We developed a new ecotoxicological test method using native test species, eggs of Ephemera orientalis, and five kinds of industrial wastewater were tested to validate the test method. The water samples were collected in Jun 2006 from the following industries: pesticide, metal plating, PCB, leather1, and leather2. Wastewater and effluent were diluted by distilled water, respectively, to prepare various concentrations, 100, 50, 25, 12.5, 6.3, 3.1, and 0%. For the egg bioassay, 20 freshly laid eggs (<24 h old) were exposed to test solutions in a Petri dish ($52{\times}12mm$) at $20^{\circ}C$ with photoperiod of 16 h light and 8 h dark for 14 days. The median egg hatching concentrations (EHC50) were estimated using Probit analysis. All EHC50s of wastewater were less than 3.1%, which meant very high ecotoxicity except for the wastewater of PCB industry having 6.1% of EHC50. Among the effluents, the least toxic effluent was from pesticide industry having 58% of EHC50, while the effluent of leather2 was the most toxic having 7.3% of EHC50.
In order to suggest correct direction of researches on Miscanthus spp. which are promising bioenergy crop, authors had reviewed and summarized various literature about botanical taxonomy, morphology and present condition of breeding, cultivation and utilization of miscanthus. Among the genus of Miscanthus which are known 17 species, the most important species are M. sinensis and M. sacchariflorus which origin are East Asia including Korea, and M. x giganteus which is inter-specific hybrid of tetraploid M. sacchariflorus and diploid M. sinensis. Miscanthus is superior to other energy crops in resistance to poor environments including cold, saline and damp soil, nitrogen utilization efficiency, budget of input energy and carbon which are required for producing biomass and output which are stored in biomass. The major species for production of energy and industrial products including construction material in Europe, USA and Canada is M. x giganteus which was introduced from Japan in 1930s. In present, many breeding programs are conducted to supplement demerits of present varieties and to develop "Miscanes" which is hybrid of miscanthus and sugar cane. In Korea, the researches on breeding and cultivation of miscanthus were initiated in 2007 by collecting germplasms, and developed "Goedae-Uksae 1" which is high biomass yield and "mass propagation method of miscanthus" which can improve propagation efficiency in 2009. In order to develop "Korean miscanthus industry" in future, the superior varieties available not only domestic but also foreign market should be developed by new breeding method including molecular markers. Researches on production process of cellulosic bio-ethanol including pre-treatment and saccharification of miscanthus biomass also should be strengthen.
Microbial metal reduction influences the biogeochemical cycles of carbon and metals as well as plays an important role in the bioremediation of metals, radionuclides, and organic contaminants. The use of bacteria to facilitate the production of magnetite nanoparticles and the formation of carbonate minerals may provide new biotechnological processes for material synthesis and carbon sequestration. Metal-reducing bacteria were isolated from a variety of extreme environments, such as deep terrestrial subsurface, deep marine sediments, water near Hydrothemal vents, and alkaline ponds. Metal-reducing bacteria isolated from diverse extreme environments were able to reduce Fe(III), Mn(IV), Cr(VI), Co(III), and U(VI) using short chain fatty acids and/or hydrogen as the electron donors. These bacteria exhibited diverse mineral precipitation capabilities including the formation of magnetite ($Fe_3$$O_4$), siderite ($FeCO_3$), calcite ($CaCO_3$), rhodochrosite ($MnCO_3$), vivianite [$Fe_3$($PO_4$)$_2$ .$8H_2$O], and uraninite ($UO_2$). Geochemical and environmental factors such as atmospheres, chemical milieu, and species of bacteria affected the extent of Fe(III)-reduction as well as the mineralogy and morphology of the crystalline iron mineral phases. Thermophilic bacteria use amorphous Fe(III)-oxyhydroxide plus metals (Co, Cr, Ni) as an electron acceptor and organic carbon as an electron donor to synthesize metal-substituted magnetite. Metal reducing bacteria were capable of $CO_2$conversion Into sparingly soluble carbonate minerals, such as siderite and calcite using amorphous Fe(III)-oxyhydroxide or metal-rich fly ash. These results indicate that microbial Fe(III)-reduction may not only play important roles in iron and carbon biogeochemistry in natural environments, but also be potentially useful f3r the synthesis of submicron-sized ferromagnetic materials.
In order to obtain scientific information for the conservation of Utricularia japonica Makino, rare plant species, we investigated life history, phenology, growth and distribution in the east coastal region of Korea. Seven habitats of U. japonica were confirmed in this study. Turion of U. japonica rests overwinter at the bottom and ripens at the water surface and then usually germinate and sprout when the water warms in spring. A single shoot grown from turion divided into several shoots. Shoot decomposition started in October and formed a new turion at the end of the decomposed shoot. Flowering period was from early July to late October. Percentage of flowering ramets was significantly low as 6.3%. U. japonica showed the fastest growth rate from April to July, the maximum growth in August and the highest biomass in October. U. japonica tended to be concentrated in larger ramets in the water depth of 50 cm~150 cm in Cheonjin lake. The main factors affecting the growth of U. japonica were water temperature and turbidity. The establishment and growth of U. japonica in Cheonjin lake were determined by responses to water temperature with seasonal change and to light conditions caused by the different plants. These conditions affect the temporal and spatial distribution of U. japonica and population change. The findings of this study would be helpful to provide the basic information needed for the conservation and restoration of U. japonica.
Recently, new ginseng cultivars having superior agricultural traits have been developed in Korea. For newly developed plant cultivars, the identification of distinctiveness is very important factors not only in plant cultivar management but also in breeding programs. Thus, eighty-five inter simple sequence repeat (ISSR) primers were applied to detect polymorphisms among six major Korean ginseng cultivars and two foreign ginsengs. A total of 197 polymorphic bands with an average 5.8 polymorphic bands and 2.9 banding patterns per assay unit across six Korean ginseng cultivars and foreign ginsengs from 236 amplified ISSR loci with an average 6.9 loci per assay unit were generated by 34 out of 85 ISSR primers. Three species of Panax ginseng including the Korean ginseng cultivars, P. quinquefolius, and P. notoginseng, could be readily discriminated using most tested primers. UBC-821, UBC-868, and UBC-878 generated polymorphic bands among the six Korean ginseng cultivars, and could distinguish them from foreign ginsengs. Sequence characterized amplified region (SCAR) marker system was introduced in order to increase the reproducibility of the polymorphism. One SCAR marker, PgI821C650, was successfully converted from the randomly amplified polymorphism by UBC-821. It showed the expected dominant polymorphism among ginseng samples. In addition, the specific polymorphism for Sunwon was generated by treating Taq I restriction enzyme to polymerase chain reaction products of PgI821C650. These results will serve as useful DNA markers for identification of Korean ginseng, especially Sunwon cultivar, seed management, and molecular breeding program supplemented with marker-assisted selection.

Although many studies have been performed to elucidate the molecular consequence of ultraviolet irradiation on an aging, little is known about the effect of natural products. Matrix metalloproteinases (MMPs) we known to play an important role in (a) photoaging. Hete we investigated the effect of $1,2,4,6-tetra-O-galloyl-{\beta}-{_D}-gluco- pyranose (1)$ and 3,4,5-trihydroxybenzoic acid (2) on the expression of MMP-1 in UVA-irradiated human skin fibroblasts (products), (on the) activity of MMP-1, and (on the) scavenging activities of free radicals. Compounds 1 and 2 were isolated from Melothria heterophylla (Cucurbitaceae). These compounds were found to scavenge free radicals and reactive oxygen species (ROS) and were measured to have the $SC_{50}$ values of $3.9{\mu}M\;and\;13.3{\mu}M$ against the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and $4.3{\mu}M\;and\; 4.0{\mu}M$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. Compounds 1 and 2 showed a dose-dependent inhibitory effect on the e):pression and activity of MMP-1 in the UVA-irradiated human skin fibroblasts. Therefore, we concluded that compounds 1 and 2 significantly inhibited MMP-1 expression at the protein level. Also, these compounds were determined to have a potent antioxidant activity. From these results, we suggest that these compounds nay be used as (a) new anti-aging agents for the photo-damaged skin.
Broad bean wilt virus 2 (BBWV2) is a species in the genus Fabavirus and family Secoviridae, which is transmitted by aphids and has a wide host range. The BBWV2 genome is composed of two single-stranded, positive-sense RNAs, RNA-1 and RNA-2. The representative symptoms of BBWV2 are mosaic, mottle, vein clearing, wilt, and stunting on leaves, and these symptoms cause economic damage to various crops. In 2019, Perilla fructescens leaves with mosaic and yellowing symptoms were found in Geumsan, South Korea. Reverse-transcription polymerase chain reaction (RT-PCR) was performed with specific primers for 10 reported viruses, including BBWV2, to identify the causal virus, and the results were positive for BBWV2. To characterize a BBWV2 isolate (BBWV2-GS-PF) from symptomatic P. fructescens, genetic analysis and pathogenicity tests were performed. The complete genomic sequences of RNA-1 and RNA-2 of BBWV2-GS-PF were phylogenetically distant to the previously reported BBWV2 isolates, with relatively low nucleotide sequence similarities of 76-80%. In the pathogenicity test, unlike most BBWV2 isolates with mild mosaic or mosaic symptoms in peppers, the BBWV2-GS-PF isolate showed typical ring spot symptoms. Considering these results, the BBWV2-GS-PF isolate from P. fructescens could be classified as a new strain of BBWV2.

Recently irregular dark-colored patches were found on the Kentucky teeing ground in a golf course in Gyunggi providence. Interestingly, blue-green algae from the leaf tissue sample containing black spot-stained symptoms were largely observed through microscopic study. In general, algae present on the upper soil surface or in the upper layer of root zone form dark brown layers of scum or crust, which invoked harmful effects to turf growth such as poor drainage, inhibition of new root development. In this observation, unlike the algae were sometime found in senescing leaves on contacted soil in July and August, the blue-green algae were detected within black spot-stained Kentucky bluegrass leaf tissues including leaf blade, ligule, auriclea as well as leaf sheath. The blue-green algae were also detected on the leaf and stem tissue adjacent to the symptomatic leaf tissues. Two species of blue-green algae, Phomidium and Oscillatoria, were greatly observed. Oscillatoria species was more commonly notified in all samples. In addition, the two species were found on a putting green showing yellow spot disease at another golf course in Gyunggi providence. The data from chemical control assay revealed that chemicals such as propiconazole, iprodione, and azoxystrobin decreased blue-green algae population and leaf spots, which finally resulted in enhanced leaf quality. All taken together, we strongly suggested that the disease-like phenomenon by blue-green algae might be very closely mediated with infection/translocation process in relation with turfgrass. It indicates that blue-green algae in turf management may play an adverse role as a secondary barrier as well as a pathogenic agent. This report may be helpful for superintendents to recognize and understand the fact that algae control should be provided more cautiously and seriously than we did previously in upcoming golf course management.

The foot-and-mouth disease virus (FMDV), a member of the Aphthovirus genus in the Picornaviridae family, affects wild and domesticated ruminants and pigs. During replication of the FMDV RNA (ribonucleic acid) genome, FMDV-encoding RNA polymerase 3D acts in a highly location-specific manner. This suggests that specific RNA structures recognized by 3D polymerase within non-coding regions of the FMDV genome assist with binding during replication. One such region is the cis-acting replication element (CRE), which functions as a template for RNA replication. The FMDV CRE adopts a stem-loop conformation with an extended duplex stem, supporting a novel 15-17 nucleotide loop that derives stability from base-stacking interactions, with the exact RNA nucleotide sequence of the CRE producing different RNA secondary structures. Here, we show that CRE sequences of FMDVs isolated in Korea from 2010 to 2017 exhibit A and O genotypes. Interestingly, variations in the RNA secondary structure of the Korean FMDVs are consistent with the phylogenetic relationships between these viruses and reveal the specificity of FMDV infections for particular host species. Therefore, we conclude that each genetic clade of Korean FMDV is characterized by a unique functional CRE and that the evolutionary success of new genetic lineages may be associated with the invention of a novel CRE motif. Therefore, we propose that the specific RNA structure of a CRE is an additional criterion for FMDV classification dependent on the host species. These findings will help correctly analyze CRE sequences and indicate the specificity of host species for future FMDV epidemics.
Along with olive flounder(Paralichthys olivaceus), black rockfish(Sebastes schlegeli) is another very popular maricultured species in Korea. As there is many difficulties in handlling live fish for aquaculturist, use of suitable anesthesia for proper handling of fish is very important in the field. In this view, the effect of $AQUI-S^{\circledR}$ has analysed for its use in the field. $AQUI-S^{\circledR}$, contains 50% isoeugenol, is a new anesthics for fish and zero-withdraw time required since it was approved as a safe additives of food. In the present study, black rockfish adult exhibted sedation effect from 5 ppm at $10^{\circ}C\;and\;15^{\circ}C$, and 7.5 ppm at $20^{\circ}C$, on the other hand, anesthesia was at least required 7.5 ppm at $10^{\circ}C\;and\;15^{\circ}C$, and 10 ppm at $20^{\circ}C$. The fish was recovered from sedation and anesthesia after approximately 5 and 10 minutes, respectively. In case of black rockfish fry, sedation was recorded from 2.5 ppm at $20^{\circ}C$, and 5 ppm at $15^{\circ}C\;and\;20^{\circ}C$. The least concentraion of anesthesia was 2.5 ppm at $10^{\circ}C$, 7.5 ppm at $15^{\circ}C$, and 5 ppm at $20^{\circ}C$. The acute toxic test showed that black rockfish adult and nv showed mortality above 12.5 and 15 ppm concentration of $AQUI-S^{\circledR}$, respectivly. In sum, $AQUI-S^{\circledR}$ is considered as useful anesthetic drug for sedation and anesthesia of black rockfish, it is highly availbale to be prevented from a varity of stress.
