Analysis of Functional Compounds on the Seeds of Sorghum (Sorghum bicolor Moench) and Their Sprouts (수수의 종자와 새싹채소의 기능성분 분석)

  • This study was done to examine antioxidant compounds, antioxidative activity and chemical properties in organically cultivated sorghum seed and their sprout, and to examine as a basic data for various application methods as an organic farming product of sorghum. The crude protein and mineral contents in the sprout had 2.5 and 1.6~9.87 folds more than sorghum seed, respectively. Total polyphenol content as antioxidant was more in the sprout showing $4,135mg\;GAE\;g^{-1}$ than the seed, whereas total flavonoid content was relatively higher in the seed showing $3.86mg\;GAE\;g^{-1}$. ABTS radical scavenging activity in the seed was observed by higher level as $15.78mg\;TE\;g^{-1}$. However, The sprouts with and without natural light were no significant difference. DPPH radical scavenging activity in the seed exhibited activity of 11 folds or more compared to that of the sprout. The highest activity was detected from the seeds and the sprouts in Donganmae. Although sorghum was no significant correlations among the varieties, it showed correlation with between the light-treated condition and antioxidation.

Screening of Medicinal Plants with Antifungal Activity on Major Seedborne Disease (주요종자전염병 억제를 위한 항균성 약용식물탐색)

  • Antifungal activity on major seedborne diasease of crops was screened by the treatment of the extracts from 50 medicinal plants in vitro and in vivo. The extracts of garlic and taxus, Rheum undulatum, Achiranthes japonica, Glycyrrhiza uralensis, Oenothera lamar kiana treated with the blotting filter paper and water agar methods inhibited the growth of Pyricularia oryzae, Alternaria sesamicola, Colletotrichum gloeosporioides, and Alternaria brassicicola among the tested plants. Antifungal activities on infected seeds by soaking methods were shown even at the dilution of the extracts by 10 times. The activity was the highest in soaking seeds at $25^{\circ}C$ for 24 hours. The effect of plant extract on seed germination was not significant as compared with untreated seed. However, early growth of seedling was increased by the treatment of extracts. The extract of taxus slightly inhibited the seed germination of radish and chinese cabbage but those of Achirunthes japonica, Glycyrrhiza uralensis and Oenthfera lamarkiana showed severe damage on the seed germination and early growth of seedling.

Antioxidative and Anti-inflammatory Effects of Extracts from Different Organs of Cirsium japonicum var. ussuriense (엉겅퀴(Cirsium japonicum var. ussuriense) 부위별 추출물의 항산화 및 항염증 효과)

  • Objective: The roots, leaves, flowers, stems and seeds of Cirsium japonicum var. ussuriense are often used in treatment of human diseases such as hemorrhage, blood congestion and inflammation. Focusing our attention on natural and bioavailable sources of antioxidants and anti-inflammation, we undertook to investigate the antioxidant and anti-inflammatory properties of Cirsium japonicum var. ussuriense used as a folk medicine in Korea. Methods: The extracts of the leaves, stems, flowers, seeds and roots from C. japonicum var. ussuriense were prepared by extracting with water or 80% ethanol. Total flavonoids and polyphenols were measured by a colorimetric assay. The free radical scavenging activity of the extract was analyzed by the DPPH (1,1-diphenyl-2-picryl hydrazyl), ABTS (2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) and Griess reagent assay. An oxidative product of nitric oxide (NO), was measured in the culture medium by the Griess reaction. The level of prostaglandin $E_2$ ($PGE_2$) was measured by enzyme-linked immunosorbent assay. The expressions of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) were measured by Western blot analysis. Results: Total flavonoid and polyphenol amounts of the leaves (CLE) and flowers (CFE) showed higher than those of the seed extract (CSE), stem extract (CSTE) and roots (CRE). CLE and CFE also showed the high antioxidant activities such as DPPH, NO-like and ABTS radical scavenging activity. An antioxidant activities of these water extracts showed higher than those of 80% ethanol extracts. We investigated the anti-inflammatory effects of CLE on lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. CLE significantly suppressed the levels of the inflammatory mediators such as NO and prostaglandin $E_2$ ($PGE_2$) in dose dependant. Furthermore, the levels of iNOS and COX-2 protein expressions were markedly suppressed by the treatment with CLE extract in a dose dependent manner. Conclusions: These results suggest that CLE water extract has a higher anoxidant and anti-inflammatory activity, these properties may contribute to the oxidative and inflammatory related disease care.

Functional Cosmetic Characteristics of Momordica charantia Fruit Extract (여주 열매 추출물의 기능성화장품 소재 특성)

  • In this research, water and ethanol extract of Momordica charantia shells, fruits and seeds were tested to see possibility as natural functional cosmetic agent. Water and ethanol extract showed 69.45 mg/g and 70.87 mg/g polyphenol concentration, respectively. Momordica charantia water and ethanol extracts did not indicate cell toxicity up to $1,000{\mu}g/ml$ concentration in MTT assay. Tyrosinase inhibition effects of water and ethanol extract were lower than arbutin, however, ethanol extract showed better DOPA oxidation inhibition effect than arbutin. Elastase inhibition effects of ethanol extract displayed similar efficacy with adenosine at higher concentrations. Solution formulations (5% extract) were stable for 28 days in both extracts, however, lotion formulation (1% extract) showed considerable variation in viscosity whereas ethanol extraction indicated relative stability. In conclusion, water and ethanol extract of Momordica charantia shells, fruits and seeds indicated strong possibility for whitening and antiwrinkle functional cosmetic ingredient.

Germination Behaviors and Seed Longevities of Three Ulmus Species in Korea (한국산(韓國産) 느릅나무속 3수종(樹種)의 종자저장성(種子貯藏性) 및 발아특성(發芽特性))

  • This study was conducted to investigate temperature effect on seed germination behaviors and seed viability changes by drying periods of three Ulmus species (Ulmus davidiana var. japonica (Rehder) Nakai, U. pumila L., and U. parvifolia Jacq.) distributed in Korea. Statistical analysis showed that temperature had a significant effect on mean germination time in three Ulmus species, but its effect on germination percentage was only shown in U. parvifolia (p<0.01). U. davidiana var. japonica and U. pumila germinated relatively consistent in all temperature conditions, especially the latter showed high germination behaviors in low temperatures (10-15$^{\circ}C$). U. parvifolia germinated well in high temperature (25$^{\circ}C$), while this species rarely germinated in low temperature conditions (10-20$^{\circ}C$). The results confirm that seeds of U. pumila and U. parvifolia are orthodox seeds which can be stored under 4-8% moisture condition, and U. davidiana var. japonica seed is recalcitrant seed which is difficult to store for a long time. In conclusion, these differences of the germination behaviors for three Ulmus species seem to be adaptive regimes of their natural habitats, respectively.

Antioxidant Activity and Identification of Lunasin Peptide as an Anticancer Peptide on Growing Period and Parts in Pepper (생육시기 및 부위별 고추의 항산화력 및 항암 Lunasin peptide의 동정)

  • The non-enzymatic anti-oxidants and lunasin peptide from the extracts of the pepper were examined in order to utilize the discovery in natural products as cancer chemopreventive agents. The DPPH (1,1-diphenyl-2-picryl-hydrazyl) free radical scavenging activity on the fruit parts of the pepper was higher than that of the seed, but the difference was low. The Inhibition activity of xanthine/ xanthine oxidase in extracts of the seed was higher than that of the fruit and that of the seed on 20 days after flowering was the highest at the growing period. These were identified as fatty acids and phenolic compounds such as 1-eicosanol, palmitic acid, linoleic acid, linolenic acid and benzonitrile. The contents of fatty acids and phenolic compounds increased according to the time passing at the growing period. Peroxidase (POD) activity of the fruit at middle stage was high than that of other growing stages and that of the seed was the highest at later growing period. Though superoxide dismutase (SOD) activities in fruit were hish by passage of Slowing stage, the activity in seed was low. Lunasin was searched from seeds of the peppers by coomassie blue staining and western blot among them and we just found lunasin peptide from extracted protein of the pepper by western blot. In addition, we observed the contents of lunasin after flowering and confirmed to appear the lunasin at 35 days after flowering. We confirmed that lunasin is complex protein of maturing seeds. 100nM lunasin peptide in pepper showed inhibition effect on colony formation in $2\~12$ cells.

Antioxidative effect of cumin seeds ethanol extract using in vitro assays and bulk oil system (쿠민 종자 에탄올 추출물이 산화방지 및 유지 산화안정성에 미치는 영향)

  • This study aimed to investigate the antioxidant ability of a cumin seeds ethanol extract (CE). The DPPH and ABTS radical scavenging activities of 0.25, 0.5, and 1.0 mg/mL of CE were found to be 24.6, 41.4, and 73.4 and 14.5, 27.2, and 50.1%, respectively (p<0.05), suggesting a dose-dependent effect. Moreover, the total phenolic content of CE was $61.0{\mu}M$ tannic acid equivalent/g extract and the FRAP value was $429{\mu}M$ ascorbic acid equivalent/g extract. In 9 hours of oil oxidation, CDA and ${\rho}-AV$ was significantly reduced to 13.4 and 59.1%, respectively, at a CE concentration of 100 ppm compared with that in the control (p<0.05). Major volatile compounds of CE were found to be ${\alpha}$-pinene, 2-butenal, cyclohexene, ${\beta}$-pinene, cis-sabinene, ${\rho}$-cymene, and limonene. These results suggest that CE containing volatile compounds has excellent antioxidant ability and oxidation stability, and thus could be used as a natural antioxidant to prevent oxidation in lipid foods.

Selection of Salt Tolerant Rapeseed (Brassica napus L.) Mutant Lines Induced by Irradiation of Proton Ion Beams and Gamma Ray (양성자 및 감마선처리에 의한 유채 내염성 변이계통 선발)

  • This study was carried out to develop salt tolerant varities of rapeseed (Brassica napus L.) which can be grown in the high salty reclaimed land. The seeds of three varieties 'Naehan', 'Tammi', and 'Halla' were treated by proton ion beams and gamma rays with 0 to 2,000 Gy. For the selection of salt tolerant lines, emergence and survival rate, and growth characteristics of $M_2$ to $M_4$ generations were investigated in the Saemangeum reclaimed fields with the different salt concentrations. The lines with potential salt tolerance were selected in the $M_4$ generation and tested indoor for their growth characteristics. There was no significant changes in the soil pH for $M_4$ generation during growth period. However, soil EC was higher in early spring than sowing period (mid October). In $M_4$ generation test, the seeds of original and selected line showed high rates of emergence and survival, as determined one month after sowing. After wintering, however, the original varieties showed the significant reduction in the survival rate, while the selected lines showed a higher survival rate and good growth, leading to the completion of their life cycle. Consequently we selected 9 lines from $M_4$ generation with better performance in growth and yield. Soil EC was $2.8{\sim}4.3dS{\cdot}m^{-1}$ during $M_4$ generation growth period. The laboratory test of the lines selected from $M_4$ generation was made for their salt tolerance potential. The selected lines showed higher chlorophyll and proline contents than the original varieties. There was also no significant difference in the emergence rate of seed between the original and selected varieties. In 200 mM natural sea salt, the $N{\gamma}600-21-1-641$ line derived from 'Naehan' was the highest in growth rate, leaf chlorophyll and proline contents. $T{\gamma}800-20-2-461$ line derived from 'Tammi' didn't show significant difference in growth rate compared to original variety in 200 mM and withered in 250 mM like other lines as time passed. $H{\gamma}200-7-1-740$ line showed similar growth and chlorophyll content compared to its original variety.

Effect of Immature Rhizome Productivity according to Harvest Times-based Treatment Method for Seed Production of Gastrodia elata (천마 종자 생산을 위한 수확시기별 처리방법에 따른 자마 생산력 효과)

  • This study was investigated the effect of immature rhizome production according to harvest times-based treatment method for seed production of Gastrodia elata. The results revealed that when the tuber weight of G. elata harvested in spring (GEHS) was ${\geq}100g$, the rate of artificial fertilization, protocorm formation, and immature rhizome formation was 90.9%-94.8%, 3.1%-5.4%, and 10.1%-15.3%, respectively. When G. elata harvested in fall (GEHF) was treated at a low temperature for 4 weeks or more, the rate of artificial fertilization, protocorm formation, and immature rhizome formation was 70.4%-87.6%, 2.2%-2.6%, 8.7%-9.5%, respectively. Therefore, to produce seeds and immature rhizomes, GEHS must have tubers of more than 100 g, whereas GEHF requires breaking dormancy by low-temperature treatment for 4 weeks or more. Compared with those of GEHS, the rate of artificial fertilization, protocorm formation, and immature rhizome formation was lower in GEHF; however, it was higher than those in the natural germination state. Thus, it can be expected that G. elata can be produced throughout the year by ensuring that the seeds and immature rhizomes of G. elata are produced using a constant tuber weight and by breaking dormancy with low temperature treatment.

  • A number of sedative alkaloids were isolated from Sanjoin(酸棗仁), the seeds of Zizyphus vulgaris Lamark var. spinosus Bunge (Rhamnaceae) which is an important Chinese medicinal material used to treat insomnia and sometimes to treat sleepiness. Those compounds were designated as Sanjoinine-A, B, C, D, etc. depending on the order of increasing polarity.
