The Crystal Structure of One Natural Compound Cyclo-(1,10-Docandiamino-11,20-Docanedioic) Amide (1,12-Diazacyclodocosane-2,11-Dione)

  • 1,12-diazacyclodocosane-2,11-dione was first isolated from a plant Phyllanthus niruri Linn. Its structure has been determined by means of spectroscopy methods and X-ray crystallography. Two peptide groups in the big ring (lactam) are the main factors influencing intermolecular contacts. The hydrogen-bond interaction of these hydrophilic groups is observed in the crystal structure. Meanwhile, C-H···O hydrogen bonds in molecules contribute to the formation of the whole crystal. These two kinds of hydrogen-bond form six- member rings among molecules. This compound crystallizes in the triclinic space group P-1 with a= 9.588(1) $\AA$, b= $9.850(1)\AA$, c = $11.810(1)\AA$, $\alpha=$ 68.18(1)$^{\circ}C$ , $\beta=$ 84.98(1), $\gamma$ = 86.03(1)$^{\circ}C$ , V = $1030.66(17)\AA3$ , Z = 2. A disorder of five-member carbon chain in the whole ring is observed in the title compound. The bond angle 105.8(4) is determined for a extreme configuration C(14)-C(15)-C(16), and 117.7(10) for another extreme configuration C(14')-C(15')-C(16'). In this crystal, two molecules are tied each other by short intermolecular hydrogen bonds, the oxygen atom being tied by hydrogen bond to nitrogen atom of another two molecules. The NMR and IR spectral data coincides to the structure of the compound.

Duplex Surface Treatments of Plasma Nitrocarburizing and Plasma Oxidation of SKD 11 Steel

  • Plasma nitrocarburizing and plasma oxidizing treatments were performed to improve the wear and corrosion resistance of SKD 11 steel. Plasma nitrocarburizing was conducted for 12 h at $520^{\circ}C$ in the nitrogen, hydrogen and methane atmosphere to produce the $\varepsilon-Fe_{2-3}(N,C)$ phase. It was found that the compound layer produced by plasma nitrocarburising was predominantly composed of $\varepsilon-phase$, with a small proportion of $\gamma'-Fe_4(N,C)$ phase. The thickness of the compound layer was about $5{\mu}m$ and the diffusion layer was about $150{\mu}m$ in thickness, respectively. Plasma post oxidation was performed on the nitrocarburized samples with various oxygen/hydrogen ratio at constant temperature of $500^{\circ}C$ for 1 hour. The very thin magnetite($Fe_3O_4$) layer $1-2{\mu}m$ in thickness on top of the compound layer was obtained by plasma post oxidation. It was confirmed that the corrosion characteristics of the nitrocarburized compound layer could be further improved by the application of the superficial magnetite layer.

세균으로부터 항진균성 물질의 생산 및 특성 (Production and Chracteristics oil Antifungal agents from Bacteria)

  • 토양으로부터 분리한 균주 중 항진균 활성이 우수한 균주를 선발하여 B-1이라 명명하였다. B-1의 항균성 물질 생산에 대한 배양조건을 검토하기 위해 최적배지, 탄소원, 질소원, pH, 온도별로의 영향을 조사하였다. PDB 배지에서 항균력이 우수하였으며 탄소원$.$질소원에는 별다른 영향이 없었다. pH와 온도에서는 각각 pH 5.0, 28$^{\circ}C$에서 항균력이 우수하였다. 항균 활성은 진균에서 강하게 나타났으며 일부 세균에서도 항균력을 나타내었다. 항진균성 물질은 산, 알칼리 처리 후 잔존 활성은 70∼80%로 나타났으며 열처리시 완전히 실활되어 열에 대해 상당히 불안정하다는 것을 확인하였다. 항균성 물질위 용매 전용성을 위해 TLC를 통하여 분석하였으며 plate assy를 실시하여 항균성물질을 확인하였다. 항진균성 물질의 대량생산을 위해 기본배지인 PDB를 직접 실험실 내에서 제조하였으며 시판의 PDB와 비교하였을때 항균성 물질 생산면에서나 가격면에서 더 나은 결과를 보였다.

  • This study was carried out in order to analyzed the changes in nitrogen containing taste compounds of three different types of traditional Korean soy sauce with varing ripening period, and to investigates correlation between sensory characteristics and taste compounds contents via contents assay and sensory evaluation of soy sauce samples. Total nitrogen, ammonia type nitrogen and amino type nitrogen contents showed the highest value in Kyupjang. 17 kinds of free amino acid was detected in Chungjangs and 16 kinds of free amino acid was detected in Kyupjang. Nucleotides and their related compounds detected were hypoxanthin, xanthin, IMP, AMP, Inosine, ADP. Free amino acid and nucleotides and their related compounds contents were highest in Kyupjang. Nitrogen related compounds content of high concentration soy sauce ripened over 150 days increased similarly with Kyupjang. In the sensory evaluation of soy sauce taste, Chungjang samples acquired the highest score in the offensive taste test while Kyupjang marked highest score for sweet taste, nutty taste, taste preference. High concentration soy sauce ripend over 150 days yielded the result similar to that of Kyupjang. Sweet taste showed positive correlation with nitrogen compounds. The materials that showed positive correlation with nutty and traditional soy sauce taste and taste preference were nitrogen compound, IMP, AMP. Except for tryptophan, all free amino acid showed positive correlation with nutty and traditional soy sauce taste. Particulary, taste preference correlated to lysine, aspartic acid and glutamic acid.

BCF WIN을 이용한 Acetanilide의 생물농축특성 평가 (Estimated Bioaccumulation properties of Acetanilide using BCFWIN)

  • Acetanilide is a High Production Volume Chemical, which is produced about 2,300 tons/year in Korea as of 1998 survey. Most is used as an intermediate for synthesis of pharmaceuticals and dyes. The chemical is one of seven chemicals, which are under the frame of OECD SIDS program sponsored by National Institute of Environmental Research of Korea. Regarding the information on the environmental fate. bioconcentration is one of important factor to estimate the environmental tranfer. However, measurement of bioconcentration needs high expense and time. For this reason, OECD recommends to use BCFWIN model to estimate bioconcentration of organic chemicals, BCFWIN estimates the bioconcentration factor (BCF) of an organic compound using the log octanol-water partition coefficient (Kow) of the compound. Structures are entered into BCFWIN through SMITES (Simplified Molecular Input Line Entry System) notations. The BCFWIN method classifies a compound as either ionic or non-ionic. ionic compounds include carboxylic acids, sulfonic acids and salts of sulfonic acids, and charged nitrogen compounds (nitrogen with a + 5 valence such as quaternary ammonium compounds). All other compounds are classified as non-ionic. In this study, bioaccumulation of acetanilide was estimated using BCFWIN model based on SMIIES notation, chemical name data and partition coefficient as one of environmental fate/distribution of the chemical elements.

Increased Production of Ginsenoside Compound K by Optimizing the Feeding of American Ginseng Extract during Fermentation by Aspergillus tubingensis

  • The ginsenoside compound K (C-K) is widely used in traditional medicines, nutritional supplements, and cosmetics owing to its diverse pharmacological activities. Although many studies on C-K production have been conducted, fermentation is reported to produce C-K with low concentration and productivity. In the present study, addition of an inducer and optimization of the carbon and nitrogen sources in the medium were performed using response surface methodology to increase the C-K production via fermentation by Aspergillus tubingensis, a generally recognized as safe fungus. The optimized inducer and carbon and nitrogen sources were 2 g/l rice straw, 10 g/l sucrose, and 10 g/l soy protein concentrate, respectively, and they resulted in a 3.1-fold increase in the concentration and productivity of C-K (0.22 g/l and 1.52 mg/l/h, respectively) compared to those used before optimization without inducer (0.071 g/l and 0.49 mg/l/h, respectively). The feeding methods of American ginseng extract (AGE), including feeding timing, feeding concentration, and feeding frequency, were also optimized. Under the optimized conditions, A. tubingensis produced 3.96 mM (2.47 g/l) C-K at 144 h by feeding two times with 8 g/l AGE at 48 and 60 h, with a productivity of 17.1 mg/l/h. The concentration and productivity of C-K after optimization of feeding methods were 11-fold higher than those before the optimization (0.22 g/l and 1.52 mg/l/h, respectively). Thus, the optimization for the feeding methods of ginseng extract is an efficient strategy to increase C-K production. To our knowledge, this is the highest reported C-K concentration and productivity via fermentation reported so far.

IQF 굴(Crassostrea gigas) 복합엑스분을 이용한 굴 소스의 가열향기 성분 (Volatile Flavor Constituents of Cooked Oyster Sauce Prepared from Individually Quick-frozen Oyster Crassostrea gigas Extract)

  • The pacific oyster Crassostrea gigas has a desirable taste and flavor that differs from those of other fish and shellfish. In order to develop a high value-added product from individually quick-frozen oyster extract (IQFOE), we prepared an oyster sauce from IQFOE and characterized its volatile compounds using vacuum simultaneous steam distillationsolvent extraction / gas chromatography / mass spectrometry. The moisture, crude protein, crude ash, salinity, pH and volatile basic nitrogen contents of the oyster sauce were 60.6%, 8.2%, 9.2%, 9.3%, 5.7 and 21.0 mg/100 g, respectively. Seventy-six volatile compounds were detected in the cooked odor of the oyster sauce. These volatile compounds included 14 esters, including ethyl acetate, 13 nitrogen- containing compounds, including 2,4,6-trimethyl pyridine, 13 acids, including hexadecanoic acid, 12 alcohols, including ethyl alcohol and 6-methyl heptanol, 6 alkanes, 5 aldehydes, including benzaldehyde, 5 ketones, including 1-(2-furanyl)-ethanone, 4 furans, including 2-furancarboxaldehyde and 2-furanmethanol, 3 aromatic compounds, including d-limonene, and 1 miscellaneous compound. Esters, acids and nitrogen-containing compounds, and alcohols were the most abundant compounds in the odor of the cooked oyster sauce, with some aldehydes, ketones, and furans.

Nitrogen Translocation and Dry Matter Accumulation of Direct Seeded Rice in No Tillage Rice-Vetch Cropping

  • Uptake, assimilation and translocation of nitrogen and dry matter assimilation and translocation in ten rice cultivars were observed in no-till direct-sown rice-vetch cropping system. There was a large degree of variation in N-uptake, grain yield, nitrogen translocation efficiency and dry matter assimilation and translocation in tested rice cultivars. Forty kg N/ha base, as compound fertilizer (21-17-21% of N-P-K) three weeks after sowing and 30 kg N/ha top-dressed at panicle initiation stage as in the form of (NH$_4$)$_2$$CO_2$ was applied. ‘Newbounet’, ‘Daesanbyeo’, and ‘Hwayeongbyeo’ showed higher translocation efficiency. The contribution of pre-heading dry matter assimilates to grain ranged from 33% to 99% of dry grain weight. Dry matter of ‘Calrose 76’ was lower than Newbounet but N content was higher in Calrose 76 than Newbonnet. By maturity, N content in vegetative parts declined considerably more than dry matter, vegetative and reproductive parts, N translocation efficiency, and N harvest index. Nitrogen translocation efficiency was greater in ‘Nonganbyeo’, Daesanbyeo, and Newbounet. Grain N concentration was positively correlated with N concentration or N content of the vegetative parts at heading in Nonganbyeo, ‘Dasanbyeo’, ‘Dongjinbyeo’, and Newbonnet. These results indicated that the greater amount of dry matter and N accumulated before heading stage, the higher translocation rates of dry matter to grain and the greater net losses at maturity.

Protoberberine의 고급지방산 유도체합성 및 활성연구(V) (Synthesis of Protoberberine Derivatives and Their Biological Activities)

  • Irradiation of phenolbetaine in a stream of nitrogen produced 8,14-cycioberbine[1]. Compound[1] was treated with 10% HCl solution to give the 8-hydroxycycloberbine[2] in 67.7% yield. Subsequently addition of ethylchloroformate to the compound[2] gave rise to the 8-hydroxy-7-ethylcarboxy-9, 10-dimethoxy-2, 3-methylenelioxy-13-oxo-norochotensane[3] in 78% yield. Treatment of the compound[3] with bis-(2-chloroethyl)amine then lead to the 7-bis(2-chloroethyl)carbamyl-norochoteneare[4]. On the other hand the compound[5], which is the 8-methoxynorochotensane, was derived when compound[1] was treated with methanol in a few drops of BF. Treatment of the compound[6], and the compound[7], 7-bis(2-chloroethyl)-carbanyl-8-methoxy-norocheyensane, was then synthesized by reaction of the compound[6] with bis(2-chloroethyl) amine. In the other synthetic pathway when compound[5] was treated with $POCl_3$ in dried benzene, 13-chloro-6-ene-norochetensane[8] with 42% yield was formed. Finally the 13-bis-(2-chloroethyl) amino-8-methoxy-norochotensane[9] was produced when we treated the compound[8] with bis-(2-chloroethyl) amine. In another pathway, reaction between phenolbetaine which is the precursor of the compound[1] and benzoylchloride in dried chloroform gave us the 5,6,7 trihydro-2, 3-methylene-dioxy-9-chloromethyl-10, 11-dimethoxyphenylisoquinoline-8-benzoate[10] in 73% yield. The results of biological activities for these compounds are also presented in Table I and II.

배에서 추출한 Phenolic Compound가 Streptozotocin으로 유발된 고혈당 생쥐에 미치는 영향 (Effects of Pear Phenolic Compound on the STZ-treated Mice for Induction of Diabetes)

  • 배에서 추출한 phenolic compound가 streptozotocin(STZ)응 투여하고 고혈당을 유발시킨 생쥐에 미치는 영향을 밝히고자, 생쥐의 혈당, 혈중 creatinine, BUN의 변화 및 insulin-면역 조직화학적 검색과 췌장섬 $\beta$-세포의 전자현미경관찰을 통한 미세구조 변화를 관찰하였다. 실험군은 정상적인 동물 사료를 식이토록 한 대조군, 사료에 phenolic compound(PA군, 13 mg/g/kg/day; PB군,90 mg/kg/day)를 혼합하여 6주 동안 섭식하게 한 실험군으로 구분하였다. 대조군의 혈당 농도는 4주부터 높게 나타났으며, PA군의 혈당은 대조군에 비하여 유의성 (p<0.05)있게 감소하였으며, 특히 PB군에서는 4주부터 감소하기 시작하여 6주까지 유의성 (p<0.05)있게 감소하였다. BUN과creatinine의 농도는 대조군에 비하여 실험군에서 다소 감소하였으나 유의성은 없었다. STZ을 투여한 대조군의 췌장섬은 대부분 파괴되어 insulin-면역조직화학적 반응을 보인 세포들이 거의 관찰되지 않았으나, PB군에서는 다수의 췌장섬이 관찰될 뿐만 아니라 인슐린-면역조직화학 반응이 양성으로 관찰되었다. 전자현미경관찰 결과 대조군의 $\beta$-세포에서는 인슐린 함유 과립들이 소수 관찰되었으나 PB군에서는 이들 과립들이 다수 관찰되었다. 이상의 결과로 보아 phenolic compound를 섭식한 실험군 생쥐는 STZ에 의해서 손상된 췌장섬이 회복 또는 재생되어 $\beta$-세포의 인슐린 분비가 복원되 어 가고 있다고 사료되었다.
