Antioxidative Activities of the Ethyl Acetate Fraction from Heated Onion (Allium cepa)

  • Heated onion juice was partitioned using the solvents hexane, chloroform, ethyl acetate, and butanol. The ethyl acetate fraction showed the strongest scavenging effect on the ABTS radical. The antioxidant activities of the ethyl acetate fraction from raw and heated onion (120, 130, and $140^{\circ}C$) were evaluated using radical scavenging assays. Radical and nitrite scavenging activities were higher in heated onion than raw onion, and the higher the temperature of heat treatment, the greater the radical and nitrite scavenging activities. Heated onion ($140^{\circ}C$, 2 hr) was more effective than raw onion, having higher DPPH radical scavenging (5.7-fold), hydroxyl radical scavenging (6.4-fold), superoxide radical scavenging (2.3-fold), hydrogen peroxide scavenging (11.8-fold), and nitrite scavenging (4.3-fold) activities. Onion increased its physiologically active materials after heating, and in this regard, heated onion can be used as biological material for the manufacture of health foods and supplements.

Nitrite Scavenging and Superoxide Dismutase-like Activities of Herbs, Spices and Curries (향신료 및 Curry의 Nitrite 분해와 Superoxide Dismutase 유사활성)

  • The superoxide dismutase(SOD)-like activities for 26 kinds of herbs and spices and 10 kinds of instant curry products were determined by measuring their abilites to reduce nitroblue tetrazolium. All samples showed the SOD-like activities. Rosemary, cassia, tarragon, allspice, oregano, bay leaves, basil, marjoram, thyme and star anise had higher activities than $10^5\;unit/g$ and clove had highest activity of $232,143{\pm}19.989\;unit/g$. The SOD-like activities for 10 kinds of instant curry products were in the range of $400{\sim}700\;unit/g$ when measured after heat treatment at $100^{\circ}C$ for 10 min. The water extracts of spices, herbs and curries were obtained by heat treatments of $25^{\circ}C$ for 60 min or $100^{\circ}C$ for 10 min, and their nitrite scavenging activity was measured at different pH conditions(1.2, 4.2 or 6.0). The nitrite scavenging activities were higher at acidic pH. However, the effects were not different from two heat treatments. The water extracts from cassia, bay leaves, allspices, oregano, staranise, rosemary, clove and tarragan had high nitrite scavenging activity(>90%) when they were measured at pH 1.2, and those from clove was highest $(97.58{\pm}0.88%)$. The pure curry used as raw materials for instant curry products had the nitrite scavenging activity in the range of $50{\sim}60%$ at pH 1.2 and the activity was not changed during the aging period$(0{\sim}12weeks)$. The ten brands of instant curry products had the nitrite scavenging activities of $12{\sim}28%$ at pH 1.2

Antioxidative Activities and Nitrite-scavenging Abilities of Some Phenolic Compounds (일부 페놀성 화합물의 항산화효과 및 아질산염 소거능)

  • This study investigated the antioxidant and synergistic effects and nitrite scavenging ability of some phenolic compounds(catechin, rutin, quercetin and naringin), The electron donating abilities of naringin, quercetin, rutin and catechin were 6.7%, 92.8%, 87.6% and 92.21%, respectively, The antioxidant activities in O/W emulsion substrates were in order of rutin > quercetin > catechin > naringin. The antioxidant effect of rutin was stranger than that of BHT or ${\alpha}$-tocopherol. ${\alpha}$-tocopherol showed synergistic effect with catechin and quercetin, but ascorbic acid not showed effect. The nitrite scavenging abilities of catechin, quercetin, rutin and naringin were 99.9%, 98.6%, 25.5% and 0.2%, respectively. The nitrite scavenging abilities of quercetin and actechin were very potent as compared with those of BHT and ascorbic acid.

Evaluation of Antioxidant and Nitrite Scavenging Activity of Seaweed Extracts (해조류 추출물의 항산화 및 nitrite 소거활성 평가)

  • In the course of study for the development of functional food ingredients from seaweeds having useful biological activities, the in-vitro antioxidant and nitrite scavenging activities of the methanol extracts prepared from 35 different seaweeds (17 phaeophyta, 11 rhodophyta and 7 chlorophyta) were determined. At $500\;{\mu}g/ml$ concentration of the methanol extracts, Ecklonia cava, Ecklonia stolonifera, Eisenia bicyclis (Kjellman) Setchell, Ishige foliacea, I. okamurai, Sargassum confusum, S. fulvellum, S. yamade and Zostera marina showed 60% more DPPH and ABTS scavenging activity. The exceptions were found in I. okamurai and Z. marina, which showed 45% and 53% ABTS scavenging activity, respectively. The correlation coefficient between DPPH and ABTS scavenging activities was 0.855, suggesting that the 9 different seaweeds extracts could scavenge anion and cation radicals concurrently. In reducing power, only E.cava, E.stolonifera and E.bicyclis showed above 0.88 ($Abs_{700}$). In a while, in nitrite scavenging activity at $100\;{\mu}g/ml$ concentration of the methanol extracts, 19 different seaweeds extracts including I. foliacea, I. okamurai, S. confusum, S. fulvellum, and S. yamade showed 60% more activity. Calculation of $IC_{50}s$ of DPPH, ABTS and nitrite scavenging activities of 9 different seaweeds extracts further showed that I. foliacea and S. yamade, besides E.cava, E.stolonifera and E.bicyclis, have strong antioxidant and nitrite scavenging activity. These results suggest that the selected 9 different seaweeds could be developed as functional food ingredients and I. foliacea and S. yamade have potential as novel natural sources of antioxidant and nitrite scavenger.

Analysis of Biological Activities of Medicinal Mushrooms (약용버섯의 생리활성 분석)

  • This study was carried out to observe the antioxidative activity, fibrinolytic activity, nitrite savenging ability and adenosine content of three medicinal mushrooms. Among water and ethanol extract of each mushroom, water extract of Inonotus obliquus (Czech) showed the highest antioxidative activity $(57.1{\mu}g/ml)$, nitrite scavenging ability (52.04%), fibrinolytic activity (86.8%) and adenosin content $(94.3{\mu}g/g)$, whereas nitrite scavenging ability of ethanol extract of Phellinus linteus showed higher than that of water extract. Apart from the above statements, water was effective than ethanol as extracting solvent in general. These results suggest that Inonotus obliquus (Czech) showed higher activities such as antioxidative activity, nitrite scavenging ability, fibrinolytic activity and adenosin content and water was effective as solvent except for nitrite scavenging ability.

Different of Total Phenolics and Flavonoids, Radical Scavenging Activities and Nitrite Scavenging Effects of Momordica charantia L. According to Cultivars (여주 품종별 폴리페놀, 플라보노이드 함량과 라디칼 소거활성 및 아질산염 소거능)

  • This study was carried out in order to determine the biological activities such as antioxidant activities in nine cultivars of Momordica charantia L. (Korea, China, Japan and Philippine native cultivars). Their antioxidant activities were measured using DPPH free radical scavenging, ABTS cation radical scavenging, and Nitrite scavenging ability. The highest total polyphenol content (16.82 ${\mu}g/m{\ell}$) measured in the native Korea cultivar, and this value was 4.0 ${\mu}g/m{\ell}$ higher than that of the 'Verde Beuhas' cultivar (12.82 ${\mu}g/m{\ell}$). The 'Peacock' cultivar had the highest total flavonoid amount which was 4.38 ${\mu}g/m{\ell}$. The free radical scavenging activity using DPPH method was the highest in the native China cultivar ($RC_{50}$ = 102.6 ${\mu}g/m{\ell}$). ABTS cation radical scavenging activity according to cultivars was significantly higher in 'Peacock'. Nitrite scavenging ability showed the most remarkable effect at the pH 1.2, exhibited to 81.5${\sim}$86.9% by addition of ethanol extract 1${\mu}g/m{\ell}$ from Momordica charantia L. These results suggest that Momordica charantia L. had the potent biological activities, and that their activities exhibited differently depending on cultivars.

The Antioxidative, Antimicrobial and Nitrite Scavenging Effects of Schizandra chinensis RUPRECHT(Omija) Seed (오미자종자의 항산화성, 항균성, 아질산염소거능)

  • This study was carried out to examine the antioxidative, antimicrobial and nitrite scavenging effects of various solvent extracts and different solvent fractions from Schizandra chinensis RUPRECHT(Omija) seed. The results were as follows; The antioxidative activities using lard, soybean oil and linoleic acid were the highest in methanol, ethanol and methanol extract from omija seed, respectively. The free radical scavenging activity using DPPH method was the strongest in acetate fraction of methanol extract from seed. The methanol extract from omija seed had the strongest antimicrobial activities to L. plantarum, B. subtilis, E. coli and P. citrinum, while ethyl acetate extract had the strongest against S. aureus and S. typhimurium. The buthanol fraction from methanol extract had the highest antimicrobial activities, followed by B. subtilis, L. plantarum, E. coli, S. aureus and S. typhimurium. The nitrite scavenging ability was pH dependent, highest at pH 1.2 and lowest at pH 6.0. The buthanol fraction of methanol extract from omija seed had more effective nitrite scavenging ability than other fractions of extracts.

Total Phenolic Contents and Antioxidant Activities of Undaria pinnatifida and Capsosiphon fulvescens (미역과 매생이의 총 페놀함량 및 항산화성)

  • This study compared the total phenolic contents and antioxidant activities of two seaweed cultivars, Undaria pinnatifida (UP) and Capsosiphon fulvescens (CF), subjected to different drying methods. UP and CF were dried under two different conditions: vacuum drying (VD) at $20^{\circ}C$ and hot-air drying (HD) at $60^{\circ}C$. After drying, the total phenolic content, DPPH, and nitrite scavenging activities of the water extracts were determined. Total phenolic contents were 101.94 mg/100 g for UP and 171.35 mg/100 g for CF upon VD-20, and these values were significantly decreased to 67.59 mg/100 g for UP and 141.48 mg/100 g for CF upon HD-60. UP upon VD-20 and HD-60 had 46.17% and 35.20% DPPH radical scavenging activity, whereas CF upon VD-20 and HD-60 scavenged 57.73% and 35.22%, respectively. UP upon VD-20 and HD-60 had 40.36% and 40.01% nitrite scavenging activity at pH 1.2, whereas CF upon VD-20 and HD-60 scavenged 72.35% and 55.24%, respectively. Nitrite scavenging activities of UP and CF were reduced at pH 3.0.

Antimicrobial Effect, Antioxidant and Tyrosinase Inhibitory Activity of the Extract from Different Parts of Phytolacca americana L.

  • This study was conducted to clarify the antimicrobial effect, antioxidant and tyrosinase inhibitory activities of the biological composition having the Phytolacca americana, and to enhance the natural materials utilization of foods and cosmetics. The antimicrobial activities of the different parts of P. americana were evaluated using the agar diffusion test. The antimicrobial activity of P. americana was relatively high in Malassezia furfur known as a skin fungi and Vibrio parahaemolyticus compared to Escherichia coli and Staphy-lococcus epidermidis. However, the antimicrobial activity in Vibrio parahaemolyticus did not show at all parts of P. americana. Both the DPPH radical scavenging activity and ABTS radical scavenging activity have been increased with the higher concentration of methanol extract. In particular, leaf extract of P. americana exhibited the highest activity both ABTS radical scavenging activity and DPPH radical scavenging activity. The nitrite scavenging activity was decreased when the pH was changed from pH 1.2 to pH 6.0. The highest nitrite scavenging activity was exhibited from the methanol extract of fruit, followed by root, stem, and leaf at pH 1.2. However, the nitrite scavenging activity at pH of 6.0 was not almost detected. All plant parts of P. americana showed tyrosinase inhibitory activity. The highest activity was found in the stem, and followed by root, leaf, and fruit in order. These tyrosinase inhibitory activity was progressively increased in a concentration-dependent manner. In this experiment on the methanol extracts of different organ from P. americana, we confirmed that the extract of P. americana showed potent tyrosinase inhibitory activity. Taken together, we conjectured that the P. americana had the potent biological activities, therefore this plant having various functional components could be a good material for development into source of natural food additives and cosmetics.

Nitrite Scavenging Effect of Methaol Fraction Obtained from Green Yellow Vegetable Juices (녹즙추출물의 아질산염 소거능에 대한 연구)

  • The objectives of this study were to investigate biological activities such as nitrite scavenging effect of water and methanol fractions obtained from each green juice(Angelica keiskei, kale, carrot) under different pH conditions, and antioxidant vitamin contents in green juice made of green yellow vegetables. The result shows that nitrite scavenging effect of each green juice was 14.0~91.2% in model system. The methanol fraction provided higher effect than the water fraction. The nitrite degradation was the highest at pH 1.2. Nitrite scavenging effect of methanol fraction obtained from kale juice was 91.2% at pH 1.2 which was the highest among tested samples in this experiments. Consequently, extracts of kale juice were shown to provide the greatest functional properties among the vegetables tested. Contents of vitamine E and C in the extracts of kale were higher than the others.
