발효조를 이용한 Monascus anka의 적색소와 황색소의 생산

  • In order to develop the method for mass production of natural food colorant from Monascus anka, optimum cultivation conditions for producing red and yellow pigments by cultiva- ting the mold in a jar fermenter and their color characteristics were investigated. The mold produced red and yellow pigments both intracellularly and extracellularly. These pigments showed unique light absorption characteristics with maximum absorption of 494, 380, 506, and 388 nm for extracellular red pigment (ERP), extracellular yellow pigment (EYP), intracellular red pigment (IRP), and intracellular yellow pigment (IYP), respectively. Optimum conditions for producing red pigments were found to be temperature 30$\circ$C, initial pH 6.0, rice powder 3-5%, peptone 0.05%, magnesium sulfate 0.25%, aeration rate 0.1 vvm. Optimum temperature for producing yellow pigments was around 35$\circ$C which is higher than that of producing red pigments. The initial pH and rice powder concentration for producing yellow pigments were the same as those of producing red pigments. The higher concentration of nitrogen source and inorganic salt, aeration rate, the more the yellow pigments were produced. The optimum agitation speed was 100 - 300 rpm for pigment production.

  • The stability of monascus pigment produced by Monascus sp. MK2 was investigated according to light, pH, temperature, organic acid, synthetic antioxidant, and natural substance. The light stability was increased (sun light < fluorescent lamp < ultraviolet rays < dark condition) when storing the monascus pigments. Although the monascus pigments were stable in the range of pH 3.0, the degradation rate of pigment was increased at pH 11.0. The stability of monascus pigment gradually decreased as temperature increased. In addition, pigment stability was increased by adding the synthetic antioxidant and natural substance.

  • Studies on extraction of red pigment was performed to provide the basic information for the utilization of red pigment as s new source of natural food colorant. A bacterium isolated from marine resources were carried out the test for excretion of red pigment. One strain of a marine bacterium, KSR-97 showed a high production of red pigment on the medium of colloidal chitin, peptone-yeast extract with minerals. In physicochemical and sensory properties in aqueous solution of red pigment was investigated at various condition of pH, temperature, concentration of ethanol and stability of storage. Potent antioxidative of red pigment was separated by thin layer chromatograpy, silica gel chromatography and reverse high performance liquid chromatography using ODS hypersil column.

Stability of Monascin Pigment Isolated from Monascus purpureus (홍국의 황색색소 Monascin에 대한 안정성 연구)

  • The stability of monascin yellow pigment isolated from Monascus purpureus was determined over a period of storage for the wide range of pH, various metal ions and antioxidants. The absorption maximum of monascin pigment was 385 nm. Monascin pigment was more stable in acid solutions than in alkaline (pH 9 and pH 11) during storage period. It was also observed the reduction of absorption was occur after 3 days storage. The stability of monascin pigment was not changed by adding the various metal ions of the concentration of 10-4 M, however, it was unstable by adding the Zn2+, Al3+ and Fe3+ of 103- M concentration. The antioxidants. BHA, BHT, cysteine and L-ascorbic acid, have no effects on the stability of monascin yellow pigment. Thus, it may be concluded that the monascin pigment is stable and useful food additives as the natural colorant except for the alkaline food and food containing the Zn2+, Al3+ and Fe3+.

  • Study on extraction and color characteristics of mulberry fruit pigment(C3G; cyanidin-3-glucoside) was performed to increase utilization as new source of natural food colorant. C3G was extracted with 0.1 % citric acid-70% EtOH. Then it was evaporated with large scale evaporation system. After adding dextrin to C3G concentration materials, we made pigment powder with freezing dryer.

  • Natural colorant extracts were obtained by extraction from tumeric root, gardenia seeds, mugwort and green tea using water, methanol, ethanol and acetone as extractants at room temperature for 3 hours under shaking condition(180rpm) with liquor ratio(solid:solvent; 1:100). The main pigment components of tumeric root, gardenia seeds, mugwort and green tea are known to be curcumin, crocin, chlorophyll b and epigallocatechin gallate, respectively. The effects of the kind of extractant and pH on the color characteristics of natural colorant extracts were investigated. The solubility parameters of pigment components were determined to find adequate extractant. The solubility parameters of curcumin, crocin, chlorophyll b and epigallocatechin gallate were found to be 27.85, 29.40, 19.48 and $37.97(J/cm^3)^{1/2}$. As expected, solvents that have a solubility parameter similar to that of pigment component were generally found to be effective to obtain pigment extracts having high visible absorbance(A). The extract(pigment/solvent) with high visible absorbance was generally found to have low $L^*$(lightness) and high Chroma($C^*$, purity).

  • The optimum cultural conditions for production of Monascus natural pigment by Monascus purpureus MK2 were investigated in submerged culture. This strain was showed the maximum production of monascus natural pigment in the optimal medium of 3.0% wheat flour, 0.15% $NaNO_3$, 0.2% $K_{2}HPO_4$ and 0.05% $MgSO_4{\cdot}7H_{2}O$, at pH 7.0. The maximum production of this pigment was achieved at $30^{\circ}C$ for 7 day cultivation under 130 rpm shaking. At optimal condition, Monascus purpureus MK2 produced 29.10, 36.84 and 48.92 units of yellow, orange and red pigment, respectively.

Antioxidant Activity According to Each Kind of Natural Plant Pigments (식물유래 천연색소의 항산화 활성)

  • The objective of this study was to determine the effect of antioxidant enzyme activity and radical scavenging activities of thirteen kinds of natural plant pigments. The analytic method of antioxidant activities were measured by estimating DPPH free radical scavenging and nitrite scavenging ability activity. The free radical scavenging activity by method using stable free radical DPPH was the highest in the red cabbaged pigment. Addition of ethanol extract 1mg/ml from onion peel pigment displayed remarkable effect on nitrite scavenging ability about 91.9%. Antioxidative enzyme activity was evaluated in terms of superoxide dismutase(SOD), catalase(CAT), and ascorbate peroxidase(APX) activity. The bitter melon pigment had the highest SOD activity of 87.3%. The activities of CAT and APX were higher in the mulberry leave pigment compared with other natural plant pigments. In contrast, CAT activity of plant pigment samples were unaffected. These results suggest that natural plant pigment had the potent biological activities such as antioxidant enzyme activities, and that their activities exhibited differently depending on each kind of pigments.

  • Each pigments were obtained by ethyl alcohol extraction method, blue and yellow pigment from Gardenia as well as dark brown pigment from Kaoliang. Concentration of these pigments are all 60 Brix, the extraction yields were 0.68, 1.97, 0.63 %(w/w), respectively. Oil soluble natural black pigment (OSNBP) was composed of soybean oil, water, emulsifier, Gardenia blue and yellow, Kaoliang dark brown etc. Blending ratio of these was 8: 22: 42: 10: 15: 13 (w/w), this mixture was carried out homogenized. Solubility of this OSNBP in soybean oil was appeared the maximum level at about 30∼40$^{\circ}C$ range. OSNBP solubilized black oil was not reseparated at below 20$^{\circ}C$.

  • This study is that of providing a fairly practical practival guide to the use of natural pigment in food industry. A strain isolated from marine resources was carried out the production of red pigment. The pigment showed UV absorption maxima at 520 and 550 nm. The color intensity in aqueous solution was fairly stable in the ranges of pH 5~8. The strain KS-97 produced a maximum yield of red pigment at$ 25^{\circ}C$ for 72 hrs with pH 7.0. The strain KS-97 was iden-tified a Bacillus sp. based on morphological and biochemical characterization such as a rod from, motility, spore for-mation, Gram positive and catalase production. The production of red pigment indicated that the strain Ks-97 utilized at thigh concentration of colloidal chitin as carbon sources obtained maximum yield of red pigment at $25^{\circ}C$ for 72 hrs. The highest production of red pigment was observed with cultivation in medium containing 20% colloi-dal chitin, 2.5g polypeptone, 2.5g yeast extract, 1.0g $KH_2$$PO_4$, 0.01g $MgSO_4$.$6H_2$O, 0.01g $ZnSO_4$, 0.01 g $MnSO_4$(per 1).
