Generation of Isthmic Organizer-Like Cells from Human Embryonic Stem Cells

  • The objective of this study was to induce the production of isthmic organizer (IsO)-like cells capable of secreting fibroblast growth factor (FGF) 8 and WNT1 from human embryonic stem cells (ESCs). The precise modulation of canonical Wnt signaling was achieved in the presence of the small molecule CHIR99021 ($0.6{\mu}M$) during the neural induction of human ESCs, resulting in the differentiation of these cells into IsO-like cells having a midbrain-hindbrain border (MHB) fate in a manner that recapitulated their developmental course in vivo. Resultant cells showed upregulated expression levels of FGF8 and WNT1. The addition of exogenous FGF8 further increased WNT1 expression by 2.6 fold. Gene ontology following microarray analysis confirmed that IsO-like cells enriched the expression of MHB-related genes by 40 fold compared to control cells. Lysates and conditioned media of IsO-like cells contained functional FGF8 and WNT1 proteins that could induce MHB-related genes in differentiating ESCs. The method for generating functional IsO-like cells described in this study could be used to study human central nervous system development and congenital malformations of the midbrain and hindbrain.

  • 해운회사에서는 현재 고유가 및 물동량 감소 등의 경영환경변화에 대처하기 위해 경쟁력강화와 경영합리화를 구현하는 전략적 선박자원관리시스템 개발에 대한 관심이 고조되고 있다. 본 논문에서는 SAN(Ship Area Network) 환경에 적합한 선박자원관리시스템을 소개한다. 이 시스템은 선박관계 당사자 요구사항을 분석하여 선주사의 경영전략 실행에 맞도록 설계되었다. 이와 함께 선박의 전 생애주기에 걸쳐서 선박자원에 대한 정보와 지식을 관리할 수 있도록 선박 온톨로지와 기계장치 및 부품에 대한 국제표준인 STEP과 PLIB를 도입할 수 있게 시스템을 설계한 다음, 비즈니스 로직과 사용자 인터페이스를 분리하여 웹기반 ERP(SHERP)로 구현하였다. 본 논문에서 기술하고 있는 시스템은 선박과 선단 운영에 적용할 수 있는 정보시스템으로서 조선산업 서비스화를 지향하는 구체적인 사례가 될 것이다.

  • 본 연구는 아동양육시설에서 성장한 개인이 어려운 환경과 다양한 역경에도 불구하고 시설퇴소 후 인간의 발달과업을 성공적으로 성취하는 것에 주목하고. 건강한 사회구성원으로서 삶을 이루는 과정에서 중요한 레질리언스 발현 과정과 내용을 분석하였다. 본 연구는 근거이론 연구방법으로 접근했으며 아동양육시설 퇴소 후 10년 이상 독립적인 생활을 한 성인 20명이 연구 참여자로 참여했다. 분석결과는 패러다임에 의한 범주분석에서 인과적 조건은 '현실적인 미래 계획 세우기', '긍정적 의미구성', '역지사지의 이해'로 나타났고, 맥락적 조건은 '자기 성찰성', '존재론적 의지처 찾기', '원초적 가족 친밀감'으로 나타났다. 중심현상은 '내실추구의 생활철학'으로 나타났다. 중재적 조건은 '세상 앞에 당당히 서기', '세상에 대한 믿음성'으로 나타났고, 작용/상호작용은 '철저한 자기관리', '가족성장 시키기'로 나타났다. 결과는 '사회적 가족만들기', '사회적 모델링되기'로 나타났다. 이상과 같은 분석결과를 토대로 논의를 거쳐 제언을 하였다.

Development of Polymorphic Simple Sequence Repeat Markers using High-Throughput Sequencing in Button Mushroom (Agaricus bisporus)

  • The white button mushroom (Agaricus bisporus) is one of the most widely cultivated species of edible mushroom. Despite its economic importance, relatively little is known about the genetic diversity of this species. Illumina paired-end sequencing produced 43,871,558 clean reads and 69,174 contigs were generated from five offspring. These contigs were subsequently assembled into 57,594 unigenes. The unigenes were annotated with reference genome in which 6,559 unigenes were associated with clusters, indicating orthologous genes. Gene ontology classification assigned many unigenes. Based on genome data of the five offspring, 44 polymorphic simple sequence repeat (SSR) markers were developed. The major allele frequency ranged from 0.42 to 0.92. The number of genotypes and the number of alleles ranged from 1 to 4, and from 2 to 4, respectively. The observed heterozygosity and the expected heterozygosity ranged from 0.00 to 1.00, and from 0.15 to 0.64, respectively. The polymorphic information content value ranged from 0.14 to 0.57. The genetic distances and UPGMA clustering discriminated offspring strains. The SSR markers developed in this study can be applied in polymorphism analyses of button mushroom and for cultivar discrimination.

LADM 기반의 몽골 지적 데이터 모델 설계에 관한 연구 (A Study on the Design of LADM-based Cadastral Data Model for Mongolia)

  • 본 연구는 현존하는 몽골 지적 시스템의 개선을 위해서 ISO 19152, 토지행정도메인모델(LADM)의 채택을 검토하였다. 현재의 몽골 지적시스템은 토지등록만을 위해 개발되었다고 할 수 있다. 따라서 이러한 점을 보완할 수 있고 현재의 문제점을 개선할 수 있는 새롭고 포괄적인 시스템이 필요한 실정이다. LADM은 국제표준으로서 2012년도 후반에 국제표준화기구(ISO)에 의해 제정되었으며, 지적 및 토지행정정보의 모델링을 통해 효율적이고 효과적인 지적시스템 개발과 지적의 공통어(온톨로지)를 제공하는 것을 목적으로 하였다. 본 연구에서는 몽골의 지적시스템인 Land Manager에 초점을 맞추어 몽골의 지적시스템에 LADM의 적용가능성을 검토하였다. 또한, Land Manager의 데이터 모델을 LADM의 해당되는 클래스별로 분석하여 각 모델간의 차이점에 의한 데이터 모델링을 수행하였다. 마지막으로 LADM 기반의 현재 문제점을 반영할 수 있는 새로운 몽골 지적시스템의 데이터 모델을 제시하였다.

Selection of candidate genes affecting meat quality and preliminary exploration of related molecular mechanisms in the Mashen pig

  • Objective: The aim of this study was to select the candidate genes affecting meat quality and preliminarily explore the related molecular mechanisms in the Mashen pig. Methods: The present study explored genetic factors affecting meat quality in the Mashen pig using RNA sequencing (RNA-Seq). We sequenced the transcriptomes of 180-day-old Mashen and Large White pigs using longissimus dorsi to select differentially expressed genes (DEGs). Results: The results indicated that a total of 425 genes were differentially expressed between Mashen and Large White pigs. A gene ontology enrichment analysis revealed that DEGs were mainly enriched for biological processes associated with metabolism and muscle development, while a Kyoto encyclopedia of genes and genomes analysis showed that DEGs mainly participated in signaling pathways associated with amino acid metabolism, fatty acid metabolism, and skeletal muscle differentiation. A MCODE analysis of the protein-protein interaction network indicated that the four identified subsets of genes were mainly associated with translational initiation, skeletal muscle differentiation, amino acid metabolism, and oxidative phosphorylation pathways. Conclusion: Based on the analysis results, we selected glutamic-oxaloacetic transaminase 1, malate dehydrogenase 1, pyruvate dehydrogenase 1, pyruvate dehydrogenase kinase 4, and activator protein-1 as candidate genes affecting meat quality in pigs. A discussion of the related molecular mechanisms is provided to offer a theoretical basis for future studies on the improvement of meat quality in pigs.

Expression profiles of microRNAs in skeletal muscle of sheep by deep sequencing

  • Objective: MicroRNAs are a class of endogenous small regulatory RNAs that regulate cell proliferation, differentiation and apoptosis. Recent studies on miRNAs are mainly focused on mice, human and pig. However, the studies on miRNAs in skeletal muscle of sheep are not comprehensive. Methods: RNA-seq technology was used to perform genomic analysis of miRNAs in prenatal and postnatal skeletal muscle of sheep. Targeted genes were predicted using miRanda software and miRNA-mRNA interactions were verified by quantitative real-time polymerase chain reaction. To further investigate the function of miRNAs, candidate targeted genes were enriched for analysis using gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) enrichment. Results: The results showed total of 1,086 known miRNAs and 40 new candidate miRNAs were detected in prenatal and postnatal skeletal muscle of sheep. In addition, 345 miRNAs (151 up-regulated, 94 down-regulated) were differentially expressed. Moreover, miRanda software was performed to predict targeted genes of miRNAs, resulting in a total of 2,833 predicted targets, especially miR-381 which targeted multiple muscle-related mRNAs. Furthermore, GO and KEGG pathway analysis confirmed that targeted genes of miRNAs were involved in development of skeletal muscles. Conclusion: This study supplements the miRNA database of sheep, which provides valuable information for further study of the biological function of miRNAs in sheep skeletal muscle.

Bioinformatics Analysis of Autophagy and Mitophagy Markers Associated with Delayed Cerebral Ischemia Following Subarachnoid Hemorrhage

  • Objective : To evaluate the interactions among differentially expressed autophagy and mitophagy markers in subarachnoid hemorrhage (SAH) patients with delayed cerebral ischemia (DCI). Methods : The expression data of autophagy and mitophagy-related makers in the cerebrospinal fluid (CSF) cells was analyzed by real-time reverse transcription-polymerase chain reaction and Western blotting. The markers included death-associated protein kinase (DAPK)-1, BCL2 interacting protein 3 like (BNIP3L), Bcl-1 antagonist X, phosphatase and tensin homolog-induced kinase (PINK), Unc-51 like autophagy activating kinase 1, nuclear dot protein 52, and p62. In silico functional analyses including gene ontology enrichment and the protein-protein interaction network were performed. Results : A total of 56 SAH patients were included and 22 (38.6%) of them experienced DCI. The DCI patients had significantly increased mRNA levels of DAPK1, BNIP3L, and PINK1, and increased expression of BECN1 compared to the non-DCI patients. The most enriched biological process was the positive regulation of autophagy, followed by the response to mitochondrial depolarization. The molecular functions ubiquitin-like protein ligase binding and ubiquitin-protein ligase binding were enriched. In the cluster of cellular components, Lewy bodies and the phagophore assembly site were enriched. BECN1 was the most connected gene among the differentially expressed markers related to autophagy and mitophagy in the development of DCI. Conclusion : Our study may provide novel insight into mitochondrial dysfunction in DCI pathogenesis.

Exploring effects of different male parent crossings on sheep muscles and related regulatory genes using mRNA-Seq

  • Objective: With improvements in living standards and increase in global population, the demand for meat products has been increasing; improved meat production from livestock could effectively meet this demand. In this study, we examined the differences in the muscle traits of different male crossbred sheep and attempted to identify key genes that regulate these traits. Methods: Dubo sheep×small-tailed Han sheep (DP×STH) and Suffolk×small-tailed Han sheep (SFK×STH) were selected to determine meat quality and production performance by Masson staining. Transcriptome sequencing and bioinformatic analysis were performed to identify differentially expressed genes (DEGs) related to meat quality. The presence of DEGs was confirmed by real-time polymerase chain reaction. Results: The production performance of SFK×STH sheep was better than that of DP×STH sheep, but the meat quality of DP×STH sheep was better than that of SFK×STH sheep. The muscle fiber diameter of DP×STH sheep was smaller than that of SFK×STH sheep. Twenty-two DEGs were identified. Among them, four gene ontology terms were related to muscle traits, and three DEGs were related to muscle or muscle fibers. There were no significant differences in the number of single nucleotide mutations and mutation sites in the different male parent cross combinations. Conclusion: This study provides genetic resources for future sheep muscle development and cross-breeding research.

Effects of Culture Mechanism of Cinnamomum kanehirae and C. camphora on the Expression of Genes Related to Terpene Biosynthesis in Antrodia cinnamomea

  • The rare edible and medicinal fungus Antrodia cinnamomea has a substantial potential for development. In this study, Illumina HiSeq 2000 was used to sequence its transcriptome. The results were assembled de novo, and 66,589 unigenes with an N50 of 4413 bp were obtained. Compared with public databases, 6,061, 3,257, and 2,807 unigenes were annotated to the Non-Redundant, Gene Ontology, and Kyoto Encyclopedia of Genes and Genomes databases, respectively. The genes related to terpene biosynthesis in the mycelia of A. cinnamomea were analyzed, and acetyl CoA synthase (ACS2 and ACS4), hydroxymethylglutaryl CoA reductase (HMGR), farnesyl transferase (FTase), and squalene synthase (SQS) were found to be upregulated in XZJ (twig of C. camphora) and NZJ (twig of C. kanehirae). Moreover, ACS5 and 2,3-oxidized squalene cyclase (OCS) were highly expressed in NZJ, while heme IX farnesyl transferase (IX-FIT) and ACS3 were significantly expressed in XZJ. The differential expression of ACS1, ACS2, HMGR, IX-FIT, SQS, and OCS was confirmed by real-time quantitative reverse transcription PCR. This study provides a new concept for the additional exploration of the molecular regulatory mechanism of terpenoid biosynthesis and data for the biotechnology of terpenoid production.
