The antioxidative and neuroprotective effects of Bombusae concretio Salicea and phenolic compounds on neuronal cells (신경세포에서 천축황(天竺黃)과 페놀성 물질의 항산화 및 신경보호 효과)

  • 산화적인 스트레스가 여러가지 신경 및 비신경계에서의 병리원인으로 알려져 있다. 퇴행성 뇌질환에 대한 예방과 치료에는 항산화 방어기술이 주요대상이며 스테로이드 분자중에서 estrogen만이 산화적인 원인에 의한 신경세포사를 방어하는데 특이적인 효과를 가지고 있다. 본 연구는 천축황(天竺黃)의 항산화적 뇌신경 보호기전을 연구하는 것으로 신경세포주, 뇌세포막, 이의 산화적 정량실험법을 사용하여 천축황(天竺黃)이 갖는 항산화 및 신경보호활성이 소수성 페놀(phenolic molecules)성 물질과 유사함을 밝히게 되었다. 즉, 페놀성 물질로서 2,4,6-trimethylphenol, N-acetylserotonin, 및 5-hydroxyindole와 유사한 뇌신경 보호활성을 나타내었으며 천축황(天竺黃)은 생쥐의 N2a cell과 사람 SK-N-MC neuroblastoma cell에서 산화적인 글루탐산 독성에 대하여 보호를 하였다. 천축황(天竺黃)의 산화적 글루탐산 독성에 대한 보호활성은 과산화수소에 대한 것과 유사하였다. 이러한 항산화 활성은 $20\;{\mu}g/ml$에서, LDL의 산화적 보호 활성은 $5\;{\mu}g/ml$농도에서 발휘되었다 (최대활성은 $16\;{\mu}g/ml$). 이러한 결과는 천축황(天竺黃)이 노인성 치매에 보호효과가 있음을 시사하였다.

Anitimicrobial Effects of Zanthoxylum schinifolium Extracts against Multi-drug Resistant Acinetobacter baumannii (산초(Zanthoxylum schinifolium) 추출물의 다제내성 Acinetobacter baumannii 억제 효과)

  • The aim of this study was to investigate the possible utilization of Zanthoxylum schinifolium as a source of antimicrobial agents. The antimicrobial effects of Zanthoxylum schinifolium extracts were investigated against Acinetobacter baumannii, which is a multi-drug resistant pathogen, and 5 other pathogenic microorganisms. The hexane extract of Zanthoxylum schinifolium was more effective than the ethyl acetate, n-butanol and methanol extracts which were active against Acinetobacter baumannii 25, with minimum inhibitory concentrations(MIC) ranging from 0.8 mg/ml to 1.6 mg/ml. Tetracycline had no effect on Acinetobacter baumanniii. The hexane extract was highly active against Candida albicans IFO 6258, with an MIC of 1.5 mg/ml. In contrast, the ethyl acetate, n-butanol and methanol extracts showed no activity against the 5 pathogenic microorganisms. Furthermore, a combination of hexane extract and ethyl acetate extract was significantly more active against the 5 Acinetobacter baumannii strains than n-butanol and methanol. These results suggest that Zanthoxylum schinifolium extracts have great potential as antimicrobial compounds against multi-drug resistant pathogens, and further studies are warranted.

Effects of Jeungmiobi-tang on the Articular Cartilage Injuries Induced by Monosodium Iodoacetate in Rats (증미오비탕이 Monosodium Iodoacetate 유발 관절연골손상에 미치는 영향)

  • Objectives: This study was carried out to investigate the protective effects of Jeungmiobi-tang on the articular cartilage injuries induced by monosodium iodoacetate in rats. Methods: Twenty four rats were divided into three groups. Rats of normal group (n=8) were injected with 0.1 ml physiological saline into both knee joint cavities. In the rats of control group (n=8) and Jeungmiobi-tang group (n=8), Arthritis was induced by injecting with 0.1 ml monosodium iodoacetate (5 mg/ml) into both knee joint cavities. After the experiment, Gross and histopathological examinations on the knee joint were performed. The content of proteoglycan in articular cartilage and TNF-α and IL-1β in synovial fluid were also analyzed. Results: Grossly, Injuries to the articular cartilage surface was observed weak in the Jeungmiobi-tang group compared to the control group. Proteoglycan content in the articular cartilage was significantly higher in the Jeungmiobi-tang group than in the control group. The chondrocyte score was significantly lower in the Jeungmiobi-tang group than in the control group. Conclusion: According to these results, that Jeungmiobi-tang has protective effects on the articular cartilage injuries induced by monosodium iodoacetate in rats.

Antioxidant and Antimicrobial Activity of Zostera marina L. Extract

  • Methanol crude extract of the sea grass Zostera marina L. and organic solvent fractions (n-hexane, chloroform, ethyl acetate, n-butanol, and water) were screened for antioxidant activity (total phenolic contents, DPPH scavenging activity, and reducing power) and antimicrobial activity against three human skin pathogens, two bacteria and a yeast; Staphylococcus aureus, Staphylococcus epidermidis, and Candida albicans. Total phenolic contents and 2, 2- diphenyl-1-picrylhydrazyl (DPPH) scavenging activity were highest in the ethyl acetate fraction with 968.50 $\mu$g gallic acid equivalent per milligram of extract, and ca. 95% scavenging activity on the DPPH radicals at 10 mg $ml^{-1}$. In antimicrobial activity tests, MICs (Minimum Inhibitory Concentration) of each Zostera marina extract partitioned ranged from 1mg to 8 mg $ml^{-1}$ (extract/ 10% DMSO) against all three human skin pathogens. The MICs of the ethyl acetate and n-butanol fractions were the same with 1 mg $ml^{-1}$ against S. aureus and C. albicans. The ethyl acetate fraction of Z. marina does protect against free radicals and may be used to inhibit the growth of human skin pathogens.

Antibacterial effect on leaf-extract from Nelumbo nucifera against oral microorganism (연잎 추출물의 구강미생물에 대한 항균 효과)

  • Objectives : The purpose of the study is to investigate the activities of Nelumbo nucifera leaf extracts on Streptococcus mutans, Streptococcus mitis, Streptococcus sobrinus, Porphyromonas gingivalis, Prevotella intermedia, and Treponema denticola. Methods : The inhibitory effect of lotus leaf extracts on the growth of oral bacteria was assessed in experiments with extracts from freshly harvested and pulverized lotus leaves and bacterial cultures of dental caries. Results : The results showed that N. nucifera extracts possess antimicrobial activity on all bacterial strains. The minimal inhibitory concentration (MIC) values varied from 4 mg/ml to 10 mg/ml against antimicrobial activity. The relative growth ratio (RGR) against of N. nucifera extracts were determined as 50% in concentration of 4.0 mg/ml. The extract of N. nucifera was effective in reducing on the glucosyltransferase (GTase) activity of six strains in vitro. Conclusions : Methanol extracts of lotus leaves showed antimicrobial effects on three bacterial species causing dental caries and three bacterial species causing periodontitis, as well as inhibitory effects on GTase activity.

The Screening of Nitrite Scavenging Effect of Marine, Algae and Active Principles of Ecklonia Stolonifera

  • The nitrite scavenging effect of methanol extracts of marine algae were evaluated to discover new natural nitrite scavengers. Among the tested seaweeds, Ecklonia stolonifera, an edible brown algae, showed the strongest scavenging effect. The MeOH extract was then sequentially partitioned into $CH_2Cl_2,\;CH_2Cl_2$ insoluble interface, EtOAc, n-BuOH, and $H_2O$ layers. The EtOAc and n-BuOH fraction demonstrated high levels of nitrite-scavenging activity while the $CH_2Cl_2,\;CH_2Cl_2$ insoluble interface, and $H_2O$ fractions were inactive. A column chromatography of the EtOAc fraction through silica gel and Sephadex LH-20 yielded phloroglucinol and a new compound tentatively named phlorotannin A. The nitrite scavenging activity of phloroglucinol $(IC_{50}=3.9{\mu}g/ml)$ was more potent than that of L-ascorbic acid $(IC_{50}=65.0{\mu}g/ml)$. However, phlorotannin A $(IC_{50}=193.2{\mu}g/ml)$ showed only low levels of activity. From the above results, it is possible to suggest that both the MeOH extract and their fractions and isolated phloroglucinol and phlorotannin A obtained from E. stolonifera may be applicable as scavengers of nitrite, which is a precursor for the formation of carcinogenic N-nitroso compounds.

  • For the purpose of studies on the citric acid production, some experiments were carried out with isolated strains. The results obtained were as follows. 1) The optimal culture media of the strain M-80 in surface culture contained 140g of sucrose, 3.0g of (N $H_4$)$_2$S $O_4$, 1.5g of K $H_2$P $O_4$, 0.2g of MgS $O_4$.7$H_2O$, 3.0mg of F $e^{++}$, 1.0mg of Z $n^{++}$, 0.5N HCI to a pH of 5.0 and distilled water to 1.0 liter; and that of the strain M-315 in surface culture contained 140g of sucrose, 2.0g of N $H_4$N $O_3$, 1.0g of K $H_2$P $O_4$, 0.25g of MgS $O_4$. 7$H_2O$, 2.0mg of F $e^{++}$, 2.0mg of Z $n^{++}$, 0.05mg of C $u^{++}$, 0.5N HCI to a pH of 4.5 and distilled water to 1.0 liter. While that of the strain M-315 in submerged culture contained 140g of sucrose, 2.5g of N $H_4$N $O_3$, 1.5g of K $H_2$P $O_4$, 0.3g of MgS $O_4$. 7$H_2O$, 3.0mg of F $e^{++}$, 0.1mg of C $u^{++}$, 0.5N HCI to a pH of 4.5 and distilled water to 1.0 liter. The optimal temperature and size of inoculum were mostly 28-3$0^{\circ}C$, 10$^{7}$ -10$^{8}$ spores/50ml, respectively. 2) Through the course of citric acid production, the growth of strains had nearly been completed, pH value was rapidly decreased below 2.0 and the content of sugar was also reduced, while the accumulation of citric acid in media was remarkably begun in about 3-4 days. The yields of citric acid generally reached the maximum level in 8-10 days in surface or submerged fermentation process. 3) Methanol was effective citric acid production when they were added to fermentation media. In the case of surface culture, by addition of 2% (strain M-80), 3% (strain M-315), the yields of citric acid was increased 6.5%, 20.6%, respectively and 5.0% yield was increased by addition of 3% methanol in submerged culture media of the strain M-315. 4) Chromatography analysis of culture broth after fermentation under optimal culture conditions detected that the majority of acid in media was citric acid. 72.1mg/ml, 98.1mg/ml, of citric acid were determined in surface culture media by strains of M-80, M-315, and 59.8 mg/ml of citric acid was contained in the submerged culture media by the strain M-315. strain M-315.

Transformation of Korean Natural Zeolite to Faujasite NaX (한국산 천연 제올라이트로부터 제올라이트 NaX로의 전환)

  • The zeolite X was prepared from the Korean natural clinoptilolite, which contains some mordenite. Thermal treatment removed the clinoptilolite structure from the ore remaining mordenite. The natural clinoptilolites dealuminated with 2N-8N HCI solution and/or thermal treatment were mixed with NaCl, $NaAlO_2$ and NaOH, and reacted to zeolites X at $95^{\circ}C$ for 12~36 hrs. Maximum yield of NaX was obtained for the reactant mixture of 25 gr of natural zeolite acidtreated with 8 N HCI, together with 3.5g NaCl, 8g $NaAlO_2$ and 50 ml of 6N NaOH at $95^{\circ}C$, for 24 hrs.

Invitro antifilarial potential of the leaf extract of Oscimum sanctum on cattle filarial parasite Setaria cervi

  • The effect of aqueous and alcoholic extract of the leaves of Ocimum. sanctum was studied on the spontaneous movements of the whole worm (w.w) preparation and nerve muscle (n.m.) complex of Setaria cervi (S. cervi) and on the survival of microfilariae (m.f.) in vitro. Both the extracts caused inhibition of the spontaneous motility of the w.w. and n.m. complex of S. cervi characterized by initial stimulation followed by reversible paralysis, aqueous extract at a higher concentration showed immediate effect and irreversible paralysis. The concentration required to inhibit the movements of n.m. complex was $1/4^{th}$ for aqueous and $1/3^{rd}$ for alcoholic extract compared to that for the w.w., suggesting a cuticular permeability barrier. On the m.f. the lethal concentration (LC 50 and LC 90) were 35 and 50 ng/ml for aqueous whereas, 60 and 85 ng/ml for alcoholic extracts respectively.

Detection of residual antibiotics by TLC and EEC-4 plate method in slaughtered pigs (도축돈에서 TLC와 EEC-4 plate법을 이용한 항생물질 잔류조사)

  • The antibiotic residues of the urine, the liver, the lung, the kidney and the spleen in slaughtered pigs at Kyongbuk province were detected by TLC(505 kit) and EEC-4 plate method. 1. The positive rate of residual sulfamethazine which was detected by 505 kit in the urine (n=200) was 0.0%. 2. The positive rate of residual sulfamethazine which was detected by EEC-4 plate in the urine (n=126), the liver(n=98), the kidney(n=72), the spleen (n=68) and the lung(n=48) were 63%, 49%, 36%, 34% and 24%, respectively. 3. By EEC-4 plate method, the positive detection rates of the urine were 53.0% in BS(pH 6.0), 29.0% in BS(pH 7.2), 11.5% in BS(pH 8.0) and 13.0% in ML(pH 8.0) medium, that of the liver 41.5% in BS(pH 6.0), 22.0% in BS(pH 7.2), 6.5% in BS(pH 8.0) and 5.0%, in ML (pH 8.0) medium, that of the lung 21.0% in BS(pH 6.0), 9.5%, in BS(pH 7.2) and 8.5% in BS(pH 8.0) medium, and that of the kidney 31.5% in BS(pH 6.0), 14.5% in BS(pH 7.2), 20.0% in BS(pH 8.0) and 3.0% in ML(pH 8.0) medium. In the spleen, only in BS(pH 6.0) medium the positive rate was detected as 33.5 %. 4. The positive rates of samples which shown TLC-positive detected by EEC-4 plate method were 53.9% in no band, 77.8% in one band, 80.9% in two bands, 66.7% in three bands, respectively. In conclusion, the EEC-4 plate method could be applied for the detection of residual antibiotics in samples which shown as out of standard Rf values by TLC-method (SOS kit).
