Appropriate nitrogen application enhances saponin synthesis and growth mediated by optimizing root nutrient uptake ability

  • Background: Cultivation of medicinal crops, which synthesize hundreds of substances for curative functions, was focused on the synthesis of secondary metabolites rather than biomass accumulation. Nutrition is an important restrict factor for plant growth and secondary metabolites, but little attention has been given to the plasticity of nutrient uptake and secondary metabolites synthesis response to soil nitrogen (N) change. Methods: Two year-field experiments of Sanqi (Panax notoginseng), which can synthesize a high level of saponin in cells, were conducted to study the effects of N application on the temporal dynamics of biomass, nutrient absorption, root architecture and the relationships between these parameters and saponin synthesis. Results: Increasing N fertilizer rates could improve the dry matter yields and nutrient absorption ability through increasing the maximum daily growth (or nutrient uptake) rate. Under suitable N level (225 kg/ha N), Sanqi restricted the root length and surface and enhanced the root diameter and N uptake rate per root length (NURI) to promote nutrient absorption, but the opposite status of Sanqi root architecture and NURI was found when soil N was deficient. Furthermore, increasing N rates could promote the accumulation of saponin in roots through improving the NURI, which showed a significant positive relationship with the content of saponin in the taproots. Conclusion: Appropriate N fertilizer rates could optimize both root architecture and nutrient uptake efficiency, then promote both the accumulation of dry matter and the synthesis of saponins.

  • To remove nitrogen compound from wastewater six kinds of bacillus were isolated from sludge. Each bacillus was identified as B. subtillis $I{cdot}II$, B. cereus $I{cdot}II$, B. anthracis, B. circulans. The test of effect of nutrient and cofector on the nitrogen removal showed that peptone, yeast extract, magnesium, iron, and calsium accerated the efficiency of nitrogen removal. In syringe test aerobic nitrification and denitrification was occured.

Nutrient variations from swine manure to agricultural land

  • Objective: Swine manure in Korea is separated into solid and liquid phases which are composted separately and then applied on land. The nutrient accumulation in soil has been a big issue in Korea but the basic investigation about nutrient input on arable land has not been achieved in detail. Within the nutrient production from livestock at the national level, most values are calculated by multiplication of the number of animals with the excreta unit per animal. However, the actual amount of nutrients from swine manure may be totally different with the nutrients applied to soil since livestock breeding systems are not the same with each country. Methods: This study investigated 15 farms producing solid compost and 14 farms producing liquid compost. Composting for solid phase used the Turning+Aeration (TA) or Turning (T) only methods, while liquid phase aeration composting was achieved by continuous (CA), intermittent (IA), or no aeration (NA). Three scenarios were constructed for investigating solid compost: i) farm investigation, ii) reference study, and iii) theoretical P changes (${\Delta}P=0$), whereas an experiment for water evaporation was conducted for analyzing liquid compost. Results: In farm investigation, weight loss rates of 62% and 63% were obtained for TA and T, respectively, while evaporation rates for liquid compost were 8.75, 7.27, and $5.14L/m^2{\cdot}d$ for CA, IA, and NA, respectively. Farm investigation provided with the combined nutrient load (solid+liquid) of VS, N, and P of 117.6, 7.2, and $2.7kg/head{\cdot}yr$. Nutrient load calculated from farm investigation is about two times higher than the calculated with reference documents. Conclusion: The nutrient loading coefficients from one swine (solid+liquid) were (volatile solids, 0.79; nitrogen, 0.53; phosphorus, 0.71) with nutrient loss of 21%, 47%, and 29%, respectively. The nutrient count from livestock manure using the excretion unit has probably been overestimated without consideration of the nutrient loss.

호흡률법에 의한 하수의 질산화성 질소화합물 추정 (Estimation of Nitrifiable Nitrogen Compounds in Municipal Wastewater by Respirometry)

  • Nitrogen compounds in municipal wastewater can be divided into biodegradable and nonbiodegradable fractions with biodegradability. Biodegradable nitrogen compounds can be removed through biological nitrification and denitrification processes, and nonbiodegradable nitrogen compounds affect the effluent quality of biological nutrient removal processes. The amount of nitrifiable nitrogen compounds, which are the sum of ammonia and biodegradable organic nitrogen, has been estimated by respirometry. Respirometry shows good estimation of the concentration of nitrifiable nitrogen when a synthetic sample of ammonium chloride is dosed. The estimated concentration of nitrifiable nitrogen compounds in municipal wastewater is close to ammonia concentration in municipal wastewater, but it is lower than that for the synthetic sample. If nitrogen assimilated into cell synthesis of nitrifiers and heterotrophs is considered, the total amounts of nitrifiable nitrogen compounds, which are nitrified and assimilated, could be more accurately estimated. The concentration of nitrifiable nitrogen compounds, which are biodegradable, is about 31 mg N/l, and this is 119% of ammonia and 94% of total nitrogen. Ammonia, nitrate, biodegradable organic nitrogen, and nonbiodegradable nitrogen are about 79%, 1%, 15%, and 5% of the total nitrogen in municipal wastewater, respectively.

Residual frying oil in the diets of sheep: intake, digestibility, nitrogen balance and ruminal parameters

  • Objective: The objective of this study was to evaluate the intake and nutrient digestibility, nitrogen balance and ruminal ammonia nitrogen in lambs of diets containing different levels of residual frying oil. Methods: Levels of 0, 20, 40, 60, and 80 g/kg dry matter (DM) base of residual frying oil in the diets of lambs were evaluated. Five castrated lambs with initial body weights of $36.8{\pm}3.3kg$, distributed in a Latin square ($5{\times}5$) design, were used. Results: There was a decreasing linear effect on the intake of DM, organic matter (OM), crude protein (CP), neutral detergent fiber (NDF), total carbohydrates (TCH), and nonfibrous carbohydrates (NFC). There was an increased linear intake of ether extract (EE). The apparent digestibility of DM, OM, CP, NDF, TCH, and NFC, as well as urine nitrogen excretion, nitrogen balance and ruminal parameters, were not influenced by different levels of residual frying oil in the diet. EE digestibility presented a crescent linear effect. Conclusion: It can be concluded that the addition of residual frying oil to the diets of sheep can affect nutrient intake without affecting the digestibility of most nutrients (with the exception of EE), nitrogen balance and ruminal ammonia nitrogen concentration.

Introduction, Development, and Characterization of Supernodulating Soybean Mutant -Nitrate Inhibition of Nodulation and Nitrogen Fixation in Supernodulating Soybean Mutant-

  • Inhibition of nodule formation and nitrogen fixation by soil nitrogen, primarily nitrate, is well known in legume plants. The present study was undertaken to evaluate the effect of ${NO_3}^-$ on the nodulation, nitrogenase activity, and growth of supernodulating soybean mutant and its wild type. A greenhouse study was conducted to compare two of supernodulating mutants, 'SS2-2' and 'nts 382', with the normal nodulating cultivar 'Sinpaldalkong 2' when grown in a 1-l styroform cup filled with sand, and fertilized with five levels of ${NO_3}^-$ (0, 2, 4, 8, and 12 mM). During the growth period, each plant was supplied two or three times a week with 50 mL of nutrient solution. Supernodulating soybean mutants, SS2-2 and nts 382, showed more nodules and nodule mass, and greater $C_2\;H_2$ activity than the wild type, Sinpaldalkong 2, regardless of the level of exogeneous nitrogen supply. On the other hand, total dry weight of SS2-2 mutant, which was smaller than Sinpaldalkong 2, did not respond to the various ${NO_3}^-$-N levels. This suggested that supernodulating SS2-2 mutant could maintain fairly high total dry weight at the low ${NO_3}^-$-N level, even in the absence of exogeneous ${NO_3}^-$-N in the nutrient solution. From the reduced top growth and high nitrogen fixing ability of supernodulating mutants, it was surmised that supernodulating mutant could potentially protect agricultural environments from pollution through the reduction in nitrogen fertilization as well as maintain fairly high yield with increasing planting density.

  • Production of biodiesel from microalgae is dependent on the microalgal lipid content and free fatty acid composition. Both lipid and free fatty acid are regulated by nutrient sources. In this study, newly developed mutant Chlamydomonas reinhardtii with higher lipid content was investigated for the effect of nutrient limitation. Nitrogen $NO_3{^{-}}$ and phosphate $PO_4{^{3-}}$ were limited for nutrient starvation during the cultivation. Under nutrient starvation, total lipid content level was increased to 27~33% and C16:0 fatty acid content constituted over 31~43% of total fatty acid. Interestingly, we also found that the expression of fatty acid desaturase (FAD7) was decreased when nutrients were starved.

Elution Behavior of Nutrient Salts from Sediment and its Impact on Water Bodies

  • This paper describes the influence of nutrient salts eluted from the bottom of a closed water area where polluted sediment has been deposited by inflowing river water. The elution pattern was monitored at our experimental facility. Both the sediment pore water and water above the bottom were sampled using a dialyzer sampler (peeper). The pore water of the eutrophicated sediment contained a large amount of nutrient salts, and the effect of elution was confined to a limited area of the bottom surface. The nutrient concentration of the sediment pore water was closely related to both the water temperature and dissolved oxygen (DO) concentration. The eluted nutrients from the sediment provided a source for phytoplankton and algae growth. This experimental data indicated that the water quality of the surface was not directly connected to the eluted nutrient salts, while it was indirectly affected by the total ecosystem, including all the organisms within an area and their environment.

간헐폭기 활성슬러지공정에서 C/N비와 질소제거의 관계 (Relationship Between C/N Ratio and Nitrogen Removal in Intermittently Aerated Activated Sludge System)

  • In this research, Effect of C/N ratio on nutrient removal in intermittently aerated activated sludge system(IAASS) was investigated with dormitary, building and swine wastewater. Three types (2-stage, 4-stage, modified) of IAASS were operated. Time interval of aeration/nonaeration in IAASS was 1hr/1hr. In treatment of Dormitary wastewater(BOD/T-N ratio : 4.4), Building wastewater (BOD/T-N ratio : 3.14) and swine wastewater(BOD/T-N ratio : 3.84), Nitrogen removal efficiency of 80, 70 and 90.4% was achieved, respectively. Nitrogen removal in IAASS was a great influenced on influent C/N ratio, efficient nitrogen removal was achieved at BOD/T-N ratio over 4. In IAASS operation, $\Delta $BOD mg/L/$\Delta $ nitrogen mg/L ratio was about 4-6. Simultaneous removal of organic, nitrogen and phosphorus in IAASS can achieved. And influent organic was efficiently utilized in denitrification. IAASS could be one of the best alternative process for the retrofit of conventional activated sludge system for the removal of nutrients.

섬진강 하구역에서 영양염의 하구내 거동과 플럭스 (Estuarine Behavior and Flux of Nutrients in the Seomjin River Estuary)

  • In order to estimate the nutrient flux of the Seomjin River into the coastal waters of South Sea, and to understand the estuarine reactions during mixing between river water and seawater, we collected surface water along the salinity gradient in the Seomjin River estuary from Mar. 1999 to Apr. 2001. We found that nitrate and silicate were delivered by fluvial input, while phosphate was, supplied from disposed wastes in the Gwangyang Bay. Mean annual flux of dissolved inorganic nitrogen (DIN), phosphate and silicate into the Gwangyang Bay was estimated 10.9 molesㆍsec$^{-1}$(4,820 tonnesㆍyr$^{-1}$), 0.07 molesㆍsec$^{-1}$(68 tonnesㆍyr$^{-1}$), 13.3 molesㆍsec$^{-1}$(11,747 tonnesㆍy$^{-1}$), respectively. An evident removal of phosphate, silicate and ammonium at the mid-salinity zone during the dry season was attributed to the active uptake of phytoplankton, and consequently nutrient flux into the Gwangyang Bay was low. Whereas, during the flood season in summer, conservative or additional distribution of the nutrients was observed in the estuary. As a rsult nutrient flux into the Gwangyang Bay was maintained high. High concentrations of chlorophyll a and the active removal of nutrient during the dry season at the mid-salinity zone suggest that nutrient distribution in the Seomjin River estuary was mainly controlled by biological processes and nutrient fluxes into the Gwangyang Bay might be significantly modified of by the primary production.
