In-situ high-pressure and ex-situ high temperature-pressure experiments of natural beryl ($Be_3Al_2Si_6O_{18}$, P6/mcc) from two different localities (beryl-A and beryl-B) were studied using pure water as pressure transmitting medium. Compared to the previous study using a mixture of methanol:ethanol medium in 4 : 1 by volume, pressure- and temperature-induced chemical and structural changes under water medium are expected to be different. The derived bulk moduli are 111(7) GPa, $K{_0}^{\prime}=73(7)$; 110(9) GPa, $K{_0}^{\prime}=65(8)$ for beryl-A and beryl-B, respectively. We observe densifications in volume compression, which appear to be attributed to the phase transitions of water to ICE VI and ICE VII around 1.0 GPa and 2.5 GPa, respectively.
The antifungal activity of ethylenediaminetetraacetic acid (EDTA), an antioxidative food preservative, was investigated against Saccharomyces cerevisiae. At the general condition for antifugal test, EDTA exhibited the minimum inhibitory concentration (MIC) of 200 ${\mu}g/ml$ and the minimum fungicidal concentration (MFC) of 6,400 ${\mu}g/mL$, As seen with the antibacterial activity, the antifungal activity of EDTA was greatly decreased by high inoculum size, acidic medium, and cation ($Ca^{++},\;or\;Mg^{++}$) added to medium. On the other hand, when EDTA was combined with polygodial isolated from natural food spice, they showed strong synergism on the antifungal activity. Further, the diminishing antifungal activity of EDTA by high inoculum size, acidic medium, and cation ($Ca^{++}\;or\;Mg^{++}$) added to medium was considerably improved by the combination with polygodial.
We have developed a reliable and high-frequency genetic transformation and regeneration system via somatic embryogensis of Eleutherococcus sessiliflorus. Embryogenic callus obtained from seed were co- cultivated with Agrobacterium tumefaciens strain EHA101/pIG121Hm harboring genes for intron-$\beta$-glucoronidase(GUS), kanamycin and hygromycin resistance. Following co-cultivation, two types of samples(fine embrogenic calli and early globular embryo clusters) were cultivated on Murashige and Skoog(MS) medium containing 1 mg/L2.4-D for 3day in dark. Transient expression of GUS gene was found to be higher in the early globular embryo clusters than in the embryogenic calli. Also, co-cultivated period affected expression of GUS gene; the best result was obtained when globular embryo clusters were co-cultivated with Agrobacterium for 3 days. Subsequently, this callus transferred to selective MS medium containing 1mg/L2.4-D, 50mg/L kanamycin or/and 30mg/L hygromycin and 300mg/L cefortaxime. These embryogenic calls were subcultured to the same selection medium at every 2 weeks intervals. Approximately 24.5% of the early globular embryos co-cultivated with Agrobacterium for 3days produced kanamycin or/and hygromycin-resistant calli. Transgenic somatic embryos were converted into plantlets in half strength MS medium supplemented with 3mg/L GA$_3$ kanamycin and were confirmed by GUS histochemical assay and polymerase chain reaction analysis. Genomic Southem blot hybridization confirmed the incorporation of NPT II gene into the host genome.
The strain DGUM 5051, an antagonistic bacterium against apple-bitter rot causing Glomerella cingulata, was isolated from soil in Kyongju. Based on the morphological and physiological characteristics, the bacterium was identified as Pseudomonas sp. and named as Pseudomonas sp. DGUM 5051. The optimal pH and temperature for cell growth were pH 6.0 and $30^{\circ}C$, whereas those for antifungal activity were pH 7.0 and $24^{\circ}C$, respectively. Among the complex media tested, brucella medium, brain heart infusion medium and Luria-Bertani medium were good for both cell growth and antifungal activity. The high antifungal activity was found in the mineral salts medium, in which sucrose, $KNO_3$ and $K_2HPO_4$ were used as sources of carbon, nitrogen and phosphorus, respectively.
Rauvolfia serpentina (L.) Benth. ex Kurz is a medicinal plant and an endangered tropical rainforest plant species. Since the field cultivation that aims to fulfill the industrial needs is never accomplished, tissue culture appears to be the most feasible way to improve the quality and quantity of R. serpentina. This experiment used two kinds of explants (roots and shoots) to induce optimal root formation in different combinations of auxin and photoperiod. Each explants exhibited different responses on given treatments. Differentiated root could be produced from explants cultured in IBA 20 mg/L with and without light. The highest number of roots, root length and root weight induced from shoot explants were effective on MS medium containing IBA 20 mg/L and incubated under dark condition, while highest total weight (callus and root) from root explants cultured on MS medium supplemented 10 mg/L IBA and 10 mg/L NAA and incubated under day length (11/13 hr). The root induced from shoot explants produced the highest major alkaloid content. The highest content of ajmaline (2.17 ppm fresh weight) and reserpine (1.30 ppm fresh weight) were observed in shoot explants cultured in MS medium containing combination of IBA 10 mg/L and NAA 10 mg/L and incubated under dark condition, yohimbine (1.47 ppm fresh weight) was in the shoot explants cultured in MS medium containing NAA 20 mg/L and incubated under day length, while serpentine was absent.

The production of pullulan by Aureobasidium pullulans HP-2001 was investigated under various ratios of glucose as carbon source and yeast extract as the nitrogen source, Highest conversion rate (productivity) of glucose to pullulan was 40.0% when concentrations of glucose and yeast extract were 5% and 0.15%, respectively. Maximal production of pullulan was 29.3g/1 when the concentration of glucose was 8%(w/v) and that of yeast extract was 40:1. On basis of the result that production of pullulan was found in a medium which concentration of glucose as carbon source was up to 20%(w/v), Aureobasidium pullulans HP-2001 seemed to overcome the catabolite repression. Conversion rate of pullulan from 20%(w/v) of glucose was 11.1%.
We examined effects of water temperature, salinity, irradiance, and different media on the growth of the toxic dinoflagellate Alexandrium tamarense (HYM9704), which was isolated from Jinhae Bay, Korea. The ranges of temperature and salinity in which the strain was able to grow were $10{\sim}20^{\circ}C$ and $20{\sim}34$ psu, respectively. These values were in accordance with those observed in situ. The maximum growth rates of axenic A. tamarense (HYM9704) was $0.25d^{-1}$ at $15^{\circ}C$, 30 psu, and $100{\mu}Em^{-2}s^{-1}$. The temperature affected the growth rates of axenic A. tamarense more significantly than the salinity. The type of culture media did not affect the growth rates of axenic A. tamarense. The strain in N-limited and P-limited media went into the stationary phase faster than that in T1 and T1/2 medium.

A numerical study on the melting process inside an isothermal spherical capsule is made. It is assumed that the phase change medium of its solid phase is heavier than the liquid phase and therefore the unmelted solid core is continuously moving downward on account of gravity forces. Such a gravity-assisted melting is commonly characterized by the existence of a thin liquid film below the solid core. The present study is motivated to present a full-equation-based analysis of the influences of the initial subcooling and the natural convection on the fluid flow and heat transfer characteristics associated with the gravity-assisted melting. In the light of the solution strategy, the present study is substantially distinguished from the existing works in that the complete set of governing equations in both the melted and unmelted regions are resolved without subdivision of the solution domains. For example, the liquid film region and the upper melted region are treated here as one domain and thus obviating laborious efforts to couple them. Numerical results are obtained by varying the Rayleigh numbers and the degree of subcooling. For the range of parameters examined, the presence of subcooling was found to impede the melting rate. The dropping velocity of the unmelted solid core was observed to affect the natural convection in the liquid significantly. When compared with the available experimental data, much improved prediction was achieved.
To investigate the role of induction on CGTase production for alkalophilic Bacillus firm us var. alkalophilus H609, the constitutive mutants that form a halo around its colonies at non-inducible AG agar media containing amylose and glucose were selected. The selected constitutive mutants could produce CGTase in the range of 18.9 to 28.8 units/ml $\cdot A_{600}$ in the alkaline basal medium, and finally a constitutive mutant Bacillus firmus var. alkalophilus CM46 was selected. The constitutive nature of CM46 was also confirmed in protein level using SDS-PAGE. The effects of induction and catabolite repression for both parent strain Bacillus firmus var. alkalophilus H609 and constitutive mutant CM46 were also compared by adding soluble starch and glucose during cultivation. The selected mutant CM46 was a non-inducible but a catabolite regulated type mutant. Even though inductive regulation was released, the specific CGTase activity defined as CGTase activity per cell concentration was not increased compared with that of parent strain. The cell growth and CGTase production patterns of constitutive mutant Bacillus firmus var. alkalophilus CM46 were compared with the parent strain to identify CGTase production characteristics.
