Immunostimulating Activity of Polysaccharides from Mycelia of Phellinus linteus Grown under Different Culture Conditions

  • Polysaccharides were extracted from mycelia of Phellinus linteus grown under different culture conditions. The in vitro immunostimulating activity was measured by plaque-forming cell (PFC) assay. The activity of the polysaccharides was different from that of mycelia from which was extracted. The number of PFC's ranged from 40 to 600 depending on the media. When P. linteus was cultured on a medium with mannose or starch as a sole carbon source, the fungus produced polysaccharide with the highest activity of 960 PFC. Activity was therefore increased by $50%$ compared with polysaccharide which was extracted from mycelia grown on medium with glucose. pH had little effect on the change in activity. All polysaccharides on media with different pH stimulated about 600 PFC. These results suggest that activity could be increased by polysaccharide modification through changes in physiological conditions.

  • 파(Allium fistulosum L.)의 자방을 0.5mg/L NAA와 0.5mg/L BA 또는 0.5mg/L kinetin을 조합처리한 MS배지에서 배양하였을 때, 자방의 표면으로부터 화아발생 캘러스가 유도되었다. 화아발생 캘러스의 유지는 0.5mg/L NAA와 0.5mg/L kinetin이 첨가된 배지에서 계대배양 하였을 때 양호하였다. 배양 3-4주 후 화아들은 화아발생 캘러스로부터 형성되었지만, 계대배양을 거듭할수록 화아의 발생률이 감소하였다. 화아의 발생 단계별 조직학적 관찰 결과 돔형의 분열조직이 넓게 퍼지면서 자연상태의 꽃과 같이 외측에 화판, 내측에 심피와 배주가 기원된 후 성숙한 꽃으로 발달하였다.

  • The quantitative analyses of the phosphatase activity in the endometrium of the rat ovariectomized on Day 2 of pregnancy was carried out in comparison with the intact one, in order to investigate the hormonal dependency of the uterus prior to the implantation, and to study the phosphatase activity in the endometrial tissues in vitro incubated in different acidity of the medium. The results obtained were as follows: 1. The activity of the total phosphatase was the highest at Day 3 of pregnancy of the intact animals irrespective of acidity of the medium. However, the ovariectomized rat showed its peak somewhat delayed. The time of the highest activity of the enzymes was matched with the time of high secretion of the ovarian hormones. 2. The activity of acid phosphatase in the endometrium was twice or four times as much high as that of neutral or alkaline phosphatase, respectively. 3. The activity of alkaline phosphatase was rather steady in Day 3 through Day 5 of the pregnancy of the rat intact or ovariectomized but with low level compared to those of other phosphatase. 4. The present re~lt indicated more important role by $Mg^{2+}$-dependent phosphatase than by $K^+$-dependent one for the preparation for decidualization.

Isolation of Bacillus sp. SHL-3 from the Dry Soil and Evaluation of Plant Growth Promoting Ability

  • Excess use of chemical fertilizer causes soil acidification and accumulation of salt, and thus might bring to desertification of soil. To overcome this problem, it needs limited usage of chemical fertilizer and increased usage of natural fertilizer as an alternative. In this study, dry soil-borne Bacillus sp. SHL-3, which was isolated from arid and barren soil, with plant growth promoting activity was isolated for identification and to determine optimal culture condition. A bacterial strain SHL-3 had the IAA productivity ($5.16{\pm}0.10mg\;L^{-1}$), ACC deaminase activity ($0.36{\pm}0.09$ at 51 hours) and siderophore synthesis. It was identified as genus Bacillus sp.. Also, optimal culture condition of SHL-3 were $20^{\circ}C$ and pH 7 in LB medium. Bacillus sp. SHL-3 had up to 4% salt tolerance in the medium. We evaluated the plant growth promotion ability of SHL-3 using yam (Dioscorea japonica Thunb.). As a result, Bacillus sp. SHL-3 was effective on the increase of the shoot length (202.4% increase for 91 days). These results indicate that Bacillus sp. SHL-3 can serve as a promising microbial resource for the biofertilizers of soil.

Modern Cause and Effect Model by Factors of Root Cause for Accident Prevention in Small to Medium Sized Enterprises

  • Background: Factors related to root causes can cause commonly occurring accidents such as falls, slips, and jammed injuries. An important means of reducing the frequency of occupational accidents in small- to medium-sized enterprises (SMSEs) of South Korea is to perform intensity analysis of the root cause factors for accident prevention in the cause and effect model like decision models, epidemiological models, system models, human factors models, LCU (life change unit) models, and the domino theory. Especially intensity analysis in a robot system and smart technology as Industry 4.0 is very important in order to minimize the occupational accidents and fatal accident because of the complexity of accident factors. Methods: We have developed the modern cause and effect model that includes factors of root cause through statistical testing to minimize commonly occurring accidents and fatal accidents in SMSEs of South Korea and systematically proposed educational policies for accident prevention. Results: As a result, the consciousness factors among factors of root cause such as unconsciousness, disregard, ignorance, recklessness, and misjudgment had strong relationships with occupational accidents in South Korean SMSEs. Conclusion: We conclude that the educational policies necessary for minimizing these consciousness factors include continuous training procedures followed by periodic hands-on experience, along with perceptual and cognitive education related to occupational health and safety.

Adventitious root culture for secondary metabolite production in medicinal plants: A Review

  • Medicinal plants are high-value natural resources that have been used as precautionary drugs by many people globally. The increasing global demand for bioactive compounds from medicinal plants has led to the overexploitation of many valuable species. One widely used approach to overcome this problem is the use of adventitious root cultures as a propagation strategy. This review examines the scientific research published globally on the application of adventitious root cultures for many medicinal plants. Adventitious roots generated under aseptic environments in suitable phytohormone-augmented medium exhibit high growth rates and production of important secondary metabolites. Parameters such as medium properties and composition, growth hormone type, and elicitation strategies for in vitro grown adventitious roots of medicinal plants, are the main topics discussed in this review. We also examine current developments in bioreactor system cultivation for plant bioactive compounds using adventitious root cultures, a technology with possible commercial applications, via several studies on adventitious root culture of medicinal plants in which bioreactor systems play a role. In conclusion, the development of adventitious root cultures for medicinal plants is highly useful because of their capability for vegetative propagation and germplasm preservation.

Free vibration analysis of nonlocal viscoelastic nanobeam with holes and elastic foundations by Navier analytical method

  • This manuscript is dedicated to deriving the closed form solutions of free vibration of viscoelastic nanobeam embedded in an elastic medium using nonlocal differential Eringen elasticity theory that not considered before. The kinematic displacements of Euler-Bernoulli and Timoshenko theories are developed to consider the thin nanobeam structure (i.e., zero shear strain/stress) and moderated thick nanobeam (with constant shear strain/stress). To consider the internal damping viscoelastic effect of the structure, Kelvin/Voigt constitutive relation is proposed. The perforation geometry is intended by uniform symmetric squared holes arranged array with equal space. The partial differential equations of motion and boundary conditions of viscoelastic perforated nonlocal nanobeam with elastic foundation are derived by Hamilton principle. Closed form solutions of damped and natural frequencies are evaluated explicitly and verified with prestigious studies. Parametric studies are performed to signify the impact of elastic foundation parameters, viscoelastic coefficients, nanoscale, supporting boundary conditions, and perforation geometry on the dynamic behavior. The closed form solutions can be implemented in the analysis of viscoelastic NEMS/MEMS with perforations and embedded in elastic medium.

Molecular Cloning and Expression of the $\beta$-Xylosidase Gene (xylB) of Bacillus stearothermophilus in Escherichia coli

  • The second $\beta$-Xylosidase gene (xylB) from Bacillus stearothermophilus was isolated from the genomic library, cloned into pBR322, and subsequently transferred into Escherichia coli HB101. Six out of 10, 000 transformants were selected from the selective LB medium supplemented with p-nitrophenyl-$\alpha$-L-arabinofuranoside (pNPAf) and ampicillin ($50\mu g$/ml) based on their ability to form a yellow ring around the colony. One of the clones was found to harbor the recombinant plasmid with 5.0 kb foreign DNA, which was identical to the $\alpha$-L-arabinofuranosidase gene (arfI) previously cloned in this lab, while the other five had 3.5 kb of the foreign DNA. Southern blotting experiments confirmed that the 3.5 kb insert DNA was from B. stearothermophilus chromosomal DNA. A zymogram with 4-methylumbelliferyl-$\alpha$-L-arabinofuranoside as the enzyme substrate revealed that the cloned gene product was one of the mutiple $\alpha$-L-arabinofuranosidases produced by B. stearothermophilus. Unlike the arfI gene product, the product of the gene on the insert DNA (xylB) showed an activity not only on pNPAf but also on oNPX suggesting that the cloned gene product could be a bifunctional enzyme having both $\alpha$-L-arabinofuranosidase and $\beta$-xylosidase activities.

공간적 접근성 및 통행비용을 고려한 천연가스 충전소 최적 입지선정 모형 (A Model of Location Decisions of Natural Gas Filling Station Considering Spatial Coverage and Travel Cost)

  • 본 연구에서는 천연가스버스 충전소 위치 선정의 객관성과 효율성을 확보하기 위한 노력의 일환으로 공간적 접근성과 통행 비용을 고려한 천연가스 충전소 입지선정 모형을 개발하였다. 본 연구에서 제시한 충전소 입지선정 모형은 고정식 충전소를 대상으로 하였으며, 2단계로 구성되었다. 1단계에서는 천연가스버스의 노선 기종점으로부터 이동거리 제약조건을 만족하는 충전소 입지 대안들을 Heuristic Algorithm을 통해 선정하였으며, 모형의 2단계에서는 변형된 Transportation Problem을 이용하여 충전소 입지대안별 최소 통행비용을 산출하고 이를 충전소의 설치 및 운영비용과 비교분석하여 충전소 최적 입지대안을 도출하였다. 본 모형의 적용가능성 분석을 위해 수도권내의 대표적 중소도시인 안양시를 대상으로하여 천연가스버스 수요량에 따른 충전소의 입지와 규모를 산출하였다. 본 연구를 통해 객관적이고 합리적인 천연가스 충전소 입지선정 방법론의 기틀을 제시함으로써 향후 천연가스버스의 원활한 보급에 기여할 수 있을 것으로 판단된다.

Culture Parameters for Nonactin Production by Streptomyces viridochromogenes JM-4151

  • Nonactin is the parent compound of a group of ionophore antibiotics, that known as the macrotetrolides. In previous report, in th course of screening superoxide radical-generating compounds from microbial sources, we first screened Streptomyces viridochromogenes JM-4151 that produces nonactin. It was proved that nonactin is superoxide radical-producing compound. In present study, we examined the optimal culture conditions of nonacin. Th optimal culture conditions for nonactin production were as follows: 1% soluble starch, 1% yeast extract, 0.2% ammonium nitrate, 0.06% magnesium sulfate, 0.2% calcium carbonate, initial pH 7.0 at 28$^{\circ}C$ for 96 h. The highest nonactin production was achieved in the production medium of initial pH7.0 at 28$^{\circ}C$ for 96h. The threshold level of dissolved oxygen was found to be above 33.2% at 28$^{\circ}C$ when 1% soluble starch was used as a carbon source. These results suggest that S. viridochromogenes JM-4151 might be a possible strain for industrial nonactin producer.
