HCV는 HIV등과 함께 수혈이나 혈장된 획물질을 통하여 감염되는 주요 바이러스이다. 주로 혈액이나 혈장에서 HCV에 대한 항체를 검출함으로서 HCV의 감염을 방지하고 있다. 그러나 바이러스에 감염되었으나 항체가 생성되기 이전이나 항체의 양이 적은 경우에는 HCV의 검출이 어렵다. 따라서 핵산중폭시험(nucleic acid amplification tests, NAT)을 이용한 HCV 유전자를 검출하려는 시도들이 진행되고 있다. 이 연구의 목적은 혈장분획물질에서 HCV RNA를 검출할 수 있는 핵산증폭시험 방법을 개발하는 것이다. 5종류의 PCR primer를선별하여 실험에 이용하였다. 혈장분획물질의 HCV RNA 추출에는 컬럼 방법을 이용하는 것이 유용한 것으로나타났다. 핵산중폭시험의 결합 온도는 $48^{\circ}C$가 가장 적절한 것으로 나타났다. 또한 2차 PCR의 경우, 1차 PCR 산물 $1{\mu}l$와 30 pmol의 primer즐 사용하였을 때 높은 민감도와 특이성을 보이는 것을 알 수 있었다. 혈장 분획물질에 HCV를 주입하여 혈장중폭시험을 수행한 결과, 100 IU/ml까지 검출 할 수 있었다. 한편 근육주사용항체(IMIG)의 경우 핵산중폭시험을 통한 검출한계는 100IU/ml로 COBAS amplicor HCV2.0의 500 IU/ml 이상의 검출한계보다 민감도가 더 높은 것으로 나타났다. 이러한 결과들로 보아 본 실험에 이용된 핵산증폭시험이 혈장분획물질에서 HCV RNA를 검출하는데 유용한 방법으로 사용될 수 있을 것으로 생각된다.
배경: 결핵은 전세계적으로 단일 원인균으로는 가장 높은 치사율을 가진 질병이다. 결핵의 치료에 있어서 마이코박테리아를 빠르고 정확하게 확인하는 것이 필수적이다. 본 연구에서는 임상 객담 검체의 도말 슬라이드에서 결핵균과 비결핵 항산균의 감별을 위해 Peptide Nucleic Acid (PNA) Probe를 이용한 FISH assay를 평가하고자 한다. 방법: 결핵균과 비결핵 항산균의 16s rRNA를 타겟으로 한 PNA probe를 합성하였다. 각 probe의 특이도는 표준 균주인 Mycobacterium tuberculosis ATCC 13950, M. kansasii ATCC 12479, 임상 검체에서 분리된 마이코박테리아 3종(M. abscessus, M. avium, and M. intracellulare) 그리고 호흡기계에서 흔히 분리되는 10종류의 박테리아를 이용하였다. Centers for Disease Control and Prevention (CDC)의 Acid fast bacili (AFB) 염색 기준상 trace 이상인 128개의 임상 객담 검체를 이용하여 Probe의 성능을 평가하였다. PNA FISH와 항산균 염색결과는 CDC 기준에 따라 단계를 분류하고 서로 비교하였다. 결과: 결핵균과 비결핵 항산균 특이 PNA probe는 결핵균과 M. kansasii 표준 균주 그리고 임상검체에서 분리된 3종의 마이코박테리아에 대해 특이적인 반응을 보였고 다른 박테리아와 교차반응을 보이지 않았다. 또한 89개의 결핵균 배양 양성 객담 검체와 29개의 비결핵 항산균 배양 양성 객담 검체에 각각 특이적인 반응을 보였고 CDC 기준에 따라 분류한 PNA FISH와 AFB염색 결과는 2+ 이상의 검체에서 서로 잘 일치하였다. 결론: PNA FISH 방법은 임상 객담 검체에서 결핵을 진단하고, 항산균과 비결핵 항산균을 구분하는데 있어서 민감하고 정확한 결과를 보였다.
Purpose: This review aims to introduce aptamers and the methods of its development to improve the sensitivity and selectivity to target bacteria. In this review, we have highlighted current developments and directions in the pathogen detection based on aptamers. Background: Aptamers, the specific nucleic acid sequences, can bind to targets with high affinity and specificity. Some of researches on the use of aptamers for the detection of pathogen have been reported in recent years. Aptamers have more applicability than antibodies for the development of pathogen detection using biosensor; such as easy to synthesis and labeling, lack of immunogenicity, and a low cost of production. However, only few reports on the development and use of aptamers for the detection of pathogen have been published. Review: Aptamers specific to pathogen are obtained by whole-cell systematic evolution of ligands by exponential enrichment (SELEX) process. SELEX process is composed of screening random oligonucleotide bound with target cells, multiple separation and amplification of nucleic acids, final identification of the best sequences. For improving those affinity and selectivity to target bacteria, optimization of multiple separating process to remove unbounded oligonucleotides from aptamer candidates and sorting process by flow cytometry are required.
The purpose of this study was to detect Mycobacterium bovis in cattle(serum, milk, lung, lymph node) by PCR. Nineteen samples from 7 skin test-positive cattle were analyzed. The amplified band of IS6110 by PCR was detected from 2 samples in lung and Iymph node. But the sensitivities of the present methods for detecting M bovis in milk and serum are deficient. Because the PCR sensitivity has been shown to be hindered by the method used to isolate the nucleic acid target. PCR-based methods have the potential to be faster, more accurate, and the most efficient means of detecting M bovis. The detection of M bovis by PCR will contribute to the more efficient detection and control of tuberculosis.
In biometric and biomedical applications, a special transporting mechanism must be designed for the ${\mu}$TAS (micro total analysis system) to move samples and reagents through the microchannels that connect the unit procedure components in the system. An important issue for this miniaturization and integration is microfluid management technique, i.e., microfluid transportation, metering, and mixing. In view of this, this study presents an optimal fuzzy sliding-mode control (OFSMC) design based on the 8051 microprocessor and implementation of a complete microfluidic manipulated system implementation of biochip system with a pneumatic pumping actuator, a feedback-signal photodiodes and flowmeter. The new microfluid management technique successfully improved the efficiency of molecular biology reaction by increasing the velocity of the target nucleic acid molecules, which increases the effective collision into the probe molecules as the target molecules flow back and forth. Therefore, this hybridization chip was able to increase hybridization signal 6-fold and reduce non-specific target-probe binding and background noises within 30 minutes, as compared to conventional hybridization methods, which may take from 4 hours to overnight. In addition, the new technique was also used in DNA extraction. When serum existed in the fluid, the extraction efficiency of immobilized beads with solution flowing back and forth was 88-fold higher than that of free-beads.
바이로이드는 매우 작은 RNA 분자로 구성되어 있으며, 외피 단백질이 없고 오로지 식물에만 감염되어 질병을 유발한다. 바이로이드 감염 질병을 예방하거나 진단하는 것은 상당히 어려운 일이며, 이는 병징이 초기에는 발견되지 않고 수확기에 접어들어서 발견되기 때문이다. 한편, 혈청학적인 방법은 식물 병원체를 검출하기 위해 주로 사용되었으나 바이로이드는 핵산인 RNA로만 구성되어 있기 때문에 이 방법으로 검출할 수가 없다. 때문에 바이로이드를 검출하기 위해 주로 사용되는 방법은 분자 생물학적인 방법으로, 초기에는 바이로이드의 분자적인 크기와 구조적 특징을 이용한 겔 전기 영동 방법이 주로 사용되었다. 그 후에는 역전사 반응과 중합효소 연쇄반응을 접목시킨 역전사 중합효소 연쇄반응(RT-PCR) 방법이 활용되었고, 그에 대한 효율적인 결과 확인을 위해 형광 물질을 도입한 실시간 역전사 중합효소 연쇄반응(Real-time RT-PCR)이 도입되었다. 그러나 그들은 온도를 변화시키기 위한 값비싼 기기와 전문적인 인력이 필요함으로 현장에서는 활용되기가 어렵다. 최근 개발된 고리 기반의 등온 증폭법(Loop-mediated isothermal amplification)의 경우, 온도의 변화가 필요 없어 비싼 온도 조절 기기가 필요하지 않다. 또한 매우 높은 증폭 효율을 지니며 반응 시간이 짧은 등의 여러 장점을 지니고 있기에 최근 현장 진단용 기술에 도입되고 있다. 이러한 배경으로, 이 총설에서는 바이로이드 유발 질병에 대하여 요약하고 그에 대한 검출 및 진단 방법에 대한 연구 동향에 대하여 기술하였다.

Background: Diagnostic methods for pulmonary tuberculosis (TB) have recently advanced. The aim of this study was to evaluate the changes in TB diagnostic tests that prompted the initiation of anti-TB treatment over time in South Korea, an industrialized country with an intermediate TB burden. Methods: Patients diagnosed with pulmonary TB in the first halves of 2005 and 2013 at a tertiary referral hospital were included. Diagnostic methods that prompted the initiation of anti-TB treatment were compared between the 2 groups of patients. Results: A greater proportion of patients were diagnosed with pulmonary TB using bronchoscopy in 2013 than in 2005 (26.7% vs. 6.6%, respectively; p<0.001), while the proportion of patients clinically diagnosed with pulmonary TB was lower in 2013 than in 2005 (24.7% vs. 49.0%, respectively; p<0.001). Additionally, more patients started anti-TB treatment based on positive polymerase chain reaction (PCR) results for Mycobacterium tuberculosis DNA in 2013 than in 2005 (47.3% vs. 7.9%, respectively; p<0.001). Conclusion: The initiation of treatment for pulmonary TB in South Korea has become more frequently based on PCR and the use of bronchoscopic specimens.

If rumen bacteria can be manipulated to utilize nutrients (i.e., ammonia and plant cell wall carbohydrates) more completely and efficiently, the need for protein supplementation can be reduced or eliminated and the digestion of fiber in forage or agricultural residue-based diets could be enhanced. However, these approaches require a complete and accurate description of the rumen community, as well as methods for the rapid and accurate detection of microbial density, diversity, phylogeny, and gene expression. Molecular ecology techniques based on small subunit (SSU) rRNA sequences, nucleic acid probes and the polymerase chain reaction (PCR) can potentially provide a complete description of the microbial ecology of the rumen of ruminant animals. The development of these molecular tools will result in greater insights into community structure and activity of gut microbial ecosystems in relation to functional interactions between different bacteria, spatial and temporal relationships between different microorganisms and between microorganisms and reed panicles. Molecular approaches based on SSU rRNA serve to evaluate the presence of specific sequences in the community and provide a link between knowledge obtained from pure cultures and the microbial populations they represent in the rumen. The successful development and application of these methods promises to provide opportunities to link distribution and identity of gastrointestinal microbes in their natural environment with their genetic potential and in situ activities. The use of approaches for assessing pupulation dynamics as well as for assessing community functionality will result in an increased understanding and a complete description of the gastrointestinal communities of production animals fed under different dietary regimes, and lead to new strategies for improving animal growth.

Mycobacterium tuberculosis (MTB) continues to be one of the main causative agents of tuberculosis (TB); moreover, the incidence of nontuberculous mycobacteria (NTM) infections has been rising gradually in both immunocompromised and immunocompetent patients. Precise and rapid detection and identification of MTB and NTM in respiratory specimens are thus important for MTB infection control. Molecular diagnostic methods based on the nucleic acid amplification test (NAAT) are known to be rapid, sensitive, and specific compared to the conventional acid-fast bacilli (AFB) smear and mycobacterial culture methods. In the present study, the clinical performances of three commercial molecular diagnostic assays, namely TB/NTM PCR (Biocore), MolecuTech Real MTB-ID® (YD Diagnostics), and REBA Myco-ID® (YD Diagnostics), were evaluated with a total of 92 respiratory specimens (22 AFB smear positives and 67 AFB smear negatives). The sensitivity and specificity of TB/NTM PCR were 100% and 75.81%, respectively. The corresponding values of MolecuTech Real MTB-ID® and REBA Myco-ID® were 56.52% and 90.32%, and 56.52% and 82.26%, respectively. TB/NTM PCR showed the highest sensitivity; however, the concordant rate was 10% compared with sequence analysis. Although MolecuTech Real MTB-ID® showed lower sensitivity, its specificity was the highest among the three methods. REBA Myco-ID® allowed accurate classification of NTM species; therefore, it was the most specific diagnostic method. Of the three PCR-based methods, MolecuTech Real MTB-ID® showed the best performance. This method is expected to enable rapid and accurate identification of MTB and NTM.
Human Sapovirus (HuSaV) is one of the major causes of acute gastroenteritis in humans, and it is used as a molecular diagnostic technique based on polymerase chain reaction (PCR) from humans, food, shellfish, and aquatic environments. In this study, the HuSaV diagnosis technique was used in an aquatic environment where a number of PCR inhibitors are included and pathogens, such as viruses, are estimated to exist at low concentration levels. HuSaV-specific primers are improved to detect 38 strains registered in the National Center for Biotechnology Information (NCBI). The established optimal condition and the composition, including the RT-nested PCR primers and SL® Non-specific reaction inhibitor, were found to have 100 times higher sensitivity based on HuSaV plasmid than the previously reported methods (100 ag based on HuSaV plasmid 1 ng/μL). Through an artificial infection test, the developed method was able to detect at least 1 fg/μL of HuSaV plasmid contaminated with total nucleic acid extracted from groundwater. In addition, RT-nested PCR primer sets for HuSaV detection can react, and a positive control is developed to verify false positives. This study is expected to be used as a HuSaV monitoring method in the future and applied to the safety response to HuSaV from water environments.
