Treatment conditions for the induced diploid gynogenesis, a maternal-exclusive form of artificial parthenogenetic reproduction, were optimized in bagrid catfish (Leiocassis ussuriensis, Siluriformes). Optimal amounts of ultraviolet (UV) irradiation for the genetic inactivation of spermatozoa in bagrid catfish and Pseudobagrus fulvidraco were proven to be ranged from 3,600 to 4,800 $ergs/mm^2$ based on the examination of viability of embryos and haploid incidence. Haploid embryos were restored to diploidy by preventing the extrusion of the second polar body using cold shock treatment. Thermal treatments (4 or $6^{\circ}C$ for 30, 40 or 50 min) were carried out 3, 5 or 7 min post insemination. Best scores for embryo viability (38.6% of total eggs taken) and incidence of normal diploidy (87.9% of hatched larvae) were observed at the embryo group treated at $4^{\circ}C$ for 40 min, 5 min after insemination. Restoration of gynogenetic diploidy was confirmed based on the absence of haploid syndrome, cell size and/or nucleolar organizing region (NOR) counts.
Dispersal polymorphism in insects Is a kind of adaptive strategy of the life history together with the diapause, consisting of the “long-winged or alate forms” of migratory phase and the “short-winged or apterous forms” of stationary phase. Dispersal polymorphism is a polymorphism related with the flight capability, and has three categories ; the wing polymorphisms, flight muscle polymorphisms, and flight behavior variations. Phase variation is another type of dispersal polymorphism varying in morphology, physiology and wing forms in response to the density of the population. The dispersal migration is a very adaptive trait that enables a species to keep pace with the changing mosaic of its habitat, but requires some costs. In general, wing reduction has a positive effect on the reproductive potential such as earlier reproduction and larger fecundity The dispersal polymorphism is a kind of optimization in the evolutionary strategies of the life history in insects; a trade-off between the advantages and disadvantages of migration. Wing polymorphism is a phenotypically plastic trait. Wing form changes with the environmental conditions even though the species is the same. Various environmental factors have an effect on the dispersal polymorphisms. Density dependent dispersal polymorphism plays an important role In population dynamics, but it is not a simple function of the density; the individuals of a population may be different in response to the density resulting different outcomes in the population biology, and the detailed information on the genotypic variation of the individuals in the population is the fundamental importance in the prediction of the population performances in a given environment. In conclusion, the studies on the dispersal polymorphisms are a complicated field in relation with both physiology and ecology, and studies on the ecological and quantitative genetics have indeed contributed to understanding of its important nature. But the final factors of evolution; the mechanisms of natural selections, might be revealed through the studies on the population biology.
PURPOSE. These days, mesenchymal stem cells (MSCs) have received worldwide attention because of their potentiality in tissue engineering for implant dentistry. The purpose of this study was to evaluate various growth inducing factors in media for improvement of acquisition of bone marrow mesenchymal stem cells (BMMSCs) and colony forming unit-fibroblast (CFU-F). MATERIALS AND METHODS. The mouse BMMSCs were freshly obtained from female C3H mouse femur and tibia. The cells seeded at the density of $10^6$/dish in media supplemented with different density of fetal bovine serum (FBS), $1{\alpha}$, 25-dihydroxyvitamin (VD3) and recombinant human epidermal growth factor (rhEGF). After 14 days, CFU-F assay was conducted to analyze the cell attachment and proliferation, and moreover for VD3, the 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay was additionally conducted. RESULTS. The cell proliferation was increased with the increase of FBS concentration (P<.05). The cell proliferation was highest at the density of 20 ng/mL rhEGF compared with 0 ng/mL and 200 ng/mL rhEGF (P<.05). For VD3, although the colony number was increased with the increase of its concentration, the difference was not statistically significant (P>.05). CONCLUTION. FBS played the main role in cell attachment and growth, and the growth factor like rhEGF played the additional effect. However, VD3 did not have much efficacy compare with the other two factors. Improvement of the conditions could be adopted to acquire more functional MSCs to apply into bony defect around implants easily.

A study on optimizing the friction welding of copper(C1100) to aluminium(A1050) for developing the electrical sleeve was experimentally carried out and also on real-time nondestructive evaluation of the friction weld quality (strength) was accomplished by acoustic emission technique. The results obtained are summarized as the following ; 1) The heating upset $U_1$(mm) or total upset U(mm) tends to increase according to the increase of heating time $t_1$(sec). The relations between $U_1$ and $t_1$ or U and $t_1$are computed as follows when n=2000rpm, $P_1$=4, $P_2$=8kgf/$mm^2$, and $t_2$=6sec. U=1.6$e^{0.39t_1}$$U_1$=3.65$e^{0.25t_1}$. 2) It was notified that the proper welding conditions by considering on both strength with more than 100% joint effieciency and toughness are heating time of 1.5-2.25 sec under n=200rpm, $P_1$=4, $P_2$=8kgf/$mm^2$, $t_2$=6sec. 3) It was confirmed that both AE total counts(N, counts) and the weld tensile strength (${\sigma}$, kgf/$mm^2$) of the welded joints increase as the increase of heating time, respectively, the relations between N and $t_1$, ${\sigma}$ and $t_1$ are computed from data points by regression analysis using the least square method as follows in case of the above proper condition ; N=50108+23917(ln $t_1$)${\sigma}$$=11.85+2.06(ln $t_1$). 4) Both empirical and calcularated equations of relationship between .sigma. and N are very coincident with a high reliability, as the following in case of the above proper welding condition ; Calculated : ${\sigma}$=0.00008N+7.5 Empirical :${\sigma}$= $8.17e^{0.0000072N}$. 5) It was confirmed that the real-time nondestructive weld strength evaluation for friction welding of copper(C1100) to aluminium(A1050) could be possible by acoustic emission technique.
To improve the productivity of peroxidase (POD) of cell line SP-47 derived from cell suspension cultures of sweet potato (Ipomoea batatas (L) Lam.cv White Star), we optimized culture conditions including the composition and concentration of plant growth regulators and carbon source, and the cell inoculum size. When one g (fr wt) of cells was inoculated into 50 mL TL medium supplemented with l mg/L 2,4-D and 30g/L sucrose in 300 mL Erlenmeyer flask at 25$^{\circ}C$ in the dark (100rpm), the POD activity per g cell dry wt was maximized to be about 6,800 units after 25 days of subculture, which was about 30 times higher than that of intact roots of horseradish plants grown in the greenhouse, but the cell growth was maximum after 15 days of subculture. The protein content per g cell dry wt maintained almost plateau and after 25 days of subculture decreased as culture Proceeded further whereas the POD specific activity (unit/mg protein) was about two times higher after subculture and continuously increased from 12 days to the end of cultures (40 days). The POD isozyme patterns showed almost the same regardless of cell growth stage, but some acidic isozymes were slightly increased after 25 days of subculture. These results indicate that POD activity in suspension cultures of sweet potato is closely associated with cell growth and stresses derived from cell culture renditions and medium depletion. Due to its high POD activity the SPL47cell line seems to be suitable for the mass production of POD.
Parameters related to dissolved oxygen for the production of pullulan by Aureobasidium pullulans HP-2001 were optimized in 7 l and 100 l bioreactors. The optimal concentrations of glucose and yeast extract for the production of pullulan were 50.0 and 2.5 g/l, respectively, and its conversion rate from glucose was 37% at a flask scale. The optimal initial pH of the medium and temperature for cell growth were 7.5 and $30^{\circ}C$, whereas those for the production of pullulan were 6.0 and $25^{\circ}C$. The optimal agitation speed and aeration rate for cell growth were 600 rpm and 2.0 vvm in a 7 l bioreactor, whereas those for the production of pullulan were 500 rpm and 1.0 vvm. The production of pullulan with an optimized agitation speed of 500 rpm and aeration rate of 1.0 vvm was 18.13 g/l in a 7 l bioreactor. Maximal cell growth occurred without inner pressure, whereas the optimal inner pressure for the production of pullulan was 0.4 kgf/$cm^2$ in a 100 l bioreactor. The production of pullulan under optimized conditions in this study was 22.89 g/l in a 100 l bioreactor, which was 1.38 times higher than that without inner pressure.
Cyclotrimethylene trinitramine (RDX) is a high explosive commonly used for military applications. Submicronization of RDX particles has been a critical issue in order to alleviate the unintended and accidental stimuli toward safer and more powerful performances. The purpose of this study is to optimize experimental variables for drowning-out crystallization applied to produce submicron RDX particles. Effects of RDX concentration, anti-solvent temperature and anti-solvent mass were analyzed by the central composite rotatable design. The adjusted determination coefficient of regression model was calculated to be 0.9984 having the p-value less than 0.01. Response surface plots based on the central composite rotatable design determined the optimum conditions such as RDX concentration of 3 wt%, anti-solvent temperature of $0.2^{\circ}C$ and anti-solvent mass of 266 g. The optimum and experimental diameters of RDX particles were measured to be $0.53{\mu}m$ and $0.53{\mu}m$, respectively. The regression model satisfactorily predicts the average diameter of RDX particles prepared by drowning-out crystallization. Structure of RDX crystals was found to be ${\alpha}$-form by X-ray diffraction analysis and FT-IR spectroscopy.
Coptis chinensis Franch is a valuable traditional oriental medicinal plant used for the treatment of various diseases. The major factors affecting the content of bioactive compounds and the relationship between bioactive compounds and antioxidant capacities of Coptis chinensis Franch were poorly understood. Thus, effects of the solvent, temperature, and extraction time on the extraction yields of bioactive compounds and the antioxidant activity of C. chinensis Franch extracts were investigated in this work. Our cluster analysis indicated that the hydroalcoholic solvent (50% ethanol : 50% water) at $35^{\circ}C$ for 30 min (extract time) was the best extraction condition for a factory use because the highest level of bioactive compounds and antioxidant activities was achieved. Multiple linear regression analysis revealed that total phenolic content (TPC) contributed to the 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) scavenging activity, while both total alkaloid content (TAC) and total flavonoid content (TFC) were responsible for ferric reducing antioxidant power (FRAP) activity. These results indicated that extraction conditions controlled the yield of bioactive compounds and the antioxidant activity of C. chinensis Franch, which can provide important information for the industrial extraction.
This study was carried out to optimize the conditions for the extraction of antioxidative materials from licorice root, Glycyrrhiz glabra. Chipped licorice roots were extracted with several solvents and their antioxidative activities were tested to determine the optimal extraction solvent. Among the solvents tested, 95% ethanol gave the highest free radical scavenging activity, and was therefore chosen as the optimal extracting solvent. The optimum extraction temperature and time were $20^{\circ}C$ and 12 hr, respectively. Next, the free radical scavenging activity of the ethanol extract was compared with that of other known antioxidants such as ${\alpha}-tocopherol$, butylated hydroxytoluene (BHT) and butylated hydroxyanisole (BHA). Ethanol extract of licorice root had greater antioxidative activity than ${\alpha}-tocopherol$ and a similar level to that of the two synthetic antioxidants (BHA and BHT). Moreover, the antioxidative activity of the ethanol extract was inhibited neither by heat treatment at $180^{\circ}C$ for 30 min nor by treatment at extreme pH. These findings suggest that ethanol extract of G. glabra may be useful as a natural antioxidant.

In 3-dimensional (3D) integrated package, thermal management is one of the critical issues due to the high heat flux generated by stacked multi-functional chips in miniature packages. In this study, we used numerical simulation method to analyze the thermal behaviors, and investigated the thermal issues of 3D package using TSV (through-silicon-via) technology for mobile application. The 3D integrated package consists of up to 8 TSV memory chips and one logic chip with a interposer which has regularly embedded TSVs. Thermal performances and characteristics of glass and silicon interposers were compared. Thermal characteristics of logic and memory chips are also investigated. The effects of numbers of the stacked chip, size of the interposer and TSV via on the thermal behavior of 3D package were investigated. Numerical analysis of the junction temperature, thermal resistance, and heat flux for 3D TSV package was performed under normal operating and high performance operation conditions, respectively. Based on the simulation results, we proposed an effective integration scheme of the memory and logic chips to minimize the temperature rise of the package. The results will be useful of design optimization and provide a thermal design guideline for reliable and high performance 3D TSV package.
