In order to develop a new microbial fungicide for the control of vegetable diseases using endophytic bacteria, a total of 260 bacterial strains were isolated from fresh tissues of 5 plant species. After they were cultured in broth media, their antifungal activities were screened by in vivo bioassays against Botrytis cinerea(tomato gray mold), Pythium ultimum(cucumber damping-off), Phytopkhora infestans(tomato late blight), Colletotrichum orbiculare(cucumber anthracnose), and Blumeria graminis f. sp. hordei(barley powdery mildew). As the results of screening, 38 bacterial strains showed potent antifungal activities against at least one of 5 plant pathogens. A bacterial strain EB072 displayed potent disease control activities against 3 plant diseases. Among the bacterial strains with a potent antifungal activity against cucunlber anthracnose, three bacterial strains, EB054, EB151 and EB215, also displayed a potent in vitro antifungal activity against C. acutatum, a fungal agent causing pepper anthracnose. A bacterial strain EB215 obtained from roots of cucumber was identified as Burkholderia cepacia based on its physiological and biochemical characteristics and 165 rRNA gene sequence. An antifungal substance was isolated from the liquid cultures of B. cepacia EB215 strain by ethyl acetate partitioning, repeated silica gel column chromatography, and invitro bioassay, Its structural determination is in progress by various instrumental analyses.

This study discusses the long-term antifungal effect and antifungal performance of Hwangtoh mortars with various natural antifungal substances on five types of mold: Chaetomium globosum, Aspergillus niger, Aureobasidium pullulans, Gliocladium virens, and Penicillium pinophilum, which can be easily detected in the indoors and outdoors of buildings in damp environments. The antifungal performances of various natural antifungal substances extracted from Marjoram, Phytoncide, Thyme, Ginkgo leaves, and Chitosan (oligosaccharide) were investigated on the five types of mold, as a basic experiment. Using the natural antifungal substances selected for the basic experiment, antifungal mortars were made, and their antifungal performance and long-term antifungal effects were also investigated. The results clearly showed that the marjoram extract and their associated mortars had excellent antifungal performance. Also, their long-term antifungal effects were outstanding and at an equivalent level to those of the mortars with organic chemical antifungal agents. The optimum addition rate of the marjoram extract in the mortar was 5% by mass of binder. It was also shown that the phytoncide extract in the mortar needed an addition rate of more than 10% according to the mass of binder, considering the long-term antifungal effect and the antifungal performance.
Hyungbangjihwang-Tang (HT), an Oriental herbal formula, has been known to play a role which helps to recover vigor of human in the Orient. In this study, antifungal substance (HTI) was purified from the ethyl-acetate extracts of HT by using $SiO_2$ column chromatography and HPLC, and the antifungal effects of HTI and its mode of action were investigated. By using a broth micro-dilution assay, the activity of HTI was evaluated against fungi. HTI showed antifungal activities without hemolytic effect against human erythrocytes. To confirm antifungal activity of HTI, we examined the accumulation of intracellular trehalose as stress response on toxic agents and effect on dimorphic transition in Candida albicans. The results demonstrated that HTI induced the accumulation of intracellular trehalose and exerted its antifungal effect by disrupting the mycelial forms. To understand its antifungal mode of action, cell cycle analysis was performed with C. albicans, and the results showed HTI arrested the cell cycle at the S phase in yeast. The present study indicates that HTI has considerable antifungal activity, deserving further investigation for clinical applications.

An Actinomycete strain isolated from Mt. Dea-Dun had a strong antifungal activity. The culture brith produced by isolated strain showed only antifungal activity against fungi with the exception of yeast and bacteria. It was heat stable, dissolved in ehtylacetate. The concentrated antifungal agent showed cytotoxicity against HEP-2 and HeLa as tumor cell line, and showed weak cytotoxicity against VERO 36 as normal cell line. Morphological and physiological characteristics were tested with isolated strain. The spore color of isolated strain was gray. It had a short chain and produced brown colored lytic substance in yeast extract-malt agar. The cell wall of isolated strain was composed of meso-DAP, and we suggested it as genus Actinomadura. In the existing of chemical inhibitor, isolated strain grew on the condition of 0.0001% crystal violet, 0.1% phenol, 0.01% sodium azide and 10% sodium chloride. Carbon utilization of isolated strain was shown that glucose, sucrose, manitol and sodium citrate were well utilized.

The object of this study was to characterize Bacillus strains GB-017 and GB-0356, which produce antifungal substances, especially for plant pathogens. In addition, this study was undertaken to characterize the culture conditions required for the production of antifungal substances and to document some of the properties of the antifungal substance produced by these soil-isolated strains. Strains GB-0365 and GB-017 were found to be bacillus-shaped, gram-positive and motile, and to inhibit Botrytis cineria, Fusarium sp., Pythium sp., and Rhizoctonia solani. Antagonistic activity was maintained up to pH 9.0, and the antifungal activity was stable to heat at 80$^{\circ}C$ for 1 h. Antifungal substances were separated and purified using ion exchange and adsorption columns including WK-I0(H$\^$+) (pH 7.0), HP20 column (pH 3.0) and IPA (pH 3.0). and IPA. Its UV absorption spectrum showed major peaks at 231 and 259 nm, corresponding to polyene and lactone. A fast atom bombardment mass spectrum (FAB MS) showed a highest peak at 441 m/z and major peaks at 192, 205, and 370 m/z.
From study of antifungal actions on the rice sheath blight by using the extract of Ginkgo biloba outer seedcoats, we found that the extracts of Ginkgo biloba outer seedcoats of all treatment concentrations had inhibited the rice sheath blight. Among them, the most effective concentration was 250 mg/l at which the growth of microbe was 26 mm and even at the packaging test, when sprayed the G. biloba outer seedcoats at the level of 250 mg/l, the damage rate of the rice sheath blight was identified as 13%. As a result investigating the antifungal activity by separating polysaccharides from G. biloba outer seedcoats, it showed that the clear zone of 14 mm or more was formed at the concentration of 250 mg/l or higher. Based on these results, we concluded that the G. biloba outer seedcoat is a natural substance with the antifungal activity on the rice sheath blight.

This study tested the antifungal compound obtained from a medicinal plant, Dioscorea quinqueloba Thunb., in order to search the possibility of practical application of this product in agriculture through evaluating its activity using the citrus fruits. The extract of D. quinqueloba Thunb., which has the strongest antifungal activity, was selected as a candidate among 101 plant extracts. Based on this examination concerning antifungal activity of the product on Penicillium digitatum in vitro, it was confirmed its effect of mycelial growth inhibition showed over 87% at 0.5 mg/ml concentration. This natural product showed the stability of the substance, as it was not significantly influenced by pH, temperature, or ultraviolet radiation. While citrus fruits were stored at room temperature, P. digitatum was inoculated into them in order to prepare a similar environmental conditions with epidemic occurrence of the mold. As the result of our investigation, the disease preventive effects of the active antifungal substance evidenced a 100% at 0.5 mg/ml. When the phytotoxicity of the selected natural product on citrus at 2 mg/ml was assessed, we noted no toxic effects. Based on the superior preventive effects from this natural product extracted from the plant, it is presumed to be very useful in agricultural applications for the control of green mold, P. digitatum, which has been occurred often the biggest problem in the storage of citrus fruits.

An important aspect of indigenous medicine is the ability of traditional societis to manipulate the environment for their own benefit, for example for their health care. In Indonesia, this indigenous medicine is called Jamu. Jamu mostly consists of a mixture of herbals of the genus Curcuma, Zingiberaceae. There are 19 species of curcuma grown in Indonesia. Eleven of them are popular in the jamu preparations. Ethnopharma-cological surveys have shown that 50% of these species are used for post partum protection, dismenorrhea, 30% are used for the treatment of stomache and as cosmetics, 20% for the treatment of various diseases such as fever, worms, asthma, etc. Chemical studies show that they contain curcuminoids, volatile oils, flavonoids, starch, and resinous substance. Pharmacological studies of extracts and isolated bioactive compounds have shown that they have a broad pharmacological activity such as antibacterial, antifungal, anti-inflammatory, choleretic, antihepatotoxic, antitumor, antioxidant, and antihyperlipidemic activity.
Rust causes significant losses in the yield and quality of various crops. The development of new effective and environmentally benign agents against the pathogen is of great interest. In the course of searching a natural antifungal compound from medicinal plants, we found that the methanol extract of Angelica gigas roots had a potent control efficacy against wheat leaf rust (WLR) caused by Puccinia recondita. The antifungal substance was isolated from the methanol extract by silica gel column chromatography, alumina column chromatography and $C_{18}$ preparative HPLC. It was identified as decursinol angelate by EI-MS and $^1H$-NMR data. In in vivo test, decursinol angelate effectively suppressed the development of WLR and red pepper anthracnose (RPA) among the 6 plant diseases tested. In addition, the wettable powder-type formulation of ethyl acetate extract of A. gigas roots significantly suppressed the development of WLR. The crude extract containing decursinol angelate and the chemical appear to be a potential candidate for control of WLR. In addition, this is the first report on the in vivo antifungal activity of decursinol angelate against WLR as well as RPA.
In oder to select an antifungal substance-producing antagonistic bacterium against Fusarium oxysporum casuing fusarium wilt on tomato, strain BK4 was isolated from local soil of Gyeoungbuk and was identified as Bacillus thuringiensis by 16s rDNA analysis, biochemical test, and Mcirolog TM 3.0 System. The antibiotic of B. thuringiensis BK 4 was highly produced at $30^{\circ}C$ in nutrient broth (pH 9.0). The crude antibiotic was even stable at $121^{\circ}C$ and more stable at slight alkalic condition than acid condition. It was also remained $50{\%}$ activity at pH 3.0. B. thuringiensis BK4 showed the inhibition of spore germination and the biocontrol ability against F. oxysporum causing fusarium wilt of tomato in vivo test. According to these results, B. thuringiensis BK4 was enough to use with a microbial agent for biocontrol against fusarium wilt.
