지유 발효추출물의 항세균 및 항산화 활성 (Antibacterial and Antioxidant activities of Bio-fermented Sanguisorbae Radix Extract)

  • Objectives : The objective of this research was to investigate the antibacterial and antioxidant activities of bio-fermented Sanguisorbae Radix extract.Methods : The Sanguisorbae Radix extract was fermented byStreptococcus thermophilus,and their products was tested for antibacterial activity against pathogenic microorganisms namely,Bacillus cereus,Bacillus subtilis,Staphylococcus aureus,Escherichia coli,Vibrio parahaemolyticusandSalmonella typhimuriumby paper disc diffusion method and the antioxidant activities of extract was evaluated by five different assays as electron donating ability(EDA), superoxide dismutase(SOD)-like activity, polyphenol, flavonoid contents and nitrite scavenging ability.Results : The bio-fermented Sanguisorbae Radix extract was safe from heat. Antibacterial activity of fermented Sanguisorbae Radix extract appeared relatively highly againstBacillus cereusandStaphylococcus aureusand didn't show any difference. EDA in comparison to Vitamin C showed over 90% activity at about the same time of Sanguisorbae Radix extract expressed highly. SOD activity showed 15% in fermentation before and after. The nitrite scavenging ability of Sanguisorbae Radix extract before and after fermentation showed higher numerical value over 70% in pH 2.5 than that of butylated hydroxytoluene(BHT). But SOD activity, EDA and nitrite scavenging ability were not different between the Sanguisorbae Radix extract before and after fermentation. Total polyphenol content expressed over about 20 mg/g, and that of the Sanguisorbae Radix extracts was increased than that of the fermented Sanguisorbae Radix extracts.Conclusions : The results suggest the usefulness of developing functional materials using antioxidant active Sanguisorbae Radix extract was fermented bySalmonella typhimuriumwith high polyphenol contents and nitrite scavenging ability.

Immune Cells Activity, Cytotoxicity and Nitrite Scavenging Activity of Extracts from Several Resource Plants

  • This study was conducted to explore the immune activity, anticancer activity and nitrile scavenging activities of methanol extracts from the various organs of four Korean resource plants. The immune responses from both human T and B cell line was significantly enhanced in the cell growth compared to control while the cell growth was influenced at a certain period of culture. The results revealed that the cell growth of both human T and B cell was altered in a time dependent manner. Among tested several resource plants, the flower extract of E. japonicum demonstrated a pronounced cytotoxicity against HCT-116 cell with an IC50 value $132.08{\mu}g\;ml-1$. The flower extract from Corylopsis coreana had a promising scavenging activity against pH 1.2 compared to other species. Taken together, the studied resource plants have influenced significantly in response to immunity and also have the potential cytotoxicity and nitric scavenging activities. However, the species E. japonicum exhibited the pronounced activities from several resource plants. The result from this investigation suggests that the extracts of studied resource plant could be an addition to basic medicine for some diseases.

여주 품종별 폴리페놀, 플라보노이드 함량과 라디칼 소거활성 및 아질산염 소거능 (Different of Total Phenolics and Flavonoids, Radical Scavenging Activities and Nitrite Scavenging Effects of Momordica charantia L. According to Cultivars)

  • This study was carried out in order to determine the biological activities such as antioxidant activities in nine cultivars of Momordica charantia L. (Korea, China, Japan and Philippine native cultivars). Their antioxidant activities were measured using DPPH free radical scavenging, ABTS cation radical scavenging, and Nitrite scavenging ability. The highest total polyphenol content (16.82 ${\mu}g/m{\ell}$) measured in the native Korea cultivar, and this value was 4.0 ${\mu}g/m{\ell}$ higher than that of the 'Verde Beuhas' cultivar (12.82 ${\mu}g/m{\ell}$). The 'Peacock' cultivar had the highest total flavonoid amount which was 4.38 ${\mu}g/m{\ell}$. The free radical scavenging activity using DPPH method was the highest in the native China cultivar ($RC_{50}$ = 102.6 ${\mu}g/m{\ell}$). ABTS cation radical scavenging activity according to cultivars was significantly higher in 'Peacock'. Nitrite scavenging ability showed the most remarkable effect at the pH 1.2, exhibited to 81.5${\sim}$86.9% by addition of ethanol extract 1${\mu}g/m{\ell}$ from Momordica charantia L. These results suggest that Momordica charantia L. had the potent biological activities, and that their activities exhibited differently depending on cultivars.

Investigation of n-Butanol and Ethyl Acetate Extracts from Thermal Treatment Yam (Dioscorea batatas DECNE.) for their Antioxidant Activities

  • n-Butanol and ethyl acetate extracts of thermal treatment yam (Dioscorea batatas $D_{ECNE.}$) belonging to the family Dioscoreaceae were measured for their radical scavenging activity and lipid peroxidation inhibition ability. In this study, ethyl acetate extract showed the most potent antioxidant activity evaluated by ferrous ion chelating activity and NO radical scavenging activity. Nevertheless, n-butanol extract was more effective in inhibiting linoleic acid peroxidation. A significant difference between n-butanol extract and ethyl acetate extract in nitrite scavenging activity ${\beta}$-carotene bleaching assays could not be found. Also, the results of this study showed that thermal treatment yam could be used as easily accessible source of natural antioxidants and as a possible food supplement.

미역과 매생이의 총 페놀함량 및 항산화성 (Total Phenolic Contents and Antioxidant Activities of Undaria pinnatifida and Capsosiphon fulvescens)

  • This study compared the total phenolic contents and antioxidant activities of two seaweed cultivars, Undaria pinnatifida (UP) and Capsosiphon fulvescens (CF), subjected to different drying methods. UP and CF were dried under two different conditions: vacuum drying (VD) at $20^{\circ}C$ and hot-air drying (HD) at $60^{\circ}C$. After drying, the total phenolic content, DPPH, and nitrite scavenging activities of the water extracts were determined. Total phenolic contents were 101.94 mg/100 g for UP and 171.35 mg/100 g for CF upon VD-20, and these values were significantly decreased to 67.59 mg/100 g for UP and 141.48 mg/100 g for CF upon HD-60. UP upon VD-20 and HD-60 had 46.17% and 35.20% DPPH radical scavenging activity, whereas CF upon VD-20 and HD-60 scavenged 57.73% and 35.22%, respectively. UP upon VD-20 and HD-60 had 40.36% and 40.01% nitrite scavenging activity at pH 1.2, whereas CF upon VD-20 and HD-60 scavenged 72.35% and 55.24%, respectively. Nitrite scavenging activities of UP and CF were reduced at pH 3.0.

Antioxidant and Xanthine Oxidase Inhibition Activities of Cynomorium songaricum Extracts

  • In this study, we evaluated the antioxidant activities and xanthine oxidase inhibition effects of water and ethanol extracts of Cynomorium songaricum. The ethanol extract of C. songaricum (EE) contained more phenolic and flavonoid compounds than the water extract (WE). The antioxidant activities of the extracts were increased as the concentration of the extract increased. The WE has better effectiveness than the EE for DPPH free radical scavenging activity and nitrite scavenging ability. The nitrite scavenging abilities of WE were 90.02% ($EC_{50}$ 653.15 ${\mu}g$/mL) at conditions of pH 1.2 and 2,000 ${\mu}g$/mL, and 84.34% ($EC_{50}$ 817.17 ${\mu}g$/mL) at pH 3.0. The EE has more effective SOD-like activity and XO inhibition than WE. The SOD-like activity of EE was 81.47% at a concentration of 2,000 ${\mu}g$/mL, $EC_{50}$ was 951.70 ${\mu}g$/mL. The xanthine oxidase inhibition of the EE, with an $EC_{50}$of 112.47 ${\mu}g$/mL, is greater than that of ascorbic acid, which was 192.50 ${\mu}g$/mL (p<0.05). These results suggest that the C. songaricum is a potentially useful antioxidant source for the development of nutraceuticals and medicines.

쑥갓 스테롤배당체의 아질산염소거작용 및 SOD 유사활성 (Nitrite Scavenging Ability and SOD-like Activity of a Sterol Glucoside form Chrysanthemum coronarium L. var. spatiosum)

  • 쑥갓의 항산화 활성을 알아보기 위해 쑥갓 총 메탄올추출물의 아질산염 소거작용과 SOD 유사활성을 측정하였다. 아질산염 소거작용과 SOD 유사활성을 가지고 있는 총 메탄올 추출물로부터 클로로포름, n-부탄올 및 물 분획물을 제조하여 각각의 활성을 측정하였다. pH가 1.5일 때의 클로로포름 분획물의 $IC_{50}$값은 39ppm으로 가장 우수한 아질산염 소거작용을 보였으며, vitamin C와 chlorogenic acid의 $IC_{50}$값이 15ppm과 36ppm일 때 클로로포름 분획물은 양성대조군인 chlorogenic acid와 동등한 소거작용을 보였다. 아질산염 소거작용에 대하여 유의성 있게 높은 활성을 나타낸 클로로포름 분획물을 silica gel 컬럼크로마토그래피와 sephadex LH-20 컬럼크로마토그래피를 이용하여 분획의 활성성분인 compound I을 분리하였다. 분리된 compound를 $^{1}H-NMR$$^{13}C-NMR$ spectral data를 통하여 구조를 동정한 결과 compound I은 ${\beta}-sitosterol-O-{\beta}-D-glucoside$임을 확인하였다. Compound I과 그 aglycone인 ${\beta}-sitosterol$의 아질산염 소거작용과 SOD 유사활성은 다소 차이가 있었다. 양성대조군으로 사용한 vitamin C와 chlorogenic acid의 $IC_{50}$값이 pH 1.5에서 각각 15ppm과 36ppm일때 compound I의 $IC_{50}$값이 335ppm, ${\beta}-sitosterol$$IC_{50}$값이 41ppm으로 가수분해된 aglycone이 아질산염 소거작용이 훨씬 높았다. SOD 유사활성의 경우 vitamin C와 chlorogenic acid의 $EC_{50}$ 값은 38ppm, 449ppm으로 이와 비교시 ${\beta}-sitosterol$과 배당체의 $EC_{50}$ 값은 1,291ppm과 2,000ppm 이상으로 나타났다.

청미래 덩굴잎(Smilax china) 추출물의 전자공여능, 아질산염 소거능 및 항균효과 (Electron Donating Abilities, Nitrite Scavenging Effects and Antimicrobial Activities of Smilax china Leaf)

  • 청미래덩굴잎의 ethanol 추출물로부터 얻은 용매별 각 분획물의 전자공여능, 아질산염소거능 그리고 항균활성을 측정한 결과 다음과 같다. 각 용매별 분획의 수율은 극성이 높은 수용성 분획과 methanol분획이 각각 63.73%와 26.40%로 높게 나타났다. 전자공여능은 ethyl acetate 분획에서 가장 높게 나타났으며 10분간 반응시켰을 때 약 81%정도 달했다. 반면 chloroform분획에서는 약 26%로 가장 낮았다. 아질산 염소거능은 ethyl acetate와 butanol분획에서 가장 높게 나타났다. 항균활성은 모든 분획의 2.5%와 5.0%의 농도에서 Gram(+)균인 Listeria monocytogenes와 Staphylococcus aureus에 대해 항균작용이 나타났으나 2.5%농도에서는 Gram (-)균인 Escherichia coli O157:H7과 Salmonella Typhimurium에 대해서는 전혀 활성이 없었으며 5.0%농도에서는 ethanol 추출물, ethyl acetate와 물분획에서 아주 낮은 활성이 측정되었다.

대나무 에탄올추출물의 항산화 효과 및 아질산염 소거작용 (Antioxidative Activity and Nitrite Scavenging Ability of Ethanol Extract from Phyllostachys bambusoides)

  • 본 연구는 왕대나무 줄기의 생리황성 및 기능성을 검토하기 위해서 왕대나무 줄기를 에탄올로 추출하여, 추출물의 항산화 효과와 아질산염 소거능을 측정하였다. 그 결과, 왕대나무 줄기 에탄올 추출물의 전자공여능($RC_{50}$)은 $116.75\;{\mu}g/mL$로 나타났고 SOD 유사활성은 추출물(0.92 mg/mL)을 첨가하였을 때 43.88%로 가장 높게 관찰되었으며, linoleic acid에 대한 항산화력은 추출물($50\;{\mu}g/mL$)을 첨가하여 TBA값을 측정한 결과 배양시간 4일과 6일 경과 후 각각 74.76%, 54.48% 감소율을 보임으로써 효과적인 항산화 효과를 나타내었다. 아질산염 소거능은 추출물(0.2 mg/mL)을 첨가하였을 때 pH 1.2 조건에서 43.02%로 가장 높게 나타났으며 pH가 증가할수록 감소하였다. 이상의 결과를 종합하면 왕대나무 줄기의 에탄올 추출물은 항산화 효과와 아질산염 소거능이 있는 물질을 함유한 것으로 판단된다. 추후, 왕대나무의 항산화 효과와 아질산염 소거능을 나타내는 원인물질에 대한 연구가 진행되어야 할 것이다.

Some Physiological Activities Observed from Ethanol Extracts of Germinated Specialty Rough Rice

  • This study analyzed the some of the physiological activities of 70% ethanol extracts of specialty rough rice ('Goami2', 'Keunnunbyeo', and 'Heugkwangbyeo' rough rice cultivars) before and after germination. The physiological activities of the extracts were analyzed for their inhibition of linoleic acid peroxidation, hydrogen peroxide radical scavenging activity, superoxide dismutase-like activity, xanthine oxidase inhibitory activity, and nitrite scavenging effects. The most active rough rice cultivar was 'Heugkwangbyeo' and germinated 'Heugkwangbyeo' rough rice was more effective than ungerminated rough rice, having higher inhibition activity of linoleic acid peroxidation (1.2-fold), hydrogen peroxide radical scavenging activity (1.4-fold), superoxide dismutase-like activity (1.2-fold), and xanthine oxidase inhibitory activity (1.1-fold). The nitrite scavenging effect was increased after germination (2.1-fold) and decreased as pH increased. According to the results obtained in this study, 70% ethanol extracts from germinated 'Heugkwangbyeo' rough rice can be used for industrial applications such as the functional food materials.
