cellulose(CMC) could serve as an inducer for an enzyme synthesis. In T. roningi, Asp. niger, and Strptomyces sp., the optimum temperature of CNCase on CMC-culture medium was 50.deg. but temperature around 40.deg.C was found to be optimum for the activities of CMCase prepared from P.ehrysogenum. The optimum temperature for Avicelase activitiles on Avicel-culture media of T. roningi and P. chrysogenum was $40{\circ}C$ whereas temperature $50{\circ}C$ was found to be optimum for Avicelase from A.niger and Streptomyces sp. The optimal activities of these CNCase and Avicelase prepared from. T. ronigi, Pen.chrysogenum and Streptomyces sp. were found similarly to be at pH's around 5.4 and 6.0 while pH 4.8 was optimum for the activities of CMCase and Avicelase from A.niger, indicating that A.niger in acidic media would yield an enzyme of high activity.
