Laying performance of two pure-line Korean native ducks at different house types (토종오리 순종 2계통의 사육형태에 따른 산란능력)

  • This work was conducted to evaluate the laying performance of Korean Native Ducks (KND) Pure-Line (PL). A total of 400 female pullets were hatched from PL to keep in National Institute of Animal Science, Korea. Twenty weeks old PL female ducks were divided into 4 groups (5 replications/group, 20 head/replications). Four groups were compared in a $2{\times}2$ factorial arrangement with strain (White strains, WW; Celadon strains, CC) and house type (flat house, cage house). On first egg laying day, CC strain laid eggs earlier compared to WW strain (P<0.01), but there was no significant difference between flat and cage house. Egg weight (EW) and body weight (BW) at first egg laying day have no difference among strains and houses. BW of CC strain was higher than WW strain at the age of 30 week (P<0.01). Weekly feed intake (FI) have no difference between flat and cage house, but FI of WW strain was high compared to CC strain during 20~30 weeks. There was no significant difference on EW between CC and WW strains, but EW was low at flat house compared to cage house (P<0.05). Weekly egg productions of WW strain and flat house were lower than CC strain and cage house, respectively (P<0.01). Finally, These results can provide the information that needed to introducing new strains.

Forklift Weight Measurement System using Anchor Bolt Type Strain Gauge Sensor (Anchor 볼트 형태의 Strain Gauge 센서를 이용한 지게차 적재 중량 측정 시스템)

  • The most frequent type of safety-accident in industry is the overturning of forklift. The leading cause of this accident is overload in forklift. Thus, it is needed to measure the weight on board of forklift. The most common method is based on load cell, and this method has the merit of high accuracy. However, high price is the disadvantage of this method. In this paper, we propose the new measurement system of the weight on board of forklift based on the strain gauge sensor, which has the disadvantage of low accuracy. The differentiation of the proposed system is that the shape of the strain gauge sensor customized for anchor bolt of forklift in order to improve the accuracy and durability. In system four strain gauge sensors are inserted into four anchor bolts. The test result shows that 1% error of measurement is obtained in the proposed anchor bolt type strain gauge sensors.

Effects of strain on the optical and magnetic properties of Ce-doped ZnO

  • The magnetic and optical properties of Ce-doped ZnO systems have been widely demonstrated, but the effects of different strains of Ce-doped ZnO systems remain unclear. To solve these problems, this study identified the effects of biaxial strain on the electronic structure, absorption spectrum, and magnetic properties of Ce-doped ZnO systems by using a generalized gradient approximation + U (GGA + U) method with plane wave pseudopotential. Under unstrained conditions, the formation energy decreased, the system became stable, and the doping process became easy with the increase in the distances between two Ce atoms. The band gap of the systems with different strains became narrower than that of undoped ZnO without strain, and the absorption spectra showed a red shift. The band gap narrowed, and the red shift became weak with the increase of compressive strain. By contrast, the band gap widened, and the red shift became significant with the increase of tensile strain. The red shift was significant when the tensile strain was 3%. The systems with -1%, 0%, and 1% strains were ferromagnetic. For the first time, the magnetic moment of the system with -1% strain was found to be the largest, and the system showed the greatest beneficial value for diluted magnetic semiconductors. The systems with -3%, -2%, 2%, and 3% strains were non-magnetic, and they had no value for diluted magnetic semiconductors. The ferromagnetism of the system with -1% strain was mainly caused by the hybrid coupling of Ce-4f, Ce-5d, and O-2p orbits. This finding was consistent with Zener's Ruderman-Kittel-Kasuya-Yosida theory. The results can serve as a reference for the design and preparation of new diluted magnetic semiconductors and optical functional materials.

Breeding of King Oyster Mushroom, Pleurotus eryngii with a High Yield and Earliness of Harvest Trait and Its Sensory Test (큰느타리(새송이)버섯 다수확 속성 품종 육종 및 관능평가)

  • Two strains Pleurotus eryngii 'Aeryni' and 'Na' carrying superior traits of a pileus and a earliness of harvest were selected to improve previously bred strains by single crosses. New hybrid, Aeryni 3 (Aeryni10 ${\times}$ Na5) showed superiority to other hybrids in yield, fruit body shape and days for harvest. The new strain, Aeryni 3 was harvested earlier than Keunneutari No. 2 by 2~3 days, and yielded 110.5 g/bottle (850 mL) which was 108% of that of Keunneutari No. 2. The ratio of diameter of pileus and stipe was 1.8 indicating that new stain will be likely low damage rate of fruit body during a distribution, and that was better than 2.1 of Keunneutari No. 2. A sensory test of taste of the new strain showed that 84.7% of evaluation panels selected "very good" while that of Keunneutari No. 2 was 55.5%. In purchasing intent test, 86.9% of panels will buy the new stain whereas 46.8% will buy Keunneutari No. 2 implicating that the new strain will likely be more marketability than previously bred strain.

Diffusible and Volatile Antifungal Compounds Produced by an Antagonistic Bacillus velezensis G341 against Various Phytopathogenic Fungi

  • The aim of this study was to identify volatile and agardiffusible antifungal metabolites produced by Bacillus sp. G341 with strong antifungal activity against various phytopathogenic fungi. Strain G341 isolated from four-year-old roots of Korean ginseng with rot symptoms was identified as Bacillus velezensis based on 16S rDNA and gyrA sequences. Strain G341 inhibited mycelial growth of all phytopathogenic fungi tested. In vivo experiment results revealed that n-butanol extract of fermentation broth effectively controlled the development of rice sheath blight, tomato gray mold, tomato late blight, wheat leaf rust, barley powdery mildew, and red pepper anthracnose. Two antifungal compounds were isolated from strain G341 and identified as bacillomycin L and fengycin A by MS/MS analysis. Moreover, volatile compounds emitted from strain G341 were found to be able to inhibit mycelial growth of various phytopathogenic fungi. Based on volatile compound profiles of strain G341 obtained through headspace collection and analysis on GC-MS, dimethylsulfoxide, 1-butanol, and 3-hydroxy-2-butanone (acetoin) were identified. Taken together, these results suggest that B. valezensis G341 can be used as a biocontrol agent for various plant diseases caused by phytopathogenic fungi.

Characteristics and breeding of a new variety Pleurotus eryngii, Saegonji (큰느타리버섯 신품종 '새곤지' 육성 및 특성)

  • We bred a new strain of Pleurotus eryngii having few number of fruit body per bottle. It's name is 'Saegonji' and it was bred by mating monokaryotic strain isolated from E12-176 and monokaryotic strain 'aerini No.3' in Mushroom Research Institute, Gyonggi province A.R.E.S. The characteristics of a new strain 'Saegonji' are as follows. The optimum temperature for mycelium growth was from 23 to $26^{\circ}C$ on PDA medium and those for the primodium formation and the growth of fruit body were about $15^{\circ}C$. The period from spawn innoculation to harvesting required around 51 days at $20^{\circ}C$. The number of fruit body per bottle was 12.8 and it was 9.3 less than 'Keunneutari No.2'. The length was 135.8 mm. It was longer than 'Keunneutari No.2', The color of cap was white grey, while that of 'Keunneutari No.2' was grey. The yield was about 159 g per bottle(1100cc) and it was same as Keunneutari No.2.

Use of Bioluminescent Indicator Acinetobacter Bacterium for Screening and Characterization of Active Antimicrobial Agents

  • Because of the need for new antimicrobial substances with novel mechanisms of action, we report here the use of an Acinetobacter reporter system for high-throughput screening of active antimicrobial agents. The bioreporter Acinetobacter strain DF4/PUTK2 carrying luciferase genes luxCDABE was chosen because of its ecological importance and it is widespread in nature. This bioreporter is genetically engineered to emit light constitutively that can be measured in real time by luminometry. Hence, this reporter system was employed to determine the bacteriostatic actions of spent-culture supernatants derived from twelve bacterial isolates. Out of the results, the strongest bioluminescence inhibitory effect of the supernatants was recorded with Bacillus cereus strain BAC (S5). Subsequently, ethyl acetate extracts of extracellular products of strain BAC (S5) were separated by a thin-layer chromatography (TLC). Based on the bioluminescence inhibitory assay, three fractions were found to have antimicrobial activity. One fraction (C) having the strongest antimicrobial activity was further purified using TLC and characterized by IR, $^1H$ NMR, mass spectrometry, SDS-PAGE, and amino acid composition analysis. The results predicted the presence of 2-pyrrolidone-S-carboxylic acid (PCA) and the octadeconic-acid-like fatty acid. Fraction C also demonstrated a broad inhibitory activity on several Gram-negative and Gram-positive bacteria. In conclusion, the Acinetobacter reporter system shows great potential to be a reliable, sensitive, and real-time indicator of the bacteriostatic actions of the antimicrobial agents.

Comparison of Karasek's Job Content Questionnaire and Korea Occupational Stress Scale (Karasek의 Job Content Questionnaire와 Korea Occupational Stress Scale의 비교 연구)

  • According to the report of the WHO, workers have been exposed to much job strain such as job load, responsibility, role, interpersonal conflict etc. In Korea, studies on job strain started to become active from 1990s and now hundreds of studies are actively under going or publishing so that the studies are contributing to development and improvement of job strain. Representative measurement models of job strain are Job Strain Model of Karasek, Job Stress Model of NOISH, Korea Occupational Stress Scale, JSQ(Job Stress Questionnaire), K-OSI(Korea Version of Occupational Stress Inventory) etc. (Lee Kwan-Suk, 2012 ; KOSHA, 2003). Among them, Job Strain Model of Karasek had been loved by many researchers of job strain before Korea Occupational Stress Scale was developed. Job Strain Model of Karasek had been fitted to Korean style and then, used to analyze job strain of Korean people so that this Scale highly contributed to seeking relationship with cardiovascular disease, musculoskeletal disease caused by job, smoking, drug, alcohol poisoning, and pulse(Lee Kwan-Suk, 2012). But as this Model was studied and developed based on foreign culture and life pattern, a model fit to Korea was developed to measure job strain for Korean people, which is Korea Occupational Stress Scale now most frequently used in measuring job strain. Accordingly, after this study made questionnaire survey about same population using the two methods used most frequently in measuring job strain, the study investigated what features appeared, what correlations appear between two models, and comparatively analyzed characteristics each independent and dependent variable. Based on this, the study aimed to exactly express job strain of Korean people. The subjects of the study were a population of 233, and Karasek's Questionnaire and KOSS's Questionnaire were surveyed at the same time. The results were analyzed by statistical program to obtain significant difference between two models. Four particular groups were divided with Job Strain Model of Karasek and the four particular groups were measured with Korea Occupational Stress Scale. And job strain come from combination of two models was measured, with which new comparative analysis method was suggested.

In Situ Spectroscopy in Condensed Matter Physics

  • Recently, many state-of-art spectroscopy techniques are used to unravel the mysteries of condensed matters. And numerous heterostructures have provided a new avenue to search for new emergent phenomena. Especially, near the interface, various forms of symmetry-breaking can appear, which induces many novel phenomena. Although these intriguing phenomena can be emerged at the interface, by using conventional measurement techniques, the experimental investigations have been limited due to the buried nature of interface. One of the ways to overcome this limitation is in situ investigation of the layer-by-layer evolution of the electronic structure with increasing of the thickness. Namely, with very thin layer, we can measure the electronic structure strongly affected by the interface effect, but with thick layer, the bulk property becomes strong. Angle-resolved photoemission spectroscopy (ARPES) is powerful tool to directly obtain electronic structure, and it is very surface sensitive. Thus, the layer-by-layer evolution of the electronic structure in oxide heterostructure can be investigated by using in situ ARPES. LaNiO3 (LNO) heterostructures have recently attracted much attention due to theoretical predictions for many intriguing quantum phenomena. The theories suggest that, by tuning external parameters such as misfit strain and dimensionality in LNO heterostructure, the latent orders, which is absent in bulk, including charge disproportionation, spin-density-wave order and Mott insulator, could be emerged in LNO heterostructure. Here, we performed in situ ARPES studies on LNO films with varying the misfit strain and thickness. (1) By using LaAlO3 (-1.3%), NdGaO3 (+0.3%), and SrTiO3 (+1.7%) substrates, we could obtain LNO films under compressive strain, nearly strain-free, and tensile strain, respectively. As strain state changes from compressive to tensile, the Ni eg bands are rearranged and cross the Fermi level, which induces a change of Fermi surface (FS) topology. Additionally, two different FS superstructures are observed depending on strain states, which are attributed to signatures of latent charge and spin orderings in LNO films. (2) We also deposited LNO ultrathin films under tensile strain with thickness between 1 and 10 unit-cells. We found that the Fermi surface nesting effect becomes strong in two-dimensions and significantly enhances spin-density-wave order. The further details are discussed more in presentation.
