Transactivation potential of the C-terminus of human ALG-2 (Human ALG-2 C-말단의 전사활성화 능력 분석)

  • ALG-2 (apoptosis linked gene-2) is a 22 kDa calcium-binding protein necessary for apoptosis induced by various stimuli in lymphocyte. The transactivation of human ALG-2 was assessed in yeast as a fusion protein with the DNA binding domains (DBDs) of LexA. The C-terminal of hALG-2 (93-191 amino acid) exhibited transacitivation of the reporter gene, LacZ, whereas the full-length hALG-2 (1-91 amino acid) and its N-terminal (1-98 amino acid) did not. The transactivation of LacZ reporter was driven more strongly (more than 2.7-fold increase) by the C-terminus of hALG-2 than by the B42, as a positive control for transactivation. Hence, our data suggested a possible regulatory role of the N-termini of hALG-2 upon transactivation.

Description of Vishniacozyma terrae sp. nov. and Dioszegia terrae sp. nov., Two Novel Basidiomycetous Yeast Species Isolated from Soil in Korea

  • Two strains, YP344 and YP579 were isolated from soil samples in Pocheon City, Gyeonggi Province, South Korea. The strains YP344 and YP579 belong to the genus Vishniacozyma and Dioszegia, respectively. The molecular phylogenetic analysis showed that the strain YP344 was closely related to Vishniacozyma peneaus. Strain YP344T differed by four nucleotide substitutions with no gap (0.70%) in the D1/D2 domain of the LSU rRNA gene and 16 nucleotide substitutions with 8 gaps (5.76%) in the ITS region. On the other hand, the strain YP579T varied from the type strain of the most closely related species, Dioszegia zsoltii var. zsoltii, by 6 nucleotide substitutions with four gaps (1.64%) in the D1/D2 domain of LSU rRNA gene and 26 nucleotide substitutions with 14 gaps (8.16%) in the ITS region. Therefore, the name Vishniacozyma terrae sp. nov. and Dioszegia terrae sp. nov. are proposed, with type strains YP344T (KCTC27988T) and YP579T (KCTC 27998T), respectively.

Teratological Study of LBD-001, a Recombinant Human Interferon $\gamma$, in Rabbits

  • LBD-001, a recombinant human interferon $\gamma$ produced by genetically engineered yeast as a host system, was intravenously administered to pregnant female rabbits (New Zealand White strain) from day 6 to 18 of gestation at dose levels of $0.35 \times 10^6$, $0. 69 \times 10^6$, and $1.38 \times 10^6$ I.U./kg/day. Hydrocortisone sodium succinate (0.3 mg/kg/day) was also given in the same way. Teratological effects of the test agents on the organogenesis of fetuses and the development of offsprings (F1 rabbits) were investigated. The results were as followings: (1) No significant changes by the treatment of LBD-001 or hydrocortisone sodium succinate were observed in the body weights, the food and water consumption, the lactating or nurshing behaviors, and the autopsy of the pregnant rabbits. (2) No significant changes in the resorption rate, the fetal organogenesis, and the normal develpoment of offsprings (F1) by the treatment of LBD-001 or hydrocortisone sodium succinate were detected. The results show that LBD-001 at the dose of $1.38 \times 10^6$ I.U./kg/day or less and hydrocortisone sodium succinate at the dose of 0.3 mg/kg/day are neither teratogenic in the organogensis of the fetuses and the development of the offsprings (F1) nor toxic to the mother rabbits.

A Recipe Development and its Biological Study for The Enriched Bread (영양 강화 빵의 제법 연구)

  • The nutrients such as protein, calcium, iron and all three of them, either by natural or chemical substances were added to the recipes of yeast breads. Acceptability test was carried for the each formulated bread before selecting eight experimental diets. Sixty albino rats were separated into ten dietary groups including eight enriched breads and two control diets of plain breads from the market, which were used as the carbohydrate sources of the each basal diet. Experimental regime was seven weeks. The growth rate, hematology, total serum protein and organ weights we compared. Generally animals on the enriched bread showed the better growth rate than the rats fed control diet. Diet H enriched protein, iron and calcium by natural foods, brought up the highest weight gain among all of the rats. Hemoglobin concentrations of the rats on the diet H and G (enriched with three nutrients by chemicals) were 14.3 and 14.2 gram % comparing with other two control groups (12.9 and 13.1 gram%). Rats on diet K (fed nothing but diet H enriched bread) showed the significantly higher growth rate, total serum protein and hemoglobin level than these on the diet L (fed only plain bread from the market).

Enzymological Evaluation of Bowel Inflammation Inhibitory Activity and Intestinal Microbial Flora Improvement by Enzymatic Hydrolysate of Schizandrae Fructus (오미자 효소가수분해물에 의한 장내 세균총 개선 및 장관 염증 억제 효과의 효소학적 평가)

  • Schizandrae Fruits has been used as a traditional Oriental medicine for treatment of many stress-induced diseases. In the present study, we investigated inhibitory activity of enzymatic hydrolysate of Schizandrae fructus (SC-EX) in growth of tested intestinal microorganism and activity of bowel inflammation related enzyme. SC-EX was added to the proteose peptone-yeast extract-fildes (PYF) media to investigation the effect on the growth of type culture of intestinal microorganism. The growth of lactic acid bacteria such as Bifidobacterium species and Lactobacillus species was accelerated by more than 3% concentration of SC-EX. But, growth of harmfulness bacteria such as E.coli, Clostridium sp. Staphylococcus sp. Streptococcus sp. was inhibited by more than 3% concentration of SC-EX. Also, SC-EX was exhibited a concentration-dependent inhibitory activity of the bowel inflammation related enzymes. The SC-EX was showed 76% and 92% inhibitory activity of 5-lipoxygenase and cyclooygenase at 5% additional concentration respectively. Our results indicated that SC-EX may possess improvement effect on the intestinal flora and Anti-inflammatory effect on the bowel.

Ethanol Fermentation of Hemicellulose Hydrolyzate Using High-Level Inocula of a Pachysolen tannophilus NRRL Y-2460 (Pachysolen tannophilus의 고동도 Inocula를 이용한 Hemicelluose hydrolyzate의 알코올 발효)

  • High-level yeast inocula was investigated as a means of overcoming the inhibition problem in ethanol fermentation of hemicellulose hydrolyzate. When the inoculum exceeded 25g dry cells/liter, the fermentation proceeded completely to the end within 24 hours. Furfural was taken up by Pachysolen tannophilus and catabolized to furfuryl alcohol. Thus inhibitory effect of furfural component was less adverse toward ethanol production than that of non-furfural components in hemicellulose hydrolyzate. The specific ethanol productivity in the fermentation of hemicellulose hydrolyzate was 14% of that of simulated media containing 41.8g xylose and 2.3g furfural per liter.

Requirement of Bni5 Phosphorylation for Bud Morphogenesis in Saccharomyces cerevisiae

  • In budding yeast, G2/M transition is tightly correlated with bud morphogenesis regulated by Swel and septin that plays as a scaffold to recruits protein components. BNI5 isolated as a suppressor for septin defect is implicated in septin organization and cytokinesis. The mechanism by which Bni5 regulates normal septin function is not completely understood. Here, we show that Bni5 phosphorylation is required for mitotic entry regulated by Swel pathway. Bni5 modification was evident from late mitosis to G1 phase, and CIP treatment in vitro of affinity-purified Bni5 removed the modification, indicative of phosphorylation on Bni5. The phosphorylation-deficient mutant of BNI5 (bni5-4A) was defective in both growth at semi-restrictive temperature and suppression of septin defect. Loss of Bni5 phosphorylation resulted in abnormal bud morphology and cell cycle delay at G2 phase, as evidenced by the formation of elongated cells with multinuclei. However, deletion of Swel completely eliminated the elongated-bud phenotypes of both bni5 deletion and bni5-4A mutants. These results suggest that the bud morphogenesis and mitotic entry are positively regulated by phosphorylation-dependent function of Bni5 which is under the control of Swel morphogenesis pathway.

Production of High Viscous Hyaluronic Acid Complex from Klebsiella sp. L-10 NTG 50 (Klebsiella sp. L-10의 NTG 50 변이주에 의한 고점성 히아루론산 복합체의 생산)

  • Klebsiella sp. L-10 was treated with physical and chemical mutagens, and one of the NTG mutant which increased hyaluronic acid complex yield 2.5 folds was selected. The yield of hyaluronic acid complex from Klebsiella sp. L-10 NTG 50 mutant reached maximum level I the YPD medium containing 0.1% yeast extract, 3% Bacto-tryptone, 3% dextrose, each 30mM of $K_2HPO_4$ and $KH_2PO_4$ (pH 6.0-6.5) with shaking culture at $37^{\circ}C$ for 24 hrs, and 2900mg of hyaluronic acid complex per litre of culture was produced under the above condition.

Effect of Ginseng Saponin Fractions on Phagocytosis and Chemotaxis of Murine Macrophages (대식세포의 식세포활동과 화학주성에 대한 인삼분획물의 영향)

  • The phagocytosis and chemotaxis of murine macrophages after treated with saponin fractions are investigated. Phagocytic appearance against yeast was photographed by dying with Wright-Giemsa. Phagocytic activity of peritoneal macrophage was invreased in diol saponin treatment by 48% and was decreased in total saponin treatment by 35%. The ingestion of alveolar macrophage was increased by 50% maximally. Peritoneal chemotactic activity was shown in 17% increases and only diol saponin had effect in alveolar macrophage by 16%. According to SDS-PAGE method the contents of actin did not show any alterations.

Stability of Monascus Pigment Produced by Monascus sp. MK2 (Monascus sp. MK2 균주가 생산하는 홍국색소의 안정성)

  • The stability of monascus pigment produced by Monascus sp. MK2 was investigated according to light, pH, temperature, organic acid, synthetic antioxidant, and natural substance. The light stability was increased (sun light < fluorescent lamp < ultraviolet rays < dark condition) when storing the monascus pigments. Although the monascus pigments were stable in the range of pH 3.0, the degradation rate of pigment was increased at pH 11.0. The stability of monascus pigment gradually decreased as temperature increased. In addition, pigment stability was increased by adding the synthetic antioxidant and natural substance.
