Apparent Dominance of Regenerated Primary Production in the Yellow Sea

  • The Yellow Sea is known to be a very productive region in terms of fisheries. However, its trophic status seems to be highly variable, ranging from oligotrophic to eutrophic, based on new production (NP) values. The NP and regenerated production (RP) values estimated from $^{15}N$-labelled nitrate and ammonium uptake in spring (April 1996) and winter (February 1997) during this study ranged from 0.05 to 19.8 mg $N m^{-2} d^{-1}$ and from 0.1 to 22.8 mg $N m^{-2}d^{-1}$, respectively. Our measurements and earlier observations suggested that NP in the Yellow Sea varied over the four orders of magnitude (range 0.05-180.9 mg $N m^{-2} d^{-1}$) temporally and spatially, and that RP (range 0.1-507.5 mg $N m^{-2}d^{-l}$) based on ammonium predominated during most period of the year, except in winter when both productions were low. The significant nitrogen uptake by phytoplankton below the euphotic zone and episodic entrainment of phytoplankton from below the euphotic zone into the euphotic zone, and nitrite excretion and dissolved organic nitrogen release during nitrate uptake might explain the apparent dominance of RP in the Yellow Sea.

  • 황해 남부해역에서 1993년 4월 25일부터 30일까지 안정동위원소인 /SUP 15/N 질 산염과 암모니아를 이용하여 질소 신생산과 재생산을 측정하였다. 질소생산은 155∼ 688 mg N m/SUP -2/d/SUP -1/의 값을 보였는데 이는 중영양 또는 부영양 해역의 특성 이다. 탄소량으로 한산하면 C:N Redfield비 5.7:1을 가정하였을 때 881∼3909 mg C m/SUP -2d/SUP -1/에 해당된다. 기초생산 중 신생산이 차지하는 비율인 f-ratio의 범 위는 0.12∼0.26으로 기초생산의 74∼88%가 수층 내에서 순환되는 암모니아에 의하여 유지된다는 것을 시사한다. 이러한 낮은 f-ratio 값은 빈영양 또는 중영양 해역의 특 징이다. 영양염의 농도는 예상했던 바와 달리 이 해역에서 의 조사기간에는 생산성을 조절하는 중요한 요인이 아니었으며, 정점간 생산성의 차이는 수괴에 따른 생물 현존 량의 차이에 주로 기인하는 것으로 보인다.

Printing Properties of Novel Regenerated Cellulosic Fibers

  • The reactive printing properties of regular viscose rayon and a new regenerated cellulosic fiber (en Vix^{\textregistered}$) which was prepared from cellulose acetate fiber was investigated in a comparative manner. From the results, it was found that en Vix exhibited better printing properties than regular viscose rayon. It showed stable final color yields, irrespective of the amount of thickener, hence reproducibility of printing of en Vix is expected to be excellent. In addition, urea requirements were less for the printings on en Vix than for the corresponding printing on viscose rayon. Therefore, en Vix is also expected to reduce the amount of the urea which causes environmental problems in dyehouse effluent.

동해 극전선역의 영양염류 순환과정 IV. Clorophyll a 분포, 신생산 및 질산염의 수직확산 (Regeneration Processes of Nutrients in the Polar Front Area of the last Sea IV. Chlorophyll a Distribution, New Production and the Vertical Diffusion of Nitrate)

  • 한국 동해 중부해역의 생$\cdot$화학적 특성에 대한 조사가 1995년 10월 11일부터 18일까지 동해 중북부해역의 31개 정점에서 연구선인 탐양호를 이용하여 수행되었다. chlorophyll a의 농도분포 및 신생산과 수온약층으로부터의 질산염의 수직확산이 측정되었다. chlorophyll a의 농도 범위는 검출한계 이하에서 $0.72{\mu}g/\ell$로서 대부분의 정점에서 최대농도는 수온약층부근에서 발견되었으나, 조사 해역의 남쪽 부분에서는 표층에서 나타났다. 이곳은 상대적으로 netphytoplankton의 비율이 높았고 우점종으로는 연안에서 종종 출현하는 Rhizesolenia sp.이었다. $^{15}N$ 질산염과 암모니아 흡수방법을 이용하여 측정한 질소 신생산과 재생산은 유광층의 적분한 질소생산은 $8.470\~72.945mg\;N\;m^{-2}\;d^{-1}$의 값을 보였으며 기초생산 중 신생산이 차지하는 비율인 f-ratio의 범위는 $0.03\~0.72$로, 기초생산의 $3\~72\%$가 수직확산 등에 의하여 공급되는 질산염에 의해서 유지되고 나머지 $28\~97\%$가 수층내에서 순환되는 암모니아에 의하여 유지된다는 것을 보여주었다. 이러한 f-ratio 값은 극도의 빈영양 해역에서 부영양 해역의 특징을 포함한다. 정점간 생산성과 f-ratio의 차이는 전선의 위치와 섬 (울릉도)와 연관된 해저지형에 관계되어 있는 것으로 보인다. 즉 전선역에서는 생산성과 f-ratio가 높게, 밖에서는 낮게 나타났으며 울릉도 부근에서 가장 높은 f-ratio를 보였다. 수직확산계수는 두 가지 방법으로 구하였는데, King and Devol(1979)의 식에 나타난 수직 안정도로부터 계산한 Kz-1과 new nitrogen 요구량으로부터 계산한 Kz-2이다. Kz-2는 $0.11\~0.55\;cm^2/s$ 의 범위로 이전의 다른 연구들에 의해 보고된 값에 비해서 낮았다.

Electrochemical Regeneration of FAD by Catalytic Electrode Without Electron Mediator and Biochemical Reducing Power

  • We created a new graphite-Cu(II) electrode and found that the electrode could catalyze FADH$_2$ oxidation and FAD reduction coupled to electricity production and consumption, respectively. In a fuel cell with graphite-Cu(II) anode and graphite-Fe(III) cathode, the electricity was produced by coupling to the spontaneous oxidation of FADH$_2$ Fumarate and xylose were not produced from the enzymatic oxidation of succinate and xylitol without FAD, respectively, but produced with FAD. The production of fumarate and xylose in the reactor with FAD electrochemically regenerated was maximally 2- 5 times higher than that in the reactor with FAD. By using this new electrode with catalytic function, a bioelectrocatalysts can be engineered; namely, oxidoreductase (e.g., lactate dehydrogenase) and FAD can function for biotransformation without an electron mediator and second oxidoreductase for cofactors recycling.

  • For breeding of a new rootstock for eggplant production, somatic hybrids between two species, Solanum integrifolium and S. sanitwongsei were obtained through protoplast fusion. The former species has been commonly used for rootstock for eggplant production in Japan. Eggplants on these rootstocks are more productive than ungrafted plants, but are susceptible to bacterial wilt caused Ralstonia solanacearum. While the latter species is resistant, the growth of eggplants on this rootstock is rather slow and low yield. Protoplast of both species were isolated from cotyledons, and inactivated with iodoacetamide or UV-irradiation, then fused electrically. The fused products were then cultured. Regenerated plantlets were then transplanted on soil then maintained in a green house. The plants were classified into four groups. Those in the first group showed morphological characters intermediate of the parentalspecies. The plants bore fruit with viable seeds. The plants showed a chromosome number of 2n=48, the sum of those of the parental species, and are suggested to be symmetric fusion products. While plants in the other groupswas less vigorous and showed chromosome number 2n= 68 to 72 suggesting asymmetric fusion products by genomic in situ hybridization(GISH). Isozyme pattern of shikimate dehydrogenase (SKDH; EC 1.1.1.25), isocitrate dehydrogenase (IDH; EC 1.1.1.41) and phosphoglucomutase (PGM; EC 2.7.5.1) showed that 24 regenerated plants in three groups were somatic hybrids. Analysis of random amplified polymorphic DNA (RAPD) showed that 43 S. integrifolium-specific and 57 S. sanitwongsei-specific bands were all found in 24 plants. Both somatic hybrids and its S1 plants were found to be resistant to bacterial wilt, and eggplant grafted these plants using for rootstocks were more productive than grafted mother plants. Now, S1 progenies are used for commercial eggplant production in Osaka Prefecture.

약(葯)배양 기법 이용한 이탈리안 라이그라스 식물체 생산 (Production of New Regenerated Plant by Anther Culture of Italian Ryegrass)

  • 이탈리안 라이그라스(Lolium multiflorum Lam.) 계통 A의 약(葯) 배양을 실시하여 약(葯) 유래의 식물체를 생산하였다. MS 기본배지에 sucrose 30 g/$\ell$, NAA 2 30 mg/$\ell$, kinetin 1 30 mg/$\ell$을 첨가한 캘러스 유도배지에서 배양 20일만에 약(葯) 유래의 캘러스를 유도하였다. 캘러스 유도율은 9.2%였으며, 캘러스의 평균 크기는 8.6mg/callus/anther 였다. MS 기본배지에 sucrose 30 g/$\ell$, NAA 1 mg/$\ell$, kinetin 2 mg/$\ell$을 첨가한 재분화 배지에 캘러스를 치상하여 배양 50일에 완전히 재분화된 식물체를 얻었으며, 캘러스로부터 식물체로 재분화되는 비율은 26% 였다.

황해의 무광대에서 식물플랑크톤에 의한 질소 섭취율은 상당한가? (Is Nitrogen Uptake Rate by Phytoplankton below the Euphotic Zone in the Yellow Sea Considerable?)

  • 황해 대륙붕 해역의 전 수주(whole water column)의 질소 섭취율 중 무광대(aphotic zone)내 식물플랑크톤에 의한 질소 섭취율의 기여도를 파악하기 위하여 상부의 유광대(euphotic zone)와 하부의 무광대에서 식물플랑크톤에 의한 질소 섭취율의 크기를 측정하였다. 1997년 5월과 11월에 유광대와 무광대에서 일정량의 해수를 채취하여 선상에서 식물플랑크톤의 질산염과 암모늄 섭취율 측정을 $^{15}N$으로 표지하여 추적하였다. 전 유광대내의 질산염 및 암모늄 섭취율은 각각 1.8${\sim}$15.3 mg N $m^{-2}$ $d^{-1}$과 5.0${\sim}$132.2 mg N $m^{-2}$ $d^{-1}$의 범위이며, 대체로 암모늄 섭취율이 질산염 섭취율보다 우세하였다(1.9${\sim}$19.4배). 전 수주내의 질산염 및 암모늄 섭취율은 각각 2.9${\sim}$22.0 mg N$m^{-2}$ $d^{-1}$과 15.7${\sim}$175.5 mg N$m^{-2}$ $d^{-1}$의 범위를 나타냈다. 전 수주내의 총 질소 섭취율 중 무광대의 식물플랑크톤에 의한 질소 섭취율이 차지하는 비율은 질산염의 경우에 13.0${\sim}$86.2%, 암모늄의 경우에는 13.8${\sim}$67.8%로서, 무광대에서 식물플랑크톤에 의한 질소 섭취가 상당함이 발견되었다. 본 연구의 결과는 무광대내에서 상당한 양으로 활발하게 질소를 섭취하는 식물플랑크톤이 물리적 작용들(예를 들면, 와류 혼합, 수온약층의 수직이동 등)에 의해 유광대로 다시 편입(entrainment)될 경우 유광대의 물질 순환에 크게 기여할 가능성이 있음을 시사한다. 그리고, 본 연구 결과는 황해에서 신생산(new production)의 크기가 질산염 섭취로만 추정될 경우 과소평가될 가능성이 있으며, 암모늄 섭취에 기초한 재생산(regenerated production)의 일부가 신생산에 포함되어야 함을 제시한다.

Recent advances in seaweed seedling production: a review of eucheumatoids and other valuable seaweeds

  • Modern seaweed farming relies heavily on seedlings from natural beds or vegetative cuttings from previous harvests. However, this farming method has some disadvantages, such as physiological variation in the seed stock and decreased genetic variability, which reduces the growth rate, carrageenan yield, and gel strength of the seaweeds. A new method of seedling production that is sustainable, scalable, and produces a large number of high-quality plantlets is needed to support the seaweed farming industry. Recent use of tissue culture and micropropagation techniques in eucheumatoid seaweed production has yielded promising results in increasing seed supply and growing uniform seedlings in large numbers in a shorter time. Several seaweed species have been successfully cultured and regenerated into new plantlets in laboratories using direct regeneration, callus culture, and protoplast culture. The use of biostimulants and plant growth regulators in culture media increases the seedling quality even further. Seedlings produced by micropropagation grew faster and had better biochemical properties than conventionally cultivated seedlings. Before being transferred to a land-based grow-out system or ocean nets for farming, tissue-cultured seedlings were recommended to undergo an acclimatization process to increase their survival rate. Regular monitoring is needed to prevent disease and pest infestations and grazing by herbivorous fish and turtles during the farming process. The current review discusses recent techniques for producing eucheumatoid and other valuable seaweed farming materials, emphasizing the efficiency of micropropagation and the transition from laboratory culture to cultivation in land-based or open-sea grow-out systems to elucidate optimal conditions for sustainable seaweed production.

In vitro Multiplication and Corm Production of Freesia hybrida 'Sunny Gold'

  • Freesia has been an important worldwide cut flower because of its fragrance, long vase life and the wide color range of the flower. The conventional propagation methods by seeds and corms have many disadvantages such as shorter inflorescences with fewer numbers of florets, a reduction in cut flower quality and the accumulation of plant viruses in corms by successive cultivation. Therefore, the conventional propagation systems in Freesia needs to be replaced with tissue cultures to overcome the disadvantages. This study explored an efficient multiplication protocol using the combination of plant growth regulators (PGRs) for developed cultivar 'Sunny Gold'. The combination between 6-benzylaminopurin (BA) and α-naphthalene acetic acid (NAA) did not produce new shoots but developed enlarged roots. BA only treatments and the combination between BA and kinetin treatments were effective on shoot multiplication. The highest average number of shoots was 5.3 in the presence of 3 mg/L BA and 0.5 mg/L kinetin. To produce corms and cormlets, proliferated shoots were subcultured on 1/2 Murashige and Skoog (MS) medium supplemented with 90 g/L sucrose, 1 g/L charcoal and 7 g/L plant agar and placed at 4℃ in the dark for 6 months. The small size of corms and comlets were produced. The average number of regenerated comlets was 2.75 per shoot. The results showed that shoot multiplication is more efficient than cormlet regeneration for in vitro freesia proliferation.
