Early Responses of Planted Quercus serrata Seedlings and Understory Vegetation to Artificial Gap Treatments in Black Locust Plantation (아까시나무림에서 인공 숲틈 처리에 대한 졸참나무 식재목 및 하층식생의 초기 반응)

  • Black locust (Robinia pseudoacacia) stand is representative lowland exotic plantation with low ecological quality and arrested succession in South Korea. To facilitate succession and restore natural vegetation, small canopy gaps (${\sim}57m^2$), which can modify minimally structural variables and reduce restoration related disturbances on stand, was established in the black locust stand, and oak (Quercus serrata) seedlings were introduced in the gap. Two types of varying levels were introduced for gap creation; cutting (C) and girdling (G) on canopies. Understory removal (CU and GU) treatment was applied as subtypes of structural modification. Growth (diameter, height and leaf area) of target species and responses (species composition, diversity and coverage) of understory community were monitored during study years (2007~2008). Canopy openness was different significantly among treatments but not for light availability. Based on the result of logistic regression, growth of height and leaf area of seedlings were significant variables on seedling survival. Height and leaf area of seedlings were increased during study years, although radial growth was reduced. During study years, there were no significant differences in species composition and diversity, and total coverage increased about 20%. Increase of resources by gap creation and understory removal likely affect growth of target species. Small gap creation was effective to reduce understory responses in composition and diverstiy. Synthesized, growth of target species and responses of understory community to small canopy gap creation exhibited, in short term, possibility of utilization in alternative forest restoration and management option. Long-term monitoring is necessary to certificate effect of artificial gap creation on forest restoration.

A Study on Antioxidant and Anti-inflammatory Effects Based on Analysis of Functional Components of Cornus officinalis Siebold & Zucc. (산수유의 채취 부위에 따른 기능 성분 분석과 항산화 및 항염증 효과에 관한 연구)

  • Cornus officinalis Siebold & Zucc. is traditionally used as an edible and medicinal plant in many countries in East Asia. Previous studies have shown the pharmacological potential of extracts and components of C. officinalis, but comparative analysis of the composition of the leaf, stem, and fruit extracts has been insufficient to date. In the present study, the content of active antioxidant and anti-inflammatory ingredients was verified in different C. officinalis parts (under-ripe sansuyu, ripe sansuyu, seed, leaf, stem, and dried sansuyu). One active component, morroniside, was high in fruit (under-ripe and ripe sansuyu), while loganin was high in fruit (under-ripe sansuyu) and cornin was high in seeds. Total polyphenol contents were highest in fruit (ripe sansuyu) and flavonoids were highest in leaves. DPPH radical scavenging activity was highest in leaves, followed by seeds and then ripe sansuyu extract. The anti-inflammatory efficacy of leaf extracts of C. officinalis (LCO) was further investigated by measuring their effects on levels of nitric oxide (NO) and the pro-inflammatory cytokines interleukin (IL)-1β and IL-6 in RAW 264.7 macrophages. Non-cytotoxic concentrations of LCO effectively decreased the lipopolysaccharide (LPS)-induced expression of inducible NO synthase, resulting in decreased NO production. LCO also significantly suppressed LPS-induced production and expression of IL-1β and IL-6. Taken together, the present findings suggest that C. officinalis leaves have potential as natural materials for the development of antioxidant and anti-inflammatory agents.

Sex Ratios, Size and Growth Variation, and Spatial and Age Distribution between the Sexes in Natural Populations of Three Species of Dioecious Rhus(Anacardiaceae) (자웅이주성(雌雄異株性) 옻나무속(屬) 3수종(樹種)의 자연집단(自然集團)에서 성비(性比)와 성간(性間) 생장량(生長量) 및 공간적(空間的) 분포(分布))

  • Sex ratios, and patterns of tree size and growth variation, resource allocation, spatial and age class distribution between the sexes were investigated in natural populations of the sexual trees, R. trichocarpa and R. sylvestris, and the mainly asexual, clonal tree, R. javanica of the dioecious Rhus (Anacardiaceae) distributed in Korea. Sex ratios for three species exhibited a significant degree of female bias, but among the populations, sex ratios were seen to vary quite widely. The measurement of tree size and annual increment of male trees in R. trichocarpa and R. sylvestris were significantly higher than those of female trees, but not significantly different in R. javanica. In all of the species, flowering branch number per individual and inflorescence number per branch of males outnumbered those of females. Branch number per individual, rachis(leaf) number per branch and rachis(leaf) number per inflorescence were more in females than in males. These results were considered as reproductive efforts to increase the pollen supply in males and the fruit production in females. Spatial distribution analysis in two different populations of R. trichocarpa indicated that males and females were randomly distributed in space, but seedlings were clumped around parental trees. Analysis of age class distributions between the sexual reproduction trees, R. triclaocarpa and R. sylvestris, and the asexual, clonal tree, R. javanica showed a different distribution in frequencies of males and females in each age class. These results showed that sexual and asexual reproduction tree species had almost different preference of habitats, and different sex ratio and annual growth.

Development of the Trichomes in Floating Leaves of Salvinia Species (생이가래속(Salvinia) 부유엽 모용의 분화발달)

  • Salvinia is an aquatic plant forming dimorphic leaves that have been modified into floating and submerged leaves. A air of floating leaves plays an important role for the floating and photosynthesis while the submerged leaves, which are lim and long, have the form and function of root. Many aquatic plants develop trichomes in the epidermis but in Salvinia, richomes grow densely in the epidermis of the dimorphic leaves. The present study examined the differentiation pattern of trichomes developing in the floating leaves of S. natans and S. molesta by scanning and transmission electron microscopy. Trichomes developing in the floating leaves of Salvinia showed very different patterns. In S. natans, they were arranged in a V-shape form, having 20${\sim}$25 rows at $18{\sim}25^{\circ}$ on both sides of the lamina divided by the midrib in the floating leaf. In each row, 8${\sim}$10 oval-shaped cells, $200{\sim}290{\mu}m$ in length, were arranged in a spiral fashion. Four trichomes of this form made a trichome unit, but their apical parts were separated from one another and developed into the so-called 'knuckle-crane' type. On the other hand, in S. molesta, trichomes differentiated in a unique pattern quite different from those of S. natans. At the early stage of differentiation, trichomes protruded from the epidermis and then 4${\sim}$6 cylindrical cells grew $400{\sim}600{\mu}m$ long and the four trichomes formed as an unit. The four grouped trichomes were interconnected through their apex and developed in the 'egg-beater' type. Then $300{\sim}600{\mu}m$ long multi-cellular stalk cells grew and protruded out of the epidermal surface from the basal part of the trichomes. Such a structural characteristic of trichomes is considered to play a very important role along with the aerenchyma tissue in the leaf mesophyll tissue for the floating of Salvinia on the water surface.

Antioxidant and Anti-inflammatory Effects of Ficus erecta var. sieboldii Leaf Extract in Murine Macrophage RAW 264.7 Cells (좁은잎천선과나무(Ficus erecta var. sieboldii) 잎 추출물이 대식세포 RAW 264.7 세포에서 미치는 항산화 및 항염증 효과)

  • In this study, a preliminary evaluation of the antioxidant and anti-inflammatory activity of the Ficus erecta var. sieboldii (Miq.) King (FES) leaf extract has been performed to assess its potential as a natural resource for food and medicinal materials. FES was extracted using 70% EtOH and then fractionated sequentially using n-hexane, $CH_2Cl_2$, EtOAc, and n-BuOH. To screen for antioxidant and anti-inflammatory agents effectively, the inhibitory effect of the FES extracts on the production of oxidant stresses (DPPH, xanthine oxidase, and superoxide) and pro-inflammatory factors (NO, iNOS, COX-2, $PGE_2$, IL-6, and $IL-1{\beta}$) in the murine macrophage cell line RAW 264.7 activated with lipopolysaccharide (LPS) was examined. Among the sequential solvent fractions of FES, the $CH_2Cl_2$ and EtOAc fractions showed decreased production of oxidant stresses (DPPH, xanthine oxidase and superoxide), and the hexane and $CH_2Cl_2$ fractions of FES inhibited the production of pro-inflammatory factors (NO, iNOS, COX-2, and $PGE_2$). The $CH_2Cl_2$ fraction also inhibited the production of pro-inflammatory cytokines ($TNF-{\alpha}$, IL-6, and $IL-1{\beta}$). These results suggest that FES has a significant effects on the production of oxidant stresses and pro-inflammatory factors and may be used a natural resource for antioxidant and anti-inflammatory agents.

The Trend and Achievements of Forest Genetics Research in Abroad (선진국(先進國)에 있어서의 임목육종연구(林木育種硏究)의 동향(動向))

  • The trend and achievements of forest genetics research in abroad were investigated through observation tours and reference work and following facts were found to be important aspects which should be adopted in the forest genetics research program in Korea. Because of world wide recognization on the urgency of taking a measure to reserve some areas of the representative forest type on the globe before the extingtion of such forest type as the results of continuous exploitations of the natural forests to meet the timber demand all over the world, it is urgently needed to take a measure to reserve certain areas of natural stand of Pinus koraiensis, Pinus parviflora, Pinus densiflora f. erectra, Abies koreana, Quercus sp., Populus sp., etc. as gene pool to be used for the future program of forest tree improvement. And the genetic studies of those natural forest of economic tree species are also to be performed. 1. Increase of the number of selected tree for breeding purpose. Because of the fact that the number of plus tree at present is too small to carry out selection program for tree improvement, particularly for the formation of source population for recurrent selection of parent trees of the 2nd generation seed orchard it is to be strongly emphasized to increase the number of plus tree by alleviating selection criteria in order to enlarge the population size of plus trees to make the selection program more efficient. 2. Progeny testing More stress should be placed on carrying out progeny testing of selected trees with open pollinated seeds. And particular efforts are to be made for conducting studies on adult/juvenile correlation of important traits with a view to enable to predict adult performances with some traits revealed in juvenile age thus to save time for progeny testing. 3. Genotype-environment interaction Studies on genotype and environment interaction should be conducted in order to elucidate whether the plus trees selected on the good site express their superiority on the poor site or not and how the environment affect the genotype. And the justification of present classification of seed distribution area should be examined. 4. Seed orchard of broad leaf tree species. Due to the difficulty of accurate comparison of growth rate of neighbouring trees of broad leaf tree species in natural stand, it is recommended that for the improvement of broad leaf trees a seedling seed orchard is to be made by roguing the progeny test plantation planted densely with control pollinated seedlings of selected trees. 5. Breeding for insect resistant varieties. In the light of the fact that the resistant characteristics against insect such as pine gall midge (Thiecodiplosis japonensis U. et I.) and pine bark beetle (Myelophilus pinipera L.) are highly correlated with the amount and quality of resin which are known as gene controlled characteristics, breeding for insect resistance should be carried out. 6. Breeding for timber properties. With the tree species for pulp wood in particular, emphasis should be placed upon breeding for high specific gravity of timber. 7. Introduction of Cryptomeria and Japanese Cypress In the light of the fact that the major clones of Cryptomeria are originated from Yoshino source and are being planted up to considerably north and high elevation in Japan, those species should be examined on their cold resistance in Korea by planting them in further northern part of the country.

Estimation of Fresh Weight and Leaf Area Index of Soybean (Glycine max) Using Multi-year Spectral Data (다년도 분광 데이터를 이용한 콩의 생체중, 엽면적 지수 추정)

  • Soybeans (Glycine max), one of major upland crops, require precise management of environmental conditions, such as temperature, water, and soil, during cultivation since they are sensitive to environmental changes. Application of spectral technologies that measure the physiological state of crops remotely has great potential for improving quality and productivity of the soybean by estimating yields, physiological stresses, and diseases. In this study, we developed and validated a soybean growth prediction model using multispectral imagery. We conducted a linear regression analysis between vegetation indices and soybean growth data (fresh weight and LAI) obtained at Miryang fields. The linear regression model was validated at Goesan fields. It was found that the model based on green ratio vegetation index (GRVI) had the greatest performance in prediction of fresh weight at the calibration stage (R2=0.74, RMSE=246 g/m2, RE=34.2%). In the validation stage, RMSE and RE of the model were 392 g/m2 and 32%, respectively. The errors of the model differed by cropping system, For example, RMSE and RE of model in single crop fields were 315 g/m2 and 26%, respectively. On the other hand, the model had greater values of RMSE (381 g/m2) and RE (31%) in double crop fields. As a result of developing models for predicting a fresh weight into two years (2018+2020) with similar accumulated temperature (AT) in three years and a single year (2019) that was different from that AT, the prediction performance of a single year model was better than a two years model. Consequently, compared with those models divided by AT and a three years model, RMSE of a single crop fields were improved by about 29.1%. However, those of double crop fields decreased by about 19.6%. When environmental factors are used along with, spectral data, the reliability of soybean growth prediction can be achieved various environmental conditions.

Growth, Rice Yield and Edible Quality of Rice under Naturally Reseeded Chinese Milk Vetch Cropping System (자운영 지속재배시 벼 생육, 수량 및 미질)

  • Growth, milled rice yield and edible quality of rice in naturally reseeded Chinese milk vetch(CMV)-rice cropping system was compared with those in rice mono cropping on silty loam soil in Milyang from 2006-2008. Practicing natural reseeding technology recorded high CMV reseeding stand ranging from 565-805 plants $m^{-2}$ and resulting in the production of 13.0-17.0 kg N/10a from the CMV plant biomass which is greater than the recommendation rate of 9 kgN/10a. The plant height of rice plant grown in natural reseeding field is shorter at tillering stage but it was similar to the rice mono cropping at later stage. Dry matter production had similar trend to plant height. On the other hand, the leaf color in naturally reseeded CMV-rice cropping system was similar to the rice mono cropping up to panicle heading stage but it was high at mature stage, indicating that the nitrogen was provided by the CMV decomposition until later stage of rice. The yield components such as culm number $m^{-2}$ was greater and 1,000-brown rice weight was heavier than those of rice mono cropping but the ripened grain ratio was lower in naturally reseeded CMV-rice cropping system. Milled rice yield of naturally reseeded CMV-rice cropping system was similar to that of rice mono cropping. However, head rice percentage of milled rice was lower due to low ripened grain ratio. This result indicates that natural CMV reseeding technology can completely replace chemical fertilizer in CMV-rice cropping system.

Effect of Foliar Sprays of CaCl2 for Improving Fruit Quality of (복숭아 )

  • To evaluate the effectiveness of the foliar spray of calcium on "Baekdo" peach fruit, we carried out experiments in the orchards. The sprays were applied with $CaCl_2\;(Ca:\;400mg.kg^{-1})\;and\;CaCl_2$ with adjuvants (amino acid, $2g.kg^{-1}$; phytic acid, $2ml.kg^{-1}$ and wood vinegar, $2ml.kg^{-1}$) for four times from June 12 through July 4 at weekly intervals. The fruits and leaves were evaluated for Ca content, firmness and incidence of Brown rot caused by Monilinia fructicola at harvest To evaluate fruit quality included Ca content, firmness natural decay during the storage, the fruits were stored at room temperature for 14 days. The Ca content in leaf and fruit flesh at harvest was significantly increased (P=0.05) in $CaCl_2$ + amino acid treatment among $CaCl_2$ treatments. However, there was not significant Ca content in fruit peel. The firmness of flesh increased significantly (P=0.05) in $CaCl_2$ + amino acid treatment. The natural decay (Rhizopus stolonifers) during storage at the room temperature for 14 days, the fruit treated with $CaCl_2$ + wood vinegar exhibited lowest (P=0.05) incidence. Also, the firmness of the fruit during storage was firmer with treated $CaCl_2$ than untreated fruit. In the treatments of $CaCl_2$ + phytic acid and $CaCl_2$ + amino acid, it was possible to reduce incidence of Brown rot caused by M. fructicola most effectively in the field. In addition, inoculation with M. fructicola in fruits was also the most effective treatment for inhibiting disease development in vitro. These results suggested that the foliar spray of $CaCl_2$ with adjuvants increased the content of Ca and firmness of the fruits positively. It also inhibited the natural decay and the Brown rot effectively.

Ethanol Extracts of Mori Folium Inhibit Adipogenesis Through Activation of AMPK Signaling Pathway in 3T3-L1 Preadipocytes (3T3-L1 세포에서 상엽이 유발하는 AMPK signaling pathway를 통한 adipogenesis 억제에 관한 연구)

  • Mori Folium, the leaf of Morus alba, is a traditional medicinal herb that shows various pharmacological activities such as antiinflammatory, antidiabetic, antimelanogenesis, antioxidant, antibacterial, antiallergic, and immunomodulatory activities. However, the mechanisms of their inhibitory effects on adipocyte differentiation and adipogenesis remain poorly understood. In the present study, we investigated the inhibition of adipocyte differentiation and adipogenesis by ethanol extracts of Mori Folium (EEMF) in 3T3-L1 preadipocytes. Treatment with EEMF suppressed the terminal differentiation of 3T3-L1 preadipocytes in a dose-dependent manner, as confirmed by a decrease in the lipid droplet number and lipid content through Oil Red O staining. EEMF significantly reduced the accumulation of cellular triglyceride, which is associated with a significant inhibition of pro-adipogenic transcription factors, including sterol regulatory element-binding protein-1c (SREBP-1c), peroxisome proliferator-activated receptor-${\gamma}$ ($PPAR{\gamma}$), and CCAAT/enhancer-binding proteins ${\alpha}$ ($C/EBP{\alpha}$) and ${\beta}$ ($C/EBP{\beta}$). In addition, EEMF potentially downregulated the expression of adipocyte-specific genes, including adipocyte fatty acid binding protein (aP2) and leptin. Furthermore, EEMF treatment effectively increased the phosphorylation of the AMP-activated protein kinase (AMPK) and acetyl CoA carboxylase (ACC); however, treatment with a potent inhibitor of AMPK, compound C, significantly restored the EEMF-induced inhibition of pro-adipogenic transcription factors and adipocyte-specific genes. These results together indicate that EEMF has preeminent effects on the inhibition of adipogenesis through the AMPK signaling pathway, and further studies will be needed to identify the active compounds in Mori Folium.
