BACKGROUND: For sustainable agriculture, there are various agricultural practices including low input. Over the last few decades high input of chemical fertilizer and compounds results in environmental pollution and deterioration of soil fertility. Soybean (Glycine max L.) is well known eco-friendly crop due to their symbionts. Soybean has a relationship with nitrogen fixation bacteria called rhizobia. In this research work, we investigated effects of soybean cultivation on soil microorganism activities. METHODS AND RESULTS: Experiments were conducted in pots and potato cultivation was used as reference. Soil chemical properties were analyzed considering soil nutrient over cropping period. For the soil microbial community analysis, dehydrogenase activity analysis (DHA) analyzed along with denaturing gradient gel electrophoresis. The results showed that higher soil organic matter in the soybean cultivation soil than in the potato cultivation soil. Available $P_2O_5$ concentration increased gradually in both pots but showed higher value in the potato cultivation soil. DHA value implying microbial activities showed higher value in the soybean cultivation soil over all cropping period. CONCLUSION: The cause of high microbial activity in the soybean cultivation soil was considered to the effects of some specific microorganisms related to soybean cultivation. Therefore, the availability of soybean cultivation for sustainable agriculture should be encouraged in terms of microorganism community activity in soil.

This experiment was conducted to evaluate the effects of cutting in field and solution culture. Periodical samplings of alfalfa in cutting and uncutting plots were taken to measure nodule development and nodule activity. Regrowth of plant and nodule development after shoot cutting by different heights and nodule removal at different levels were investigated in solution culture of alfalfa plant. 1. Nodule weight in the field was reduced 30% after the first cutting and 25% after the second cutting, but during the following 30 days after second cutting, there was no significant difference between cutting and uncutting plots. 2. Specific nodule activities of cutting plots at the beginning of June and at the beginning of September were 80% and loo%, higher than those of uncutting plots respectively. Total nodule activities of cutting plots in late August and early September were 40% higher than those of uncutting plot. The decrease of nodule activity can be prevented by cutting at flowering stage. 3. The decrease of nodules in solution culture when 50% of the shoot was cut, was as much as that when shoot was not cut or flower buds were removered. But when 90% of the shoot was cut, the number of the nodules were decreased more remarkably than the above treatments. New nodules, when 90% of the shoot was cut, were reformed slowly and did not grow fully until 15 days after cutting.
The genus Paenibacillus is a new group of bacilli separated from the genus Bacillus, and most of species have been isolated from soil. In the present study, we collected 450 spore-forming bacilli from the roots of winter crops, such as barley, wheat, onion, green onion, and Chinese cabbage, which were cultivated in the southern part of Korea. Among these 450 isolates, 104 Paenibacillus-like isolates were selected, based on their colony shape, odor, color, and endospore morphology, and 41 isolates were then finally identified as Paenibacillus spp. by 16S rDNA sequencing. Among the 41 Paenibacillus isolates, 23 were classified as P. polymyxa, a type species of the genus Paenibacillus, based on comparison of the 16S rDNA sequences with those of 32 type strains of the genus Paenibacillus from the GenBank database. Thirty-five isolates among the 41 Paenibacillus isolates exhibited antagonistic activity towards plant fungal and bacterial pathogens, whereas 24 isolates had a significant growth-enhancing effect on cucumber seedlings, when applied to the seeds. An assessment of the root-colonization capacity under gnotobiotic conditions revealed that all 41 isolates were able to colonize cucumber roots without any significant difference. Twenty-one of the Paenibacillus isolates were shown to contain the nifH gene, which is an indicator of $N_{2}$ fixation. However, the other 20 isolates, including the reference strain E681, did not incorporate the nifH gene. To investigate the diversity of the isolates, a BOX-PCR was performed, and the resulting electrophoresis patterns allowed the 41 Paenibacillus isolates to be divided into three groups (Groups A, B, and C). One group included Paenibacillus strains isolated mainly from barley or wheat, whereas the other two groups contained strains isolated from diverse plant samples. Accordingly, the present results showed that the Paenibacillus isolates collected from the rhizosphere of winter crops were diverse in their biological and genetic characteristics, and they are good candidates for further application studies.
To investigate the effects of barley straw on microflora, acetylene reducing activity, enzyme activity and sugar in relation to nitrogen fixation in submerged soil. The obtained results were summarized as follow: Of the nitrogen fixing microorganisms, the number of Azotobactor tended to increase with the application of barley straw as the rice grew. The number of Clostridia were increased at the tillering stage of plant and decreased thereafter, and that of Blue-green algae tended to increase at the heading stage and to decrease thereafter. On the other hand, the number of Blue-green algae tended to increase by the application of barley straw. Acetylene reducing activity was decreased in the heading stage and increased in the harvesting stage. There was no difference of acetylene reducing activity between the application of barley straw and control. In submerged soil treated with barley straw, enzyme activity of ${\beta}$-glucosidase was increaded significantly but that of phosphatase was not entirely affected. Of the change of enzyme activity, the observation of ${\beta}$-glucosidase was increased at the heading stage and decreased thereafter, and the activity of phosphatase tended to decrease in the submerged soil when rice plants were not cultured and to increase in the submerged soil when rice plants were cultured. Protease tended to increase in the heading stage and increase in the tillering stage and heading stage with the application of barley straw. The change of sugar was decreased and hexose was increased in the tillering stage with the application of barley straw.
Rhizobium japonicum isolates from all around Korea could be classified into two groups, i.e., acid producing fast-growers with 2.4 hour mean generation time and non-acid producing slow-growers in yeast extract-mannitol medium with 13.1 hour mean generation time. Tested fast-growers were higher in 6-phosphogluconate dehydrogenase activity than slow-growers were and used sucrose as carbon source whereas slow-growers did not. Fast-grower R4, R257, R278, showed tolerance even in 0.5M NaCl or above and the growth of all the strains tested were inhibited at below pH 4.5. Relative symbiotic activities of nitrogen fixation for these isolated with Glycine max cv. Jangyeobkong (commercial soybean cultivar mostly cultivated in Korea) ranged 0.1 to 2.0 comparing to that of R. japonicum L-259 (NRRL), without regard to their growth rate.
A full understanding of the interdependence of leaf nitrate (($No_3$ ̄) metabolism and symbiotic nitrogen($N_2$) fixation in legume crops is needed to help maximize the use of both N sources as well as to improve forage quality through the inhibition of leaf nitrate accumulation. The present work examines the effects of added nitrate, the level of which are 0,2,4,8 and 12mM, on the nodule formation and leaf nitrate utilization and on the possibility of inducing nitrate-toxicity to livestocks in two alfalfa varieties, ' Vernal ' of grazing type and ' Victoria ' of hay type. Higher level of exogeneous nitrate resulted in the increased above-ground dry weight. Nodulation was inhibited severely when more than 8mM NO$_3$ ̄ was supplied to alfalfa plants, and leaf nitrate reductase reached a maximunm at 4mM nitrate supply. The $V_{max}$of nitrate reductase in leaves of Vernal was similar to that of Victoria, whereas the $K_m$ of Vernal was higher than that of Victoria. High accumulation of leaf nitrate, $4{\times}10^{-5}$ g/g leaf fresh weight, was shown at 12mM nitrate supply, which was thought to be not enough to induce nitrate-toxicity to livestocks.icity to livestocks.

This study was conducted to investigate both plant growth-promoting and plant disease- controlling activities of bacterial strains isolated from soil. All the isolated strains were able to grow at various temperatures. All the strains, except ANG40, showed antagonistic effects against various phytopathogenic fungi. This antagonism can be ascribed to the production of siderophores and antibiotic substances. In addition, all the strains showed abilities such as nitrogen fixation, phosphate solubilization, and siderophore production. These results suggest that nitrogen, phosphorus, and iron can be converted into forms that can be easily absorbed by the plants for their growth. Analysis of the growth-promoting properties revealed that ANG51 produced 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase and indole-3-acetic acid (IAA) both of which are related to ethylene production. In contrast, the other strains were found to have only IAA-producing ability. Therefore, this study suggests that Pseudomonas geniculata ANG3, Exiguobacterium acetylicum ANG40, and Burkholderia stabilis ANG51, which were selected through analysis of comparative advantages for both plant growth promotion and disease-controlling activity, may be used as biological agents.
As the amount of pesticides consumption increases in agriculture, the side effects of pesticides on soil microorganisms have become an essential part in safety evaluation of pesticide for continued soil fertility. In order to establish the method of safety assessment of pesticides in Korea, a series of tests were carried out. Among the paddy pesticides Fujione (Isoprothiolane, fungicide), Ortran (Acephate, insecticide), and Machete (Butachlor, herbicide) were chosen and the effects of above three pesticides on Korean paddy soil microorganisms were studied. The measurements of pesticide effects on the cycling of carbon ($CO_2$ production), nitrogen fixation ($C_2H_2$ reduction), nitrification, and dehydrogenase activity were carried out. These measurements were complemented by evaluation of pesticide effects on viable microbial numbers. Although Fujione reduced the fungi numbers and inhibited nitrogen cycling activities somewhat, no significant adverse effects were show by any of the tested pesticides in our study.
An acid tolerant R. meliloti "YA03" which was characterized through both acidified tube and pot soil experiment, was examined on its inoculation effect to alfalfa[Medicago sativa (L.)]cv. Vernal on hilly acid soil. It was conducted for 2 years to evaluate inoculation residual effect and there were two-fertilizer factors of nitrogen(0.8kg/10a) and lime(0,162kg/10a)as urea and $Ca(OH)_2$, respectively. The results obtained were summarized as follows : 1. In the 1st-year experiment, YA03 inoculated without nitrogen and lime application showed higher symbiotic effectiveness than check strains, YA026 and Rm2011 in nodule mass, $N_2$-fixing activity, and dry matter yield, but with N 8kg/10a application lower effectiveness than Rm2011. 2. In the 2nd-year experiment, Alfalfa dry matter yield by YA03 inoculation with N application(without lime) was 74% higher than that(276 kg/10a) in the 1st year, and the value showed 166%, 20%, and 21% increments as compared with noninoculated control(180kg/10a). YA026(399kg/10a), and Rm2011(397kg/10a), respectively. 3. In the 2nd-year experiment of N applicated plote(without lime), especially, YA03 showed higher shoot nitrogen(7.7kg N/10a/Yr.) fixed and soil rhizobial population($1{\times}10^4\;cells/g.\;soil$) than check strains. 4. On the basis of lime application, application effect of three major fertilizers with YA03 inoculation on alfalfa yield and soil rhizobial population was in the order of $N+P_2O_5+K_2O>P_2O_5+K_2O>P_2O_5+K_2O$. 5. On the whole, alfalfa yield by acid tolerant R. meliloti YA03 inoculation on hilly acid soil was enhanced with nitrogen application, and besides it appeared 44% higher with lime than without lime.

Bacillus velezensis is an important plant growth-promoting rhizobacterium with immense potential in agriculture development. In the present study, Bacillus velezensis Lle-9 was isolated from the bulbs of Lilium leucanthum. The isolated strain showed antifungal activities against plant pathogens like Botryosphaeria dothidea, Fusarium oxysporum, Botrytis cinerea and Fusarium fujikuroi. The highest percentage of growth inhibition i.e., 68.56±2.35% was observed against Fusarium oxysporum followed by 63.12 ± 2.83%, 61.67 ± 3.39% and 55.82 ± 2.76% against Botrytis cinerea, Botryosphaeria dothidea, and Fusarium fujikuroi, respectively. The ethyl acetate fraction revealed a number of bioactive compounds and several were identified as antimicrobial agents such as diketopiperazines, cyclo-peptides, linear peptides, latrunculin A, 5α-hydroxy-6-ketocholesterol, (R)-S-lactoylglutathione, triamterene, rubiadin, moxifloxacin, 9-hydroxy-5Z,7E,11Z,14Z-eicosatetraenoic acid, D-erythro-C18-Sphingosine, citrinin, and 2-arachidonoyllysophosphatidylcholine. The presence of these antimicrobial compounds in the bacterial culture might have contributed to the antifungal activities of the isolated B. velezensis Lle-9. The strain showed plant growth-promoting traits such as production of organic acids, ACC deaminase, indole-3-acetic acid (IAA), siderophores, and nitrogen fixation and phosphate solubilization. IAA production was accelerated with application of exogenous tryptophan concentrations in the medium. Further, the lily plants upon inoculation with Lle-9 exhibited improved vegetative growth, more flowering shoots and longer roots than control plants under greenhouse condition. The isolated B. velezensis strain Lle-9 possessed broad-spectrum antifungal activities and multiple plant growth-promoting traits and thus may play an important role in promoting sustainable agriculture. This strain could be developed and applied in field experiments in order to promote plant growth and control disease pathogens.
