Normal human skin, constantly challenged by environmental micro-organisms, has an innate ability to fight invading microbes through antimicrobial peptides. These peptides, described in both plant and animal kingdoms are able to inactivate a broad spectrum of micro-organisms. Mammalian defensins constitute one of the most common antimicrobial peptide family. Among the three human beta-defensins hBD1, hBD2 and hBD3 produced in epithelia, only hBD2 and hBD3 are inducible and additionally have been described as expressed by differentiated keratinocytes at site of inflammation and infection. The aims of these studies were to define a cell culture model in which the basal production of hBD could be detected and up-regulated in order to enhance skin auto-protection against micro-organisms. A specific Polymerase Chain Reaction method have been developed for hBD2 and hBD3 mRNA detection in non-differentiated monolayer keratinocytes cell culture. We have been able to demonstrate that in vitro, hBD2 and hBD3 expression in normal human keratinocytes could be detected and enhanced by TNF-alpha and IFN-gamma, in hypercalcic culture conditions. This research opened the possibility of the development of cosmetic active compounds, able to induce the expression of skin natural antibiotic peptides responsible about microflora ecology of the skin.
Apoptosis, or programmed cell death, is a genetically regulated pathway that is altered in many cancers. Overexpression of bcl-2 leads to resistance to apoptosis and promotes tumorigenesis. To determine the effect of bcl-2 antisense treatment in human lung cancer cell lines, a 20 mer full phosphorothioate oligonucleotide (ODN) targeted at the coding region of the bcl-2 mRNA was synthesized. Western blot analyses were used to examine bcl-2 protein level in five human non-small cell lung cancer (NSCLC) cell lines (NCI-H226, SK-MES-1 NCI-H358, NCI-H522 and NCI-Hl 299) and four human small cell lung cancer (SCLC) cell lines (NCI-H69, NCI-H4l7, HCC-2108 and SW2). Three out of five NSCLC (NCI-H226, SK-MES-1 and NCI-Hl 299) and all of SCLC cell lines expressed Bcl-2 protein. Treatment of these cell with antisense ODN for 48 hours reduced their viability and Bcl-2 protein level. As a conclusion, bcl-2 antisense treatment appears reduction of the Bcl-2 protein levels and cytotoxic effect including apoptosis in human lung cancer cell lines.
Objectives In human myeloid leukemia cells, there are no specific features of apoptosis compared with apoptosis in other cell types. Solanum nigrum L.(SNL) is a deciduous tree, which is widely distributed in Korea with reported anti-tumor, anti-inflammatory and non-specific immune-enhancing properties. Although the plant has been clinically used for treating a variety of diseases, its bioactive ingredients are unknown and its mode of action potential has never been investigated. Thus anti-tumor property of methanol extract was investigated. Methods In this study, anti-tumor property of methanol extract was investigated by determining its in vitro growth-inhibitory effects on human myeloid leukemia cells. XTT proliferation assay, DNA fragmentation, immunoblot analysis, densitometric analysis were used. Results 1. The methanol fraction of the extracts of SNL induced mitochondria-mediated apoptosis in human myeloid leukemia cells. 2. The methanol fraction exhibited relatively higher cytotoxic activity in a dose-dependent manner than chloroform, and hexane fraction. 3. Typical ladder profile of Oligonucleosomal fragments were appeared. 4. The secreted cytosolic cytochrome C level was increased by treatment of methanol fraction. Conclusions Methanol fraction of SNL is capable of inducing apoptosis in human myeloid leukemia cells.

Pectin lyase-modified red ginseng extract (GS-E3D) is a newly developed ginsenoside Rd-enriched ginseng extract. This study was designed to investigate the skin safety of GS-E3D. Single oral toxicity, single dermal toxicity, bovine corneal opacity and permeability (BCOP) assay, skin irritation test with $SkinEthic^{TM}$ human epidermis model, skin sensitization local lymph node assay, and human patch test, were examined. The oral and dermal $LD_{50}$ value of GS-E3D was over 2,000 mg/kg in rats. GS-E3D was identified as a non-irritant to skin in BCOP assay, human epidermis models, and patch test from the 32 human subjects. The skin sensitization potential of GS-E3D was less than 25% in local lymph node assay. These results indicate that GS-E3D can be used as a safe ingredient without adverse effects in various skin care products.
Using GIS and socio-economical data the relationship between human activities and global environmental change Is Analysed from the view point of food productivity and CO2 emission. Under the assumption that the population problem, the food problem and global warming due to energy consumption can be stabilized through managing land use, impacts of human activities such as consumption of food, energy and timber on global environment changes, and global population capacity are Analysed using developed system dynamics model in the research. In the model the world is divided into two groups: OECD countries and the others. Used global land use data set Is land cover map derived from satellite data, and potential distribution of arable land is estimated by the method of Clamor and Solomon which takes into consideration spatial distribution of climate data such as precipitation and evapotranspiration. In addition, impacts of CO2 emission from human activities on food production through global warming are included in the model as a feedback. The results of the analysis for BaU scenario and Toronto Conference scenario are similar to the results of existing models. From the result of this study, the human habitability in 2020 is 8 billion people, and CO2 emission in 2020 based on BaU Scenario and on Toronto Scenario is 1.7 and 1.2 times more than the 1986's respectively. Improving spatial resolution of the model by using global data to distribute the environmental variables and sauce-economical indices is left for further studies.

This paper suggests a fast human detection algorithm that can be applied to a high-resolution CCTV camera. Human detection algorithms, which used a HOG detector show high performance in the region of image processing. On the other hand, it is difficult to apply to real-time high resolution imaging because of its slow processing speed in the extracting figures of HOG. To resolve this problems, we suggest how to detect humans into two stages. First, candidates of a human region are found using background subtraction, and humans and non-humans are distinguished using a HOG detector only. This process increases the detection speed by approximately 2.5 times without any degradation in performance.

Natural human epidermal growth factor (nhEGF) was purified from pregnant human urine by benzoic acid adsorption, DEAE-Sepharose ion exchange, and immunoaffinity chromatography. The purified nhEGF was further separated into four fractions using Bondapak C$_{18}$ HPLC system. Following characterization by Western blot analysis and double immu- nodiffusion, we found that each fraction corresponds to four derivatives of the nhEGF. For biological analysis of nhEGF, we optimized the labeling time and serum concentration for the incorporatioin of 5-bromo-2'-deoxy uridine (BrdU), a non-radioactive alternative for [$^{3}$H]-thymidine uptake, into NIH 3T3 cells. The DNA synthesis of NIH 3T3 cells was gradually increased at the nhEGF concentrations between 0.1 - 10 ng/ml in the Dulbecco's Modified Eagles Medium (DMEM) containing 0.2% Fetal calf serum (FCS). When we assayed the biological activity of four fractions, the activity of the second fraction was superior to that of the others. Based on the results from the HPLC analysis spiked with recombinant human epidermal growth factor (rhEGF) and amino acid sequencing, we concluded that the second fraction was nhEGF and the other three fractions were the derivatives of nhEGF. In addition, the proportion of nhEGF was approximately 46% is compared with that of the other three derivatives.
This study assessed the stabilization of fluorine (F)-contaminated soil using calcium hydroxide (Ca(OH)2) and the consequent changes in human health risk. The bioavailable F decreased to 3.5%, (i.e., 57.9 ± 1.27 mg/kg in 6% Ca(OH)2-treated soil sample) from 43.0%, (i.e., 711 ± 23.4 mg/kg in control soil sample). This resulted from the conversion of water-soluble F to stable calcium fluoride, which was confirmed by XRD spectrometry. Soil ingestion, inhalation of fugitive dust from soil, and water ingestion were selected as exposure pathways for human health risk assessment. Non-carcinogenic risks of F in soils reduced to less than 1.0 after stabilization, ranging from 4.2 to 0.34 for child and from 3.0 to 0.25 for adult. Contaminated water ingestion owing to the leaching of F from soil to groundwater was considered as a major exposure pathway. The risks through soil ingestion and inhalation of fugitive dust from soil were insignificant both before and after stabilization, although F concentration exceeded the Korean soil regulatory level before stabilization. Our data suggested that substantial risk to human health owing to various potential exposure pathways could be addressed by managing F present in soil.
Human Aichivirus A1 (HuAiV-A1) is a waterborne human pathogenic virus classified as Picornaviridae and Kobuvirus. In this study, we developed a method that can detect about 35 minutes faster with the same detection sensitivity level than the previously reported HuAiV-A1 diagnostic RT-PCR primer. The RT-PCR primer sets developed in this study are capable of detecting HuAiV-A1 at a level of about 100 ag and formed 563 bp amplification product. In addition, the RT-nested PCR method was able to amplify 410 bp using the RT-PCR product as a template. The detection sensitivity of our method was 10 times higher than the method with the highest detection sensitivity to date. Therefore, the detection method of HuAiV-A1 developed in this study is expected to be used in the water environment in which a small amount of virus exists. Also, this detection method is expected to be used as HuAiV-A1 diagnostic technology in both clinical and non-clinical field.

The issue of the relationship between environmental degradation and human health has been widely addressed by medical doctors. However, economists have sparsely debated it. The release of carbon dioxide (CO2) into the air can cause several environmental problems and, thus, it can affect human health. Therefore, it is imperative to examine the effect of CO2 emissions on life expectancy in the D-8 countries (Malaysia, Indonesia, Bangladesh, Nigeria, Egypt, Iran, Pakistan, and Turkey) from 1992 to 2017. The panel ARDL method is employed and, then, the PMG estimator is selected. The results show that economic growth, population growth and health expenditure can significantly and positively affect life expectancy, but CO2 emissions can have a significant and negative effect on life expectancy. Since, the major findings reveal that life expectancy can be explained by CO2 emissions. Hence, it is important to formulate policies on reducing CO2 emissions so that life expectancy will not be affected. Energy diversification policies should be formulated or improved in some countries. This is to ensure that the countries are not highly dependent on non-renewable energy that can harm the environment. The government should increase its expenditure on the health sector to save more lives by extend human lifespan.
