  • Methanolic extracts of fresh ginseng, white ginseng and red ginseng were found to have a biological antioxidant activity against ethanol induced lipid peroxidation in the mouse liver. This antioxidant activities were repressed by the addition of ferric ion to the Korean ginseng in the process of its extraction.

추출에 따른 갈매보리수 잎 추출물의 항산화 효능 (Effect of Extraction Conditions on In Vitro Antioxidant Properties of Extracts from Hippophae rhamnoides Leaves)

  • This study investigated optimal extraction conditions for application of Hippophae rhamnoides leaves as a natural antioxidant. Hippophae rhamnoides leaves were extracted using ethanol at various concentrations (0, 30, 50, and 70%) and extraction times (4, 8, and 12 hrs) at $70^{\circ}C$ and then evaluated for extraction yield (22.51%), total phenolic contents (84.21~149.25 mg/g), total flavonoid contents (63.10~124.14 mg/g), as well as antioxidant activities DPPH, ABTS radical scavenging activity, reducing power, and FRAP. Antioxidant activities (DPPH, ABTS radical scavenging activity, reducing power) were correlated with total phenolic and flavonoid contents. 70% EtOH extracts for 12 h at $70^{\circ}C$ showed the highest total phenolic and flavonoid contents with strong antioxidant activities, and that the leaves of Hippophae rhamnoides could be considerd as a candidate of new functional antioxidant agents. This extract could be a good source for natural antioxidants and good health food. This work offers would be useful for chemical and biological studies of natural plants and its products.

  • Oxygen is necessary to sustain life, but reactive oxygen species (ROS) produced by oxygen metabolism can cause mutations and toxicity. ROS can damage cellular macromolecules, leading to oxidative stress, which can accelerate cell death and aging. ROS generated in food affect the taste, color, and aroma of food, and high levels of ROS in meat can cause spoilage. Superoxide dismutase (SOD) plays an important role in scavenging ROS in food and reducing their toxicity to organisms. SOD exerts its antioxidant effect by catalyzing the breakdown of O2-• to H2O2. As a natural antioxidant, SOD has the ability to regenerate and maintain its activity over a long period of time without depletion, unlike chemical antioxidants that may have side effects or stability issues. This antioxidant effect of SOD has great potential in a variety of industries, and in the food industry it can be utilized to improve product quality and provide safe and healthy products to consumers. By disrupting the SOD2 and SOD3 genes in Candida albicans, we studied the effects of SOD2 and SOD3 genes on the antioxidant system, suggesting its potential as a natural antioxidant.

땃두릅(Aralia continentails)의 항산화 성분 (Antioxidant Components from Aralia continentalis)

  • The root of Aralia continentalis Kitagawa (Araliaceae) have been used as an analgesic and fever remedy, and for treatment of rheumatism in Chinese medicine, whereas the young leaves are used for ingredient of salad. Antioxidant activity of the young leaves of A. continentails was determined by measuring lipid peroxide produced when a mouse liver homogenate was exposed to the air at $37^{\circ}C$, using 2-thiobarbituric acid (TBA) and by evaluation the radical scavenging activity on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. Chromatographic separation of active fraction led to the isolation of six flavonoids, among which quercetin, hyperoside and kaempferol showed strong antioxidant activities, while 6"-O-acetyl astragalin, astragalin and trifolin were inactive. Adenosine, oleanolic acid 28-O-glucosyl ester and salsoloside C methyl ester isolated from the somewhat active BuOH fraction exhibited no antioxidant activities.

Natural Compounds with Antioxidant Activity: Recent Findings from Studies on Medicinal Plants

  • Reactive oxygen species potentially cause damage to cellular components including lipids, protein and DNA; this oxidative damage plays a key role in the pathogenesis of neurodegenerative disease, cardiovascular disease, metabolic disease and cancer. On the basis of the oxidative stress hypothesis, a number of studies have been performed to search for an efficient and safe antioxidant. Although in vitro studies have provided promising results, only a limited number of natural and synthetic antioxidants have been developed for clinical application due to their low efficacy and side-effects. Thus, the discovery of new antioxidants with marked efficacy and safety has attracted worldwide attention in recent decades. Since plants are recognized as important sources of natural antioxidants, our research has focused on the discovery of new naturally occurring antioxidants from medicinal plants. The purpose of this review is to open a new prospect in the field of search for natural antioxidants from medicinal plants by summarizing our recent findings. Using in vitro bioassay systems such as 2,2-diphenyl-1-picrylhydrazyl, superoxide radical scavenging tests and lipid peroxidation models, we have tested over than 350 species of medicinal plants for their antioxidant activity and selected several of them for further investigation. During the research on the discovery of effective natural antioxidants from the medicinal plants selected, we have isolated several new and known antioxidant compounds that include stilbene glycosides, phenolic glycosides, flavonoids, oligostilbenes, and coumarins. Our results suggest that the presence of antioxidant compounds in the medicinal plants might be associated with the traditional use to treat inflammation, cardiovascular disease and various chronic diseases.

한국 전통발효식품인 청국장에서 분리한 Bacillus methylotrophicus에 의한 항산화물질의 생산 (Antioxidant Production by Bacillus methylotrophicus Isolated from Chungkookjang, Korean Traditional Fermented Food)

  • Although antioxidant activities of Korean traditional fermented foods were reported by many researchers, study on antioxidant activity of microorganism originated from Korean traditional fermented foods was little. Therefore, we improved condition for antioxidant production by a bacterium isolated from home-made Chungkookjang. We selected a bacterial strain, which showed the highest antioxidative activity, from Chungkookjang and then named GJ. The selected GJ strain was identified as Bacillus methylotrophicus by alignment data of 16S rRNA gene nucleotide sequences. Improved medium compositions for DPPH radical scavenging activity were 0.25% sucrose, 1% peptone, 0.01% $MgSO_4{\cdot}7H_2O$ and initial pH 6.5, respectively. Optimal culture conditions were $30^{\circ}C$, 200 rpm and 4% inoculum volume, respectively. In improved conditions, DPPH radical scavenging activity of GJ reached to 91% in a short time. The strain GJ also possessed ACE inhibition and other antioxidative activities; ACE inhibition activity (49.4%), ABTS radical scavenging activity (99.8%), metal chelating activity (67.9%), SOD-like activity (36.5%) and reducing power ($A_{700}$ = 5.982) were observed, respectively. Therefore, our results suggest that B. methylotrophicus GJ strain may be potential candidate for functional foods, cosmetic products for anti-aging and medicine for diseases caused by oxidative stress.

Antioxidative Properties and Flavonoids Contents of Matured Citrus Peel Extracts

  • We assessed various antioxidant activities, such as 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, 2,2'-azino-bis 3-ethylbenz-thiazoline-6-sulfonic acid (ABTS) radical, and reactive oxygen species (ROS) radical scavenging effect, along with antioxidant capacity, of soybean oil with added Citrus species peel extracts (CPEs). These extract oils showed higher radical scavenging effects than grape fruit seed extract, the natural antioxidant agent, did. When CPEs were added to soybean oil, they showed peroxide value (POV) and acid value (AV) increasing inhibition effects. Furthermore, none of the CPEs showed any cytotoxicity over the tested concentration range of 0.01-100 ppm. The major flavonoid contents of Citrus junos, as determined by HPLC, were naringin ($7.5\;{\mu}g/mg$) and neohesperidin ($7.5\;{\mu}g/mg$), and those of Citrus unshiu were narirutin ($3.13\;{\mu}g/mg$) and hesperidin ($1.97\;{\mu}g/mg$). However, the aglycone form was not found. This study showed that CPEs might be a potent source of natural antioxidant, without any toxic effects.

Stachys riederi var. japonica Extract Reduces Cytochrome C Release from Mitochondria in UVA-irradiated Human Dermal Fibroblasts

  • This study was performed to investigate the cytoprotective effects of Stachys riederi var. japonica ethanol extract (SREE) to control oxidative stress induced by UVA-irradiation by examining antioxidant capacity and gene expression of cytochrome c using human dermal fibroblasts. The total polyphenolics and flavonoids in the SREE were 41.2 and 25.4 mg/g, respectively. At concentrations of 500 and $1000{\mu}g/mL$, the electron-donating ability of SREE was 48.6% and 82.0%, respectively, and the 2,2'-azino-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity was 62.3% and 78.8%, respectively. These findings showed that SREE has a fairly good antioxidant capacity. As determined by an MTT assay, the maximum permissible level for treating SREE to human dermal fibroblasts was shown to be over $200{\mu}g/mL$. SREE ($200{\mu}g/mL$) significantly decreased cytochrome c mRNA and protein expression by 31.1% (p<0.001) and 38.8% (p<0.01), respectively. These findings suggest that SREE may protect human skin cells against mitochondrial-dependent apoptosis. Therefore, SREE seems to be a natural antioxidant to protect cells against oxidative stress induced by UVA-irradiation.
