Chlorogenic Acid, an Antioxidant Principle from the Aerial Parts of Artemisia iwayomogi that Acts on 1,1-Diphenyl-2-picrylhydrazyl Radical

  • The antioxidant activity of Artemisia iwayomogi was determined by measuring the radical scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical. The methanol extract of A. iwayomogi showed strong antioxidant activity, and thus fractionated with several solvents. The antioxidant activity potential of the individual fraction was in the order of ethyl acetate > n-butanol > water > chloroform > n-hexane fraction. The ethyl acetate and n-butanol soluble fractions exhibiting strong antioxidant activity were further purified by repeated sitica get and Sephadex LH-20 column chromatography. Antioxidant chlorogenic acid was isolated as one of the active principles from the n-butanol fraction, together with the inactive components, 1octacosanol, scopoletin, scopolin, apigenin $7, 4^{I}$-di-O-methylether, luteolin $6, 3^{I}$-di-O-methylether (jaceosidin), apigenin methylether (genkwanin), 2, 4-dihydroxy-6-methoxyacetophenone $4-O-{\beta}-$D-glucopyranoside and quebrachitol. The antioxidant activity of chlorogenic acid was comparable to that of L-ascorbic acid, which is a well known antioxidant.

Comparison of Biological Activities of Extracts from Different Parts and Solvent Fractions in Cornus kousa Buerg (산딸나무의 부위별 추출물 및 용매 분획물의 생리활성 비교)

  • Several extracts of different parts and solvent fractions of Cornus kousa were obtained and their functional material contents, antioxidant activities and tyrosinase inhibition effects were determined. Content of total polyphenols and flavonoids contents in flower were 169.638 $mg{\cdot}g^{-1}$ and 25.418 $mg{\cdot}g^{-1}$, respectively, which were much higher than those of other parts. Also, flower extracts showed the strongest effects on DPPH and ABTS radicals scavenging and ferrous ion chelating. In flower, leaf, and stem extracts, inhibition effects on peroxidation of linoleic acid determined by ferric thiocyanate(FTC) method were higher than a synthetic antioxidant, BHT. Tyrosinase inhibition activities were shown only in flower extract. Flower and leaf extracts, showing high biological activities in various system, were successively reextracted with n-hexane, chloroform, ethylacetate and n-butanol. Total polyphenol contents of water fractions were higher than any other solvent fractions in both flower and leaf, 67.006 $mg{\cdot}g^{-1}$ and 67.739 $mg{\cdot}g^{-1}$, respectively. But total flavonoid contents were higher in ethyl acetate fraction for flower extract and butanol fraction for leaf extract. Among the solvent fractions, the highest efficiency of free radical scavenging activities was obtained in ethyl acetate fraction for flower extract and n-butanol fraction for leaf extract. Tyrosinase inhibition activities were higher in water fraction for both flower and leaf extracts, 49.24% and 31.8%, respectively.

  • Black fermented garlic includes many pharmacological components. Therefore, in this study, black fermented garlic wine was manufactured and its flavor compounds were investigated difference of aging chips from America and France. The fermented wine was stored at $10^{\circ}C$ for 6 months. GC/MS was used for the flavor components analysis. Wine using American chip contained 2-methyl-1-propanol, 3-methyl-1-butanol, 2-methyl-1-butanol, acetaldehyde, butanoic acid, octanoic acid, 1,1-diethoxyethane, and allyl methyl sulfide. 1-Propanol, 2-methyl-1-propanol, 3-methyl-1-butanol, acetaldehyde, acetic acid, propanoic acid, butanoic acid, octanoic acid, 2-heptanone, 1,1-diethoxyethane, N-amino32-hydroxypropanamidate, n-butylamine, and chloroacetonitrile were detected as major flavor compounds using France chips. Especially, the wine contained allyl methyl sulfide that was resulted from black fermented garlic. There were more compounds that smell like fruit in the wine using American chips relatively. And allyl methyl sulfide was detected only in the wine using America chips. Whereas acetic acid was detected only in the wine using France chips.

Effects of Butanol Isomers on the Mixed Micellization of TTAB/Brij 35 Mixed Surfactant Systems (TTAB/Brij 35 혼합계면활성제의 미셀화에 미치는 부탄올 이성질체들의 효과)

  • critical micelle concentration (CMC) and the counter ion binding constant (B) in a mixed micellar state of the trimethyltetradecylammonium bromide (TTAB) with the polyoxyethylene (23) lauryl ether (Brij 35) at 25oC in water and in aqueous solutions of butanol isomers were determined as a function of 1 (the overall mole fraction of TTAB) by the use of electric conductivity method and surface tensiometer method. Various thermodynamic parameters (Xi, i, Ci, aiM, and Hmix) were calculated by means of the equations derived from the nonideal mixed micellar model. The results say that the effects of butanol isomers on the micellization of TTAB/Brij 35 mixtures have been in the order of n-butanol>iso-butanol>t-butanol> water.

Purification and Properties of Protease Inhibitor from Streptomyces sp. SK-862 (방선균이 생성하는 단백질 가수분해효소 저해물질의 정제 및 특성)

  • A strain of Streptomyces sp. SK-862, isolated from soil in Wonju city, was able to prodce a biologically active substance that has a strong inhibitory activity against proteolsis by trypsin. The inhyibitory substance was extracted by n-butanol, and then purified by the adsorption chromatography followed by the reverse-phase high performacne liquid chromatography. The purified substance was stable over the pH range from 2 to 10, but was unstable when treated at 8$0^{\circ}C$ for 60 min. This substance was soluble in water, methanol, ethanol nd butanol, but insoluble in chlorofrom and ethylacetate. The Rf value of the purified substance on the thin layer chromatography were 0.56 in n-butanol : methanol : water(5 : 3 : 1v/v) solvent system compare dto 0.23 in ethanol : ammonium hydoxide : water(8 : 1 : 1v/v) solvent system. This substance has maximum absorption at 259 nm. The chemical reaction of the substance was negative for sugar but positive for ninhydrine and iodine reaction.

Inhibitory Effects of Artemsia capillaris Thumb. on ${\alpha}-Glucosidase$ and ${\alpha}-Amylase$

  • This study was carried out to investigate inhibitory effect of extracts from Artemisia capillaris Thumb. on maltase, sucrase, ${\alpha}-amylase$, nonspecific ${\alpha}-glucosidase$, and postprandial hyperglycemia. Methanol extract and organic solvent (n-hexane, ethyl acetate, butanol, aqueous) fractions from the medicinal herb were determined for the inhibitory activities against maltase, sucrase and ${\alpha}-amylase$. The methanol extract from A. capillaris strongly inhibited maltase (57%) and ${\alpha}-glucosidase$ (72%) at the concentration of 100 ${\mu}g/m{\ell}$. Among the four fractions (n-hexane, ethyl acetate, butanol, aqueous) examined, the butanol fraction from A. capillaris showed potent inhibitory effects on maltase (73%), sucrase (33%), and ${\alpha}-amylase$ (75%) at the concentration of 100 ${\mu}g/m{\ell}$. The butanol fraction from Artemisia capillaris also exhibited significant reductions (20%) of blood glucose elevation in mice loaded with maltose. These results suggest that the extract from Artemisia capillaris can be used as a new nutraceutical for inhibition on postprandial hyperglycemia

  • The mechanical degradation of poly (-vinylacetate) in several mixed solvents (dioxane-n-butanol, dioxane-sec-butanol, dioxane-ethyleneglycol, dioxane-kerosene.) was studied using the capillary flow method. The velocity constant of scission reaction (k) and the limited degree of polymerization (g) were compared at the same value of [${\eta}$], which is considered as the parameter of molecular dimension of polymers in solution. As result, (k) did not change much, even if the species and the volume fraction of poor (non-) solvents changed, while the value of (g) changed according to the species of poor solvents and the value of [${\eta}$]. From the facts described above, It follows that the limited degree of polymerization (g) were affected by the composition and distribution of mixed solvent molecules around the polymer chain, and the value of $\alpha$ (at [${\eta}$] = $KM^a$) in the polymer solution.

  • AMX is one of extremely high mutagenic compound produced from the reaction of the chlorine and the organic compound during the disinfection of tap water. In this paper, the chemical derivatization of MX with 2% sulfuric acid-methanol, iso-propanol, sec-butanol and n-butanol derivatives for the GC/MS analysis were tested or compared. Protective effects of Betula platyphylla var. japonica extracts against the cellular damage induced by reactive oxygen species

  • In our present study, total methanol extracts prepared from B. platyphylla var. japonica showed a significant increase in cell proliferation upon the induction of oxidative stress by hydrogen peroxide or $\gamma$-ray irradiation. Total methanol extracts were fractionated into five separate preparations i.e. n-hexane, dichloromethane, ethylacetate, n-butanol and water fractions. Among these, the ethylacetate and butanol fractions of B. platyphylla var. japonica showed the highest protective effects against oxidative stress induced by hydrogen peroxide. These fractions also showed strong protective effects against $\gamma$-ray irradiation. When we evaluated the cytotoxicity of these fractions, the butanol fraction showed no effects in a colony formation assay. In addition, the butanol fraction showed a cell proliferation activation effect evidenced by significant increase in the colony formation of $\gamma$-ray irradiated cells. Both a radical scavenging activity and clonogenic activity assay suggested that the mechanism behind this protective effect against reactive oxygen species may be due to the radical scavenging and cell proliferation activity of B. platyphylla var. japonica extracts.

The Measurement of Lower Flash Points For Binary Mixtures (이성분계 혼합물의 하부인화점 측정)

  • The flash point is an important indicator of the flammability of a chemical. In this study, the flash points for the n-propanol+n-butanol and n-propanol+acetic acid systems were measured by Tag open-cup apparatus. The experimental data were compared with the values calculated by the Raoult's law and optimization method based on van Laar and UNIQUAC equations. The calculated values by optimization method were found to be better than those based on the Raoult's law.
