Studies on the Saponins in the Shoot of Aralia Elata (II) -Identification of the Saponins- (두릅나무 순의 Saponin에 관한 연구 (II) - Saponin 의 동정 -)

  • We determined the structure of main saponin which extracted from the shoot of Aralia Elata. The results were as follows. 1. The main aglycons and suger of the total saponins of Nr2 sample were identified as oleanolic acid and hederagenin, and glucose, arabinose and rhamnose. A probable new aglycon was isolated and inferred as 1, 3-methylenedioxy-3-dehydroxyoleanolic acid. 2. One compound of Fh saponin (named as Elatoside $Fh_2$) which was obtained first in this species was elucidated as 3-O-$({\alpha}-L-arabinopyranosyl(1{\rightarrow}2)-{\beta}-D-gluco-pyranosyl)$-28-O-${\beta}-D-glucophyranosyl$ oleanolic acid on the basis of chemical and spectral evidence of IR, $^1H$, $^{13}C-NMR$ and MS.

Solubilization of Oleanolic Acid and Urolic Acid by Cosolvency

  • Solubilities of oleanolic acid and ursolic acid in aqueous surfactant solutions, liquid polyethylene glycols (PEG), and solvents of various polarity were measured. The results showed that the solutes were slightly or moderately solubilized in the surfactant solutions and the liquid PEGS. It was also revealed that the solutes were slightly soluble in the solvent of either extreme polarity or nonpolarity, but moderately soluble in solvents of intermediate polarity of which solubility parameters are around 10. The solubility parameters of these solutes were calculated from the group contribution to be 10.2 for both of them. Of the solvents tested, tetramethylurea was exceptionally effective in solubilizing the solutes. The solutes were also moderately soluble in the aqueous cosolvents containing tetramethylurea. This suggests that the mixed systems of tetramethylurea could be employed for the solubilization in the formulation of these compounds as an aqueous system.

  • From the Paeoniae Radix, gallic acid methyl ester, (+)-catechin, paconiflorin, albiflorin, paeonol, ${\beta}-sitosterol$ and campesterol, benzoic acid, oleanolic acid, hederagenin and ${\beta}-sitosterol$ 3-O-glucoside were isolated and characterized by means of spectral data. Oleanolic acid and hederagenin have been isolated for the first time from the genus Paeonia.

  • Chemical constituents of fruits, leaves and barks of Acanthopanax senticosus forma inermis were studied. Their fruits have higher contents of crude ash, crude protein, crude fat, fructose and glucose than those of other Acanthopanax species, and contained larger amount of glutamic acid and malic acid among amino acid and organic acid, respectively. The compounds identified from their barks and leaves, were $\beta$-sitosterol and stigmasterol, sesamin, savinin, syringaresinol diglucoside, oleanolic acid, chiisanoside and polyacetytene ($C_9H_{10}O_2$, mp. 62~63).

Isolation of epi-Oleanolic Acid from Korean Mistletoe and Its Apoptosis-Inducing Activity in Tumor Cells

  • A triterpene was isolated as a cytotoxic principle from the dichloromethane extract of Korean mistletoe (KM; Viscum album colora turn) by repeated silica gel chromatography and recrystalli-zation. In in vitro analysis of cytotoxic activity using various human and murine tumor cell lines, the dichloromethane extract of KM was highly cytotoxic against these cells. We isolated the most active compound, referred to VD-3, from the dichloromethane extract of KM. The VD-3 was shown to be less cytotoxic to normal cells (murine splenocytes). From the identification of the chemical structure of VD-3 by spectral data and chemical synthesis, the compound was proven to be epi-oleanolic acid. Tumor cells treated with VD-3 showed a typical pattern of apo-ptotic cell death, such as apparent morphological changes and DNA fragmentation. These results indicate that epi-oleanolic acid is an important compound responsible for antitumor activity of KM.

Identification of Inhibitory Effect on Streptococcus mutans by Oleanolic Acid (Oleanolic acid(OA)의 Streptococcus mutans에 대한 저해효과)

  • Among endogenous oral microflora, Streptococcus mutans plays a critical role in dental plaque formation, which mainly contributes to the development of caries and periodontal disease. Phytochemicals are plant-derived chemical compounds that have been studied as beneficial nutrients to human health. The purpose of this study was to determine the effects of phytochemicals against S. mutans. Among them, oleanolic acid (OA) and 5-(hydroxymethyl)-2-furfural (HF) from Thomson seedless raisins were tested for anti-microbial effects against various clinically important bacteria. OA inhibited the growth of Gram-positive bacteria, but not Gram-negative bacteria. However, HF did not display any antibacterial effect against any of the strains tested. OA also exhibited inhibitory effects in surface adherence and biofilm formation of S. mutans. The results suggest that OA can be utilized as a potential anti-plaque and anti-caries agent by controlling the physiological characteristics of S. mutans on teeth.

Antimicrobial Effect of Oleanolic Acid and Ursolic Acid against Streptococcus downei (Oleanolic acid 및 Ursolic Acid의 Streptococcus downei에 대한 항균작용)

  • Oleanolic acid (OA) and ursolic acid (UA) are triterpenoid compound present in many plants. This study examined the antimicrobial activity of OA and UA against Streptococcus downei. The antimicrobial activity was evaluated by the minimal inhibitory concentration (MIC) and time kill curves. The MIC values of OA and UA for S. downei isolated from the Korean population were $8{\mu}g/ml$. OA and UA had a bactericidal effect on S. downei ATCC $33748^T$ above $2{\times}MIC$, $16{\mu}g/ml$ and $8{\mu}g/ml$, respectively. The results suggest that OA and UA can be used in the development of oral hygiene products for the prevention of dental caries.
