Sargassum coreanum extract alleviates hyperglycemia and improves insulin resistance in db/db diabetic mice

  • BACKGROUND/OBJECTIVES: The goal of this study was to examine the effect of Sargassum coreanum extract (SCE) on blood glucose concentration and insulin resistance in C57BL-KsJ-db/db mice. MATERIALS/METHODS: For 6 weeks, male C57BL/KsJ-db/db mice were administrated SCE (0.5%, w/w), and rosiglitazone (0.005%, w/w). RESULTS: A supplement of the SCE for 6 weeks induced a significant reduction in blood glucose and glycosylated hemoglobin concentrations, and it improved hyperinsulinemia compared to the diabetic control db/db mice. The glucokinase activity in the hepatic glucose metabolism increased in the SCE-supplemented db/db mice, while phosphoenolpyruvate carboxykinase and glucose-6-phosphatase activities in the SCE-supplemented db/db mice were significantly lower than those in the diabetic control db/db mice. The homeostatic index of insulin resistance was lower in the SCE-supplemented db/db mice than in the diabetic control db/db mice. CONCLUSIONS: These results suggest that a supplement of the SCE lowers the blood glucose concentration by altering the hepatic glucose metabolic enzyme activities and improves insulin resistance.

Effects of acute reducing salt supplementation on cardio-respiratory function, blood pressure and serum nitric oxide production in elite players

  • The purpose of this study was to evaluate changes of body composition, cardio-respiratory function in ventilation threshold (VT) and maximal state exercise, systolic (SBP) and diastolic blood pressure (DBP) and serum nitric oxide (NO) production during acute reducing salt (RS) supplementation in college elite athletes. Variables of cardio-respiratory function during rest, ventilation threshold and maximal exercise was not shown a significantly difference between RS supplementation and non-supplementation, there was shown a significant increase in ventilation threshold time (p<0.05) and exhaustion time (p<0.05) during RS supplement compared to non-supplement. SBP and DBP were not shown a significant difference between RS supplement and non-supplement. This result suggests that acute intake of RS is not increased a blood pressure. Serum NO production was not significant difference in the RS supplement group, but it was shown a significantly increased levels (p<0.01, vs. recovery 30 min.) immediately after maximal exercise in the non-supplement group. This result suggests that acute intake of RS have important role in inhibition of serum NO production during maximal exercise. Conclusively, This study suggest that acute intake of RS was not influence in body composition variables, but it was positive effect in ventilation threshold time, exhaustion time, maintenance of blood pressure and inhibition of serum NO production in maximal treadmill exercise.

A Study on Vitamin C Content of Nutrition Emphasized Products (영양강조표시제품 중 비타민 C 함량 조사)

  • Objectives: Vitamin C has various functions such as antioxidative effect and supporting absorption of iron (Fe). Aim of this present study was to provide vitamin C nutrition information and to briefly evaluate absorption interaction of vitamin C and Fe content of vitamin C emphasized products. Methods: Vitamin C emphasized foods including beverages, cereal, snacks, chocolate products, other cocoa products, and sugary products were examined by HPLC. Fe contents in samples after dry-ashing were examined by ICP. Results: Vitamin C content ranges in various products tested were the following: beverages (n=11) $20.15{\pm}0.08{\sim}845.41{\pm}6.07mg$, cereal (n=11) $52.50{\pm}0.23{\sim}262.50{\pm}0.07mg$, snacks (n=1) $50.00{\pm}0.25mg$, chocolate products (n=1) $311.73{\pm}2.44mg$, other cocoa products (n=1) $311.73{\pm}2.44mg$, other sugary products (n=2) $52.50{\pm}0.23{\sim}262.50{\pm}0.07mg$. Vitamin C (n=27) analysis values ranged from 82 to 450% of the labeled value. Vitamin C content in vitamin C emphasized food (n=6) was estimated 7.7 times~56.6 times more than Fe content. Conclusions: Analyzed samples ranged more than 80% of the labeled value in vitamin C emphasized products, which complied with food labeling regulation. But, beverages (n=3), cereal (n=4), chocolate products (n=1) were 2 times more than the labeled value. To provide accurate nutrition information, food manufactures should supervise nutrition labeling and understand the interactions between nutrients. Also, consumer should decide about the adequate amount of nutrient intake by thoroughly checking nutrition labeling.

A Study on Fat Content in Commercial Retort Foods - Crude Fat, Saturated Fatty Acid and Trans Fatty Acid - (시판 레토르트식품의 지방함량 조사 - 조지방, 포화지방, 트랜스지방산 중심으로 -)

  • The aim of this study was to provide nutrition information to consumers by analyzing crude fat, saturated and trans fatty acids in commercial retort foods (n=70). The following sauce products of curries (n=21) and black-bean-sauces (n=16), other sauces (n=17) and instant cooking foods (n=16) were collected. Crude fat contents were quantified with the Rose-Gottlieb method using acid digestion. While saturated and trans fatty acids were examined by gas chromatography with a flame ionization detector (FID). Crude fat, saturated and trans fatty acid content ranges were $0.47{\pm}0.42{\sim}12.80{\pm}0.07g/100g$, $0.24{\pm}0.02{\sim}17.41{\pm}0.41g/100g$, $0.00{\pm}0.00{\sim}0.46{\pm}0.05g/100g$, respectively. Maximum recovery of analysis values was crude fat (119.7%), saturated fat (119%) and trans fatty acid (90%) compared the actual amounts based on the reference value indicated on the nutrition label. The analyzed samples were found to be compliant with nutrition label standard, because the contents of crude fat, saturated fatty acid, trans fatty acid were less than 120% of the reference value indicated on the nutrition label in retort foods. Therefore, the nutrition information on retort foods available to consumers was found to be trustworthy.

  • In this study we examined effects of a new dietary supplement on the lipid level and body weight. The efficacy of this weight-reduction supplement, based on natural ingredients consisting of Phaseolus vulgaris, Garcinia cambogia, and microstalline cellulose, was investigated by randomized, placebo-controlled double-blind study. The 36 subjects were assigned evenly into weight-reduction supplemented (Active) and placebo groups (Placebo). The supplement could reduce the absorption of different types of sugar from the gastro-intestinal tract. A significant difference in weight reduction was shown in the active group (3.5 kg). Body composition measurements indicated that about 34% of fat loss in the active group could be achieved. These findings suggest that this dietary supplement could reduce body weight and fat gains, and its inhibitory effects might lead to obesity improvement.

Evidence of Memory Improvement by Phosphatidylcholine Supplement at Fetus and Neonate -Studies of Basal Forebrain Cholinerge Neuronal Activities- (태생기 및 신생기의 Phosphatidylcholine 보충기 기억력 향상에 미치는 영향 -전뇌기저부의 Choline성 신경세포 활성에 관한 연구-)

  • To investigate the effect of dietary phosphatidylcholine(PPC) supplement on memory improvement, biochemical study on the brain, and morphometric studies on the cholinergic neurons in the rat basal forebrain were undertaken. The pregnancy rats were divided into the normal control, the choline deficient and the PPC supplemental groups according to quantity of the PPC in diet. According to choline deficiency and PPC supplement after birth, the neonate rate of the normal control group were subdivided into the control diet(N-N) and the PPC supplied (N-S) groups, the choline deficient group were subdivided into the continually deficient (D-D), the control diet(D-N) and the PPC supplied groups(D-S), and the PPC supplemental group were subdivided into the control diet (S-N)and the continually supplied (S-S)group. The PPC supplemented diet was added 2% egg PPC in AIN 76 formula diet. PPC concentrations and cholinesterase(CE) activities were measured in the serum, the liver and the brain, respectively. Immunohistochemical stains for choline acetyltransferase(ChAT) was employed for the morphological and morphometric studies. The maze test was undertaken to evaluate memory improvement. PPC concentration and CE activities in the serum, liver and the brain were high in the PPC supplemental groups and low in the choline deficient groups. ChAT immunoreactivity neurons at the medial septal diagonal bond complex and the basal forebrain nucleus of Meynert were reduced in the choline deficient groups. Average failure rate for the maze test was the lowest in the S-S group and the highest in the D-D group. Insufficient choline suppley during the neuronal development would result in cholinergic neuronal damage, which could be prevented by adequate PPC supplement. It is consequently suggested that PPC supplement may be effective on memory improvement by maintaining the cholinergic neuronal activity in the basal forebrain of the rats.

The Effect of Supplementation of Persimmon Leaf Extract on Lipid and Antioxidant Metabolism in Rats Fed a High-cholesterol Diet

  • Fruits and vegetables reportedly have a protective effect against hyperlipidemia and oxidative disease. Accordingly, this study aimed to investigate the lipid-lowering effect and antioxidative capacity of persimmon leaf extract (PLE) in rats fed a high-cholesterol diet. Male rats were fed a high-cholesterol (1% wt/wt) or high-cholesterol diet supplemented with Lovastatin (0.02% wt/wt) or PLE (0.2% wt/wt) for 5 weeks. The concentration of plasma total cholesterol was significantly lower in the PLE group than in the lovastatin group. However, the concentration of plasma HDL-cholesterol and the ratio of HDL-cholesterol/total-cholesterol (%) were significantly higher in the PLE group than in the control group. The PLE supplement also significantly lowered the contents of hepatic cholesterol and triglyceride. In comparing fecal sterol contents, the PLE group saw a significant increase of both neutral and acidic sterol compared to the other groups. The PLE supplement significantly lowered plasma GOT and GPT activity, which ave indices of hepatic toxicity. Plasma TBARS concentration was significantly lower in the PLE group than in the control group, while hepatic TBARS level was not significantly different between the groups. In a comparison of hepatic antioxidant parameters, SOD, catalase and GSH-Px activity were significantly higher in the PLE group than in the control group. However, the PLE supplement significantly towered antioxidant enzyme activity in the erythrocyte. Furthermore, these results suggest that supplementation of PLE promoted the excretion of fecal sterols, thereby leading to decreased absorption of dietary cholesterol. In addition, PLE may play an important role in regulating antioxidative capacities by altering SOD and ChT activity.

Climacteric and Menopausal Women's Beliefs on Daily Meals and Food Supplements - A Focus Group Interview Study - (경남지역 일부 갱년기.폐경기 여성의 식사와 건강보조식품에 관한 신념 - 포커스그룹 심층면접 연구 -)

  • The aim of the study was to explore the current status as well as personal views, attitudes, and beliefs regarding daily meal consumption (DM) and food supplement use (FS) in conjunction with the improvement of health condition of the women. Eight focus group interviews were performed and the interview material was condensed systematically with the aim to extract core meanings related to DM, FS, and menopause-associated health. Participants were 40 in number and showed ages ranging from 45 and 60 years with various menopausal status. Current status and beliefs about DM, resources of purchase motivation of FS, types of FS that are currently used, and perceived effects and personal beliefs about FS are discussed. Theme content analysis revealed 3 themes for beliefs about DM, 5 themes for beliefs pertaining FS, and 4 themes for the association between DM and FS. Non-dietary factors such as positive mental attitude and exercise appeared to be also important to maintain good health. The bottom line message from this study may be that proper nutrition through daily meals is essential for good health, while food supplement are used merely to supplement the diet. Findings from this study may deepen our understanding of how women who translate their lifespan through "menopause" perceive the roles and meaning of DM and FS, suggesting health professionals need to monitor and evaluate DM and implement strategies targeting the improvement of daily meal quality of middle and older aged women.

Quercetin Supplement is Beneficial for Altering Lipid Metabolism and Antioxidant Enzyme Activities in the Middle of Ethanol Feeding in Rats

  • The current study examined the effect of quercetin supplements on the lipid-lowering and antioxidant metabolism in ethanol-fed rats. The control $group(E_8)$ received ethanol only diet for 8 wks, whereas the other group$(E_8Q_4)$ received a diet including quercetin supplementation(0.05% wt/wt) for 4 wks while on the ethanol diet for 8wks. The hepatic alcohol dehydrogenase activity was significantly higher in the $E_8Q_4$ group than in the $E_8$ group. Supplementation with quercetin significantly elevated the HDL- cholesterol concentration, the HDL-C/total-C ratio, and lowered the atherogenic index(AI) compared to the $E_8$ group. The hepatic triglyceride and cholesterol contents were significantly lowered by the quercetin supplement compared to those of the control group. The hepatic HMG-CoA reductase and ACAT activities of the $E_8Q_4$ group were significantly lower than those of the $E_8$ group. The overall potential for antioxidant defense was significantly enhanced by the quercetin supplement, as indicated by a decrease in plasma and hepatic TBARS levels. The hepatic GSH-Px and G6PD activities were significantly higher in the $E_8Q_4$ group compared to the $E_8$ group. The current results suggest that dietary quercetin leads to the inhibition of HMG-CoA reductase and ACAT, which in turn lowers cholesterol levels and normalizes antioxidant enzyme activities.
