Enzymatic Synthesis of β-Glucosylglycerol and Its Unnatural Glycosides Via β-Glycosidase and Amylosucrase

  • ${\beta}$-Glucosylglycerol (${\beta}-GG$) and their derivatives have potential applications in food, cosmetics and the healthcare industry, including antitumor medications. In this study, ${\beta}-GG$ and its unnatural glycosides were synthesized through the transglycosylation of two enzymes, Sulfolobus shibatae ${\beta}$-glycosidase (SSG) and Deinococcus geothermalis amylosucrase (DGAS). SSG catalyzed a transglycosylation reaction with glycerol as an acceptor and cellobiose as a donor to produce 56% of ${\beta}-GGs$ [${\beta}$-$\text\tiny{D}$-glucopyranosyl-($1{\rightarrow}1/3$)-$\text\tiny{D}$-glycerol and ${\beta}$-$\text\tiny{D}$-glucopyranosyl-($1{\rightarrow}2$)-$\text\tiny{D}$-glycerol]. In the second transglycosylation reaction, ${\beta}$-$\text\tiny{D}$-glucopyranosyl-($1{\rightarrow}1/3$)-$\text\tiny{D}$-glycerol was used as acceptor molecules of the DGAS reaction. As a result, 61% of ${\alpha}$-$\text\tiny{D}$-glucopyranosyl-($1{\rightarrow}4$)-${\beta}$-$\text\tiny{D}$-glucopyranosyl-($1{\rightarrow}1/3$)-$\text\tiny{D}$-glycerol and 28% of ${\alpha}$-$\text\tiny{D}$-maltopyranosyl-($1{\rightarrow}4$)-${\beta}$-$\text\tiny{D}$-glucopyranosyl-($1{\rightarrow}1/3$)-$\text\tiny{D}$-glycerol were synthesized as unnatural glucosylglycerols. In conclusion, the combined enzymatic synthesis of the unnatural glycosides of ${\beta}-GG$ was established. The synthesis of these unnatural glycosides may provide an opportunity to discover new applications in the biotechnological industry.

Effects of Pinitol Supplementation and Strength Training on Anaerobic Performance and Status of Energy Substrates in Healthy Young Men

  • To assess the effect of pinitol supplementation and strength training for two weeks on the anaerobic capacity during and after exercise, and improvement of glucose metabolism during the recovery period of muscular fatigue with repeated acute bouts of cycling exercise, a total of 24 healthy young men were recruited and randomly and equally divided into three groups; pinitol supplementation group (PSG), placebo group (PLG), and control group (CON). Using a randomized double-blinded design, subjects in PSG were provided pinitol supplement, consumed orally 1.2 g/day, and participated in the resistance exercise program and cycling exercise for two weeks. Subjects in PLG underwent the same protocol as those in PSG but consumed the same amount of placebo. No supplementation and exercise program was given to CON. Before and after the intervention, all subjects were tested for their anaerobic capacities evaluated by Wingate test twice separated by 30 min. During the test, peak anaerobic power (PP), mean anaerobic power, total work, and fatigue index were evaluated During resting and recovery, blood samples were drawn and plasma pinitol, myo-inositol, chiro-inositol, insulin, free fatty acid, glucose, and lactate levels were analyzed After two weeks, PP and relative PP of the second biking were improved from the first biking in PSG only (p<0.05). No changes were found in all other variables of Wingate test in all groups. No statistical differences between groups and pre- and post-intervention were observed in concentrations of pinitol, myo-inositol, and chiro-inositol, but pinitol concentration was higher during recovery compared to the baseline in all groups and testings (p<0.05). Lactate level during recovery was higher than the resting level, but no other blood parameters were significantly changed. In conclusion, two weeks of pinitol supplementation in conjunction with short duration of anaerobic training in healthy young men did not induce any obvious benefits in terms of anaerobic capacity and energy metabolism Individual and/or population susceptibility may be one factor responsible for adopting pinitol supplementation.

Dynamical Expression of MicroRNA-127-3p in Proliferating and Differentiating C2C12 Cells

  • MicroRNAs (miRNAs) are highly conserved, short non-coding RNAs that regulate gene expression at the posttranscriptional level. Although many miRNAs are identified in muscles and muscle cells, their individual roles are still not fully understood. In the present study, we investigated a muscle highly-expressed miRNA, miR-127-3p, in C2C12 myoblasts and tissues of goats with different muscle phenotypes (Boer vs Wushan black goats). Our results demonstrated that i) miR-127-3p was extensively expressed in tissues of goats; ii) miR-127-3p was higher expressed in muscle, spleen, heart, and skin in the muscular goats (Boer goats) than the control (Wushan black goats). Then we further characterized the dynamical expression of miR-127-3p, MyoD, MyoG, Myf5, Mef2c, and Myosin in the proliferating and differentiating C2C12 myoblasts at day of 0, 1, 3, 5, and 7 in culture mediums. Especially, we found that miR-127-3p was significantly higher expressed in the proliferating than differentiating cells. Our findings suggest that miR-127-3p probably plays roles in the proliferation and differentiation of myoblasts, which further underlies regulation of muscle phenotype in goats.

The difference of castration timing of Korean Hanwoo bulls does not significantly affect the carcass characteristics

  • It is already well known that castration improves marbling quality but exact timing of castration is still highly debated in beef cattle production industry. After castration, blood hormonal changes occur in steer and objective of this study was to investigate the effects of growth hormone (GH) levels on adipocyte differentiation in stromal vascular cells (SVCs) and transdifferentiation into adipocytes in C2C12 myoblasts. Total GH concentrations were measured via enzyme-linked immunosorbent assay (ELISA) in 24 male calves and 4 female calves. Cell proliferation, cellular triglyceride (TG) accumulation, and the cell's lipolytic capability were measured in C2C12 myoblasts and SVCs. Myogenic, adipogenic, and brown adipocyte-specific gene expression was measured via real-time polymerase chain reaction (PCR) using SYBR green. Serum GH levels were the highest in late-castrated calves. Treatment with 5 ng/mL GH resulted in greater TG accumulation as well as increased CCAAT-enhancer-binding protein (C/EBP)α and peroxisome proliferator-activated receptor (PPAR)γ expression compared to that after treatment with 15 ng/mL GH. Treatment with 5 ng/mL GH also resulted in lower myogenin (myo)G and myoD expression compared to that after treatment with 15 ng/mL GH. The expression of bone morphogenetic protein (BMP) 7 after treatment with 5 ng/mL GH was higher than that after treatment with 15 ng/mL GH. But carcass characteristics data showed no significant difference between early and late castrated steers. Therefore, our results indicate that castration timing does not seem to be inevitable determinate of carcass qualities, particularly carcass weight and marbling score in Hanwoo beef cattle.

Design of the Wide-Band Printed-Monopole Antenna Using Parasitic Elements (기생 소자를 이용한 광대역 프린트 모노폴 안테나의 설계)

  • In this paper, we have proposed a printed monopole antenna using parasitic elements. The broadband characteristics of the antenna is obtained by using the printed monopole and parasitic elements. To confirm the broadband characteristics of the antenna, of have designed and fabricated the proposed antenna. The return loss and radiation patterns are measured in the frequency range of$0.8{\sim}1.2\;GHz$. The measured results show that the proposed antenna has return loss less than -10 dB in the operating frequency band and the radiation pattern is the dipole-like patterns. The antenna gain varies from 1.7 to 4.6 dBi in the operating frequency band. Thus, the proposed antenna can be used for the broadband repeater antenna $0.806{\sim}0.960\;GHz$(TRS-800, CDMA and GSM-900).

  • Polycystic ovary syndrome (PCOS) is a common endocrine and metabolic disorder among reproductive-age women. As a leading cause of anovulatory infertility, it complicates fertility treatments, including in vitro fertilization. The widely accepted 2003 Rotterdam diagnostic criteria for PCOS include sub-phenotypes based on variations in androgen excess, ovulatory dysfunction, and polycystic ovarian morphology. In this systematic review, we examined the impacts of inositol and vitamin D on fertility in PCOS. Adhering to the Preferred Reporting Items for Systematic Reviews and Meta-Analyses 2020 guidelines, we used relevant keywords to comprehensively search databases including PubMed, Google Scholar, and MDPI. From an initial pool of 345 articles, 10 met the inclusion criteria. The articles suggest that vitamin D and inositol, particularly myo-inositol and D-chiro-inositol, may represent therapeutic options for PCOS. Vitamin D influences ovarian follicular development, glucose regulation, and insulin sensitivity. When combined with metformin therapy, it is associated with improved menstrual regularity and ovulation. Inositol is crucial for cellular signaling, energy metabolism, glucose regulation, and fertility. This systematic review underscores the importance of investigating inositol and vitamin D within a PCOS management strategy, given the disorder's prevalence and impacts on fertility and metabolic health. Although these agents show promise, additional research could clarify their mechanisms of action and therapeutic benefits. This review emphasizes the need for exploration of effective treatments to improve the quality of life among individuals with PCOS. Inositol and vitamin D represent potential options, but more studies are required to elucidate their roles in the management of this condition.

  • Production of camptothecin ((PT) from callus cultures of Camptotheca acuminata Decne was affected by light and culture conditions. Among the culture media tested, modified B5 medium containing 3% (w/v) sucrose, 2 mg/L B,4-D, 2 times of MS medium vitamins, 500 mg/L casein hydrolysate, 250 mg/L myo-inositol, 0.05% (w/v) activated charcoal, and 0.15% (w/v) gelite was used for callus induction . The highest cell growth and CPT production were obtained in dark and green light condition, respectively. Photoperiod has no effect on cell growth and CPT production. Both cell growth and CPT production were also influenced by combination ratio of red and blue light .Cell growth and CPT production were the highest in the ratio of red and blue light,90:10.

  • In this paper, the infinite time optimal regulation problem for weakly coupled bilinear systems with quadratic performance criteria is obtained by a sequence of algebraic Lyapunov equations. This is the new approach is based on the successive approximation. In particular, the order reduction is achieved by using suitable state transformation so that the original Lyapunov equations are decomposed into the reduced-order local Lyapunov equations. The proposed algorithms not only solve optimal control problems in the weakly coupled bilinear system but also reduce the computation time. This paper also includes an example to demonstrate the procedures.

  • To develop a new functional agent for cosmetics, we investigated the anti-aging activities in fibroblasts of casuarictin. The anti-aging effect of casuarictin in CCD-986sk cell was as follows: it inhibited ROS expression increased by Ultraviolet B and suppressed pro-collagen expression. Also, casuarictin had inhibited Matrix metalloproteinase-1 expression. Therefore, the results suggested that casuarictin has considerable potential as a cosmetics ingredient with a anti-aging effect.
