Vertebrate retinal neurons, like brain tracts farm complex synaptic relations in the enter and inner plexiform layers which ape equivalent to the central nervous system nuclei. The effects of $\gamma-aminobutyric$ acid(GABA) and glycine on retinal neurons were explored to discern the mechanisms of action of neurotransmitters. Experiments were performed in the superfused retina-eyecup preparation of the channel catfish, Ictalurus punctatus, using intracellular electrophysiological techniques. The roles of GABA and glycine as inhibitory neurotransmitters are well established in the vertebrate retina. But, we found that the depolarizing action of GABA and glycine on third-order neurons in the catfish retina. GABA and glycine appeared to act on retinal ueurons based on the observations that (1) effects on photoreceptors were not observed, (2) horizontal cells were either hyperpolarized $({\sim}33%)$ or depolarized $({\sim}67%)$, (3) bipolar cells were all hyperpolarized (4) amacrine and ganglion cells were either hyperpolarized $({\sim}37%)$ or depolarized $({\sim}63%)$, (5) GABA and glycine may be working to suppress presynaptic inhibition. The results suggest that depolarization of third-order neurons by GABA and glycine is due to at least two mechanisms; a direct postsynaptic effect and an indirect effect. Therefore, in the catfish retina, a mechanism of presynaptic inhibition or disinhibition including the direct postsynaptic effect may exist in the third-order neurons.
Bone remodeling is a continuous process of skeletal renewal during which bone formation is tightly coupled to bone resorption. Mechanical loading is an important regulator of bone formation and resorption. In recent studies, neurotransmitters such as vasoactive intestinal peptide (VIP) were found to be present inside bone tissue and have been suggested to potentially regulate bone remodeling. In this study, our objective was to use a pre-established in vitro oscillatory fluid flow-induced shear stress mechanical loading system to quantify the effect of VIP on bone resorptive activity and investigate its combined effect with mechanical loading. VIP decreased osteoclastogenesis significantly decreased RANKL/OPG mRNA ration by approximately 90%. Combined VIP and mechanical loading further decreased RANKL/OPG ratio to approximately 95%. These results suggest that VIP present in bone tissue may synergistically act with mechanical loading to regulate bone remodeling via suppression of bone resorptive activities.

Diazinon, an organophosphate pesticide, is relatively highly toxic to fish and causes vertebral malformation and behavioral change of fish at relatively low concentrations. To elucidate biochemical mechanism of the behavioral change of Oryzias latipes (killifish) caused by diazinon, the effect of the insecticide on acetylcholine esterase activities and the levels of some neurotransmitters were evaluated. Acetylcholine esterase activities in both head and body were significantly lowered at the concentration of 10 ppb of diazinon and acetylcholine contents in head tended to be upregulated with increasing concentration of diazinon. Exposure of killifish to 5000 ppb diazinon resulted in gradual decrease in acetylcholine content in body part with exposure time. Norepinephrine and serotonin concentrations in killifish head and body were highest at 1000 ppb of diazinon while neurotransmitter were relatively low in fish unexposed or exposed to lower dose of the pesticide, suggesting that increased norepinephrine and serotonin can partially account for diazinon-induced behavioral abnormality.

Various neurotransmitters are involved in regulating stress systems. In this study, we investigated the combination relieving effect of green tea extract(GTE) and L-theanine on the stress induced by electric foot shock. Four week oral administration of GTE (24 mg/kg), L-theanine (4 mg/kg) or their combination reduced the levels of dopamine, noradrenaline and corticosterone in blood, brain cortex, hippocampus, and striatum, wherease increased serotonin level. The combination of GTE and L-theanine showed much greater effects than single treatment of each component, and the effects are comparable to diazepam (2 mg/kg). Therefore, this study suggests that the combination of GTE and L-theanine may act effective and be useful for stress relieving treatment.

The effects of transmural nerve stimulation induced releasing neurotransmitters on the changes of swine uterine smooth muscle motility were examined by polygraph through isometric force transducer. The frequency dependent relaxation and rebound contraction were revealed on precontraction with histamine by transmural nerve stimulation. The rebound contraction by transmural nerve stimulation was inhibited by nonselective ${\alpha}$-adrenergic receptor antagonist, phentolamine, and the relaxation by transmural nerve stimulation was blocked by nonselective ${\beta}$-adrenergic receptor antagonist, propranolol. The relaxation induced by nonselective ${\beta}$-adrenergic receptor agonist, isoproterenol on precontraction with histamine were the dose dependent manner and this relaxation was blocked by nonselective ${\beta}$-adrenergic receptor antagonist, propranolol in isolated uterine smooth muscle of pig. These results suggest that endogenous neurotransmitters on smooth muscle relaxation was influenced by ${\beta}$-adrenergic receptor in swine.
Cerebral monoamines play important roles as neurotransmitters that are associated with various stressful stimuli. Some components such as ginsenosides (triterpenoidal glycosides derived from the Ginseng Radix) may interact with monoamine systems. The aim of this study was to determine whether ginsenoside Rb1 can modulate levels of the monoamines such as dihydroxyphenylalanine (DOPA), dopamine (DA), norepinephrine (NE), epinephrine (EP), 3,4-dihydroxyphenylacetic acid (DOPAC), 5-hydorxytryptamine (5-HT), 5-hydroxindole-3-acetic acid (5-HIAA), and 5-hydroxytryptophan (5-HTP) in mice frontal cortex and cerebellum in response to immobilization stress. Mice were treated with ginsenoside Rb1 (10 mg/kg, oral) before a single 30 min immobilization stress. Acute immobilization stress resulted in elevation of monoamine levels in frontal cortex and cerebellum. Pretreatment with ginsenoside Rb1 attenuated the stress-induced changes in the levels of monoamines in each region. The present findings showed the anti-stress potential of ginsenoside Rb1 in relation to regulation effects on the cerebral monoaminergic systems. Therefore, the ginsenoside Rb1 may be a useful candidate for treating several brain symptoms related with stress.

Purpose : This study investigated the effects of low-intensity muscle training on neurotransmitters associated with depression and emotion regulation disorders in survivors with stroke who were diagnosed with depression or emotion regulation disorder. Methods : Survivors of chronic stroke diagnosed with depression or emotion regulation disorder (n = ) were included in the study. Concentrations of serotonin, norepinephrine, and epinephrine were measured before and after. Low-intensity muscle training was performed once a day for 50 minutes, 5 times a week for 10 weeks. Result : Levels of serotonin, norepinephrine, and epinephrine improved significantly after intervention (p < .05). Conclusion : These results confirm that low-intensity muscle training has a positive effect on neurotransmitter concentrations in survivors with stroke diagnosed with depression and emotion regulation disorders. Future studies should analyze the changes in neurotransmitters in more detail using various exercise methods and intensities to investigate their effects on depression and emotion regulation disorders.
The present study was carried out to investigate the effects of Woowhangcheongshim-won(WCW) on the extracellular concentrations of amino acid neurotransmitters(glutamate, aspartate, GABA, glycine, taurine, alanine, and tyrosine) and organic acid (lactate and pyruvate) in striatum and cerebral infarction volume in rats subjected to permanent focal cerebral ischemia induced by 2 hours of middle cerebral artery occlusion(MCAO), using intracerebral microdialysis as the sampling technique, Microdialysis probes were inserted into the lateral part of the caudate-putamen 2 hours before the experiment and microdialyzates were collected at 20min intervals and analyzed by high performance liquid chromatography, WCW significantly decreased the infarction volume with reducing focal cerebral ischemia-induced increase of extracellular glutamate, asparate, and tyrosine. On the other hand, the increase of GABA and taurine was enhanced after treatment of WCW in the ischemia-induced rats, These results suggest that WCW can produce a neuroprotective effect against cerebral ischemia by regulating extracellular excitatory and inhibitory amino acid levels in relation to the concept of excitotoxicity in brain ischemia.
Sleep is a vital, highly organized process regulated by complex systems of neuronal networks and neurotransmitters. Normal sleep comprises non-rapid eye movement (NREM) and REM periods that alternate through the night. Sleep usually begins in NREM and progresses through deeper NREM stages (2, 3, and 4 stages), but newborns enter REM sleep (active sleep) first before NREM (quiet sleep). A period of NREM and REM sleep cycle is approximately 90 minutes, but newborn have a shorter sleep cycle (50 minutes). As children mature, sleep changes as an adult pattern: shorter sleep duration, longer sleep cycles and less daytime sleep. REM sleep is approximately 50% of total sleep in newborn and dramatically decreases over the first 2 years into adulthood (20% to 25%). An initial predominant of slow wave sleep (stage 3 and 4) that peaks in early childhood, drops off abruptly after adolescence by 40% from preteen years, and then declines over the life span. The hypothalamus is recognized as a key area of brain involved in regulation of sleep and wakefulness. The basic function of sleep largely remains elusive, but it is clear that sleep plays an important role in the regulation of CNS and body physiologic processes. Understanding of the architecture of sleep and basic mechanisms that regulate sleep and wake cycle are essential to evaluate normal or abnormal development of sleep pattern changes with age. Reduction or disruption of sleep can have a significant impact on daytime functioning and development, including learning, growth, behavior, and emotional regulation.
