국내에서 분리한 20종의 Alternaria 277개 균주를 사용하여 Alternaria 진균소독인 altenuene(ALT), altertoxin-I(ATX-I) 및 tenuazonic acid(TA)의 in vitro에서의 생성능과 그 분포를 조사하였다. A. alternata와 그와 형태적으로 유사한 A. kikuchiana, A. longipes 및 A. mail는 비록 종간 및 종내의 균주간에 생성량의 차이는 매우 다양하였지만 TA를 비롯한 5종의 진균독소를 모두 생성하였다. A sesami와 A. sesamicola는 4종의 진균독소(AOH, AME, ALT, ATX-I)를 생성하였고 A. cucumerina, A. dauci, A. macrospora, A. porri, A. solani, A. tagetica와 A. zinniae와 같은 대형분생포자와 긴 beak를 형성하는 7종의 Alternaria균들은 AOH와 AME만을 생성하였다. A. brassicicola, A. helianthi, A. panax, A. radicina 및 A. raphani등 5종의 Alternaria는 5종의 진균 독소를 모두 생성하지 않았다.
Mycotoxins are a group of toxicants giving a risk potential to human health in connection with the daily food intake. Food commodities once contaminated with mycotoxins can not be detoxified by any economic means and prevention was suggested as the only measure. In order to minimize the economic loss and health hazard posed by mycotoxins and toxicoses, systematic and toxicological studies on the subject should be undertaken. Most reports in Korea were concentrated on the mycological studies of relatively easy techniques and the confirmation or quantitation of mycotoxins was rarely done. Research topics to be undertaken in future may be exemplifid below: (1) Establishing assay methods for individual or multi-residue of mycotoxins (2) Monitoring of mycotoxins for suspicious food or feed samples in Korea (3) Epidemiological survey of mycotoxicoses (4) Etiological survey of disease outbreaks associated with mycotoxins (5) Accumulation of testing method and data on the toxicity of mycotoxins (6) Legal regulation to control mycotoxins and development of their detoxification / elimination methods
The alkaloids, piperlongumine, piperine, pipernonaline, and piperoctadecalidine, isolated from Piper longum L., were found to inhibit the biosynthesis of aflatoxin $B_1$ (AF$B_1$) in Aspergillus flavus WRRC 3-90-42-12. Piperlongumine was the most active among the compounds tested, with a 96% inhibition of AF$B_1$biosynthesis at 0.2% (w/v) supplement in a potato dextrose agar (PDA) medium. The three other piperidine alkaloids, pipeline, pipernonaline, and piperoctadecalidine, also inhibited the biosynthesis of AF$B_1$. Of these three alkaloids, piperoctadecalidine exhibited a potent inhibitory activity with a 100% inhibition of AF$B_1$ production at 0.7% (w/v) supplement in a PDA medium. Therefore, piperlongumine and piperoctadecalidine could be used as antiaflatoxigenic agents in agricultural industries. To determine the antiaflatoxigenic mode of action of piperlongumine, further studies are needed.

Deoxynivalenol (DON), a Fusarium mycotoxin, causes health hazards for both humans and livestock. Therefore, the aim of this study was to investigate the metabolic profiles of the liver, serum, and urine of piglets fed DON using proton nuclear magnetic resonance ($^1H-NMR$) spectroscopy. The $^1H-NMR$ spectra of the liver, serum, and urine samples of the piglets provided with feed containing 8 mg DON/kg for 4 weeks were aligned and identified using the icoshift algorithm of MATLAB $R^2013b$. The data were analyzed by multivariate analysis and by MetaboAnalyst 4.0. The DON-treated groups exhibited discriminating metabolites in the three different sample types. Metabolic profiling by $^1H-NMR$ spectroscopy revealed potential metabolites including lactate, glucose, taurine, alanine, glycine, glutamate, creatine, and glutamine upon mycotoxin exposure (variable importance in the projection, VIP > 1). Forty-six metabolites selected from the principal component analysis (PCA) helped to predict sixty-five pathways in the DON-treated piglets using metabolite sets containing at least two compounds. The DON treatment catalyzed the citrate synthase reactions which led to an increase in the acetate and a decrease in the glucose concentrations. Therefore, our findings suggest that glyceraldehyde-3-phosphate dehydrogenase, citrate synthase, ATP synthase, and pyruvate carboxylase should be considered important in piglets fed DON contaminated feed. Metabolomics analysis could be a powerful method for the discovery of novel indicators underlying mycotoxin treatments.
These studies were carried out to detect the presence of mycotoxin producing fungi in various kind of grains and foodstuffs in Korea. The experiments were divided into three parts: bacteriologic, toxicologic and electron microscopic studies. The results were summarized as follows: 1. From the 133 various samples, 426 colonies of fungi were isolated. In 405 of 426 colonies, it was possible to identify 17 genera. Among the identified strains, the predominant genera were Penicillium, Aspergillus and Alternaria. 2. In cytotoxicity test, 20 strains showed mild to severe toxic effects in mice and 24 strains showed toxic effects on HeLa cells among the 107 strains of experiments. 3. In electron microscopic studies of liver cells from animal which had been treated with toxin like substances, the liver cells showed the cytoplasmic changes: dilatation of endoplasmic reticulum, swelling of mitochondria, disappearance of mitochondrial cristae, increased number of lipid and glycogen particles. Nucleus and nuclear envelope alterations were also noted. 4. In fine structure of HeLa cells treated with culture filtrates of mycotoxin producing fungi and experimental strains had been noted a certain specific changes induced by culture filtrates. These alterations showed the disappearance of golgi complex, endoplasmic reticulum vacuolization and mitochondrial changes. 5. As a mass screening, the cytotoxicity tests of HeLa cells and histopathologic study of mice liver cells treated with toxin-like substances, employed are feasible to detect mycotoxin producing fungi.
Objective: To evaluate the effect of lactic acid bacteria and storage temperature on the microbial, chemical and mycotoxin composition of corn silage. Methods: Corn was harvested at 32.8% dry matter, and chopped to 1 to 2 cm. The chopped material was subjected to three treatments: i) control (distilled water); ii) $1{\times}10^6$ colony forming units (cfu)/g of Lactobacillus plantarum; iii) $1{\times}10^6cfu/g$ of Pediococcus pentosaceus. Treatments in triplicate were ensiled for 55 d at $20^{\circ}C$, $28^{\circ}C$, and $37^{\circ}C$ in 1-L polythene jars following packing to a density of approximately $800kg/m^3$ of fresh matter, respectively. At silo opening, microbial populations, fermentation characteristics, nutritive value and mycotoxins of corn silage were determined. Results: L. plantarum significantly increased yeast number, water soluble carbohydrates, nitrate and deoxynivalenol content, and significantly decreased the ammonia N value in corn silage compared with the control (p<0.05). P. pentosaceus significantly increased lactic acid bacteria and yeast number and content of deoxynivalenol, nivalenol, T-2 toxin and zearalenone, while decreasing mold population and content of nitrate and 3-acetyl-deoxynivalneol in corn silage when stored at $20^{\circ}C$ compared to the control (p<0.05). Storage temperature had a significant effect on deoxynivalenol, nivalenol, ochratoxin A, and zearalenone level in corn silage (p<0.05). Conclusion: Lactobacillus plantarum and Pediococcus pentosaceus did not decrease the contents of mycotoxins or nitrate in corn silage stored at three temperatures.
To analyze prevalence of mycotoxins, a total of 74 feedstuff samples were collected from silos (n=37) and hoppers (n=37) in nine pig farms. Six mycotoxins were tested with commercialized ELISA kits. All samples were contaminated with four or more mycotoxins. Zearalenone was detected in all of the tested samples. Ochratoxin, deoxynivalenol and H-2/HT-2 toxin were detected in more than 90% of the samples. And also, fumonisin was positive in 89.2% of the samples from the silos, 75.2% from the hoppers, respectively. On the other hand, aflatoxin was detected in about 40% of the samples. When the behavior of lactating sows was observed, possible mycotoxicosis was suspected. It was confirmed that their feedstuffs were contaminated with high levels of mycotoxins such as ochratoxin and T-2/HT-2 toxin. After cleaning the feedline, the clinical symptoms in sows suspected with mycotoxicosis were disappeared. Although mycotoxin concentration in most of the feedstuffs was below the acceptance level, these data indicate that what are required is more monitoring and continuous management for mycotoxins in pig feedstuffs.
Our recent studies conducted in South America have shown that mycotoxin contamination of red chili peppers (RCPs) may be associated with an increased risk of gallbladder cancer (GBC). Whether this relationship exists in India, a country with a high incidence of GBC and high consumption of RCPs, is unclear. We therefore measured concentrations of aflatoxins (AFs) and ochratoxin A (OTA) in RCPs from areas of low, medium, and high incidence of GBC in India, and compared these concentrations with GBC incidence in each area. Twenty-one RCP samples were collected from nine cities (eight from a low-incidence area, five from a medium-incidence area, and eight from a high-incidence area). Concentrations of AFs and OTA were measured using high-performance liquid chromatography. No significant differences in mean concentrations of AFs and OTA were found in the three areas. AFB1 levels in the low-incidence area ($10.81{\mu}g/kg$) and high-incidence area ($12.00{\mu}g/kg$) were more than 2.2 and 2.4 times higher compared with the maximum permitted level of AFB1 in spices ($5.0{\mu}g/kg$) set by the Commission of the European Communities, or that ($4.4{\mu}g/kg$) obtained in our previous study in Chile. Our results show that the mean concentrations of mycotoxins in RCPs are similar among the three areas in India with different incidences of GBC. Further studies with human subjects are needed to evaluate any association between AFB1 and GBC.
