Development of reverse-transcription loop-mediated isothermal amplification assays for point-of-care testing of human influenza virus subtypes H1N1 and H3N2

  • Influenza A virus (IAV) is the most widespread pathogen causing human respiratory infections. Although polymerase chain reaction (PCR)-based methods are currently the most commonly used tools for IAV detection, PCR is not ideal for point-of-care testing. In this study, we aimed to develop a more rapid and sensitive method than PCR-based tools to detect IAV using loop-mediated isothermal amplification (LAMP) technology. We designed reverse-transcriptional (RT)-LAMP primers targeting the hemagglutinin gene. RNAs from reference H1N1 and H3N2 showed specific RT-LAMP signals with the designed primers. We optimized the reaction conditions and developed universal reaction conditions for both LAMP assays. Under these conditions, the detection limit was 50 copies for both RT-LAMP assays. There was no non-specific signal to 19 non-IAV respiratory viruses, such as influenza B virus, coronaviruses, and respiratory syncytial viruses. Regarding the reaction time, a positive signal was detected within 25 min after starting the reaction. In conclusion, our RT-LAMP assay has high sensitivity and specificity for the detection of the H1 and H3 subtypes, making it suitable for point-of-care IAV testing.

  • Conventional measurement methods using ultrasonic wave or x-ray, eddy current for non-destructive testing(NDT) in nuclear power plants and other industrial plants have been utilized as the method of contact with objects to be inspected. For this reason these methods require relatively much time and inspection area is limited by the location of probe or film. But holograpic interferometry which is a non-contact optical measurement method using a coherent light source has an advantage that quantative measurement can be performed at a time. In this paper a new method using realtime holographic interfreometry and image processing for detecting internal flaws of pressure vessels is presented.

  • IBM developed the MPI-IO, we call it MPI-2, on the General Parallel File System. We designed and implemented various Matrix Multiplication Benchmarks to evaluate its performances. The MPI-IO on the General Parallel File System shows four kinds of data access methods : the non-collective and blocking, the collective and blocking, the non-collective and non-blocking, and the split collective operation. In this paper, we propose benchmarks to measure the IO time and the computation time for the data access methods. We describe not only its implementation but also the performance evaluation results.

Numerical assessment of step-by-step integration methods in the paradigm of real-time hybrid testing

  • Real-time hybrid testing (RTHT) involves virtual splitting of the structure into two parts: physical substructure that contains the key region of interest which is tested in a laboratory and numerical substructure that contains the remaining part of the structure in the form of a numerical model. This paper numerically assesses four step-by-step integration methods (Central difference method (CDM), Operator splitting method (OSM), Rosenbrock based method (RBM) and CR-integration method (CR)) which are widely used in RTHT. The methods have been assessed in terms of stability and accuracy for various realistic damping ratios of the physical substructure. The stability is assessed in terms of the spectral radii of the amplification matrix while the accuracy in terms of numerical damping and period distortion. In order to evaluate the performance of the methods, five carefully chosen examples have been studied - undamped SDOF, damped SDOF, instantaneous softening, instantaneous hardening and hysteretic system. The performance of the methods is measured in terms of a non-dimensional error index for displacement and velocity. Based on the error indices, it is observed that OSM and RBM are robust and performs fairly well in all the cases. CDM performed well for undamped SDOF system. CR method can be used for the system showing softening behaviour. The error indices indicate that accuracy of OSM is more than other method in case of hysteretic system. The accuracy of the results obtained through time integration methods for different damping ratios of the physical substructure is addressed in the present study. In the presence of a number of integration methods, it is preferable to have criteria for the selection of the time integration scheme. As such criteria are not available presently, this paper attempts to fill this gap by numerically assessing the four commonly used step-by-step methods.

비접촉 초음파 탐상기법을 이용한 스폿용접부 및 탄소복합체의 내부 결함평가 (Internal Defection Evaluation of Spot Weld Part and Carbon Composite using the Non-contact Air-coupled Ultrasonic Transducer Method)

  • NAUT(Non contact Air coupled Ultrasonic Testing)기법은 초음파 탐상법 중의 하나로서 공기중 음향 임피던스의 차이로 생기는 에너지 손실을 High Power 초음파 Pulser Receiver, PRE-AMP, 고감도의 탐촉자로 보완하여 비접촉식으로 초음파 탐상을 가능하게 하는 탐상 방법이다. NAUT는 초음파의 송신 및 수신이 안정된 상태에서 이루어지므로 기존의 접촉식 탐상으로는 불가능하였던 고온, 저온의 물질이나 시험편의 표면이 거친 부분, 좁은 지점 등에서도 탐상이 가능하다. 본 연구에서는 NAUT기법의 산업체 실용여부를 알아보기 위해 자동차생산 공정에서 많이 사용하는 스폿용접부 및 CFRP 제품에 있어 상용화 연구를 통해 다음과 같은 결과를 얻었다. 본 연구에서는 NAUT기법의 사용 여부를 알아보기 위해 자동차 부품에서 많이 사용하는 스폿용접부 및 CFRP 부품의 내부결함 검출을 검출하였다. 스폿용접부에서는 초음파의 투과율이 높아 적색으로 나타났으며, 복층으로 된 부분은 투과율이 낮아 청색 화상이 나타났다. 또한 측정 속도를 결정하는 중요요소인 PRF(Pulse Repetition Frequency;송신펄스주기)에 따라 색상 선명도의 차이를 보였다. CFRP 시험편 또한 화상장치를 통해 취득된 각 화상 결과를 보고 내부 결함의 모양, 크기, 위치 등의 파악이 단시간에 가능하였다. 실험을 통해 NAUT기법과 화상화가 동시에 이루어짐을 확인하였고, 스폿 용접부와 CFRP 탐상에 NAUT의 적용이 가능한지 그 실현여부를 확인하였다.

Developing Objective Linear Scale Data from Subjective Tests for Consumer Products

  • In the consumer products industry, it has been highly desirable to develop objective test methods to replace subjective evaluation methods. In developing an objective test method, subjective evaluation data should be on a linear scale. According to Thurstone's theory of comparative judgment, a%-preference from a paired-comparison test can be converted to a linear-scale value. The required number (N) of paired-comparison tests increases dramatically as the number of products increases. This problem should be solved by classifying the total products into several subgroups consisting of 3-4 products in each group. By doing so, it can not only significantly reduce the number of required paired-comparison tests, but it can also obtain more reliable, reproducible data.

  • RFI 공정은 진공백 내부에 섬유 매트와 수지 필름을 적층하여 성형하는 OoA 공정이다. 외부에서 따로 주입되는 수지가 없기 때문에 수지 필름의 양이 섬유가 필요로 하는 양보다 적은 경우 복합재 내부에 기공 결함이 발생하며 기계적 물성이 저하된다. 이러한 이유로 제작한 복합재를 실용화하기 위해서 비파괴검사를 이용한 기공예측이 필수적으로 요구된다. 따라서 본 연구에서는 RFI 공정에서 비파괴검사 시 기준으로 사용할 수 있는 표준 기공률 시편을 제조하는 방법을 제시하였다. 표준 기공률 시편 제작 방법으로 수지 필름 두께를 조절하는 방법을 사용하였으며, 목표 기공률별 수지 필름 두께를 설정하기 위한 방법으로 섬유 압착 실험을 제시하였다. 수지 필름 두께 조절을 통하여 0%, 2%, 4%의 목표 기공률 패널을 제작했고 비파괴시험과 기공률 측정을 통하여 기공률에 따른 비파괴검사 신호 감쇠를 측정했다. 또한 인장, 면내전단, 숏빔, 압축 시편의 신호 감쇠를 통하여 기공률을 추정하였고, 기공률에 따른 기계적 물성을 평가하였다.

  • We have fuzzy hypotheses testing from Bayesian statistics with ideas from fuzzy sets theory to generalize Bayesian methods both for samples of fuzzy data and for prior distributions with non-precise parameters. Appling the principle of agreement index, the posterior odds ratio in the favor of hypotheses $H_0$ is equal to product of the fuzzy odds ratio and the fuzzy likelihood ratio. If the Posterior odds ratio exceeds the grade judgement, we accept the hypothesis $H_0$ for the degree.

  • As the function of digital products gets diverse and complex, more than 30~40% job of software developing and testing group within its development cycle, concentrates on the software testing[1]. To cope with consumers'quickly changing needs, development cycle time gets short, and the number of model and test items increase steadily This is why automating software basic function and UI(User Interface) verification is needed [4][5][6]. This paper draws many strength and weakness defines Input/Output through integrating mobile phone, DVDP, PDA, and Black-Box (Intrusive (mobile) or Non-Intrusive(DVDP, PDA)) testing systematically, which brought above the 64% average of automation rate, and ensure 80% of test coverage [4][5][6].

  • Conventional measurement methods for non-destructive testing(NDT) in nuclear power plants other industrial plants have been performed as the methods of contact with objects to be inspect, but those methods have been taken relatively much time to be inspected. Holographic interferometry which is a non-contact optical measurement method using a coherent light can overcome these demerit, and also has an advantage that the quantitative measurement of small deformation for large areas can be accomplished at a time with high precision. In this paper the comparisons of the experimental results from holographic interferometry with those form the finite element method(FEM) and the analytical solutions of elastic equation are discussed.
