Effects of Synchronization of Carbohydrate and Protein Supply on Ruminal Fermentation, Nitrogen Metabolism and Microbial Protein Synthesis in Holstein Steers

  • Three rumen-cannulated Holstein steers were fed three diets, each with a different synchrony index (SI) (LS: 0.77, MS: 0.81, and HS: 0.83), in order to examine the effect of diet on rumen fermentation, nitrogen balance, and microbial protein synthesis. Synchrony index was calculated based on the carbohydrate and crude protein fractions of each ingredient and their degradation rates. Feeding the steers diets with different SIs did not influence dry matter, crude protein, NDF, or ADF digestibility. The concentrations of total and individual VFA in the rumens of steers that were fed the two higher-SI diets were higher than in those fed the low-SI diet (p<0.05), but there was no significant difference between the two higher-SI diets. One hour after feeding, steers on the LS diet had lower ruminal pHs than did those fed the MS or HS diets (p<0.05), and animals on the LS diet generally showed higher ruminal $NH_3$-N levels than did animals on the other diets, with the 4-h post-feeding difference being significant (p<0.05). Steers receiving the LS diet excreted more nitrogen (N) in their urine than did those on the two higher-SI diets (p<0.05), and the total N excretion of those on the LS diet was also higher (p<0.05). Microbial N levels calculated from the concentration of urinary purine derivatives were generally higher when the SI was higher, with the highest microbial protein synthesis being produced by steers on the HS diet (p<0.05). In conclusion, in the current study, ingestion of a synchronous diet by Holstein steers improved microbial protein synthesis and VFA production and decreased total N output.

Influence of Urea Treatment and Soybean Meal (Urease) Addition on the Utilization of Wheat Straw by Sheep

  • The effect of ammoniation with urea and with soybean meal (SBM) as a source of urease on the nutritive value of wheat straw was evaluated in sheep. Twenty-four male Najdi lambs were used in a 3${\times}$2 factorial design, in which the animals were allocated to three straw treatments: 0% urea-treated (NT), 6% urea-treated (UT) and 2.2% urea-supplemented (US) straws. Each straw treatment was either supplemented or non-supplemented with 70 g SBM $kg^{-1}$ straw during the treatment time with urea, giving a total of six straw treatments. Each of these treatments was individually fed ad libitum to 4 lambs, together with 300 g of barley grain/head/day. Total N content of UT and US straws increased significantly (p<0.001) as compared to NT straw. The degree of urea hydrolysis, either with or without SBM addition, was nearly similar. Lambs fed either UT or US straw based diets had significantly (p<0.01) and numerically (p>0.05) higher straw DM intake (g $d^{-1}$ $kg^{-1}$ $BW^{0.75}$), compared to those fed NT straw based diet. Apparent DM or OM digestibilities increased significantly (p = 0.014) in lambs fed UT diet, and numerically (p>0.05) in lambs fed US diet as compared to those fed NT diet. Fiber (CF, NDF, ADF, cellulose and hemicellulose) digestibility increased to a similar magnitude, averaging 20.2 (p<0.001) and 7.8% (p<0.07); this corresponds to 35 (p<0.001) and 51% (p<0.001) in N digestibility and approximately 78 (p<0.017) and 105% (p<0.002) in N retention, for UT and US diets, respectively, as compared to NT diet. However, the UT diet had higher (p<0.01) fiber digestibility over the US diet. Addition of SBM tended to improve (p = 0.09) straw DM and digestible OM intakes, while significantly increasing (p<0.001) total and digestible CP intakes across all diets. Lambs fed on US diet had higher ruminal ammonia N than those fed on UT (p<0.05) or NT (p<0.001) diets. However, ruminal pH and molar proportion of the volatile fatty acids did not differ (p>0.05) among the treatment diets. This study suggests that US and UT treatments, particularly the latter, improved straw intake, digestibility and N utilization by lambs compared to NT treatment. On the other hand, addition of SBM as a source of urease had a negligible effect on urea hydrolysis.

국내 시판 주요 건제품의 영양 특성 (Nutritional Characteristics of the Major Commercial Dried Fish in Korea)

  • This study was conducted to investigate nutrition characteristics of the following major commercially available dried fish (MCDF) in South Korea: plain-dried fish [nogari (PD-N), Alaska pollock (PD-AP), red tongue sole (PD-RTS), and young tidepool gunnel (PD-YTG)], salt-dried fish [(yellow corvina (SD-YC) and red tilefish (SD-RT)], four types of boiled-dried anchovy of different sizes (BD-As), and freeze-dried fish [Pacific saury (FD-PS), Alaska pollock (FD-AP), and Katsuobushi]. The energy content of the MCDF ranged from 103.0 to 420.5 kcal per 100 g. The MCDF in nutritional and functional properties of minerals were SD-RT and Katsuobushi in calcium; none in phosphorus, sodium, or zinc; Katsuobushi in potassium and magnesium; PD-AP, SD-YC, and SD-RT in iron, PD-AP, PD-RTS, SD-YC, and SD-RT in copper; and PD-N, PD-AP, PD-RTS, SD-YC, SD-RT, FD-PS, FD-AP and Katsuobushi in manganese. The total amino acid content of the MCDF ranged from 15.85 to 71.96 g per 100 g; the major amino acids were glutamic acid, aspartic acid, lysine, and leucine. The fatty acid content of the MCDF ranged from 0.81 to 2.93 g per 100 g. The MCFSP expected in nutritional and functional properties of vitamins were PD-N, PD-RTS, FD-PS, FD-AP, and Katsuobushi in riboflavin; PD-N, PD-AP, PD-YTG, BD-As, FD-PS, FD-AP, and Katsuobushi in niacin; PD-N, PD-RTS, SD-YC, and BD-As in vitamin A; and PD-AP, PD-YTG, SD-YC, and FD-AP in vitamin E.

해양미세조류(Schizochytrium mangrovei MM103)를 이용한 발효 대두박 급이에 따른 DHA 다량 함유 육계와 계란의 생산 (Production of DHA-Rich Meats and Eggs from Chickens Fed Fermented Soybean Meal by Marine Microalgae(Schizochytrium mangrovei MM103))

  • 대두박을 해양미세조류(Schizochytrium mangrovi MM103)로 발효하여 제조한 DHA 발효 대두박을 시판 농협사료와 3%, 5%, 10% 혼합하여 육계 병아리용과 산란계용 실험 사료를 조제하였다. 실험 사료를 3주간 급여하면 DHA 발효 대두박 농도와 관계없이 육계가슴육과 다리근육 및 계란에서 지방중 DHA 농도가 시판 사료 급여에 비하여 높아졌다. DHA 발효 대두박 10% 사료를 급여한 육계 병아리 가슴살과 다리근육 및 계란에서 각각 2.21%와 2.02% 및 1.88%로 증가하였다. 6주간 실험 사육하면 가슴살과 다리근육 및 계란 지방의 DHA 함량이 각각 5.10%와 2.48% 및 3.42%로 실험사육 기간의 경과에 따라 증가하였다. 본 연구에서 DHA 발효 대두박은 DHA 함유 가금육 및 계란 생산에 이용 가능하다는 것을 나타내었다.

재고미의 가공처리에 따른 in vitro, in situ 소화율 및 발효성상 평가 (Nutritional Evaluation of Rice with Different Processing Treatments on in vitro Rumen Fermentation Characteristics and in situ Degradation)

  • 본 연구에서는 재고미의 사료가치 평가를 위해 무처리(Control)를 비롯한 Extruding, Roasting, Steam 처리구들의 영양소 성분분석, in vitro 소화율, in situ 소화율을 분석하였다. In vitro 실험에서 Extruding은 건물 소화율과 gas 발생량이 타 처리구에 비해 상대적으로 높았고, 가장 낮은 pH를 보였으며, 특히 배양 시간 초기에 소화가 신속히 발생하였다. $NH_3-N$ 함량은 배양 6 h 이후에서 Control이 Extruding, Roasting, Steam에 비해 유의적으로 높은 값을 보였고, 이는 가열처리된 단백질의 반추위 미생물의 이용성 저하에 따른 결과로 보이나, 더 정확한 결과를 위해 건물 소화율 분석뿐 아니라 CP 소화율 분석이 필요할 것으로 사료된다. VFA 발생량에서는 Extruding이 다른 처리구들에 비해 배양 6, 12 h에서 유의적으로 높았는데 이는 Extruding 처리구의 분해가 배양 초기에 주로 이어져 발생한 결과로 추정된다. 전분을 발효시키는 박테리아에 의해 높은 비율로 생산되는 propionate 증가 폭에서도 같은 경향을 보였다. In situ 소화율은 in vitro의 소화율과는 다소 차이가 있는 것으로 여겨지나, 이는 실험 방법에 대한 오차로 여겨질 수 있으며, ED(유효분해율)를 통해 in vitro 시험과 소화율의 경향성이 유사한 것을 알 수 있었다. 따라서 본 연구결과를 근거로 상대적으로 적합한 사료가공 처리방법은 무처리, Roasting 및 Steaming인 것으로 보여진다. 또한 무처리(control)의 경우 가공처리에 따른 추가비용 발생이 없으므로 경제적으로 유리할 것으로 판단된다. 재고미가 TMR의 원료로 이용될 수 있다면 국내에서 자급 가능한 사료원료로써 우수한 경제성을 지닐 수 있을 것이다.

The Plasma Level of Insulin-like Growth Factor-I (IGF-I) in Relation to Mammary Circulation and Milk Yield in Two Different Types of Crossbred Holstein Cattle

  • The objective of the present study was to determine the plasma level of insulin-like growth factor-I (IGF-I) in relation to mammary blood flow and milk yield including biological variables of relevance to milk synthesis in two different types of crossbred Holstein cattle at 3 different stages of lactation. Eight heifers were 87.5% HF and eight 50% HF animals were selected for the experiments. The three stages of lactation tested were: early lactation (30 days postpartum), mid-lactation (120 days postpartum) and late lactation (210 days postpartum). Animals in each group were fed a concentrate and rice straw treated with 5% urea as the source of roughage throughout the experiments. In early lactation, mammary blood flow and milk yield of 87.5% HF animals were significantly higher than those of 50% HF animals. In mid- and late lactation, both mammary blood flow and milk yield showed a proportional decrease from the early lactating period of 87.5% HF animals. The trends for persistency were observed in 50% HF animals as for udder blood flow and milk yield throughout the experimental periods. The plasma glucose level of the 50% HF animals was significantly higher than those of 87.5% HF animals in both early and mid-lactation. The concentrations of arterial plasma free fatty acids ($C_{16}\;to\;C_{18}$) were higher in 50% HF animals as compared with 87.5% HF animals in all periods of study. In early lactation, the concentration of plasma growth hormone (GH) of 87.5% HF animals was higher than those of the 50% HF animals, thereafter the mean level of plasma growth hormone declined in both mid- and late lactation. The concentration of plasma IGF-I of 50% HF animals was significantly higher than those of 87.5% HF animals in all stages of lactation. There were no differences among stages of lactation for the levels of plasma IGF-I, insulin and growth hormone in 50% HF animals. In 87.5% HF animals, the plasma levels of both IGF-I and insulin were lower in early lactating period while it showed an increase during mid- and late lactation. The present results indicated that the regulatory role for the higher mammary blood flow and milk yield during lactation in 87.5% HF are not mediated via the higher level of circulating IGF-I. Differences in mammary blood flow and milk yield between 50% HF and 87.5% HF animals are in part due to a higher concentration of circulating growth hormone. The lower level of circulating growth hormone in 50% HF animals would be regulated by higher levels of IGF-I, free fatty acid and glucose in plasma.

Effects of Freeze-dried Citrus Peel on Feed Preservation, Aflatoxin Contamination and In vitro Ruminal Fermentation

  • The objective of this study was to investigate antimicrobial activity, during the storage period, of animal feed and any effects on in vitro rumen digestion by supplementing different levels (5.55, 11.1, and 22.2 g/kg) of freeze dried citrus peel (FDCP) to the feed compared to untreated feed and feed treated with an antifungal agent (AA) at 0.05 g/kg. In a preservation test, feed supplemented with FDCP showed no deterioration over 21 days. Untreated feed and AA-treated feed, however, showed signs of deterioration after 16 days storage. Yellow colour and red colour, measured by spectro chromameter, decreased in the untreated and AA-treated feeds, but not in feed supplemented with FDCP. Aflatoxin was detected in untreated and AA-treated feeds at 16 days (8 ppb and 2 ppb) and 21 days (8 ppb and 4 ppb), but aflatoxin was not detected in the feed supplemented with FDCP. In a second experiment, fermentation by rumen microorganisms of FDCP (22.2 g/kg) and AA (0.05 g/kg) supplemented feeds was studied in vitro. Feeds were incubated with buffered rumen fluid for 3, 6, 9, 12, 24, and 48 h. Dry matter digestibility (DMD) and organic matter digestibility (OMD) were affected by treatment, but ammonia-N, total, and individual volatile fatty acids (VFA) were not adversely affected by treatment. In conclusion, the results indicated that FDCP might be useful for inhibiting microbial growth of animal feed during storage without disrupting rumen fermentation.

Effects of Forage Sources on Rumen Fermentation Characteristics, Performance, and Microbial Protein Synthesis in Midlactation Cows

  • Eight multiparous Holstein cows ($632{\pm}12$ kg BW; $135{\pm}16$ DIM) were used in a replicated $4{\times}4$ Latin square design to evaluate the effects of forage sources on rumen fermentation characteristics, performance, and microbial protein (MCP) synthesis. The forage portion of the diets contained alfalfa hay (AH), oat hay (OH), Leymus chinensis (LC), or rice straw (RS) as the primary source of fiber. Diets were isonitrogenous and isocaloric, and cows were fed four corn silages based total mixed rations with equivalent nonfiber carbohydrate (NFC) and forage neutral detergent fiber (NDF). Dry matter intake was not affected by the source of dietary forages, ranging from 18.83 to 19.20 kg/d, consequently, milk yield was similar among diets. Because of the numerical differences in milk fat and milk protein concentrations, 4% FCM and ECM yields were unchanged (p>0.05). Mean rumen pH, NH3-N content, and concentrations of volatile fatty acids in the rumen fluid were not affected by the treatments (p>0.05). Dietary treatments did not affect the total tract apparent digestibility of dry matter, organic matter, and crude protein (p>0.05); however, digestibility of NDF and acid detergent fiber in RS diet was higher compared with AH, OH, and LC diets (p<0.05). Total purine derivative excretion was higher in cows fed AH, OH, and LC diets compared with those fed RS diet (p<0.05), consequently, estimated MCP synthesis was 124.35 g/d higher in cows fed AH diet compared with those fed RS diet (p<0.05). The results indicated that cows fed AH, OH, LC, and RS diets with an equivalent forage NDF and NFC have no unfavourable effect on the ruminal fermentation and productive parameters.

Development of Appropriate Fibrolytic Enzyme Combination for Maize Stover and Its Effect on Rumen Fermentation in Sheep

  • In vitro studies were undertaken to develop an appropriate fibrolytic enzymes cocktail comprising of cellulase, xylanase and ${\beta}$-D-glucanase for maize stover with an aim to increase its nutrient utilization in sheep. Cellulase and xylanase added individually to ground maize stover at an increasing dose rates (0, 100, 200, 400, 800, 1,600, 3,200, 6,400, 12,800, 25,600, 32,000, 38,400, and 44,800 IU/g DM), increased (p<0.01) the in vitro dry matter digestibility and in vitro sugar release. The doses selected for studying the combination effect of enzymes were 6,400 to 32,000 IU/g of cellulase and 12,800 to 44,800 IU/g of xylanase. At cellulase concentration of 6,400 IU/g, IVDMD % was higher (p<0.01) at higher xylanase doses (25,600 to 44,800 IU/g). While at cellulase doses (12,800 to 32,000 IU/g), IVDMD % was higher at lower xylanase doses (12,800 and 25,600 IU/g) compared to higher xylanase doses (32,000 to 44,800 IU/g). At cellulase concentration of the 6,400 to 32,000 IU/g, the amount of sugar released increased (p<0.01) with increasing levels of xylanase concentrations except for the concentration of 44,800 IU/g. No effect of ${\beta}$-D-glucanase (100 to 300 IU/g) was observed at lower cellulase-xylanase dose (cellulase-xylanase 12,800 to 12,800 IU/g). Based on the IVDMD, the enzyme combination cellulase-xylanase 12,800 to 12,800 IU/g was selected to study its effect on feed intake and rumen fermentation pattern, conducted on 12 rams (6 to 8 months; $20.34{\pm}2.369$ kg body weight) fed 50% maize stover based TMR. The total volatile fatty acids (p<0.01) and ammonia-N concentration was higher in enzyme supplemented group, while no effect was observed on dry matter intake, ruminal pH and total nitrogen concentration.

Comparison of Quality Traits of Meat from Korean Native Chickens and Broilers Used in Two Different Traditional Korean Cuisines

  • With the aim of investigating the differences in the quality traits between Korean native chicken (Hanhyup, KNC) and broilers commonly used in two different traditional Korean cuisines, the chemical composition and sensory properties of breast and thigh meat from the two chicken strains were assessed. KNC for baeksuk (chicken meat braised in soup with various Oriental medicinal plants; KNL), KNC for samgyetang (similar to baeksuk but young chickens and ginseng are used; KNS), broiler for baeksuk (BL), and broiler for samgyetang (BS) were used as treatments in this study. KNL and KNS contained higher protein but lower fat content than BL and BS. The $L^*$ values of breast and thigh meat, but not the $a^*$ values, were significantly different between KNS and BS, whereas significant differences in both values were observed between KNL and BL. Compared to the other three types of chickens, KNS contained the highest total and insoluble collagen content, and KNL and BL showed higher inosine-5'-monophosphate content in their meat. Overall, KNL and KNS contributed darker, less tender meat with higher protein and less fat content together with more n-3 fatty acids, as opposed to their counterparts used for the same cuisines. Based on the results of the sensory analysis, even though there are some differences in physiochemical traits, different chicken sources do not differ in overall sensory quality. This information can help consumers to understand better the meat available for their preferred traditional cuisines.
