Phenolic Acid Changes in Mycelia of Sclerotium rolfsii After Garlic and Onion Supplementation in a Broth Medium

  • High performance liquid chromatographic (HPLC) analysis of mycelia of Sclerotium rolfsii grown in broth medium supplemented with garlic (Allium sativum) and onion (Allium cepa) was carried out to estimate qualitative and quantitative changes in phenolic acids. Several phenolic acids, such as gallic, chlorogenic; ferulic, o-coumaric and cinnamic acids were detected in varied amounts in mycelia grown on such media as compared to control. Phenolic acids represents a wide range of secondary metabolite found in the cells of plants and microbes including fungi. The growth characters of S. rolfsii in various supplements also varied from thin and transparent to thick and opaque.

  • A Fructan-degrading enzyme was partially purified from onion (Allium cepa)bulbs by a combination of ammonium sufate precipitation, concanavalin-A-Affinity chromatography, and ion-exchange and gel-filtration chromatography. The enzyme hydrolyzed sucrose more effectively than inulin and was identified as a $\beta$- fructofuranosidase (invertase). The optimum pH and temperature were pH 5.5 and 35$^{\circ}C$, respectively. The enzymehydrolyzed sucrose with a Km of 1.2mM . The soluble $\beta$-fructofuranosidase is likely glycoprotein based on its ability to bind the lectin concanavalin-A. The enzyme was heatlabie, with mose activity being lost at 5$0^{\circ}C$ in 1 hr of incubation. The onion $\beta$-fructofuranosidase was partially inhibited by ZnCl2 HgCl2 and CuSo4.

Stemphylium vesicarium on Garlic and other Allium spp. in Korea (마늘 및 기타 Allium속 식물에 발생하는 Stemphylium vesicarium)

  • More than 100 isolates of Stemphylium obtained form leaf blight lesions of garlic (Allium sativum), onion (A. cepa) and leek (A. fistulosum) were identified as Stemphylium vesicarium (Wallr.) Simm. based on various morphological characteristics rather than S. botryosum Wallr. previously reported on those plants in Korea. In controlled inoculations, the pathogen induced the disease symptoms on leaves of garlic 7 days after inoculation. Onion and leek were also artificially infected by the fungus. This is the first report of S. vesicarium causing a leaf blight of garlic, onion and leek in Korea.

The Effects of Onion(Allium cepa L.) peel Extract on UVB-induced on Skin Damage (양파외피추출물이 UVB에 손상된 피부에 미치는 영향)

  • In this study, we investigated the effects of onion(Allium cepa L.) peel extraction aplication on UVB-induced damage of mouse skin. The male C57BL/6 weeks mice were divided into three groups; the control group(Con), the UVB irradiated group(UVB) and the group treated with onion peel extract after UVB irradiation(UVB+Onion peel). Onion peel extraction were topically treated after UVB irradiation(800 $mJ/cm^2$) to dorsal skin. We were measured TEWL, melanin value, erythema index and histological of mouse skin. In the TEWL, melanin value and erythema index observation, UVB+onion peel group were decreased then in the UVB group and 120 and 168 hr groups were similar to the control group. In the histological observation, UVB+onion peel group were indicated hyperkeratosis then in the UVB. These results showed that onion peel extract as a topical application may have preventive effect against UVB-induced skin damage. Therefore onion peel extract might be good material for UVB-damage skin care.

Ameliorative effect of onion (Allium Cepa L.) flesh and peel on amyloid-β-induced cognitive dysfunction via mitochondrial activation (미토콘드리아 활성화를 통한 양파(Allium Cepa L.) 과육 및 과피의 Amyloid-β 유도성 인지손상에 대한 개선효과)

  • In this study, in order to confirm the ameliorative effects of onion (Allium cepa L.) flesh and peel on amyloidbeta (Aβ)-induced cognitive dysfunction, we evaluated their in vitro neuroprotection and in vivo cognitive functions. As the result of in vitro neuroprotection, the protective effect of the ethyl acetate fraction of onion flesh (EOF) on Aβ-induced cytotoxicity was similar to that of the ethyl acetate fraction of onion peel (EOP). In the behavioral tests, the EOF and EOP effectively improved the Aβ-induced learning and memory impairments. For this reason, it could be concluded that the EOF and EOP improved the antioxidant activities (superoxide dismutase, oxidized glutathione/total glutathione, and malondialdehyde) in brain tissue. In addition, the EOF and EOP effectively activated mitochondrial functions by protecting the mitochondrial membrane potential, ATP, mitochondria-mediated protein (BAX and cytochrome c), and caspase 3/7 activities. The EOF and EOP also improved the cholinergic system (acetylcholinesterase and acetylcholine). Therefore, we suggest that onion could be used for management of Aβ-induced cognitive dysfunction.

The Compositions and Contents of Flavonol Glycosides in Onion (Allium cepa L.) Germplasm (양파(Allium cepa L.) 유전자원의 Flavonol 배당체 조성 및 함량)

  • Flavonols as a major kind of plant secondary metabolites are known for health-promoting compounds in onions (Allium cepa L.). The objectives of this study are to determine profiles of flavonol glycosides in different 75 onion accessions. A total of five flavonols (quercetin 3,4'-diglucoside, Q34'diG; quercetin 3-glucoside, Q3G; quercetin 4'-glucoside, Q4'G; isorhamnetin 4'-glucoside, I4'G; quercetin, Q) were identified from onion accessions. In positive ion mode using LC-ESI-MS, individual flavonols were confirmed from one and two glycosylation binding with aglycone such as quercetin and isorhamnetin. Total flavonol contents were distributed in white onion (range of 0.18-6.47 mg/g DW) and purple onion accessions (range of 2.39-6.47 mg/g), respectively. The mean of flavonol contents in purple onion (4.41 mg/g) showed 1.4-fold higher than white onion (3.23 mg/g). The Q34'diG and Q4'G were considered as the major compounds of flavonol glycosides in onion accessions.

Effects of Onion (Allium cepa) Skin Extract on Pancreatic Lipase and Body Weight-related Parameters

  • The aim of the present study was to assess the effects of onion (Allium cepa) skin extract (OSE) on pancreatic lipase (PL), the key enzyme of the digestion and absorption of dietary fat in the small intestine, and to evaluate its potential for the inhibition of body-weight gain. OSE inhibited PL with an $IC_{50}$ of 53.70 mg/mL, which means as potent as 0.07635% of the activity of orlistat. At 3 and 4 hr after administration of OSE, the plasma triacylglycerol concentration was significantly lower in the OSE-treated rats than control. Body-weight gain and parametrial adipose tissue weights were significantly lower in mice fed the high-fat diet (fat comprises 31% of total calories) with 5%(w/w) OSE than in control. The results suggest that OSE may be an effective nutraceutical for the inhibition of body-weight gain.

Effect of Kalium Fertilizer on Storage Quality of Onion (Allium Cepa L.) (칼리시비량이 양파의 저장성에 미치는 영향)

  • In order to find out the storage quality as influenced by the amount of kalium fertilizer in the Onion (Allium Cepa L.) cultivation , 4 different levels of kalium fertilizer amount, 0, 11, 154 and 462 kg/ha, were applied to cv. "Changnyungdaego" with the following results. As the amount of kalium fertilizer was increased, the content of kalium was increased in the soil and plant, the progress of falldown was somewhat delayed at the level of 462kg/ha kalium fertilizer , but the plant growth and yield were not different , respectively. The rotting ratio was decreased 7.5% by October at the 462kg/ha kalium than the 0 kg/ha treatment . The sprouting and total weight loss ratio were decreased at the levels of 154kg/ha kalium fertilizer.

Application of the Molecular Marker in Linkage Disequilibrium with Ms, a Restorer-of-fertility Locus, for Improvement of Onion Breeding Efficiency

  • To analyze the linkage relationships among molecular markers recently reported to be linked to onion (Allium cepa L.) Ms, a restorer-of-fertility locus, in onion (Allium cepa L.), three single nucleotide polymorphism markers were converted into cleaved amplified polymorphic sequence (CAPS) markers based on onion transcriptome sequences and the rice genome database. Analysis of the recombinants selected from 4,273 segregating plants using CAPS and other linked markers demonstrated the jnurf13 and jnurf610 markers to perfectly co-segregate with the Ms locus. In contrast to jnurf13, the jnurf610 marker was not in perfect linkage disequilibrium with the Ms locus in diverse breeding lines. Thus, the jnurf13 marker and the marker for identification of cytoplasm types were utilized to enhance the efficiency of onion breeding through four applications. First, 89 maintainer lines containing the normal cytoplasm and homozygous recessive Ms genotypes were successfully identified from 100 breeding lines. Second, these two molecular markers were used to analyze the main sources of male-fertile contaminants frequently found in the male-sterile parental lines during F1 hybrid seed production. The majority of the contaminants contained heterozygous Ms genotypes, indicating that pollen grains harboring the dominant Ms genotype may have been introduced during propagation of the maintainer lines. Therefore, the genetic purity of the two maintainer lines was analyzed in the third application, and the results showed that both maintainer lines contained 13-21% off-types. Finally, the two markers were used to increase the seed yield potentials of two open-pollinated varieties containing sterile cytoplasms by removing the plants harboring homozygous recessive and heterozygous Ms genotypes.

Variation in Phenotypic Traits in Onion (Allium cepa L.) Germplasm Collections

  • Variations in phenotypic traits are important for onion genetic improvement. The aim of this study was to identify the phenotypic traits of temporary genetic resources and the best accessions for the development of onion breeding programs. Sixteen phenotypic traits of 79 onion accessions were studied. The descriptive statistics of phenotypic traits exhibited a high variation in onion accessions. Among the 79 evaluated accessions, 64.55% had a large bulb neck width and 44.30% had a circular bulb shape. Principal component analysis showed that six principal components (PCs) accounted for 72.65% of the total variation. The main factors contributing to PC1 were bulb weight, equatorial and bulb polar diameters, plant height, and degree of splitting into bulblets, whereas those contributing to PC2 were the bulb color of the epidermal cells of the fleshy scales and color of the dry skin on the bulb. The accessions were classified into three groups-clusters 1, 2, and 3. Cluster 2 accessions were the most suitable for selecting large and circular bulb-shaped onion cultivars. The phenotypic variation observed in this study may help to select potential accessions for breeding new onion cultivars.
