Development of Ecotoxicological Standard Methods using Early Life Stage of Marine Rotifer Brachionus plicatilis and Benthic Copepod Tigriopus japonicus (윤충류 Brachionus plicatilis 및 저서 요각류 Tigriopus japonicus의 초기 생활사를 이용한 해양생태독성시험 방법에 관한 연구)

  • Marine ecotoxicological standard method was applicated using marine rotifer and benthic copepod as primary consumer of marine ecosystem. Marine rotifer, Brachionus plicatilis and benthic copepod, Tigriopus japonicus were designated as standard test species with the endpoints of 24hr neonate mortality (24hr $LC_{50}$) and 48hr population growth (48hr $EC_{50}$) for rotifer, and 48hr nauplius mortality (48hr $LC_{50}$) for benthic copepod. Tests method was referred to those of ASTM (American Society for Testing and Materials) with the replacement of test species which are widely distributed in Korean waters. The two species showed a wide tolerance on salinity ($5{\sim}35\;psu$) and can be easily cultured in small space even they were not as sensitive as in the mortality test using the nauplius of marine invertebrates. However, these species revealed the significant "concentrationresponse relationship" tested with ocean disposal wastes, and reproducibility using cadmium as standard reference material between laboratories. Accordingly, these two species have good potential as test species for marine ecotoxicological test species. Also, we strongly recommend that at least one of these species be included in the test species of "battery test" in marine bioassay.

On the Feeding Behavior of Zooplankton in Lake Soyang (소양호에서 동물 플랑크톤의 섭식작용에 관한 연구)

  • Zooplankton feeding was investigated with epilluorescence microscope in Lake Soyang in August 1991. Zooplankton. which ingested fluorescence bead or fluorescently labeled bacteria (FLB). was regarded as bacterivore. The algavores wert. easily distinguished with autofluorescence of chlorophyll in gut. Copepoda nauplius and Copepodids. 7'hermocyclop.s spp, Pleosomcl spp. Brachionus spp were algavores. and DuphnB hpp. Bosmincr spp. Kerutrlla spp and Hrxuthru spp werc identified as bacterivc~res.T he mixo\ory was not detected. The percentages of algavores and bacterivores in Lake Hoyang were 65 7% and 34.3%. respectively. The bacterivorous zooplankton had trend to ingcst the beads bigger than 0.5 pm. Use of 0.5 pm bead as grazing tracer gave similar estin~ates of ingestion to FLR.

Production of Mullet (Chelon lauvergnii) Seedlings (가숭어, Chelon lauvergnii 종묘생산)

  • At the Poryoung coast, the spawning season of the mullet, Chelon lauvergnii lasts from May to June. A combination of rotifer, Rotifer nauplii and synthetic feed supported significantly (P < 0.05) better survival and growth of 1-day old mullet larvae during 70-day rearing experiment than the other feeds consisting of rotifer or rotifer + Artemia nauplii. Significantly higher growth and survival were also observed, when the larvae were fed on rotifer, that were harvested from culture enriched with masimaTR, or Artemia nauplii from culture enriched with cuttlefish liver oil, At the feeding density of 97-98 individuals /200 l, the mullet larvae displayed maximum growth and survival in aquarium each with 100 larvae /200 l.

Marine Meiobenthic Faunal Communities of the Sediments near Dokdo in the East Sen, Korea (독도 주변 해역에 서식하는 중형저서생물)

  • Meiobenthic community structure near Dokdo was investigated during two survey periods of June, 1999 and October 2000. Meiobenthos were separated from the sediment core samples collected from eight stations. A total of 19 meiobenthic animal groups were identified from those core samples. The most abundant meiobenthic animals belonged to Phylum Nematoda in both seasons and all stations. Sarcomastigophorans, nauplius of crustaceans, and benthic harpactiocoids were the next abundant meiobenthos. Vertical distribution of meiobenthic animals showed the highest individual numbers in the surface sediment layers of 0-1cm depth with a steep decreasing trend with depth for the three sampling stations (A0, A9', A19) located in the continental slope. Horizontal distribution of meiobenthic animals for the study area near Dokdo showed lower density at the sampling stations near Dokdo and the lowest density of meiobenthos in A19 (depth-2261 m). For size distribution, analysis showed that animals which fit onto the mesh size of 0.125mm were abundant. This study suggested that the continental shelf near Dokdo have a more unstable environment fo meiobenthos than the continental slope.

Characteristics of Meiobenthic Community Inhabiting Sandy Sediment in the Yellow Sea, Korea (서해 장봉도 인근 사질 퇴적물에서 서식하는 중형저서동물 군집 특성)

  • The community structure of meiobenthos was studied at 16 stations within sandy tidal and subtidal zones in Jangbongdo in the Yellow Sea, Korea from Aug. 2006 to Jan. 2007. Meiobenthic organisms were collected by three core samples, with a 3.6 cm diameter, from each sediment sample taken with a Smith-McIntyre Grab. Mean grain size of study stations ranged from $1.49{\phi}$ to $3.55{\phi}$. Composition of sand ranged from 80.38% to 99.89%. There was reduction in total abundance and biomass of meiobenthos from summer to winter. Total densities of meiofauna ranged between 17 inds./10 $cm^2$ and 853 inds./10 $cm^2$. Nematodes, gastrotricha, nauplius and harpacticoids appeared as major taxa in decreasing order. This study shows that major taxa comprised 90 percent of total abundance. Most meiofaunal organisms are concentrated in the upper sediment layers and the total abundance and biomass of organisms in the tidal zone is higher than the subtidal zone.

Investigation of marine bacteria for the food of Tigriopus japonicus Mori(Harpacticoida) (동물플랑크톤 Harpacticoid, Tigriopus japonicus Mori 배양에 유효한 해양세균의 탐색)

  • In order to fine some proper bacterial food for Tigriopus japoncus, bacterial flora of the tide pool inhabited by the copepod has been isolated and tested as bacterial food. Food effect and optimum density of the bacteria in terms of survival rate of the copepod was measured in the larval and the adult stages. Among the 264 strains of isolated bacteria, Acinetobacter spp. Moraxella, spp., Flavobacterium spp. and Pseudomonas spp. were certified as effective food for the copepod. According to the experimental results, Acinetobacter spp. AG-3 was the most effective food for all the stages from nauplius to adult, while Moraxella spp. and Flavobacterium spp. were effective for copepodite stage, and Pseudomonas spp. for the adult stage only. The optimum density of bacteria for the food was about $10^6\;cell/ml$, which was the same average density of bacteria in the tide pool.

Development and Reproduction of Eucyclops serrulatus(Copepoda: Cyclopoida) in the Laboratory Culture (실험실에서 배양한 Eucyclops serrulatus(검물벼룩목 요각류)의 생식과 성장에 관한 연구)

  • Four cyclopoids were tested for laboratory culture. Among these animals, only Eucyclops serrulatus was successfully cultured in the laboratory. Under the laboratory culture condition, nine kinds of diets were tested for the suitability of nauplius development. Development and reproduction of E. serrulatus were also investigated. Chlamydomonas reinhardii was the only one which could induce complete development from nauplii to adults. It was found that E. serrulatus had relatively short generation time, could produce lots of progenies, and could be handled easily, allowing to obtain many individuals during a short period. With these characteristics, this species may be a candidate for a good test organism for evaluating freshwater pollution.

Abundances and Species Composition of Microzooplankton during Summer in Kwangyang Bay, Korea (여름철 광양만 소형동물플랑크톤의 출현량과 종조성)

  • To study the abundances and species composition of microzooplankton during summer, microzooplankton were sampled fortnightly from 22 May to 3 August in Kwangyang Bay, Korea, The dominant species were Tintinnopsis spp. and Coaonellopsis sp. of tintinnids, and eggs and nauplii of copepods, They occupied $83.8{\%}$ of the total microzooplankton. The abundance of microzooplankton on 22 May was $374.6 ind./l$. It increased to 1,276.0 ind./l and 1,234.9 ind./l in June, and then reduced to $528.2{\~}609.8 ind./l$ in July and August. The abundances and species composition of microzooplankton were affected by the predation of fish larvae.

Meiobenthic Community Structure on the Northeast Coastal Area of Jeju Island, Korea (제주 북동부 연안 조하대에 서식하는 중형저서동물 군집 변동 특성)

  • We analyzed the characteristics of meiofaunal communities in the northern part of Jeju Island and the environmental factors affecting the meiofaunal communities. For the analysis of the communities, subtidal sediments containing meiofauna were collected from 6 sites (3 shallow sites and 3 deeper sites) on the northern coast of Jeju Island in April, June, August, and October 2017. The sediment samples were collected by SCUBA using a syringe. After isolating and counting meiofauna from the sediment using a series of sieves and centrifugation, the density of the meiofauna was expressed as individuals/10 ㎠. Density of meiofauna was lower in August compared to April and June, and the density determined in October was higher than August. During April and June, the meiofaunal communities at the shallow sites (i.e., ST 1, 2, and 3) were significantly different from the meiofaunal communities at deeper sites (ST 4, 5, and 6, p < 0.01). However, such spatial variation in the meiofaunal communities was not obvious in August and October. The most dominant meiofaunal taxa were harpacticoids, followed by nematodes and nauplius larvae. These three taxa accounted for 90% of the total meiofauna density. Abundance of harpacticoids and nematodes showed a significantly negative correlation with the water depth. BIO-ENV analysis indicated that the total organic nitrogen (TON) and the water temperature exerted a significant influence over the meiofaunal communities at the sampling sites.

Comparison of the Nutritional Value of Chlorella ellipsoidea and Nannochloris oculata for Rotifers and Artemia Nauplii

  • Microalgae are widely used for mass culture of the rotifer Brachionus plicatilis in aquaculture. Since the nutritional value of the rotifer is closely related to its food, the nutritional value of its food should be known in detail. Chlorella ellipsoidea and Nannochloris oculata are re­presentative food organisms for rotifers that are easily cultured. Therefore, the nutritional values of these micro algae were examined for ultrasmall, small, and large rotifers and Artemia nauplii. Chlorella ellipsoidea contained seven times more total fatty acids than N. oculata. The three types of rotifer fed N. oculata contained more amino acids than those fed C. ellipsoidea. However, the total fatty acids of the rotifers fed each microalga species differed according to the type of rotifer. Newly hatched Artemia nauplii contained more protein and had a higher dry weight than those fed microalgae for 6 h. As with the rotifers, the Artemia nauplii fed N. oculata contained more protein and amino acids than those fed C. ellipsoidea, while the reverse was true for the total fatty acid content. Our results suggest that N. oculata is a good supply of protein, while C. ellipsoidea is a good source of lipids as food organisms for rotifers and Artemia nauplii in aquaculture.
