Seven rc mutant and seven normal male birds (Rhode Island Red suie, RIR) were used in this study to determine the effects of rc mutation on semen characteristics, testosterone profile and spermatogenic tissues. All birds were randomly selected at week 12 of age and housed in individual cages and were fed and watered ad libitum. The birds were exposed to a 14L:10D light cycle during experiment. Semen were collected at weeks 22 to 23 from each bird twice a week and evaluated for semen volume (SV), sperm concentration (SC), total sperm count (TSC), percent of sperm motility (%SM), dead sperm (%DS), and sperm metabolic activity (SMA). To determine the testosterone concentration (TC) in plasma, blood was collected at weeks 12, 16 and 18. Testicular tissue were collected, processed and evaluated for semineferous tubule diameter (STD), round spermatid number (RSN), percent elongated sperm (%ES) and semineferous tubules length (STL). Body weight (BW), comb weight (CW) and testes weight (TW) were weighted at the end of experiment (week 23). The SV, TSC and %SM were significantly higher in normal birds but the %DS was higher in blind birds (p<0.05). The SC did not differ significantly between the two groups but its value was higher in normal birds. The sperm metabolic activity in the first h of collection did not differ significantly between the two groups but after 24 h, the level of SMA in normal group was significantly higher (p<0.05). The level of TC did not differ significantly between the two genotype groups but normal birds had higher TC in all collections except the last one. The STD, RSN, %ES and STL in normal birds were higher when compared to blind birds but the differences were insignificant except for ES percent. The BW, CW and TW between the two groups did not differ significantly but the weights were higher in normal group compared to blind birds. Statistical analysis of semen characteristics, testosterone profile and histological factors were indicated detrimental effects of rc mutation in prepubertal RIR blind male birds due to lack of light.
Purpose : These studies were undertaken to evaluate the effects of Allii tuberosi Semen (ATS) on the spermatogenic abilities such as concentration, motility and morphological normality of sperm from the testis and the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. Materials and Methods : We used the 8-week-old mice and administered the 0.2 ml extract solution of ATS in the different concentration (0.1 mg/ml, 1 mg/ml, 10 mg/ml and 100 mg/ml) once a day for 60 days. The control group was administered the distilled water in the same way. After the administration of each extract solution, we examined the number of total, motile and normal sperm, the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. Also we observed the histological changes of isolated testis. And we compared to the testicular tissue especially seminiferous tubules between control and treated group by histochemical methods. Results : The concentration of total sperm, the motility and normality of spermatozoa were significantly increased in ATS groups, especially in 1 and 10 mg/ml groups, compared to control group. In the histological analysis of the testicular tissues, the enlargement of testicular lobe diameter and apparent vasculogenesis between testicular lobes were observed in the ATS groups compared to the control group, respectively. Also, the activity of hyaluronidase was significantly increased in the ATS groups compared to the control group. In the antioxidant activity analysis, the activity of testicular peroxidase was significantly increased in the ATS groups compared to the control group, especially in 1 mg/ ml group. The activity of testicular catalase was increased in ATS groups. Conclusion : This study shows that ATS has the beneficial effect on the concentration, morphology and motility of sperm, the activities of sperm hyaluronidase and testicular peroxidase. We can suggest that ATS extract solution be useful for the treatment of male sexual dysfunctions and infertility.

Methods : We used the 2-month-old mice and administered the extract solution of Morindae officinalis Radix in the different concentration once a day for 60 days. The control group was administered the normal saline in the same way and duration. We examined the number of total, motile and normal sperm from the cauda epididymis, the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. Also we observed changes of isolated testis before and after administration of Morindae officinalis Radix extract solutions the mice. And we compared to the testicular tissue especially seminiferous tubules between control and treated group by histochemical methods. Results : The significant dose-dependent differences were observed in the concentration of total sperm, the motility and normality of spermatozoa of Morindae officinalis Radix extract solution administered groups compared to the control group, respectively. In the histological analysis of the testicular tissues, the enlargement of testicular lobe diameter and apparent vasculogenesis between testicular lobes were observed in the Morindae officinalis Radix extract solution administered groups compared to the control group, respectively. Also, the activity of hyaluronidase was significantly increased in the Morindae officinalis Radix extract solution administered groups compared to the control group. In the antioxidant activity analysis, the activity of testicular peroxidase was significantly increased in the Morindae officinalis Radix extract solution administered groups compared to the control group, respectively. With an increase in the number of people suffering from ageing-related diseases in our rapidly aging society, interests in natural products such as maca(Lepidium meyenii), which has properties of enhancing cognition and sexual performance, have increased. This study was conducted to assess the effects of 7 weeks of maca extract supplementation(0.5~2.0 g/kg BW) on scopolamine-induced amnesia in mice and on sperm count in male mice. All doses of maca supplementation significantly protected against scopolamine-induced amnesia as determined by a Morris water maze, but not according to passive avoidance tests. Maca supplementation did not affect acetylcholinesterase activity in the whole brain, nor the testicular sperm count of male mice. This study suggests that maca may have some neuroprotective properties in mice, which will be further examined by future studies.
This study was performed to investigate the reproductive characteristics of the cloned Hanwoo bulls produced by SCNT. The semen ejaculated from the cloned bulls (C-38 and C-39) and normal Hanwoo bull was properly measured the volume, the number of sperm, and the viability of frozen-thawed sperm. The sperm activity was analyzed using computer assisted sperm analysis (CASA). To analyze fertilizing ability of the cloned bulls, in vitro fertilization and artificial insemination were performed using the frozen-thawed semen. There were no differences in semen volume, sperm concentration, and the viability of frozen-thawed sperm between cloned bulls and normal bull. The difference was statistically significant in total motility, curvilinear velocity (VCL), straight-line velocity (VSL), and average-path velocity (VAP) of both cloned bulls compared to those of normal Hanwoo bull, respectively (p<0.05). The cleavage and blastocyst development rate were not different between the groups. five cloned cows were artificially inseminated using the frozen-thawed semen of C-38, two of them became pregnant. Two second generation calves (one male and one female) were produced. Based on these results, the cloned Hanwoo bulls showed normal reproductive abilities of semen parameters and sperm activity to their comparators and produced cloned calves, although there are some individual differences on the parameters.
The Jeju horse has been raised for centuries in Jeju island. Recently, as the number of this indigenous horse has been dropped dramatically, this breed became Natural Monument #347 to conserve and multiply this endangered breed. To provide the basic information for AI, sexual activity and semen characteristics in Jeju horse were investigated. Jeju horse semen was collected using Missouri style artificial vagina from fertile stallion.\\`she number of mount per ejaculation was 2..3$\pm$1.8, and the ejaculation time was 27.0$\pm$12.5 seconds. The total volume and gel-free volume of semen was 47.8$\pm$26.7 ml and 42.7$\pm$27.4ml, respectively, and the concentration of sperm and the total number of spermatozoa per ejaculation was 200.7$\pm$112.9$\times$10$^{6}$ ml and 7.6$\pm$3.9$\times$10$^{9}$ ml, respectively. The percentage of motile sperm and the number of live spermatozoa per ejaculation was 75.0$\pm$18.2% and 6.1$\pm$3.4$\times$10$^{9}$ ml, respectively, and the pH of gel-free semen was 7.3$\pm$0.2. The total percentage of abnormal sperm was 31.5%, and the percentage of sperm with abnormal head, midpiece and tail was 9.5$\pm$11.7%, 7.0$\pm$4.0% and 15.0$\pm$15.0%, respectively.
This study was performed to investigate the characteristics within ages and freezing tolerance of spermatozoa in Jindo Dog. Experimental animals were selected 12 herds within 1~8 year's old and collected semen for 2 times in a week. Collected semen was evaluated whole volume and sperm number with CASA system (SIAS, Medical Supply, Korea). Then seminal plasma were separated and diluted with modified Tris-egg yolk extender and added 4, 6 and 8% glycerol for 4 times to final concentration and equilibrated for 1.5 hrs. Before and after freezing, equilibrated semen were evaluated the survival rates. Total volume of sperm at 1~2 year old group is as $5.2{\times}10^8\;cells/ml$ largest and there were no significance among groups. The motility of 1~2 year old group is highest as 90.9% and there were significance among groups. Abnormal sperm showed similar among groups. The survival rate in terms of pre-freezing and post-freezing were decreased all levels of glycerol and reveled 87.0% to 64.5% in 4%, 87.5% to 51.9% in 6% and 73.4% to 29.7% in 8%, there were significant difference among the groups (p<0.05). These results suggest that the optimal sperm-freezing methods in Jindo Dog are utilized with modified Tris egg-yolk extender with 4% glycerol and were improve the reproductive activity by these methods.
