유전자보유 계통수를 이용한 Archaea와 Proteobacteria 분류

  • 염기서열 분석이 완료된 9종의 고세균 (Archaea)과 15종의 단백세균 (Proteobacteria)에 대하여 유전자보유 유무와 16S rRNA에 의한 계통수를 neighbor joining method와 통계적 의미를 갖는 bootstrap method (n=1000)를 이용하여 분석하였다. 보존적 COG와 각 미생물 보유 ortholog수에 대한 비율은 4.60% (Mezorhizobium loti)와 56.57% (Mycoplasma genitalium) 사이로 종에 따라서 공통 유전자의 보유정도가 차이를 보이는 것으로 독특한 유전자를 탐색할 수 있는 가능성을 제시하는 결과로 사료되었다. Archaeabacteria와 Proteobacteria 그리고 Firmicutes모두 유전자보유 계통수와 16S rRNA 계통수가 일치하는 부분과 일치하지 않는 부분으로 나뉘어진다는 것을 알 수 있었다.

  • Mycoplasma hyopneumoniae is among the most prevalent and important infectious agents associated with porcine respiratory disease complex. The airway dagame caused by M. hyopneumoniae adversely affect the pulmonary host defense mechanisms and may lead to secondary bacterial infections. Culture is considered to be the "gold standard" for diagnosis but this is a very slow and labor-intensive procedure. Isolation of M. hyopneumoniae is complicated by its fastidious nature and extremely slow growth. Thirty days of incubation may be necessary to detect the organism in primary broth cultures. The purposes of the study were (ⅰ) to develop nested PCR for the detection of M. hyopneumoniae for the detection of M. hyopneumoniae DNA in the formalin-fixed, paraffin-embedded lung tissues from experimentally and naturally infected pigs and (ⅱ) to compare the utility of nested PCR with in situ hybridization.   • This study was undertaken to establish reliable diagnostic-procedures for the microbiological monitoring of laboratory animals. Murine(mice and rats) antibodies against hemagglutinating virus of Japan(HVJ), mouse hepatitis virus(MHV) and Mycoplasma pulmonis(Mp) were detected sensitively and specifically in experimentally and naturally infected animals' sera by an indirect enzyme-linked immunosorbent assay(ELISA), using urease conjugated antimurine immunoglobulin. The sensitivity and specificity of the complement fixation test which has been apllied widely for serodiagnosis of HVJ, MHV and Mp infections were apparently lower than those of ELISA. From these results, the ELISA was found to be available for the serodiagnosis of HVJ, MHV and Mp infections in mice and rats.

영남지방 도축돈의 Mycoplasma 폐렴조사 및 분리균에 대한 약제 감수성 (Survey on mycoplasmal pneumonia of swine in Youngnam area and antimicrobial susceptibility of Mycoplasma hyopneumoniae isolated from Slaughter pigs)

  • The present study was carried out to investigate the prevalence of mycoplasmal pneumonia of slaugter pigs in Youngnam area during the period from 1995 to 1997. The prevalence and pathomorphology of gross lung lesions were studied from 682 slaughter pigs in 8 swine herds. Gross lesions of pneumonia were recorded in the lungs of 442(64.8%), from 367 out of them(83.0%) were diagnosed as mycoplasmal pneumonia. Microbiological examination was performed with 197 lungs with gross lesions of mycoplasmal pneumonia of slaughter pigs from 8 different swine herds. M hyopneumoniae, P multocida, A pleuropneumoniae, Streptococcus spp, Corynebacterium spp, and H parasuis were detected in 24. 4%, 48.2%, 2.5%, 11.2%, 3.6%, and 1.0% of the pneumonic lungs, respectively. A total of 48 strains of M hyopneumoniae was investigated for their in vitro susceptibility to 8 antibiotics. Among the drugs tested, lincomycin, oxytetracycline, tiamulin and tylosin showed the high activity in minimal inhibitory concentration(MIC) of $0.04{\sim}5{\mu}g/ml$ while erythromycin showed low activity in MIC values($1.25{\sim}{\geq}40{\mu}g/ml$).

도축돈의 폐렴병소에서 분리한 세균의 항생제 감수성 (Antibiotic susceptibility of microorganisms isolated from Pneumonic lungs of slaughtered pig in northern Chungnam area)

  • A total of 315 microorgainisms were isolated from 256 pneumonic lung samples of pig in abattoirs of northern Chungnam area during February to November 1994, and Identified 97 strains as Pusteurella multocida, 89 strains as Staphylococcus spp, 54 strains as Streptococcus spp, 22 strains as Mycoplasma spp, 21 strains as Escherichia coli, 18 strains as Haemophilus parsuis, 11 strains as Corymebacteroi, pyogenes, and 3 strains as Acrinobacillus spp by Gram's and Dienes stain, and biological properties test Involved API system. After that, they were examined anti biotic susceptibility for ampicillin(AM), cephalothin(CF), chloramphenicol(CP), erythromycin(EM), kanamycin(KM), gentamicin(GM), neomycin(NM), penicillin(PC), streptomycin(SM), tetracycline(TE), tiamulin(TIA), tylosin(TYL), methicillin(DP), colistin(CL) and trimet hoprim(SXT). In antibiotics susceptibility test, 293 isolates except Mycoplasma spp 22 strain were highly susceptible to DF(79.2%) and AM(76.2%), but resistant to PC(14.0%), NM(19.5%) and KM (23.2%) The multiple drug resistant patterns were noted in most isolates, whereas only 7 isolates resistant to single drugs.

  • Today swine respiratory disease is one of the most important diseases because of its economic losses and severe infection nationwide, and swine society as well as veterinary service are trying to prevent the diseases in Korea. This study would like to obtain some information useful for the control of the diseases. A total of 174 lung specimens with lesion consisted of 3 sorts; 60 were collected from nursey pigs requested for diagnostic service from March of 2006 to October of 2007, 58 finishing pigs and 56 sows were selected from slaughterhouse from September to November 2007. In the detection test of pathogens by PCR, porcine circovirus 2 (PCV2), porcine reproductive and respiratory syndrome virus (PRRSV) and Mycoplasma hyopneumoniae were positive in 95.4%, 31.6%, and 20.1%, respectively. Double infection rate with PCV2 and PRRS was 30.4%, PCV2 and M hyopneumoniae was 19.5%, triple infection with PCV2, PRRS and M hyopneumoniae was 5.7%, respectively.

  • Mycoplasma pneumoniae strain CL-s attached to broth-covered surfaces was examined sequentially during growth from single cells for morphologic and ultrastructural changes using several different electron microscopic techniques. Changes in morphology revealed both round and spindle shapes and observation of cell transitions suggested some type of morphological cycle. The round to-ovoid cells observed in the early stages of growth appeared to be viable, and morphologically and ultrastructurally different from the spherical fors which were produced during the latter stage of growth. The spindle segments were detected appeared to be structurally the same as the terminal cored structure seen in thin sections and may be a growing point or an attachment site of the cell. A tubular structure was observed in the core of the terminal structure and a microtubule-like element appeared to bridge between some spindle segments. A matrix sunstance was observed around single cells as well in the intercellular space of the colonies prepared by critical point metrical triple-layered cytoplasmic mermbranes, surfaces, of which appeared to be structurally different each other, were observed in young cells, whereas symmetrical and thicker membranes were seen in older cells. Small bodies were found in 4d or older cultures and did not appear to contain any internal structures or an easily detectable unit membrane.

Current perspectives on atypical pneumonia in children

  • The major pathogens that cause atypical pneumonia are Mycoplasma pneumoniae, Chlamydophila pneumoniae, and Legionella pneumophila. Community-acquired pneumonia (CAP) caused by M. pneumoniae or C. pneumoniae is common in children and presents as a relatively mild and self-limiting disease. CAP due to L. pneumophila is very rare in children and progresses rapidly, with fatal outcomes if not treated early. M. pneumoniae, C. pneumoniae, and L. pneumophila have no cell walls; therefore, they do not respond to β-lactam antibiotics. Accordingly, macrolides, tetracyclines, and fluoroquinolones are the treatments of choice for atypical pneumonia. Macrolides are the first-line antibiotics used in children because of their low minimum inhibitory concentrations and high safety. The incidence of pneumonia caused by macrolide-resistant M. pneumoniae that harbors point mutations has been increasing since 2000, particularly in Korea, Japan, and China. The marked increase in macrolide-resistant M. pneumoniae pneumonia (MRMP) is partly attributed to the excessive use of macrolides. MRMP does not always lead to clinical nonresponsiveness to macrolides. Furthermore, severe complicated MRMP responds to corticosteroids without requiring a change in antibiotic. This implies that the hyper-inflammatory status of the host can induce clinically refractory pneumonia regardless of mutation. Empirical macrolide therapy in children with mild to moderate CAP, particularly during periods without M. pneumoniae epidemics, may not provide additional benefits over β-lactam monotherapy and can increase the risk of MRMP.

전투경찰에서 발생한 폐렴에 있어서 Macrolide와 2세대 Cephalosporin의 치료 효과에 대한 전향적 비교 연구 (A Prospective Study for Comparing the Effects of Macrolide and Second-generation Cephalosporin on the Treatment of Pneumonia among Combat Policemen)

  • 목 적 : 집단생활을 하는 젊은 성인에서 발생한 폐렴에 관한 연구는 매우 드물며, 국내 연구는 전무한 실정이다. 이에 저자들은 전경 대원에서 발생한 폐렴의 양상에 대해 알아보고 Macrolide와 2세대 Cephalosporin의 치료 효과를 비교하여 보고자 전향적 연구를 시행하였다. 방 법 : 2003년 1월부터 2004년 4월까지 국립 경찰병원을 방문한 52명의 폐렴 환자들을 무작위로 구분하여 Cefuroxime 또는 Azithromycin을 경구 투여하였다. 폐렴 원인균의 진단을 위해 각종 배양 검사와, Mycoplasma 및 Chlamydia 항체 검사를 시행하였고, 혈액 검사와 방사선 검사 등 환자에 대한 평가는 입원 이후 1주 간격으로 2주간 시행하였다. 치료시작 2주후, 항체검사를 반복하였다. 결 과 : 객담 배양 검사를 시행한 전체 52명의 환자 중 3예(5.8%)에서 양성소견을 보였으며, Streptococcus pneumoniae 2예, Klebsiella pneumoniae 1예가 배양되었다. Mycoplasma pneumoniae 항체 검사를 시행하였던 40예 중 양성으로 판정된 것은 20예(50.0%)였으며, Chlamydia pneumoniae의 경우 4예(10.8%)에서 양성 소견을 보였다. 임상 증세와 방사선 소견 모두 호전되어 치료 성공으로 간주될 수 있는 경우는 전체 52예 중 51예로서 전체적으로 98.1%의 치료 성공률을 보였으며, Azithromycin과 Cefuroxime의 치료 효과는 유의한 차이를 보이지 않았다(96.0% vs 100%, p>0.05). Azithromycin 군 환자 중 한 예에서 초기 항생제 투여 후 임상 증세 및 흉부 방사선 소견이 호전되지 않아 항생제 처방을 변경하였으며, 2세대 세팔로스포린을 정맥 투여한 뒤 호전되었다. 결 론 : 집단생활을 하는 젊고 건강한 성인에서 발생한 폐렴에서 비정형 병원균에 의한 폐렴이 대다수를 차지하였고, 그 치료를 위해 투여한 Macrolide와 2세대 Cephalosporin 간 치료효과의 차이는 보이지 않았다.

가와사끼병 환아에서 흉부 X-선 검사의 변화와 혈중 Tumor Necrosis Factor-α에 대한 연구 (Chest X-ray Findings and Serum Tumor Necrosis Factor-αLevels in Patients with Kawasaki Disease)

  • 목 적 : 가와사끼병에서 흉부 X-선 검사에서 변화가 나타나는 빈도, 변화 양상, 염증성 cytokine과의 관계를 알아보고자 하였으며, 가와사끼병 환아에서 흉부 X-선 검사에서 이상 소견이 있을 때 염증 정도를 예측할 수 있는지 알아보기 위하여 본 연구를 시행하였다. 방 법 : 2003년 1월부터 2004년 5월까지 이화의대 동대문병원 소아과에서 가와사끼병으로 진단된 환아 84명을 대상으로 흉부 X-선 검사 결과를 동일 방사선과 전문의가 판독하였다. 흉부 X-선에서 정상 소견을 보이는 군과 비정상 소견을 보이는 두군으로 나누어, 발병 당시 각 군의 관상동맥 확장 정도, 혈청 TNF-${\alpha}$, Mycoplasma 항체, 혈색소, 백혈구, 혈소판, 적혈구 침강속도와 C 반응 단백을 비교 연구하였다. 결 과 : 대상 환아 84명 중에서 흉부 X-선 검사에서 이상 소견을 보인 경우는 43명(51.2%)이었다. 흉부 X-선 검사에서 기관지주위둘러쌈(peribronchial cuffing) 22.4%, 그물결절음영 17.7%, 과도통기(hyperaeration) 8.2%, 폐포경화 3.5%, 흉막삼출 1.2%, 무기폐 1.2%에서 관찰되었다. 흉부 X-선 검사에서 정상 소견을 보인 군과 비정상 소견을 보인 두 군에서 평균 연령, 성별과 발열 기간은 유의한 차이가 없었으며, 혈색소, 백혈구, 혈소판, 적혈구 침강 속도와 C 반응 단백도 두 군에서 유의한 차이가 없었다. 양측 관상동맥 직경은 두 군에서 유의한 차이는 없었고, 혈중 TNF-${\alpha}$ 농도도 두 군간에 유의한 차이가 없었다. 비정상 소견을 보인 군에서 Mycoplasma 항체가를 측정한 결과 12.5%에서 1 : 320 이상의 양성을 보였다. 결 론 : 가와사끼병으로 진단된 환아에서 흉부 X-선 검사의 이상 유무로 염증의 심한 정도를 판단하는 것은 제한점이 있다. 가와사끼병 환아에서 Mycoplasma와의 관련성에 대해서는 향후 더 연구가 필요하리라 생각된다.
