A Stack of Recessive Alleles of Kunitz Trypsin Inhibitor, Lectin, and Stachyose in Soybean (콩에서 쿠니츠트립인히비터, 렉틴 및 스타키오스에 대한 열성 유전자의 집적)

  • Soybean [Glycine max (L.) Merr.] is one of the major food sources of protein, oil, carbohydrates, isoflavones, and other nutrients for both humans and animals. However, soybean seeds contain antinutritional factors, such as lectin protein, Kunitz Trypsin Inhibitor (KTI) protein, and stachyose. The objective of this research was to stack recessive alleles for development a triple recessive genotype, titilelers2rs2, with low KTI protein, lectin protein, and stachyose contents. Three parents (Gaechuck#2, PI200508, and 14G20) were used to develop the breeding population. The presence or absence of the lectin and KTI proteins was detected by western blotting. The stachyose content in mature seeds was determined by HPLC. Agronomic traits, such as plant type, plant height, maturity date, lodging, seed quality, and 100-seed weight, were evaluated for the four $F_3$ plant strains. One $F_4$ plant strain with the desired agronomical traits was selected. One new strain with the triple recessive titilelers2rs2 genotype was developed. The plant height of the new strain was 51 cm and the 100-seed weight was 31.0 g. The new strain had a yellow seed coat and yellow hilum. The stachyose content of the new strain was 3.8 g/kg. One strain developed in this research will be used to produce improved yellow soybean cultivars that are free of lectin and KTI proteins and low in stachyose content.

  • In this paper, the global study trends for material behaviors are investigated regarding the static and dynamic hardenings and final fractures of marine structural steels. In particular, after reviewing all of the papers published at the 4th and 5th ICCGS (International Conference on Collision and Grounding of Ship), the used hardening and fracture properties are summarized, explicitly presenting the material properties. Although some studies have attempted to employ new plasticity and fracture models, it is obvious that most still employed an ideal hardening rule such as perfect plastic or linear hardening and a simple shear fracture criterion with an assumed value of failure strain. HSE (2001) presented pioneering study results regarding the temperature dependency of material strain hardening at various levels of temperature, but did not show strain rate hardening at intermediate or high strain rate ranges. Nemat-Nasser and Guo (2003) carried out fully coupled tests for DH-36 steel: strain hardening, strain rate hardening, and temperature hardening and softening at multiple steps of strain rates and temperatures. The main goal of this paper is to provide the theoretical background for strain and strain rate hardening. In addition, it presents the procedure and methodology needed to derive the material constants for the static hardening constitutive equations of Ludwik, Hollomon, Swift, and Ramberg-Osgood and for the dynamic hardening constitutive equations of power from Cowper-Symonds and Johnson-Cook.

An Evaluation of Probabilistic Strain-Life Curve in Polyacetal (폴리아세탈 소재의 확률론적 변형률-수명선도 평가)

  • In order to evaluate variation of fatigue life of mechanical components including engineering plastics, it is important to estimate probabilistic strain-life curves to accurately define the variation of fatigue characteristics. This paper intends to provide new assessment of P-$\varepsilon$-N (probabilistic strain-life curves) for considering the variation of fatigue characteristics in polyacetal. The fatigue strain controlled tests were conducted under constant 50% humidity and room temperature condition by a universal testing machine at strain ratio, R=0. A practical procedure is introduced to evaluate probabilistic strain-life curves. Three probabilistic distributions were used for generating P-$\varepsilon$-N curves such as normal, 2-parameter and 3-parameter Weibull. In this study, 3-parameter Weibull distribution was found to be most appropriate among assumed distributions when the probability distributions of the fatigue characteristic were examined using chi-square and Kolmogorov-Smirnov test. The more appropriate P-$\varepsilon$-N curves for these materials are generated by the proposed method considering 3-parameter Weibull distribution.

  • Currently, potentiometric method has been employed to measure fuel amount for automobile. However, potentiometer could be malfunctioned or damaged due to the additives or dirt in fuel because a variable electric resistor is immersed in fuel. Therefore, a device employing strain sensor is designed and tested to verify the possibility as an alternative of the conventional fuel sender. Existing fuel tank has irregular cross section. Therefore, buoyancy is not increased linearly as fuel amount is increased. We design a floater that can compensate the irregular cross section of fuel tank and make buoyancy to increase linearly with increasing fuel amount. New fuel sender, comprising of strain gages on circular membrane, the above commented floater and the cover to prevent the disturbance due to sudden acceleration and deceleration, is designed and tested to replace current fuel sender. On the other hand, we are developing strain sensor that has strain gage deposited directly on the circular membrane with cantilever beam.

Characterization of Streptomyces sp. AMLK-135 Producing Anti- MRSA Antibiotics

  • The present research program was conducted to characterize a strain of actinomycetes producing an anti methicillin-resistant Staphylococcus aureus (MRSA) antibiotic. Soil samples were collected from various sites in Korea and a number of actinomycetes were isolated from the soil samples by applying selective agar for actinomycetes. Among over 400 isolates, a strain (AMLK-135) producing anti-MRSA antibiotic against S. aureus TK 784 was selected. According to the morphological and physiological characteristics, the strain AMLK-135 was confirmed to belong to the genus Streptomyces. From the results of species identification with the TAXON program, the strain AMLK-135 was shown to belong to major cluster 5 (Streptomyces exfoliatus), but it had a low simple matching coefficient ($S_{SM}$ SM/) value to member organisms of major cluster 5. Percentage ($\%$) of strain further away of the strain AMLK-135 was low (1.9400) and it was placed further away than the outer-most members in major cluster 5. Therefore, the strain AMLK-135 was identified as a new species of the genus Streptomyces.

High Strain Rate Compression Behavior of EPP Bumper Foams (변형률 속도에 따른 EPP Foam의 대변형 동적 압축 특성에 관한 연구)

  • Bumper is designed to protect the automotive frame without damage at low velocity. Expanded polypropylene (EPP) foam is used in the bumper as an energy absorbing material. In order to exactly predict the energy absorbing performance of the foam material under impact loading condition, it is important to use high strain rate material properties. In this study, a new apparatus for dynamic compression tests was developed to investigate the high strain rate behavior of EPP foams. Three kinds of EPP foams which have different expansion ratios were tested to investigate the quasi-static and dynamic compression behavior. Quasi-static compressions were performed at low strain rates of 0.001/s, 0.1/s and 1/s. The dynamic compressions were carried out at high strain rates of 50/s and 100/s with the developed apparatus. It was observed that the EPP foam has significant strain rate effect as compared to quasi-static behavior.

New Stress-Strain Model for Identifying Plastic Deformation Behavior of Sheet Materials (판재의 소성변형 거동을 동정하기 위한 새로운 응력-변형률 모델)

  • In sheet metal forming numerical analysis, the strain hardening equation has a significant effect on calculation results, especially in the field of spring-back. This study introduces the Kim-Tuan strain hardening model. This model represents sheet material behavior over the entire strain hardening range. The proposed model is compared to other well known strain hardening models using a series of uniaxial tensile tests. These tests are performed to determine the stress-strain relationship for Al6016-T4, DP980, and CP Ti sheets. In addition, the Kim-Tuan model is used to integrate the CP Ti sheet strain hardening equation in ABAQUS analysis to predict spring-back amount in a bending test. These tests highlight the improved accuracy of the proposed equation in the numerical field. Bending tests to evaluate prediction accuracy are also performed and compared with numerical analysis results.

Molecular Characterization of a New Hantaan Virus Howang Strain (한탄바이러스 호왕균주의 M, S 유전자 절편의 염기서열 및 분자생물학적 특성)

  • Hantaan virus Howang strain which isolated from the blood of severe case of Korean hemorrhagic fever is more virulent than HTN 76/118 and showed different RFLP from partial PCR amplifed M genome segment to established Hantaan serotype viruses. We have determined the nucleotide sequence of the M and S genome segments and compared to HTN 76/118. The M and S segment of Howang strain has 3615 and 1696 nucleotides long, respectively. The M segment sequence of Howang strain is one mucleotide shorter than HTN 76/118. The sequence data of Howang strain shows 93.5% homology to HTN 76/118. One long open reading frame, which strats from 41nt. to 3448nt. of the M segment and from 37nt. to 1326nt. of the S segment, exist to on complementary sense of the virus genome. There are no significant difference between HTN 76/118 and Howang strain on hydrophobicity of deduced polypeptides, but has slight difference on secondary structure.

Damage Detection in Floating Structure Using Static Strain Data (정적 변형률을 이용한 플로팅 구조물의 손상탐지)

  • Recently, people's desire for the waterfront space has been increasing, and more people want to spend their leisure time close to the water. This paper proposes a damage detection technique using the static strain for the floating structure. An existing damage index, in which the modal strain energy was utilized to identify possible location of damage, is expanded to apply the static strain. The new damage index is expressed in terms of the static strains of undamaged and damaged structures. After calculating damage index, the possible damage locations in the structure are determined by the pattern recognition technique. The accuracy and feasibility of the proposed method is demonstrated by using experimental strain data from a scale model of floating structure.

Strain-Sensing Characteristics of Multi-Walled Carbon Nanotube Sheet

  • In this paper, the properties of strain sensors made of spin-capable multi-walled carbon nanotubes (MWCNTs) were characterized and their sensing mechanisms analyzed. The key contribution of this paper is a new fabrication technique that introduces a simpler transfer method compared to spin-coating or dispersion CNT. Resistance of the MWCNT sheet strain sensor increased linearly with higher strain. To investigate the effect of CNT concentration on sensitivity, two strain sensors with different layer numbers of MWCNT sheets (one and three layers) were fabricated. According to the results, the sensor with a three-layer sheet showed higher sensitivity than that with one layer. In addition, experiments were conducted to examine the effects of environmental factors, temperature, and gas on sensor sensitivity. An increase in temperature resulted in a reduction in sensor sensitivity. It was also observed that ambient gas influenced the properties of the MWCNT sheet due to charge transfer. Experimental results showed that there was a linear change in resistance in response to strain, and the resistance of the sensor fully recovered to its unstressed state and exhibited stable electromechanical properties.
