Assessment of the Effects of Interactions between Climatic Conditions and Genetic Characteristics on the Agronomic Traits of Soybeans Grown in Six Different Experimental Fields

  • Soybean [Glycine max (L.) Merr.] is a species of legume native to East Asia. The interactions between climatic conditions and genetic characteristics are known to affect the agricultural performance of soybean. Therefore, the present investigation was conducted to identify the main elements affecting the agricultural performances of 11 soybean varieties/lines from China [Harbin ($45^{\circ}12^{\prime}N$), Yanji ($42^{\circ}53^{\prime}N$), Dalian ($39^{\circ}30^{\prime}N$), Qingdao ($36^{\circ}26^{\prime}N$)] and the Republic of Korea [Suwon ($37^{\circ}16^{\prime}N$), and Jeonju ($35^{\circ}49^{\prime}N$)]. The days to flowering (DTF) of soybeans with the e1-nf and e1-as alleles and the E1e2e3e4 genotype, except in 'Keumgangkong', 'Tawonkong', and 'Duyoukong', were relatively short compared to those of soybeans with other alleles. Although DTF of the soybeans was highly correlated with all climatic conditions [negative: precipitation, average temperature (AVT), accumulated temperature; positive: day-length (DL)], days to maturity and 100-seed weight of the soybeans showed no significant correlation with any climatic conditions. The soybeans with a dominant Dt1 allele, except 'Tawonkong', had the longest stem length (STL). Moreover, STL of the soybeans grown in the test fields showed a positive correlation with only DL; however, the results of our chamber test that was conducted to complement the field tests showed that STL of soybean was positively affected by AVT and DL. Although soybean yield (YLD) showed positive correlations with latitude and DL (except L62-667, OT89-5, and OT89-6), the response of YLD to the climatic conditions was cultivar-specific. Our results show that DTF and STL of soybeans grown in six different latitudes are highly affected by DL, and AVT and genetic characteristic also affect DTF and STL.

Modifications of skim milk protein by Meju protease and its effects on solubility, emulsion and foamming properties (메주 단백질 가수분해 효소가 탈지 우유의 기능성에 미치는 영향)

  • This study was attempted to investigate the effects of enzymatic modification of milk protein with protease on functional properties. The selected functional properties were solubility, emulsifying activity (EA), emulsion stability(ES), foam expansion(FE), and foam stability(FS). These properties were measu-red from pH 3.0 to pH 8.0. The proteases used in this study were iaolated from Meju(fermemted soybean) and had specific activity of 250 units/㎎ protein at pH 7.0, 1600 units of pretense was used for 1gr. of skim milk protein. Skim milk showed 30.5% degree of hydrolysis for 1 hr. and 36.4% degree of hydrolysis for 3.5 hrs. of protease treatment at pH 7.0. Solubility of native skim milk, control, 1 hr. and 3.5 hrs. groups were 3.37, 3.64, 10.21, 14.34%o at pH 4.0 respcetively. The emulsifying activity of native skim milk, control, 1 hr. and 3.5 hrs. groups were 38.8,42.0,43.0,46.7ft at pH 4.0, respectively. Enzymatic modification resulted in the increase of solubility and emulsifying activity at pH 4.0. However at pH 5.0 emulsifying activity of 1 hr. and 3.5 hr. group were lower than native skim milk and control groups. 1 hr. protease treatment was found to be most effective way of increasing foam expansion at pH 4.0 to 6.0. It was supported that, protease treated skim milk can be used to improve solubility, emulsifying activity, foam expansion at acid pH. meju protease. skim milk, solubility, emulsion, foam.

Effects of Feeding Angelica gigas By-products of Performance and Meat Quality of Korean Native Chicks (당귀 부산물의 급여가 재래닭의 생산성과 육질에 미치는 영향)

  • The objective of this experiment was to investigate the effect of feeding Angelica gigas Nakai by-products on performance and meat quality of Korean native chicks(KNC). Five hundred forty KNC were placed into 0, 0.2, and 0.4% Angelica gigas supplements with four replications between males and females. Weight gain, feed conversion ratio(FCR), breast meat lipid, fatty acid and protein contents were measured from 17 to 20 weeks old. Egg production, feed intake and FCR were examined from 22 to 29 weeks at four weeks at four weeks intervals. Basal diets based on corn and soybean meal contained 15% crude protein and 2,850㎉/kg ME for the growing period, 16.5% and 2,800㎉/kg for the laying period. Experiments were designed in a one way analysis. Weight gain of female chicks fed 0.2% Angelica gigas by-products increased significantly compared to that of other treatments for the growing period, but was not consistency in male groups. Moisture, protein, fat content were not statistically different among all treatments. Fatty acid composition(C16:0, C18:0) of chicks fed 0.2% Angelica gigas by-products was higher than other treatments. In laying period, hens fed 0.2 or 0.4% Angelica gigas Nakai by-products seemed to increase the egg production and significantly improved FCR compared to that of control(P〈0.05). Birds fed 0.4% treatment showed significantly the lowest feed intake and FCR of all treatments(P〈0.05). The results of these studies indicated that dietary supplemental Angelica gigas by-products may have a role to improve the performance of KNC.

Cricket (Gryllus bimaculatus) meal pellets as a protein supplement to improve feed efficiency, ruminal fermentation and microbial protein synthesis in Thai native beef cattle

  • Objective: Replacing soybean meal (SBM) with cricket (Gryllus bimaculatus) meal pellets (CMP) in concentrate diets was investigated for feed efficiency, ruminal fermentation and microbial protein synthesis in Thai native beef cattle. Methods: Four male beef cattle were randomly assigned to treatments using a 4×4 Latin square design with four levels of SBM replaced by CMP at 0%, 33%, 67%, and 100% in concentrate diets. Results: Results revealed that replacement of SBM with CMP did not affect dry matter (DM) consumption, while digestibilities of crude protein, acid detergent fiber and neutral detergent fiber were significantly enhanced (p<0.05) but did not alter digestibility of DM and organic matter. Increasing levels of CMP up to 100% in concentrate diets increased ruminal ammoniacal nitrogen (NH3-N) concentrations, blood urea nitrogen, total volatile fatty acids and propionate concentration (p<0.05), whereas production of methane and protozoal populations decreased (p<0.05). Efficiency of microbial nitrogen protein synthesis increased when SBM was replaced with CMP. Conclusion: Substitution of SBM with CMP in the feed concentrate mixture at up to 100% resulted in enhanced nutrient digestibility and rumen fermentation efficiency, with increased volatile fatty acids production, especially propionate and microbial protein synthesis, while decreasing protozoal populations and mitigating rumen methane production in Thai native beef cattle fed a rice straw-based diet.

Screening for Resistance to Purple Seed Stain 'Cercospora kikuchii' by Seed Inoculation Technique. (종자접종법에 의한 대두 자반병 저항성 검정에 관한 연구)

  • An attempt was made to establish a mass-screening technique for resistance to purple seed stain .disease in soybean. Seeds sterilized in 1 : 10000 'mercuric chloride for 1 minute and transffered to Petridishes containing 20ml water agar plus 50ppm of the sodium salt of 2,4-D were inoculated by placing a small agar cube containing fungal mycelium on the seed coat of each seed. A positive correlation between natural infection of purple seed :stain and purple discoloration by seed inoculation technique was highly significant and by this technique, some native soybean collections and introduced varieties were tested for resistance to the disease. Most of the soybean varieties tested were susceptible except for the varieties Hill, Harosoy and Sac, resistant comparatively.

Relationship between Fruiting Period and Agronomic Characters in Korean Local Soybean Collections (재래종대두의 결실일수군별과 제형질과의 관계)

  • The present study was conducted to evaluate agronomic characters of the soybean germplasm which was collected from the sites covering whole areas of southern Korean peninsula. A total of 2,748 Korean native soybeans was grouped for fruiting period and in this group frequency distribution of the important agronomic characters and relationships among the characters were determined. The results obtained were summarized as follows; The soybean germplasm was comprised of SeoulㆍKyonggi-do 7.5%, Kangwon-do 13.3%, Chungchongbuk-6.6%. Chungchongnam-do 12.3%, Chollabuk-do 10.7%, Chollanam-do 12.6%, Kyongsangbuk-do 20.8%, Kyongsangnam-do 15.3% and Cheju-do 0.6% in number of collections. Frequency distribution of the germplasm for fruiting period was 0.14% in Group I (below 55 days). 1.67% in Group II (56-60 days), 10.58% in (Group III(61-65 days), 23.18% in Group IV(66-70 days), 25.91% in Group V(71-75 days) 19.79% in Group Ⅵ(76-80 days), 15.1% in Group Ⅶ(81-85 days) and 3.6% in Group Ⅷ(over 85 days). The lines over 70 days in fruiting period were as much as 64.4% of germplasm. The fruiting period varied largely from 45 days to 91 days. Fruiting period was positively correlated to seed weight, days to maturity, days to flowering, plant height and number of branches per plant, respectively. While. it was negatively correlated to number of pods, lodging and virus infectivity, repectively, and in particular negative correlation was shown significantly between yield and fruiting period. Average yield was very high as much a 1.389kg/ha in fruiting period Group I, and decreased with increment of fruiting period, 1.400kg/ha in Group II. 1.384kg/ha in Group III. 1,299kg/ha in Group IV, 1,197kg/ha in Group V, 1,117kg/ha in Group Ⅵ, 967kg/ha in Group Ⅶ and 832kg/ha in Group Ⅷ.

Biochemical and Ultrastructural Trends in Proteolysis of the $\beta$-subunit of 7S Protein in the Cotyledons During Germination of Soybean Seeds

  • Antibodies raised against the purified p-subunit of $\beta$-conglycinin were used in immunohistochemical studies to monitor the pattern of $\beta$-conglycinin mobilization in the cotyledons during soybean [Glycine max (L.) Merr.] seed germination. Western blot analysis revealed that the break down of the $\beta$-subunit of $\beta$-conglycinin commenced as early as 2 days after seed imbibition (DAI). Concurrent with the degradation of the $\beta$-subunit of $\beta$-conglycinin, accumulation of 48, 28, and 26 kD proteolytic intermediates was observed from 2 to 6 DAI. Western blot analysis also revealed that the acidic subunit of glycinin was mobilized earlier than the basic subunit. The basic glycinin subunit was subjected to proteolysis within 2 DAI resulting in the appearance of an intermediate product approximately 2 kD smaller than the native basic glycinin subunit. In contrast to the major seed storage proteins, lipoxygenase was subjected to limited proteolysis and was detected even after 8 DAI. The first sign of $\beta$-conglycinin breakdown was observed near the vascular strands and proceeded from the vascular strands towards the epidermis. Protein A-gold localization studies using thin sections of soybean cotyledons and antibodies raised against the $\beta$-subunit of $\beta$-conglycinin revealed intense labeling over protein bodies. A pronounced decrease in the protein A-gold labeling intensity over protein bodies was observed at later stages of seed germination. The protein bodies, which were converted into a large central vacuole by 8 DAI, contained very little 7S protein as evidenced by sparse protein A-gold labeling in the vacuoles.

  • Thirty eight varieties with high uniformity and clearance in grain quality among 70 colored soybeans collected from nation-wide provinces were investigated for their seed morphological and agronomic traits. As for seed morphological traits, grain volumetric size, grain weight, grain length, grain thickness, . hilum length and width were studied on the tendency of statistical dispersion and variations. As a result, four groups could be classified on the basis of 100 grains weight, so as small, medium, large, and super large groups, Also, as for agronomic traits, among others, the tendency of setting in flowering date, maturing date, yield components, and stem weight, stem length were studied on the basis of statistical inter-relationships between above mentioned characteristics. From the above, bigger variations were detected in weighing characters of soybean plants than in duration characters. And the flowering and maturing days showed significant relationships to the stem length, stem weight, also stem length and stem weight to the grain yield.

Studies on the Soybean Pod Borer Damage (대두의 콩나방피해율에 관한 연구)

  • Present studies were carried out to get a basic information for biological control of soybean rod borer, Grapholitha glycinivorella Matsumura, causing main injury in soybean cultivation in Korea. 1474 native strains were cultivated in field to evaluate the pod borer damage. Pubescence density and color, maturity group, seed coat color, aphid and general leaf damage and seed damage by pod borer were investigated, and determined among their relationships. As another basic study for ecological control of insect, 6 leading varieties were planted on different planting date with 3 levels of plant spacing, on which pod borer damage and some related agronomic characters were studied. The results obtained one summarized as follows: Average injury of soybean pod borer was $5.2\%$ ranging $0-38\%$ in 1474 strains planted on 20 May. Non-significant correlations were found between pod borer damage and pubescence density and color, while pod borer damage closely related with maturity groupand aphid damage at $5\%$ level ofttests. Most severe damage was found in green seed coast color and maturity group V. Late and sparse plantings tend to have more damages to the pod borer than those of early and dense plantings. Pod setting date and period of pod maturing seemed related with pod borer damage. Bongeui and Chungbuk-baik cultivars were resistance to soybean pod borer, while Clark and Kumkang-daerip were susceptible in field tests.

Comparison of Flavor Characteristics and Palatability of Beef Obtained from Various Breeds (품종별 쇠고기의 풍미특성과 기호성 비교)

  • For comparison of beef quality, four kinds of beef (Korean native cattle beef, dairy cattle beef, imported beef, cross-bred beef) were investigated through tenderness, juiciness and flavor related components measurement and organoleptic tests. Flavor related chemical components such as NPN, IMP, free fatty acid and free amino acids were analyzed, water holding capacity, contents of hydroxyproline and intramuscular fat were measured for evaluation of beef tenderness. Instron was also used for measuring beef tenderness as an objective method. Triangle test and descriptive analysis test were conducted for comparison and evaluation of preference of various beef samples. In hardness analysis using Instron, imported and cross-bred beefs had higher value than that of Korean native cattle or dairy cattle beef. Water holding capacity and pH of Korean cattle beef was higher than that of others. The intramuscular fat content of Korean cattle beef was highest, so it was expected juicier than other beef. In flavor related compound analysis, NPN content of Korean native cattle beef was the lowest, which shows it spent the least time among sample meats after slaughter. IMP, hypoxantine and inosine were most abundant in Korean native cattle beef. In free amino acids analysis showed that the proportion of basic acid and aromatic acid content of Korean native cattle beef was highest, whereas that of sulfur containing amino acid of imported beef was highest. TBA value of Korean native cattle beef was the lowest, and analysis of fatty acid composition revealed that the proportion of unsaturated fatty acid of Korean native cattle beef was higher than imported and dairy cattle beef, but similar to cross-bred beef. Organoleptic test was performed by triangle test and descriptive analysis. In triangle test, most panelist could distinguish Korea native cattle beef from imported beef and cross-bred beef, imported beef from cross-bred beef. In descriptive analysis which relys on subjective standards of panelists, there was no difference among beef in aroma, flavor and tenderness except juiciness. Even though contents of non volatile flavor compounds in Korean native cattle beef were higher than those of other beef samples, there were no significant differences in subjective panel test. The results showed that Korean consumers do not have common standards for beef quality evaluation.
