Determination of Silybin of Nutraceutical Herbal Preparations Using HPLC-PDA

  • Silymarin is an antihepatotoxic substance isolated from the fruits of silybum mariamum. Possibly due to their antioxidant and membrane stabilizing properties, the compounds was shown to protect various organs and cells against a number of insults (Kvasnicka et al., 2003). Among the main silymarin components, [silybin($SB_A,\;SB_B$, isosilybin ($ISB_A,\;ISB_B$) silydianin (SD) and silychristin (SC)], silybin is the major pharmacologically active compound. Korean Pharmaceutical Codex (2nd ed.) describes silybin as the main substance of Cardus Marianus extract as supportive treatment of chronic inflammatory liver disorders. The aim of this work was to analyze silybin from various preparations containing cardus marianus extract, nicotinamide, and riboflavin (CNR). Nine commercial products were tested using reversed-phase HPLC-PDA assay. The limits of detection and quantification were $0.2\;{mu}g/ml$ and $1\;{mu}g/ml$, respectively. Calibration curve showed a good linearity ($r^2$=1.00000) in the range of $1{\sim}500\;{\mu}g/ml$ of silybin standard solutions.

Safety Evaluation of Ethanol Extract from Unripe Fruit of Bitter Melon (Momordica Charantia L.) in Sprague-Dawley Rats (랫드를 이용한 여주 추출물의 안전성 평가)

  • This study was performed to evaluate repeated dose oral toxicity upon administration of the test substance 1,2-benzisothiazolin-3-one for 90 days and to determine NOAEL (no observed adverse effect level) and target organs in Sprague-Dawley rats. Single, 2-week repeated, and 13-week repeated oral dose toxicity studies were conducted in Sprague-Dawley rats. The dose levels of groups were 1,250, 2,500, and 5,000 mg/kg/d. All dose groups were compared with the vehicle control group. The animals were observed for clinical signs and weekly body weight. Urinalysis, hematology, and serum biochemistry analyses were conducted. Subsequently, animals were sacrificed and subjected to histopathological examination. For the result, NOAEL of ethanol extract from unripe fruit of bitter melon had an optimal dose of 5,000 mg/kg/d and acceptable daily intake up to 3,000 mg/man. There was no target organ detected. Therefore, bitter melon, which contains a variety of bioactive substances, could be widely used as a health functional food ingredient.

Comparison of the Antioxidant Activities and Nitrite Scavenging Activity of Black Chokeberry (Aronia melanocarpa) Extracts (블랙 초크베리 추출물의 항산화 활성 및 아질산염 소거 활성)

  • The purpose of this study was to measure the antioxidant activity and bioactivity of black chokeberry (Aronia melanocarpa). The black chokeberry was known to contain many physiologically active substance, such as ascorbic acid, anthocyanin, quercetin. We extracted black chokeberry powder from four solvents such as chloroform:methanol(CM, 2:1, v/v), distilled water (DW), 70% ethanol, 70% methanol. After that, we determined anthocyanin, total phenol, flavonoid content, DPPH radical scavenging activity, ABTS radical scavenging activity, ferric reducing antioxidant power, reducing power and Nitrite scavenging activity of black chokeberry extracts and as results of comparing each extract. respectively. From the above results shows that antioxidant activity and bioactivity of black chokeberry extracts was the highest in 70% methanol (p<0.05). The results suggest that black chokeberry can be used as nutraceutical foods and natural antioxidant.

Neuroprotective Effects and Physicochemical Characteristics of Milk Fortified with Fibroin BF-7 (BF-7 강화 우유의 뇌기능보호 효과 및 물리화학적 특성)

  • The impact of storage on the neuroprotective effects against $A\beta$-induced cell death and physicochemical characteristics of milk fortified with BF-7 were investigated. The BF-7 milk exerted protection of neuronal cells SK-N-SH from amyloid beta ($A\beta$)-induced neuronal stress. Our results showed that incubation of the cell with pretreated BF-7 milk, significantly attenuated apoptotic stress by $A\beta$, considered in cell morphology and nucleus shape. The general compositions were maintained consistently in BF-7 fortified milk (BF-7 milk). The BF-7 did not make any disturbance on pH and titratable acidity. The color change was not detected, either. Also, any microorganism had not been detected with more than 7 days storage at $4^{\circ}C$. In sensory evaluation study. the average scores of each sensory attribute were quite similar with plain milk. In conclusion, our results strongly indicate that BF-7 characteristics are quite adequate to be included in milk and BF-7 milk is still working well on neuro-protection, result in enforcing our brain and delaying neurodegeneration.
