  • A highly productive fed-batch fermentation process was developed for the production of L-ornithine by using a new stabilized strain, Breviabcterium ketoglutamicum BK52. Fed-batch cultures with a continuous feeding of the complex medium were conducted on various operating conditions. The optimal concentration of phosphate in the complex medium was 2.1g/L. The optimal feeding rate of L-arginine was 0.028g/L/hr. The optimal feeding point of the complex medium was determined to be at 40 OD of the cell mass. The final L-ornithine concentrations within 64hrs of cultivation in 5 and 50 liter fermenters were 73g/L and 71g/L, respectively. The maximum overall L-ornithine productivity was 1.14g/L/hr which was about 2 times higher than that of the conventional fed-batch culture with intermittent feeding. The overall productivity of the fermentation system is remarkably improved by employing the optimized conditions, and it offers a significant potential for industrial application.

Isolation and Cultivation Characteristics of Acetobacter xylinum KJ-1 Producing Bacterial Cellulose in Shaking Cultures

  • Eight strains producing bacterial cellulose (BC) were isolated from rotten fruits and traditionally fermented vinegars. One of the isolated strains from the rotten grape in Gwangju, Korea, maintained a relatively stable BC production in shaking cultures. This isolated strain proved to be Acetobacter xylinum, based on several biochemical and morphological tests. It was shown that the slant-baffled flask was more efficient than the conventional flask for the BC production in shaking cultures. To determine the most suitable carbon and nitrogen sources for the production of BC, various compounds were examined. Fructose was found to be the most effective carbon source with an optimal concentration of 2%. Mixed carbon source (glucose:fructose=1:3) was also better than glucose or fructose alone. Optimal nitrogen source, when basal medium was used, was 10% (v/v) com steep liquor (CSL). When com steep liquor was used with a mixed carbon source (glucose:fructose=1 :3),4% CSL exhibited the best BC production. Based on these results, a defined medium was developed for the BC production by Acetobacter xylinum KJ-1. When this medium was used under optimal culture conditions, the BC production was 7.2 g/1, which was approximately 3 times higher than that with the traditional HS medium.

Establishment of In-Vitro Culture System for Enhancing Production of Somatic Cell Nuclear Transfer (SCNT) Blastocysts with High Performance in the Colony Formation and Formation of Colonies Derived from SCNT Blastocysts in Pigs

  • Although somatic cell nuclear transfer (SCNT)-derived embryonic stem cells (ESCs) in pigs have great potential, their use is limited because the establishment efficiency of ESCs is extremely low. Accordingly, we tried to develop in-vitro culture system stimulating production of SCNT blastocysts with high performance in the colony formation and formation of colonies derived from SCNT blastocysts for enhancing production efficiency of porcine ESCs. For these, SCNT blastocysts produced in various types of embryo culture medium were cultured in different ESC culture medium and optimal culture medium was determined by comparing colony formation efficiency. As the results, ICM of porcine SCNT blastocysts produced through sequential culture of porcine SCNT embryos in the modified porcine zygote medium (PZM)-5 and the PZM-5F showed the best formation efficiency of colonies in α-MEM-based medium. In conclusion, appropriate combination of the embryo culture medium and ESC culture medium will greatly contribute to successful establishment of ESCs derived from SCNT embryos.

Optimization of Monochamus alternatus media and culture period for cordycepin production in Cordyceps militaris culture using solid-state fermentation

  • In this study, we investigated the effect of solid culture medium on the production of cordycepin in Cordyceps militaris. The regression equation was expressed as follows: Y1 = 755.3-58.6625X1+4.79432E-14X2-46.6625X3-5.66269E-14X1X2-0.025X1X3+1.62475E-14X2X3-160.6625X12+0.0125X22-206.9625X32, where, Y represents the value of cordycepin content (㎍/g), X1 corresponds to the weight of M. alternatus in solid culture medium (g/bottle), X2 to the water content of the solid culture medium (%), and X3 to the culture period (day). The solid culture medium was optimized using the response surface methodology, and the optimal medium composition was as follows: the weight of M. alternatus in solid culture medium and water content were 16.2% and 100.7% (20.14 mL water/20 g solid culture medium), respectively, with a culture period of 39 days. Under these conditions, the cordycepin content of the fruiting bodies reached 150.0 ㎍/g (actual value). The supplementation of M. alternatus in solid culture for improved cordycepin content of C. militaris seems to be a promising alternative to wild and solid cultivation.

눈꽃동충하초균의 액체배양법 개선 (Improvement of Liquid Culture Methods of Paecilomyces japonica)

  • This study was carried out to improve the liquid culture methods of Paecilomyces japonica. The results show that the size of granular mycelium is smaller when the shaking speed is increased. Especially, the granular mycelium is the smallest at the shaking speed of 150rpm under the photoperiod of 12L-12D. Dry weight of mycelium was averagely 1.216 g in the Sikworm larva (SL) medium, and the weight was 2 times heavier than in the Potato dextrose (PD) medium. By adding 6 g of 6 mmbeads in the SL medium, the dry weight is increased to 1.332 g. The optimal addition of silkworm larval powder to the culture medium for gest harvest was 1.360$\pm$0.67 g in dry weight.

Optimizing Fermentation Medium Composition for Bacterial Cellulose Production by a Newly Isolated Gluconacetobacter sp. RKY5

  • In this study, we investigated the optimal medium composition for bacterial cellulose (BC) production by Gluconacetobacter sp. RKY5. Among the various kinds of carbon sources, glycerol was the most efficient as a sole carbon source and its optimal concentration for BC production was 15 g/L. The optimal concentration of yeast extract as a nitrogen source for BC production was found to be 8 g/L. $K_{2}HPO_{4}$ and acetic acid were selected respectively as a phosphate source and a secondary substrate, and both optimal concentrations were 3 g/L. The amount of produced BC was 4.59 g/L in a static culture and 6.5 g/L in a shaking culture condition with 150 rpm. These values were 2.1 and 2.7 times higher than those in a static (2.16 g/L) and a shaking (2.41 g/L) cultures using HS medium generally used for BC production.

잿빛공팜이에 대한 길항균 Bacillus Iicheniformis N1의 배양적 특성 (Cultural Characteristics of Antagonistic Bacterium, Bacillus licheniformis N1 against Botrytis cinerea)

  • This study was conducted to estimate the cultural characteristics, the production of antibiotic, and the selection of optimal media for mass culture of Bacillus licheniformis N1 isolate which was previously reported as an antagonistic bacterium to Botrytis cinerea. We investigated initial pH, temperatures and shaking speed for good cultural conditions and antibiotics production by N1 isolate. According to the results, the optimal conditions of initial pH, temperatures, and shaking speed were determined to be pH 5.0~5.5, 30~35$^{\circ}C$ and 250 rpm, respectively. Also, the optimal conditions for the antagonism by N1 isolate highly appeared in the initial pH as 5.0, and the mycelial growth inhibition was high when the substances used such as glucose or corn starch as carbon sources, and biji(soybean curd residue) flour as a nitrogen source. Furthermore, inhibitory area was significantly expanded, when 3% or 5% of corn starch was added into 5% of Biji flour as nitrogen source, were respectivley selected for mass culture of N1 isolate. Among them, 5% Biji flour medium showed higher cell density more than 10 times that in NB medium after 48 hour incubation. Therefore, the optimal medium was determined as 5% biji flour added 3~5% of corn starch for high density of cells.

  • Aspergillus terreus NRRL 1960에 의한 이다콘산의 생산에 관해 연구하였다. pH, 접종량, 배지조성 등의 최적조건을 확립하였다. 이타콘산의 최대생산 량은 37°C, pH 2.5의 조건에셔, 5% (w/v) glu­c cose, 0.5 % (w/v) $NH_4Cl$, 0.2 % (w/v) yeast ex­tract, 0.1 % (w/v) $MgSO_4$, 0.2 % (w/v) NaCI을 포함하는 배지에서 7일만에 $19.18g/\ell$를 얻을 수 있었다. 종균배양 배지로서 2 % malt extract가 적합하였다. 교반식 반응기에서 유리균체에 의한 이타콘 산의 회분식 생산은 플라스크 배양에 비해 비효율적 이었다.

돼지혈액 및 간을 이용한 사료용 효모배양과 효모배양물이 육계성장에 미치는 영향 (Utilization of Porcine Blood and Liver in Yeast Culture for Animal Diets and Effects of Yeast Culture on the Growth of Broiler Chicks)

  • 도축 후 생산되는 돼지 혈액과 간을 이용한 효모의 최적 배양조건과 효모배양물이 육계의 성장에 미치는 영향을 조사하였다. 10% 돼지 혈액 상등액에 Glucose 5%를 첨가 한 BSG 배지와 20% 간 상등액에 Glucose 5%를 첨가한 LSG배지에서 24시간 동안 효로를 배양한 결과 YEPD(Yeast Exract(DIFCO) 1%, Bacto Pepton(DIFCO) 2%, Glucose 4%)배지에서 자란 효모의 수보다 각각 4%, 10% 증가하였다. 효모수를 증가하기 위해 BSG 배지에 첨가한 Ammonium sulfate 최적 농도는 각각 24시간 배양하였을 때 100mM(1.3%)이었으며, 48시간 배양하였을 때에는 50mM(0.65%)로 나타났다. BSG 배지에서 효모배양의 최적 pH는 약 6에서 7 정도였다. LSG 배지에서 Ammonium sulfate와 Taurine을 1%씩 첨가하였을 때 첨가하지 않은 배지에 비해 효모수가 각각 18%와 9% 증가하였을 때 체중이 무첨가구에 비해 4주령에서 10% 증가하였다. 따라서 본 연구의 결과 도축부산물인 혈액과 간을 가축사료를 위한 효모배양 배지 원료로서 재활용할 수 있다는 것을 의미한다.

Optimal Culture Conditions for in vitro Propagation of Orostachys japonicus and Enhancement of Polysaccharide Production

  • Optimal culture conditions for efficient in vitro propagation and polysaccharide production of Orostachys japonicus were established. The highest growth yield was achieved in 1/2 MS medium, while the lowest growth yield was obtained in 4 MS medium. The patterns of polysaccharide formation were a little similar in all cases, but on MB5 medium, the po]ysaccharide contents of plant were higher than others. Among the modified nitrate levels, effective growth level were obtained in 1/4 N and 1/2 N. High contents of polysaccharide were obtained in 4 N. Different concentration of potassium and calcium did not improve the growth and polysaccharide production. The micropropagated shoots were successfully acclimatized artificial soils.
