In vitro model system을 활용한 마늘 추출물의 치매예방 특성 (Preventive Characteristics of Garlic Extracts Using in vitro Model System on Alzheimer's Disease)

  • 본 연구에서는 대표적인 퇴행성 신경질환인 알츠하이머성 치매에 대한 마늘 물, 100% 메탄올, 디클로로메탄 추출물들의 acetylcholinesterase (AChE) 저해 및 신경세포 보호효과를 조사하였다. 마늘 디클로로메탄 추출물은 농도 의존적으로 AChE를 저해하는 것으로 나타났으며, $IC_{50}$$36.1{\mu}g/mL$로 나타났다. MTT reduction assay를 이용해 amyloid ${\beta}$ protein ($A{\beta}$) 유도성 신경세포 독성에 대한 신경세포 보호효과를 측정한 결과, 세 가지 마늘 추출물들은 대부분 40% 미만의 세포생존율을 보였고 이 결과는 $A{\beta}$ 유도성 신경세포 독성보다 상대적으로 더 높은 세포독성을 보여주었다. LDH assay에서는 마늘 물 추출물이 37%의 LDH 방출량을 나타내 $200{\mu}M$의 vitamin C과 유사한 세포막손상 보호효과를 보였다. 마지막으로 neutral red uptake assay를 실시한 결과, MTT reduction assay와 마찬가지로 모든 마늘 추출물들에서 세포생존율의 감소를 확인하였으며 특히 디클로로메탄 추출물의 경우 현저하게 낮은 세포생존율을 나타내었다. AChE 저해활성을 갖는 마늘 디클로로메탄 추출물로부터 얻은 column fractionations에 함유된 생리활성물질을 탐색하기 위해 HPLC 분석을 실시하였으며, 마늘 98:2 fraction의 LC-MS 분석을 통하여 allyl methyl disulfide, diallyl monosulfide, diallyl disulfide로 추정되는 물질군이 확인되었다.

대식세포에서 국산약용식물의 항암 및 항Virus에 대한 효과 (Antiviral and Antitumoral Activitivies of Domestic Medicinal Plants in Macrophages)

  • In the present work, 70 extracts from 23 plants have been determined to induce cytotoxic and antiviral activities of macrophages using both MTT assay and neutral red dye uptake assay. We show that 13 extracts have induced cytotoxic activities and 5 extracts induced antiviral activity in mouse peritoneal macrophages. Among 13 extracts, macrophages treated with extracts from Salvia plebeia have demonstrated significant cytotoxicity but not antiviral activity. The present findings indicate that extracts from plants can stimulate macrophages to become resistant to virus and to kill tumor cells.

Effect of Cultural Conditions on Polysaccharide Production and its Physicochemical Properties in Cordyceps militaris

  • This study was concerned with the optimization of liquid culture conditions for mycelial growth and polysaccharide production and its physicochemical properties in Cordyceps militaris. The one factor at a time method was adopted to investigate the effects of medium composition, environmental factors and C/N ratio. Among the these varialbles, glucose 80g/L, yeast extract 10g/L, $MgSO_4{\cdot}7H_{2}O\;0.5g/L$, $KH_{2}PO_4\;0.5g/L$ were proved to be the most suitable carbon, nitrogen, and mineral sources, respectively. The optimal temperature, initial pH, working volume were identified to be $24^{\circ}C$, 7.0 and 100ml, respectively. Under the optimal conditions, the strategies in shake flask culture and 5L jar fermentor led to mycelial growth of 29.43 g/L, 28.88g/L and polysaccharide production of 2.53g/L, 6.38 g/L, respectively. Among the phisicochemical properties, relative concentrations(w/v) of total sugar, uronic acid, protein and hexoseamine were identified to be 74.07%, 1.13%, 0.91%, and 0.46%, respectively. The fraction of neutral and acidic polysaccharide were identified to be 81.9% and 18.1%, respectively.

머위 잎 추출액의 염색성 -매염제가 염색성에 미치는 영향- (A Study on the Dyeing Properties of Petasites Japonicus Leaf Extract)

  • This study were carried out to investigate the effects of mordants and mordanting methods under the various dyeing conditions such as temperature, time, pH, repetition of dyeing in the silk fabrics with Petasites japonicus leaf. It was the most high K/S values of the silk fabrics dyed with Petasites japonicus leaf according to co mordanting and used cupric sulfate, ferric sulfate as mordanting agents. K/S values of the silk fabrics dyed with Petasites japonicus leaf were increased gradually with dyeing temperature, dyeing time, repetition of dyeing was higher and the optimum pH was pH 7. Silk fabrics were colored yellowish orange and surface color(munsell value) was changed from 8.3YR to 2.0Y by using mordanting agents and those of the silk showed high tone when mordanting with cupric sulfate but decolored and darked when mordanting with ferric sulfate. Washing fastness of silk fabrics were good in 4 ~ 5 grade, so washing fastness of the silk fabrics was significantly improved when washed with the neutral detergent.

검은밀복어 간유중의 고도불포화지방산의 함량 (Contents of Polyunsatureated Fatty Acid in the Pufferfish (Lagocephalus gloveri) Liver Oil)

  • Ten specimens of pufferfish Lagocephalus gloveri(called Gumeun-mibog in Korean) were purchased at a fish market in Pusan Korea. The pufferfishes were immediately frozen packed in ice boxes transpor-ted to our laboratory and then dissected in to four parts. The tissues were homoginized after adding with chloroform: methanol mixture solution and storaged at cool and dark place to extract total lipid. The total lipid contents were 29,34∼36.54% in liver 4.95∼6.11% in intestine 1.08∼1.60% in skin and 0.23∼0.38% in muscle of the pufferfish respectively. The contents of DHA and EPA were higher in the total lipids of livers showing 15.99% DHA and 3.04% EPA. The other fatty acids in the total lipids of liver were mainly composed of palmitic acid(16:0) palmitoleic acid(16:1) stearic acid(18:0) and oliec acid(18:1) Furthermore the contents of neurtral lipids were 95.45% and those of phospholipids and glycolipids were 1.45 and 3.09% respectively. Main fattty acids of the neutral lipid were composed of palmitic acid(16:0) stearic acid(18:0) oleic acid(18:1) oleic acid(18:1) EPA(20:6) and DHA(22:6) The contents of DHA and EPA were 16.62 and 2.41% respectvely. From these results of toxicity in the raw liver the tissue was judged to be nontoxic before and after extracting of total lipid.

  • In the present study, we have evaluated cytotoxic effects of the chloroform soluble fraction of the methanolic extract of Perilla frutescens in human oral epitheloid carcinoma cells. The light microscopic study showed morphological changes of the treated cells. Cell membrane damaging activity was measured by the lactate dehydrogenase (LDH) assay and disruptions in cell organelles were determined by 3-(4,5-dime-thylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT), neutral red (NR) and sulforhodamine protein B (SRB) of colorimetric assay. These results suggest that Perilla frutescens retains a potential antitumor activity.

Robust Multi-Layer Hierarchical Model for Digit Character Recognition

  • Although digit character recognition has got a significant improvement in recent years, it is still challenging to achieve satisfied result if the data contains an amount of distracting factors. This paper proposes a novel digit character recognition approach using a multi-layer hierarchical model, Hybrid Restricted Boltzmann Machines (HRBMs), which allows the learning architecture to be robust to background distracting factors. The insight behind the proposed model is that useful high-level features appear more frequently than distracting factors during learning, thus the high-level features can be decompose into hybrid hierarchical structures by using only small label information. In order to extract robust and compact features, a stochastic 0-1 layer is employed, which enables the model's hidden nodes to independently capture the useful character features during training. Experiments on the variations of Mixed National Institute of Standards and Technology (MNIST) dataset show that improvements of the multi-layer hierarchical model can be achieved by the proposed method. Finally, the paper shows the proposed technique which is used in a real-world application, where it is able to identify digit characters under various complex background images.

Hpa I endonuclease의 정제와 특성 (Purification and Characterization of Hpa I endonuclease)

  • Hpa I endonuclease를 순수 정제하였다. 150g (wet weight)의 Haemophilus parainfluenzae로 부터 얻은 crude extract를 ammonium sulfate fractionation을 거친후 Heparin agarose, SP-sephadex, DEAE-sephadex, phosphocellulose 등 chromatography를 거쳐 최종적으로 0.2mg의 효소를 얻었다. Specific activity는 $2.2{\times}10^6units/mg$이었다. 얻어진 Hpa I endonuclease는 SDS-polyacrylamide gel에서 한개의 band를 보였고, 그 효소활성은 50mM NaCl과 pH 7.0과 7.5사이에서 가장 높았다.

Effects of Different Levels of Concentrate in Complete Rations on Nutrient Digestibilities and Ruminal Metabolites in Sheep and Growth Performance in Korean Native Bulls

  • Objective of the present study were to investigate effects of different levels of concentrate in complete rations on nutrient digestibilities and ruminal fermentation in sheep and growth performance in Korean native bulls. Increasing levels of concentrate (35, 50, 65, and 80% of complete rations) improved digestibilities of dry matter (DM), crude protein (CP) and ether extract (EE) without affecting digestibility of neutral detergent (NDF) and acid detergent fiber (ADF). Increasing levels of concentrate decreased ruminal fluid pH but increased concentrations of $NH_3-N$, propionic acid, and total volatile fatty acids (VFA). Both the disappearance rates of DM and nitrogen (N) in an in sacco study were linearly increased as the levels of concentrate in complete rations increased. Meanwhile, increasing levels of concentrate in complete rations improved growth rate and feed conversion ratio in Korean native bulls. In conclusion, the complete rations containing 80% concentrate showed better digestibility and energy supply than those of the lower levels (35, 50 and 65%) of concentrate of the rations, resulting in improved growth performance of Korean native bulls.

A New Approach to Robustly Exchange Models in Heterogeneous CAD/CAE Environment and its Application

  • The model exchange from CAD system to CAE system in valid and effective manner is the major issue of automatic analysis modelling of ship structure. However, model exchange approaches based on the neutral CAD file have resulted in invalid model exchange that could not properly reflect the characteristics of CAD model and CAE model of ship structure. This paper presents the new approach of n-to-n mapping to exchange ship structure model in heterogeneous CAD/CAE environments. In this study, the common model called 'unified ship model for analysis' to directly extract proper information from different CAD systems for ship structural analysis is proposed. Moreover, a command language based model interfacing technique to construct an idealized model for analysis job is also proposed. The proposed approach has been actually implemented in DSME CAD/CAE environment of ship structure such as TRIBON system, PATRAN system and FLUENT system. The applicability and effectiveness of the proposed approach was verified by applying it to the real analysis project for fore-body of ship and block lifting. This application results show that the proposed approach can be effectively used for heterogeneous CAD/CAE environment.
