Simple Sequence Repeat Markers Linked to Quantitative Trait Loci Controlling Seed Weight, Protein and Oil Contents in Soybean (콩에서 종실의 무게와 oil 및 단백질 함량을 조절하는 양적 형질 유전자좌와 연관된 simple sequence repeat marker)

  • Soybean [Glycine max (L.) Merr.] is an important crop, accounting for 48% of the world market in oil crops. Improvement of the quality and quantity of soybean seed constituents is one of the most important objectives in soybean breeding. Protein content and seed size are important properties to determine the quality of tofu and soy sprouts respectively. The objective of this study was to identify quantitative trait loci (QTLs) that control seed weight, protein and oil content in soybean. The 117 $F_{2:10}$ recombinant inbred lines (RlL) developed from a cross of 'Keunolkong' and 'Shinpaldalkong' were used. Narrow-sense heritability estimates based on a plot mean on seed weight, protein and oil content were 0.8, 0.78 and 0.71, respectively. Four independent QTLs for seed weight were identified from linkage group (LG) F, I and K. Five QTL for protein content were located on LG D1b, E, H, I and L. Oil content was related with six QTLs located on LG D1b, E, G, I, J and N. Protein and oil content have three common QTLs on LG D1b, E and I. Thus, we identified major loci improving soybean seed quality.

Bioactive Components and Volatile Compounds According to Illite Addition in Saururus chinensis Baill Cultivation (일라이트 시용량에 따른 삼백초 잎의 유효성분 및 향기성분 함량 비교)

  • This study was carried to investigate the effect of illite addition on the growth, bioactive components and volatile compounds of leaf on Saururus chinensis Baill cultivation in greenhouse. Illite addition on Saururus chinensis Baill cultivation resulted no significant effects on the aerial part. However, the root part was highest in 5% illite addition group. Crude oil and ash amount increased as illite additives was increased. Ca of inorganics and free sugars contents were highest in 5% and 20% illite addition, respectively. Hyperoside and isoquercetin of available component on leaf were highest in 5% illite addition group, 4.02mg/g and 4.31mg/g, respectively. The volatile compounds in Saururus chinensis Baill leaf cultured with illite addition amounts were isolated by solid-phase microextraction fiber (polydimethysiloxane $65{\mu}m$) and identifed by gas chromatogtaphy mass spectrometry. As the results, the 22 volatile compounds were identified from in Saururus chinensis Baill leaf and major volatile compounds were the ${\alpha}$-cadinol (18.50%), myristicin (16.46%), methyl-9-methyl-tetradecanoate (10.22%), and ${\gamma}$-muurolene (9.75%). Especially, the content of ${\alpha}$-cadinol was highest in 5% illite addition group and ${\gamma}$-muurolene on overall illite addition group was lower than no addition group.

Improvement of Low Temperature Property of Biodiesel from Palm Oil and Beef Tallow Via Urea Complexation (요소 착물형성에 의한 포화지방산 고함유 팜유 및 우지 유래 바이오디젤의 저온유동성 개선효과 연구)

  • Biodiesel is non-petroleum based fuel produced from vegetable oils or animal fats through transesterification. The compositions of saturated and unsaturated fatty acids in the feedstocks are important factors for biodiesel quality in terms of low-temperature fluidity and oxidative stability. The goal of this study is to improve the cold flow property of biodiesel from vegetable and animal origin containing highly saturated methyl esters (approx. 50%). In this purpose poly-saturated methyl esters in palm and tallow biodiesel were removed via urea-based fractionation and then the recovered fractions (enriched unsaturated fatty acid methyl esters) were supplemented with cold flow improvers. The highest concentration of unsaturated fatty acid methyl esters (93.8%) was obtained using a urea/fatty acid ratio of 3:1 at the crystallization temperature of $0^{\circ}C$ for 17 hours in incubation, with recovery of 71% and the addition of cold flow improver (Flozol$^{(R)}$ 515, 3,000 ppm) to the enriched poly-unsaturated fatty acid methyl esters reduced the CFPP(cold filter plugging point) of palm biodiesel from $12^{\circ}C$ to $-42^{\circ}C$. In tallow biodiesel both the enrichment of unsaturated fatty acid methyl esters (93.71%) and the addition of cold flow improver (Infineum R408, 3,000ppm) reduced the CFPP from $10^{\circ}C$ to $-32^{\circ}C$.

Studies on Yield Increase of Soybean as a Protein Source -Varietal response of yield components, protein and oil yield to different sowing dates and calcium levels in soybean- (단백자원으로서의 대두증산에 관한 연구 - 품종 파종기 및 석회시용량의 차이가 대두의 수량형질과 단백질 및 유분생육량에 미치는 영향 -)

  • To study the varital response of grain, protein and oil yield to different sowing dates and calcium levels in soybean, the most important crop as a protein source in Korea, this experiment was conducted in 1969. Three leading soybean varieties (Choongbukbaek, Ryooku#3 and Changdanbaekmok) were sown at May 22,, June 11 and July 1 under 3 different calcium levels, 0.50 and 100kg per 10a respectively. The results are summarized as follows: 1. Ryonku #3 showed the highest grain yield because of the highest number of pods per hill. In order of protein content, and oil content of grain, Choongbukbaek>Ryooku#3>Changdanbaekmok and Changdanbaekmok>Choogbukbaek>Ryooku#3 were found. Ryooku#3 also showed the highest protein and oil production per unit area owing to the highest grain yield. However. varietal differences of the grain yield the protein and the oil production per unit area varied along the different sowing dates. Interactions between varieties and sowing date on the grain yield should be considered in practice. 2. No difference in the grain yield was found between the first sowing date (May22) and the second (June11) on average. The grain yield in the plot of the third sowing date (July l)was strikingly decreased mainly by the smaller number of pods per hill. The protein and the oil content were not varied significantly by the different sowing dates. The protein and oil production per unit area were lowest in the plot of the third sowing date owing to the decreased grain yield. 3. More calcium application increased the number of branches per hill, the number of pods per hill, and the grain yield. The highest grain yield was found in the plot received 100 kg of calcium hydroxide per 10are. Protein content increased in higher calcium level, but oil content was highest in the plot of noncalcium. More protein production per unit area was gotten by more calcium application because of higher grain yield and protein content. all production increased in higher calcium level by the increased grain yield. 4. High plus (+) correlation was found between stem length and stem weight per hill stem length and grain weight, stem length and grain yield, stem weight per hill and number of pods per hill and grain weight, number of branches per hill and grain weight, number of pods per hill and grain yield, and grain weight and protein content of grain. Minus(-) correlation was recognized between number of pods per hill and grain weight.

Fatty Acid Composition Analysis of Major Oil Crops by One-Step Extraction/Methylation Method (일단계 추출 및 메칠화법에 의한 유료작물의 지방산 분석)

  • Lipid extraction preceding fatty acid methyl esters preparation for gas-liquid chromatography is time-consuming and cumbersome. We performed one-step extraction/methylation method with a mixture of methanol-heptane-benzene-DMP-H$_2$SO$_4$ without prior fat extraction. The simultaneous digestion and lipid transmethylation takes place at 8$0^{\circ}C$ in a single phase. After cooling till room temperature, two phases are formed. The upper one of the phases contains the fatty acid methyl esters ready for GLC. The fatty acid composition of major industrial crops obtained by the one step extraction/methylation method (method 1 and 2) was almost identical with the fatty acid composition of the pure fats extracted with hexane by the Soxtec instrument (method 3). Due to its simplicity, speed, and reduced organic solvent the one-step extraction/methylation method (method 1 and 2) should be useful to determine overall fatty acid composition, especially in situations where many samples have to be analyzed.

Characterization of Lipophilic Nutraceutical Compounds in Seeds and Leaves of Perilla frutescens

  • Perilla frutescens, which comprises var. frutescens and var. crispa, has been cultivated traditionally in Asian countries as an edible oil, leaf vegetable, and medicinal crop. To evaluate the lipophilic phytonutrient properties of P. frutescens, we selected 54 Perilla accessions [19 landraces of var. frutescens (FL), 22 weedy type var. frutescens (FW), 9 weedy type var. crispa (CW), 2 cultivars of var. frutescens widely cultivated for seed oil (FCS), and 2 cultivars of var. frutescens cultivated as a leaf vegetable (FCL)] and analyzed their seeds and leaves for vitamin E, squalene, and phytosterols. Among the four vitamin E isomers analyzed, ${\gamma}$-tocopherol was the major form of vitamin E in seeds, whereas ${\alpha}$-tocopherol was the major form in leaves of all types of P. frutescens. The highest total vitamin E content in seeds was present in FL ($170.0mg{\cdot}kg^{-1}$), whereas that in leaves was highest in FCL ($358.1mg{\cdot}kg^{-1}$). The highest levels of squalene in seeds and leaves were in FL ($65.5mg{\cdot}kg^{-1}$) and CW ($719.3mg{\cdot}kg^{-1}$), respectively. Among the three phytosterols, ${\beta}$-sitosterol occurred in the highest amount in both leaves and seeds of all of the crop types. Phytonutrient contents were comparatively higher in leaves than in seeds of all crop types. All of these results suggest that the consumption of leaves and seeds of Perilla crops could be beneficial to human health, as Perilla possesses considerable amounts of various lipophilic compounds.

Determination of Sesamin and Sesamolin in Sesame (Sesamum indicum L.) Seeds Using UV Spectrophotometer and HPLC

  • Sesamin and sesamolin, antioxidant lipidsoluble lignan compounds, are abundant in sesame (Sesamum indicum L.) seed oil and provide oxidative stability of oil related to sesame quality. The sesamin and sesamolin contents of 403 sesame land races of Korea were determined by HPLC analysis of methanol extract (HPLC value), and their total lignan content was compared with those by using UV-Vis spectrophotometric analysis (UV method) of methanol (UV-MeOH value) and hexane (UV-Hexane value) extracts. HPLC values of total lignan content were strongly associated with UV-Hexane (r=0.705**) and UV-MeOH (r=0.811**) values. The UV values from both the extracts were 3.8-4.7 times higher than those of HPLC values. Lignan content was overestimated by UV method because total compounds in the mixture solution were quantified by absorbing at the same ultraviolet wavelength as in HPLC method. UV method could more rapidly analyze small amount of sample with higher sensitivity of detection than HPLC method. Average contents of lignans in sesame germplasm evaluated in this study were $2.09{\pm}1.02mg/g$ of sesamin, and $1.65{\pm}0.61mg/g$ of sesamolin, respectively, showing significant variation for lignan components. The results showed that UV method for the determination of sesamin and sesamolin could be practically used as a faster and easier method than HPLC by using the regression equations developed in this study.

A New Perilla Cultivar for Edible Seed 'Dayu' with High Oil Content (기름함량이 높은 종실용 들깨 신품종 '다유')

  • 'Dayu', a perilla (Perilla frutescens (L.) Britton) cultivar for edible seed was developed by Department of Functional Crop, NICS, RDA in 2004. It was developed from a cross between a pedigree of Daeyeop as a female and YCPL1846 as a male parent in 1994. 'Dayu' could be characterized by white flower color and brown grain color. Dayu shows 127 cm in plant height, and has lodging tolerance. Maturing date of 'Dayu' was October 6, which was similar with that of 'Saeyeopsil'. This new cultivar has high oil content with 48.5% and high linolenic acid in the fatty acid composition. The grain yield potential of 'Dayu' is about 1.44 MT/ha in the regional yield trial.

Studies on the Cultivation Technique and Evaluation of Forage Resources for a New Oil Crop Chufa(Cyperus esculentus L.) (새로운 유지작물 기름골의 재배기술과 사료가치)

  • A new oil crop chufa(Cyperus esculentus var. sativus) was studied to evaluate the yield potential of tuber and productivity of leaf as a new forage crop resources. Response to nitrogen fertilizer was not clear, showing low yield over 10kg/l0a of nitrogen application. Tuber yield was higher at 50cm $\times$ 15cm and 50cm$\times$20cm of planting population of tuber. Planting at early May showed high tuber yield, and sprouting tuber planting could not increased the tuber yield. The plant height of Chufa is 126cm, and the number of tiller per $\m^2$ are 671. It is moderate to lodging, but susceptable according to the amount of field products. Fresh forage and dry matter yield of above ground of chufa were 2,135kg/10a, and 590kg/l0a, respectively. The contents of crude protein, ether extract, fiber, ash, and acid detergent fiber are 24.8%, 8.1%, 38%, 64.4%, 52.8%, and 55.9%, respectively. It was considered that the nutritive value of chufa leaf in maturing stage was so good as rice straw.

Evaluating Soil Carbon Changes in Paddy Field based on Different Fraction of Soil Organic Matter

  • Organic matter plays important roles in soil ecosystem in terms of carbon and nitrogen cycles. Due to recent concerns on climate change, carbon sequestration in agricultural land has become one of the most interesting and debating issues. It is necessary to understand behavior of soil carbon for evaluating decomposition or sequestration of organic matter and analyzing potential carbon decomposition pattern about the kinds of organic matter sources to cope with well. In order to evaluate decomposition of soil carbon according to organic material during cultivating rice in paddy field, we treated organic material such as hairy vetch, rice straw, oil cake fertilizer, and manure compost at $50{\times}50{\times}20cm$ blocks made of wood board, and analyzed carbon contents of fulvic acid and humic acid fraction, and total carbon periodically in 2013 and 2014. Soil sampling was conducted on monthly basis. Four Kinds of organic matter were mixed with soil in treatment plots on 2 weeks before transplanting of rice. The treatment of animal compost showed the highest changes of total carbon, which showed $7.9gkg^{-1}$ in May 2013 to $11.6gkg^{-1}$ in October 2014. Fulvic acid fraction which is considered to easily decompose ranged from 1 to $2gkg^{-1}$. Humic acid fraction was changed between 1 to $3gkg^{-1}$ in all treatments until organic material had been applied in 2014. From May to August in the second year, the contents of humic acid fraction increased to about $4gkg^{-1}$. The average of humic fraction carbon at treatments of animal compost was recorded highest among treatments during two years, $2.1gkg^{-1}$. The treatment of animal compost has showed the lowest ratio of fulvic acid fraction, humic acid fraction compared with other treatments. The average ratio of fulvic fraction carbon in soil ranged from 16 to 20%, and humic fraction carbon ranged from 19 to 22%. In conclusion, animal compost including wood as bulking agent is superior in sequestrating carbon at agricultural land to other kinds of raw plant residue.
