Optimization of Soybean Oil Fatty Acid Methyl Esters Preparation for Sucrose Polyesters Synthesis (Sucrose polyesters 합성에 사용하는 대두유 지방산 메틸에스테르 제조의 최적화)

  • Preparation of soybean oil fatty acid methyl esters (soybean oil FAME) through the transesterification of soybean oil with alkaline catalyst was optimized in terms of contents of residual free fatty acids (FFA) in soybean oil FAME and yield of soybean oil FAME due to the inhibitory effect of FFA on sucrose polyesters synthesis. Soybean oil FAME and residual FFA were analyzed quantitatively by simultaneous gas chromatography on a fused silica capillary column after converting the FFA in soybean oil FAME to tert.-butyldimethylsilyl (TBDMS) derivatives. Transesterification of soybean oil was successfully performed with alkaline catalyst (NaOH, 95%), which resulted in 99.1% yield of soybean oil FAME and less than 0.1% residual FFA contents under the conditions such as $30^{\circ}C$, 20min. and 6:1 molar ratio of anhydrous methanol to soybean oil.

  • Gerber (Gerbera hybrida) is a valuable ornamental species grown as a potted plant and cut flowers. However, genetic variability within the gerbera genus is very limited. So it is absolutely needed to introduce and widen genetic resources into gerbera lines by genetic transformation. For the purpose, 18 Korean gerbera lines were screened to establish Agrobacterium-mediated genetic transformation procedure. In an experiment to select Korean gerbera lines which are amenable to Agrobacterium-inoculation, 12 lines turned out to be positive in Agrobacterium-inoculation. More callus were produced from BA 2ppm, Zeatin 2ppm, IAA 0.2ppm in pre-culture and regeneration medium (2X media) but there was no difference in the frequency of GUS expression rate. In another experiment to find out optimal condition for highly efficient Agrobacterium-inoculation, petiole and leaf explants have been treated with four different pre-culture periods, two different co-culture periods and two different Agrobacterium strains. As a result, high GUS expression has been showed from petiole and leaf explants treated no pre-culture period with LBA4404 Agrobacterium tumerfaciens, 5 day co-culture period and dipping treatment.

  • The purpose of this study is to suggest a method of predicting seismic vulnerability and safety conditions of each building in a targeted area. The scope of this study includes 'developing a simulation model for precaution activities,' 'testing the validity of the developed model', From the facility point of view, target of this study is a local building system. According to the literature review, the number of earthquake prediction modeling and cases with GIS applied is extremely few and the results are not proficient. This study is conducted as a way to improve the previous researches. Statistic analyses are conducted using 348 domestic and international data. Finally, as a result of the series of statistical analyses, an adequate model is developed using optimization scale method. The ratio of correct expectation is estimated as 87%. In order to apply the developed model to predict the vulnerability of the several chosen local building systems, spatial analysis technique is applied. Gangnam-gu and Jongro-gu are selected as the target areas to represent the characteristics of the old and the new downtown in Seoul. As a result of the analysis, it is discovered that buildings in Gangnam-gu are relatively more dangerous comparing to those of Jongro-gu and Eunpyeong-gu.

  • Digital imaging for general rediography has many advantages over the film/screen systems, including a wider dynamic range and the ability to manipulate the images produced. The wider range means that acceptable images may by acquired at a range of dose levels, and therefore repeat exposures can be reduced. Digital imaging can result in the over use of radiation, however, because there is a tendency can be reduced. We investigated the actual exposure dose conditions on general radiography and a questionnaire survey was conducted with radiotechnologiest at medical institutions using digital radiology system. As a results, the dose of exposure was not controlled with patient's figure and dose optimization but was controlled by worker's convenience and image quality. Radio-technologiests often set up the exposure dose regardless of patient figure and body part to be examined. Many organizations, such as the International Commission on Radiological Protection, recommend to keep the dose as low as possible. In addition, they strongly recommend to keep the optimal but minimal dosage by proper training programs and constant quality control, including frequent patient dose evaluations and education.

  • The growing demand for customer-responsive, made-to-order manufacturing is stimulating the need for improved dynamic decision-making processes in commercial fleet operations. Moreover, the rapid growth of electronic commerce through the internet is also requiring advanced and precise real-time operation of vehicle fleets. Accompanying these demand side developments/pressures, the growing availability of technologies such as AVL(Automatic Vehicle Location) systems and continuous two-way communication devices is driving developments on the supply side. These technologies enable the dispatcher to identify the current location of trucks and to communicate with drivers in real time affording the carrier fleet dispatcher the opportunity to dynamically respond to changes in demand, driver and vehicle availability, as well as traffic network conditions. This research investigates key aspects of real time dynamic routing and scheduling problems in fleet operation particularly in a truckload pickup-and-delivery problem under various settings, in which information of stochastic demands is revealed on a continuous basis, i.e., as the scheduled routes are executed. The most promising solution strategies for dealing with this real-time problem are analyzed and integrated. Furthermore, this research develops. analyzes, and implements hybrid algorithms for solving them, which combine fast local heuristic approach with an optimization-based approach. In addition, various partitioning algorithms being able to deal with large fleet of vehicles are developed based on 'divided & conquer' technique. Simulation experiments are developed and conducted to evaluate the performance of these algorithms.

Process Optimization of ITO Film on PC Substrate Deposited by In-line Sputtering Method for a Resistive-type Touch Panel (인라인 스퍼터링에 의한 저항막 방식 터치패널용 ITO 기판 제조공정 최적화 기술)

  • Indium tin oxide(ITO) substrate is one of the key components of the touch panel and its sputtering process is dependent on the characteristics of various touch panel, such as driving type, size of panel, and the intended use. In this study, we optimized the sputtering condition of ITO film on polycarbonate(PC) by using in-line sputtering method for the application to resistive type touch panel. We varied the $O_2$/Ar gas ratio, sputtering power, pressure and moving speed of substrate to deposit ITO films at room temperature with the base vacuum of $1{\times}10^{-6}\;torr$. The sheet resistance and its uniformity, the transmittance, the thickness of the ITO film on PC substrate are investigated and analyzed. The optimized process parameters are as follows : the sheet resistance is $500{\pm}50\;{\Omega}$/□, the uniformity of sheet resistance is lower than 10%, the transmittance is higher than 87 % at 550nm, and the thickness is about 120~250. The optimized deposition conditions by in-line sputtering method can be applied to the actual mass production for the ITO film manufacturing technology.

Optimization of Wool Dyeing with Yellow Dye from Carthamus Tinctorius L. (홍화 황색소를 이용한 모염색의 최적화)

  • This study investigated the adsorption of safflower yellow dye on wool protein fiber and the optimum dyeing conditions to test color reproducibility. In addition, the effects of mordants on dye adsorption, color, fastness, and photofading rate were also studied. The prepared dye in powder form was characterized with UV-vis spectroscopy and FT-IR spectrometric analysis. The color of dyed fabrics was characterized by CIE $L^*a^*b^*$ coordinates, H V/C, and K/S values. The color reproducibility of the dyed wool fabrics was examined. The amount of dye adsorption increased and also, the shade of the dyed wool fabrics became deeper and more saturated with increasing temperature, time, and dye concentration. The maximum color strength was obtained at pH 3.0. The shade of dyed wool fabrics ranged from light yellow to dark mustard yellow as the pH of the dye bath shifted from alkaline to acidic. Color reproducibility was reliable with color differences in the range of 0.53~1.75. Fastness to dry cleaning was relatively good at 4/5 rating irrespective of mordanting. Fe and Cu mordants showed the least color change of the dyed wool fabrics after exposure to light. Mordants did not contribute to improve dye uptake and color fastness, although they made variations in color tone. Safflower yellow dye can be used satisfactorily without mordants and will not cause damage to the environment.

Fabrication of a Partial Genome Microarray of the Methylotrophic Yeast Hansenula polymorpha: Optimization and Evaluation of Transcript Profiling

  • The methylotrophic yeast Hansenula polymorpha has been extensively studied as a model organism for methanol metabolism and peroxisome biogenesis. Recently, this yeast has also attracted attention as a promising host organism for recombinant protein production. Here, we describe the fabrication and evaluation of a DNA chip spotted with 382 open reading frames (ORFs) of H. polymorpha. Each ORF was PCR-amplified using gene-specific primer sets, of which the forward primers had 5'-aminolink. The PCR products were printed in duplicate onto the aldehyde-coated slide glasses to link only the coding strands to the surface of the slide via covalent coupling between amine and aldehyde groups. With the partial genome DNA chip, we compared efficiency of direct and indirect cDNA target labeling methods, and found that the indirect method, using fluorescent-labeled dendrimers, generated a higher hybridization signal-to-noise ratio than the direct method, using cDNA targets labeled by incorporation of fluorescence-labeled nucIeotides during reverse transcription. In addition, to assess the quality of this DNA chip, we analyzed the expression profiles of H. polymorpha cells grown on different carbon sources, such as glucose and methanol, and also those of cells treated with the superoxide­generating drug, menadione. The profiles obtained showed a high-level induction of a set of ORFs involved in methanol metabolism and oxidative stress response in the presence of methanol and menadione, respectively. The results demonstrate the sensitivity and reliability of our arrays to analyze global gene expression changes of H. polymorpha under defined environmental conditions.

Microwave Digestion and Solid-Phase Extraction for Determination of Aluminum in Human Urine by Graphite Furnace Atomic Absorption Spectrometer (흑연로 원자흡수 분광법에 의한 사람 오줌 중 알루미늄의 정량을 위한 마이크로파 삭힘과 고체상 추출)

  • Microwave digestion and solid-state extraction were studied for determination of trace aluminum{Al(III)} in human urine samples. A mixed acid of nitric acid and hydrogen peroxide was added to urine samples, organic materials were destructed in a home microwave oven and dried in a drying oven. The dried residues were dissolved in a sulfuric acid solution. The solution was eluted through a XAD-4 resin column adsorbed with 8-hydroxyquinoline(Oxine, HQ). Al(III)-8-hydroxyquinolinate complex was formed in the column and eluted with 0.5 M nitric acid solution. The Al(III) eluted was determined by graphite atomic absorption spectrophotometry. Various experimental conditions of followings were investigated for the optimization : the type of acid to dissolve the residues, the amount of HQ adsorbed on the resin, the pH of sample solutions, the type and concentration of acid to elute the complex from column and so on. The contents of Al(III) in real samples were determinated by a calibration curve method. The recovery in standard spiked samples was 94~101% and the detection limit of this procedure was 0.05 ng/mL.

Optimization of Extraction Method for the Quantitative Analysis of Gallic Acid from Cornus officinallis (산수유로부터 gallic acid 추출 및 HPLC에 의한 정량분석)

  • Cornus fructus has been used as a tonic, astringent, and haemostatic agent in Korea, China, and Japan. In this study, the fruit of Cornus officinalis was treated with different osmotic pressures, pH values, heat, and ethanol percentages in order to establish optimum extraction conditions for gallic acid, an example of a hydrolyzable tannin. The extract was analyzed by HPLC and LC-MS/MS to identify the gallic acid. As a result, the highest extraction rate of gallic acid (1.57 mg/g) occurred when the Cornus fructus was extracted with 100% ethanol for 1 hr at $80^{\circ}C$. Also, when it was treated with 70% ethanol for 24 and 48 hr, contents of gallic acid were 1.35 and 1.50 mg/g, respectively.
