Protoplast Fusion between Zygosaccharomyces rouxii and Saccharomyces cerevisiae Selected from Soy Sauce Mash (간장덧에서 선별한 Zygosaccharomyces rouxii와 Saccharomyces cerevisiae와의 Protoplast 융합)

  • Protoplast susion between Zygosaccharmoyces rouxii M-12 and Saccharimyces cerevusuare M-43 were investigated for breeding of a new brewing yeat strain for soy sauce. Auxotrophic mutants of Zygosaccharomyces rouxii ZRM-83 ($Met^-,\;Thr^-$) and Saccharomyces cerevisiae SCM-46 ($Lys^-,\;Arg^-$) were selected by treatment of 3.0% ethylmethane sulfonate and nutritional complementary method. Protoplast of both strains were more effective by treatment of 0.05mg/ml zymolase 20T for 60min. Fusion effeciency was much higher by treatment of 30% PEG 6,000 for 30min and fusion frequencies were $10^{-4}{\sim}10^{-5}$. These fusants originated from two protoplasts had properties of big cell size and much DNA content.

  • In recent years, the study of metal matrix composite has increased specially, aluminum alloy, research and development are briskly progress to find new metal matrix composite. this study is following the this purpose; This study is used metal matrix composite that was produced by matrix, AC4CH. and reinforcement $A_{18}B_4O_{33}$ metal matrix composite to add $Al_2O_3,\;TiO_2$ for strengthen of binding together among the Whisker. Each Metal matrix composite is produced using the squeeze casting method. Fracture tounghness test was in accordance with the provisions of ASTM E399; Specimen was produced half-size CT specimen W=25mm, B=12.5mm, Cross head speed 0.05mm/min in room temperature. The plane strain fracture toughness $K_{IC}$ is $8.7MPa-m^{0.5}$ for $Al_{18}B_4O_{33}$/AC4CH., $9.28MPa-m^{0.5}$ for $Al_{18}B_4O_{33}$/AC4CH added $TiO_2$. and $Al_2O_3$ but AC4CH alloy was violated the critical stipulated by ASTM standard for a valid measurement of $K_{IC}$. In case of, it was performed $I_{IC}$ test instead of $K_{IC}$ based on ASTM E 1820

  • Many constitutive models for concrete have been developed to predict the nonlinear behavior of concrete members considerably. The constitutive model for reinforcing bar that include the tension stiffening effect due to the bond characteristics between steel bars and concrete is being studied but the bilinear model is generally used. It was found that the buckling of the longitudinal reinforcing bars is controlled the nonlinear behavior of hybrid precast concrete panel, which is being developed for core wall. In this study, the constitutive models that can consider the embedding and buckling effects of reinforcing bar are investigated and a new model combing these constitutive models is proposed. In order to verify the proposed model, the analysis results are compared with experimental results of the concrete wall and hybrid precast concrete panel. The analysis of embedding-effect-only modeling predicted that the deformation increases continually without the decrease in the load carrying capacity. However, the analysis results of proposed model showed good agreement with some experimental results, thus verifying the proposed computational model.

Isolation and Identification of Xylanase Secreting Yeast (Xylanase를 분비하는 효모 균주의 분리 및 성질)

  • Among the new yeast strains which were isolated from soils by incubating in the xylan containing minimal medium at 3$0^{\circ}C$, one strain(XB-33) was finally selected by the results of extracellular xylanase production test. The characteristics of XB-33 was almost consistent with those of the Cryptococcus ater. The formation of xylanase activity was induced by xylan and repressed by xylose or glucose. The xylanase was partially purified from the culture supernatant with DEAE-Sephadex A5O chromatography. The enzyme had a pH optimum for activity at 5.0 and its stability range was pH 5-7. The temperature optimum was at 5$0^{\circ}C$, but the enzyme activity was greatly lost by heating at 7$0^{\circ}C$ for 60 minutes. The hydrolysis products from xylan by crude enzyme detected by TLC, were xylose and n series of higher oligosaccharides. The Km value of xylanase was 20 (mg/ml).

Fermentation Characteristics of Exopolysaccharide-Producing Lactic Acid Bacteria from Sourdough and Assessment of the Isolates for Industrial Potential

  • Lactic acid bacteria (LAB) with antimicrobial activity and high exopolysaccharide (EPS) production ability isolated from sourdough were studied for their fermentation characteristics as potential new starter cultures. The values of pH, titratable acidity, and viable cell counts were $4.06{\pm}0.009-4.50{\pm}0.015,\;0.787{\pm}0.020%-1.172{\pm}0.018%,\;and\;8.78{\pm}0.08-8.98{\pm}0.06$ log CFU/ml, respectively. In order to select probiotics with a high survival rate in the gut, isolates were tested to assess resistance against the artificial gastric acid and bile juice. Viable LAB counts were significantly (p<0.05) affected by the acidity. At pH 2.0, the total declines in the initial bacterial counts were 4.52$\pm$0.07 log for S. thermophilus St-Body-1, >7.98$\pm$0.03 log for E. flavescens DU-10, >7.95$\pm$0.05 log for E. faecium DU-12, and 3.15$\pm$0.06 log for L. amylovorus DU-21. Among the strains, L. amylovorus DU-21 was the only strain that had bile tolerance under simulated gastrointestinal conditions. In order to improve EPS production by L. amylovorus DU-21, the influence of carbon source was studied. When glucose was used as a carbon source, EPS production dramatically increased to 17.19$\pm$0.28 g/l (p<0.05). The maximum cell growth (10.012$\pm$>0.012 log CFU/ml) and EPS production (18.71$\pm$0.19 g/l) were achieved when 15 g/l of glucose was employed as the carbon source.

Identification of 12 radiation-resistant bacterial species in the phylum Proteobacteria new to Korea

  • In 2019, after a comprehensive investigation of indigenous prokaryotic species in Korea, a total of 12 bacterial strains assigned to the phylum Proteobacteria were isolated from soil. With the high 16S rRNA gene sequence similarity (>98.8%) and formation of a robust phylogenetic clade with the closest species, it was determined that each strain belonged to independent, predefined bacterial species. This study identified two species in the family Burkholderiaceae, one species in the family Comamonadaceae, two species in the family Oxalobacteraceae, one species in the family Micrococcaceae, one species in the family Bradyrhizobiaceae, one species in the family Methylobacteriaceae, one species in the family Rhizobiaceae, one species in the family Rhodocyclaceae, and one species in the family Sphingomonadaceae. There is no official report about these 12 species in Korea, so are described as unreported bacterial species in Korea in this study. Gram reaction, basic biochemical characteristics, colony, and cell morphology are also described in the species description section.

Development of diagnostic kit(Test-MT) for the microplate latex agglutination test of toxoplasmosis in animal (Latex 응집반응을 이용한 동물의 톡소플라즈마병 진단용 kit 개발에 관한 연구)

  • The present study was conducted to develop a toxoplasma latex agglutination test antigen(Test-MT) and evaluate the toxoplasma latex agglutination(LA) test using a newly-made "Test-MT kit" by comparing with the Toxo-MT kit(Eiken chemical co, Tokyo). Also, the specifity and sensitivity test were made by comparing with IFA test and IgG-ELISA. Tachyzoite suspensions of Toxoplasma gondii(RH strain) were ultracentrifuged for 30min at $60,000{\times}g(4^{\circ}C)$ and the supernatant was used as a water-lysate antigen. Polystyrene latex particles of $1.0{\mu}m$ in diameter(Polyscience co) were used for the preparation of sensitized latex-antigen supension(Test-MT). The frequency distribution of LA titers in Test-MT showed two peaks at <1:32 and 1:128. The borderline titer for positive test in Test-MT was determined to be 1:64. But the frequency distribution of LA tites in Toxo-MT showed two peaks at <1:16 and 1:64. The positive borderline was determined to be 1:32. Agreement of reactions between Test-MT and Toxo-MT kit by LA test was shown 92.5% in bovine sera and 97.0% in swine sera, respectively. From the results obtained here it was determined that the sensitized latex-antigen, Test-MT kit, for the microtiter agglutination test prepared as same as by the procedure described in the previous paper(Suh and Lee, 1993) was useful as a highly specific, sensitive and stable immunotiteration reagent for serodiagnosis of toxoplasma infection in animal sera.

Isolation, characterization, and phylogenetic position of a new sulfur-oxidizing bacterium

  • A sulfer-oxidizing bacterium was isolated from mine wastewater and characterized. The isolate was gra-negative, rod (0.2 * 1.2-1.5.mu.m), nonmotiloe, catalase positive, and oxidase prositive. The opotimal pH and temperature for growth were 7.0 and 30.deg.C. respectively. The optimum thiosulfate concentration was 70 mM and the maximum growth rate was 0.081 hr. The major ubiquinone contained in the isolate was Q-8. The cellular fatty acid composition was $C_{16 : 0}$, $C_{18 : 1}$, $C_{17cyc}$,and $C_{19cyc}$ as nonpolar fatty acids, and 3-OH C10 : 0 and 3-OH $C_{12 : 0}$ as hydroxylated fatty acids. The isolate was a facultative chemolithoautotroph which can grow autotrophically on sodium thiosulfate and sodium sulfide and which can grow heterotrophically on yeast extract. It can also grow mixotrophically on sodium thiosulfate and yeast extract. Comparison of the 16S rRNA gene sequence of the isolate with that of Thiobacillus species and Paracoccus thiocyanatus revealed that it is closely related to T. caldus which belongs to the .betha.-subclass of the class Proteobacteria. However, the isolated could not grow at extremely low pH (pH 1-3.5). On the basis of the phenotypic, chemotaxonomic and phylogenetic data, the isolate was tentatively named Thiobacillus sp. strain C.ain C.

A Model Study on Deformability of A Transversely Isotropic Rock (평면이방성 암석의 변형특성 모델연구)

  • In the uniaxial compressive test of a single specimen of transversely isotropic rock, its 5 independent elastic constants can not be defined since maximum 4 independent strain measurements are available theoretically. In order to solve this problem, one equation proposed by Saint Venant in 19C and confirmed by Lekhnitskii through the test experiences has been used for long time. Accordign to authors' experiences, however, this equation turned out to give erroneous elastic constants in some cases. Three new equations are suggested and their compatibilities are discussed in this paper. As the results of the analyses of the models, Lekhnitskii's suggested equation is effective for the specimen with the high dip angle whereas it results in the large erred output for that with dip angle less than $25{\sim}30$. It was found that the effectivenesses of three suggested equations and their compatibilities are subject to the dip angle and not to the amounts of elastic constants. Guide map to the selection of the compatible one of those suggested equations is presented as a result of the study.

Paenibacillus elgii SD17 as a Biocontrol Agent Against Soil-borne Turf Diseases

  • Paenibacillus elgii SD17 (KCTC $10016BP^T$=NBRC $100335^T$) was recently reported as a new species. Based on its inhibitory activity to Thanatephorus cucumeris AG1-1, strain SD17 was further evaluated for its potential as a biocontrol agent against soil-borne diseases of turf grasses in Korea. P. elgii SD17 showed a broad spectrum of antimicrobial activity in vitro test and suppressed development of turf grass diseases; Pythium blight caused by Pythium aphanidermatum and brown patch caused by T. cucumeris AG1-1 on creeping bentgrass (Agrostis palustris) in the growth chamber tests. Under a condition for massive culture in a 5,000 L fermenter, P. elgii SD17 reached $6.4{\times}10^8$ spores/ml that resulted in approximately $1.0{\times}10^7$ cfu/g when formulated into a granule formulation (GR) using the whole culture broth instead of water. Using the GR formulation, biocontrol activity of P. elgii SD17 was confirmed. In the growth chamber tests, the GR formulation was effective against brown patch and Pythium blight with similar level of disease severity compared to each of the standard fungicides at the application rates of 10 g/$m^2$ or above. In the field tests, compared to each untreated control, the GR formulation also effectively controlled Pythium blight, brown patch and large patch at all the application rates of 5, 10 and 20 g/$m^2$, respectively, without significant response by the application rates. However its performance was inferior to each of the standard chemical fungicides. Based on these results, we consider this GR formulation of P. elgii SD17 as an effective biocontol agent to suppress Pythium blight, brown patch and large patch of turf grasses in Korea.
