  • 최근 수년간 polarimetry, Raman spectroscopy, near infrared (NIR) absorption spectroscopy, NIR scattering, optoacoustics 등의 방법을 통하여 비침습적으로 Glucose의 농도를 측정하려는 연구가 많이 시도되었다. 일반적으로 이들 방법은 sensitivity 와 signal-to-noise ratio가 매우 낮고 복잡한 알고리즘이 요구되어져 glucose 농도 측정에 한계가 있음이 드러났다. 본 연구에서는 polarization sensitive low coherence interferometer (PS-LCI) 기법을 이용하여 농도에 따른 stokes parameters를 측정함으로써 비침습적으로 glucose를 측정하는 것이 가능한지 알아보는데 그 목적이 있다. 근적외분광분석법과 Micro-Kjeldahl 법 간의 맥주보리 종실의 단백질함량 분석 비교 (Comparisons between Micro-Kjeldahl and Near Infrared Reflectance Spectroscopy for Protein Content Analysis of Malting Barley Grain)

  • 맥주보리 품질에서 중요한 성분인 단백질함량을 NIRS를 이용하여 신원하고 정확하게 분석하기 위해 최적의 검양식 작성에 관한 일련의 시험을 실시한 결과는 다음과 같다. 1. Micro-Kjeldahl법에 의해 분석한 단백질함량을 이용하여 작성된 NIRS의 검양식중 2095 /1941/ 2282/ 2086nm 파장으로 구성된 검양식에서 결정계수($R^2$)가 0.95로서 가장 높았다. 2. NIRS의 2095/1941/2282/2086nm 파장으로 구성된 검양식으로 '92년도에 생산된 18품종을 분석한 결과 SDD가 0.47, SEP가 0.43, r이 0.95로서 매우 우수하였으며 평균 단백질함량도 Micro-Kjeldahl법의 10.25%와 동일하였다. 3. NIRS의 2095/1941/2282/2086nm 파장으로 구성된 검양식으로 '93년에 생산된 미지의 시료 31품종을 Micro-Kjeldahl법과 비교분석한 결과 SDD가 0.69, SEP가 0.67, r(simple correlation)이 0.91이었다. 4. 본 시험에서 작성된 NIRS의 검양식을 이용할 경우 Micro-Kjeldahl 분석치와 r값이 0.91로서 고도의 유의성이 인정되었으며 bias값을 보정해 주면 단백질함량의 평균값이 Micro-Kjeldahl법과 매우 근접된 값을 얻을 수 있어서 맥주보리 육종의 초기세대에서 단기간 다량의 계통을 분석할 수 있을 것으로 기대되었다.

FAST QUANTITATIVE AND QUALITATIVE ANALYSIS OF PHARMACEUTICAL TABLETS BY NIR

  • The implementation of NIR and chemometrics in the Pharmaceutical industries is still in strong progress, both regarding qualitative and quantitative applications and beneficial results are seen. Looking at the development so far, NIR will change the pharmaceutical industry even more in the future. This presentation will address the experiences and progress achieved regarding the application and implementation of quantitative methods for determination of content uniformity and assay of tablets with less than 10% w/w of active, using Near Infrared transmittance spectroscopy in combination with PLS. Also qualitative methods for identification of the same tablets by Near Infrared reflectance spectroscopy will be discussed. Four commercial tablet strengths are formulated and produced from two different compositions by direct compression. Three different strengths are dose proportional, i.e. fixed concentration by varying in size. The aim was to replace the conventional primary methods for analysing content uniformity, assay and identification by NIR. Studies were performed on comparing transmittance versus reflectance spectroscopy for both applications on the dose proportional tablets. The model for determination of content uniformity and assay was developed to cover both coated and uncoated tablets, whereas the qualitative model was developed to identify coated tablets only. The impact of the tablet formulation, tablet size and coating, resulted in individual models far each composition The best calibration was achieved using diffuse reflectance for the identification purposes and diffuse transmittance for the quantitative determination of the active content within the tablets. As NIR in combination with other techniques opens up the possibility of total quality management within the production, the transfer of the above-mentioned models from a laboratory based approach to an at-line approach at H.Lundbeck will be addressed too.

생물학적 질소제거 공정에서 용존산소변화에 따른 미생물의 군집변화 (Changes of Microbial Community Depending on Different Dissolved Oxygen in Biological Nitrogen Removal Process)

  • 본 연구에서는 PCR-DGGE 기법을 이용하여 DO 농도에 따른 동시 질산화.탈질 반응조 내 미생물 군집 변화 양상을 규명하고자 하였다. DO 농도의 변화에 따른 eubacteria의 군집 변화 해석 실험에서, DO 농도를 2와 1 mg/L로 운전한 반응조 내의 band profile은 거의 유사하게 관찰되었으며 5종의 우점화 미생물(Uncultured Bacterium 3종, Bacillus sp. 1종, Uncultured Bacteroidetes sp. 1종)을 포함한 16종의 미생물을 동정할 수 있었다. 그리고 DO 농도 0.5 mg/L로 운전한 반응조 내의 DGGE 결과 7종의 우점화 미생물(Uncultured Bacterium 5종, Zoogloea sp. 2종)을 포함한 12종의 미생물을 동정할 수 있었으며, DO 농도 0.1 mg/L로 운전한 반응조의 경우 3종의 우점화 미생물(Uncultured Bacterium 1종, Zoogloea sp. 2종)을 포함한 11종의 미생물을 동정할 수 있었다. 반응조 내 DO 농도의 변화에 따른 $\beta$-AOB($\beta$-Ammonia Oxidizing Bacteria)의 군집 변화 해석 실험 결과, 하나의 band를 관찰할 수 있었다. 이 band는 Uncultured Nitrosomonas sp. done DNB Y20와 97%의 유사도를 갖는 것으로 나타났으며 2, 1, 그리고 0.5 mg/L의 DO 농도에서 추출한 sample에서는 선명하게 관측되었으나, 0.1 mg/L DO 농도에서 추출한 sample에서는 선명도가 현저히 감소하였다. 이는 NH$_4{^+}$-N의 질산화 양상과 상관관계가 있음을 보였다. 반응조 내 DO 농도에 따른 탈질 bacteria의 군집 변화 해석실험 결과, 다섯 개의 band(nirS를 함유하는 Uncultured organism 미생물 3종, nirK를 함유하는 Uncultured bacterium 미생물 2종)가 관측되었으며 관측된 band 중 한 band는 DO 농도가 낮아질수록 선명도가 현저히 증가하는 것을 확인할 수 있었다. 이 band에 해당하는 미생물은 86%의 유사도를 가진 Uncultured organism clone eS1 cd1 nirS gene, partial cds로, 본 연구의 탈질 반응에 직접적으로 관여 하는 미생물로 사료된다.

  • A portable near infrared (NIR) system which was newly integrated by our lab has been used to develop a non-invasive blood glucose monitoring. The portable NIR system includes a tungsten halogen lamp, a photo diode array type-InGaAs detector, and specialized reflectance fiber optic probes. The shape of probes is composed of two parts. one for illumination into sample and the other for receiving the radiation from sample. Three kinds of probes with different distance between illumination and receiving part. such as 0.03. 0.1. and 0.5mm, were investigated for optimization. (omitted)

Determination of Water Content in Ethanol by Miniaturized Near-Infrared (NIR) System

  • The miniaturized NIR (Near-infrared) spectrometer has been utilized for the determination of water content (1-19% range) in ethanol that is the most popular organic solvent in pharmaceutical industries. It has many potential capabilities that can replace the conventional analyzers especially for the on-line measurement since it is compact, versatile and cost-effective. By using two dimensional (2D) correlation spectroscopy, it was preliminarily investigated to find any unforeseen spectral distortion among the spectra collected from the miniaturized spectrometer. The 2D study revealed that the spectral variation clearly followed the variation of water concentration without any spectral distortion or abnormality. PLS (Partial Least Squares) was employed to build the calibration model and the resulting prediction performance was acceptable and stable over several days. Even though the miniaturized NIR system was evaluated to fairly simple chemical matrix, the overall study demonstrates the sufficient feasibility for diverse practical and industrial applications.

Nondestructive Internal Defects Evaluation for Pear Using NIR/VIS Transmittance Spectroscopy

  • Internal defects such as browning of the flesh and blackening and rot of the ovary of pear can be easily developed because of the inadequate environmental conditions during the storage and distribution of fruit. The quality assurance system for the agricultural product is to be settled in Korea. All defected agricultural products should be excluded prior to the distribution to enhance the commercial values. However, early stage on-line defect detection of agricultural product is very difficult and even more difficult in a case of the internal defects. The goal of this research is to develop a system that can detect and classify internal defects of agricultural produce on-line using VIS/NIR transmittance spectroscopy. And Shingo pear, which is one of the famous species of Korean pear, was used for the experiment. Soft independence modeling of class analogy (SIMCA) algorithm was employed to analyze the transmittance spectroscopic data qualitatively. On-line classification system was constructed and classification model was developed and validated. As a result, the correct classification rate (CCR) using the developed classification model was 96.1 %.

  • An integrated grader which measures soluble solid content, color and weight of fresh apples was developed by NAMRI. The prototype grader consists of the near infrared spectroscopy and machine vision system. Image processing system and an algorithm to evaluate color were developed to speed up the color evaluation of apples. To avoid the light glare and specular reflection, an half-spherical illumination chamber was designed and fabricated to detect the color images of spherical-shaped apples more precisely. A color revision model based on neural network was developed. Near-infrared(NIR) spectroscopy system using NIR reflectance method developed by Lee et al(1998) of NAMRI was used to evaluate soluble solid content. In order to observe the performance of the grader, tests were conducted on conditions that there are 3 classes in weight sorting, 4 classes in combination of color and soluble solid content, and thus 12 classes in combined sorting. The average accuracy in weight, color and soluble solid content is more than about 90 % with the capacity of 3 fruits per second.

경피약물전달을 위한 아세트아미노펜 각인 기능성 전분 기반 바이오 소재 제조 및 방출 특성 (Preparation and Release Properties of Acetaminophen Imprinted Functional Starch based Biomaterials for Transdermal Drug Delivery)

  • 본 연구에서는 mung bean starch (MBS), polyvinyl alcohol (PVA), sodium benzoate (S), glycerol (GL), melanin (MEL)을 이용하여 광열 효과가 있는 기능성 acetaminophen (AP) 각인 MBS 기반 바이오 소재를 제조하고 약물 방출 특성을 조사하였다. 제조된 AP 각인 바이오 소재의 물리화학적 특성은 FE-SEM과 FT-IR을 통해 분석하였다. 또한, NIR (near infrared) laser (1.5 W/cm2) 조사에 따른 기능성 바이오 소재의 광열 효과 및 AP 방출 특성을 조사하였다. 바이오 소재에 NIR laser를 조사하였을 때, MEL이 첨가 바이오 소재는 첨가하지 않은 바이오 소재보다 2배 이상 높은 온도상승을 보였다. 표준 버퍼 용액과 인공 피부를 사용하여 기능성 AP 각인 바이오 소재의 AP 방출 특성 조사결과, NIR laser를 조사하였을 때, MEL 첨가 바이오 소재는 첨가하지 않은 바이오 소재보다 AP 방출율이 1.2배 높은 것을 확인하였다. 이 결과로부터, 기능성 바이오 소재는 급성 진통 치료를 위한 바이오 소재로 활용될 수 있을 것으로 판단된다.
