Isolation and Characterization of Plant Growth Promoting Rhizobacteria From Button Mushroom Compost

  • An auxin-producing bacteria (strain 5-1) was isolated from button mushroom compost in Boryeong-Si, Chungcheongnam-Do. The 5-1 strain was classified as a novel strain of Enterobacter aerogenes based on chemotaxonomic and phylogenetic analyses. The isolated E. aerogenes 5-1 was confirmed to produce indole-3-acetic acid (IAA), one of the auxin hormones, using TLC and HPLC analyses. When the concentration of IAA was assessed by performing HPLC quantitative analysis, a maximum concentration of IAA of $109.9mgL^{-1}$ was detected in the culture broth incubated in R2A medium containing 0.1% L-tryptophan for 24 h at $35^{\circ}C$. Acidification of the culture was deemed caused by an increase of IAA because a negative relationship between IAA production and pH was observed. Supplementation with a known precursor of IAA production, L-tryptophan, appeared to induce maximal production at 0.1% concentration, but it reduced production at concentrations above 0.2%. To investigate the growth-promoting effects to crops, the culture broth of E. aerogenes 5-1 was used to inoculate water cultures and seed pots of mung bean and lettuce. In consequence, adventitious root induction and root growth of mung bean and lettuce were two times higher than those of the control.

아위느타리와 백령느타리의 종간교잡 품종 '크리미'의 육성 및 특성 (Breeding and characterization of 'Creamy', a new interspecific hybrid between Pleurotus ferulae and P. tuoliensis)

  • 국내에서 가장 많이 재배되고 있는 버섯은 느타리류로 일반느타리와 큰느타리(새송이) 두 품목이 대부분을 차지한다. 그 중에서 큰느타리는 자동화 시설이 확대되면서 생산량이 크게 증가하였으며, 저장성이 우수하여 수출되는 버섯의 주요 품목 중 하나로 자리잡았다. 그러나, 생산량이 점차 증가하면서 가격이 다소 하락하였고 이에 생산자는 큰느타리를 대체할 수 있는, 소비자는 버섯 시장의 선택의 폭을 넓힐 수 있는 새로운 버섯 품목 개발을 요구해 왔다. 이에 따라, 국립원예특작과학원에서는 중국 등 아시아에서 맛과 향이 우수하여 고품질로 생산되고 있는 백령느타리와 아위느타리를 종간교잡하여 '크리미'라는 품종을 개발하였다. 기존에 육성된 품종 및 수집된 유전자원을 재배하여 형태적 특성검정을 하였고, 아위느타리 품종 '비산2호(KMCC00430)' 와 백령느타리 유전자원 'KMCC00461'을 모본으로 선발한 뒤 각 모본으로부터 단포자를 분리하여 mono-mono 교잡하였다. 약 1,000 조합의 교잡을 하였고, 그 중 73계통이 교잡이 확인되어 재배시험 및 자실체특성조사를 수행하였다. 그 중 갓이 밝은 연백색이며 대가 굵고 곧아 품질이 우수한 '7773' 계통을 최종선발하였고 '크리미'라고 명명하였다. '크리미'의 균사생장 적온은 25~30℃이고, 자실체 생육온도는 16℃이다. 농가에서 대량생산시험 후 현장평가회 및 시식회를 개최해 본 결과, '크리미'의 수량이 대조구 '백황'보다 약 5% 증수되었고, 기존 아위느타리 품종 '비산2호'에 비해 식감과 맛이 더 우수하며, 아위×백령느타리 품종 '백황'보다 자실체 형태가 더 우수하여 고품질로 판매가 가능할 것이라는 평가를 얻었다. 큰느타리와 다르게 밝은 연백색인 갓을 가졌으며, 식감과 향이 더 우수한 아위×백령느타리 신품종 '크리미'가 앞으로 버섯 농가의 새로운 소득 창출과 버섯 시장의 품목 다변화에 이바지할 것을 기대해 본다.

한국(韓國)에서 9월(月)의 기상인자(氣象因子)가 송이 발생(發生)에 미치는 영향(影響)과 그 극복방안(克服方案) (Effects of Climatic Condition in September on Pine-mushroom(Tricholoma matsutake) Yield and a Method for Overcoming the Limiting Factors in Korea)

  • 송이 발생(發生)의 제한인자(制限因子)를 파악(把握)하고 극복방안(克服方案)을 개발(開發)하기 위(爲)하여 18개(個) 주요(主要) 송이산지(産地)의 생산량(生産量)과 송이 발생(發生)에 관여(關與)하는 주요(主要) 기상인자(氣象因子)(월별(月別) 강수량(降水量), 상대습도(相對濕度) 평균기온(平均氣溫), 최고(最高) 최저기온(最低氣溫), 지중온도(地中溫度), 일조율(日照率)의 관계(關係)을 지난 17년간(年間)(1978-1994)의 자료(資料)를 이용(利用)하여 분석(分析)하였다. 송이 발생(發生)에 가장 큰 영향(影響)을 미치는 인자(因子)는 지역별(地域別)로 다르지만 대체로 송이 발생기(發生期)인 9월(月)의 기상(氣象)이었으며 각(各) 인자(因子)들의 중요도(重要度)는 지역별(地域別)로 다르게 나타났다. 이 중(中)에서 1% 유의수준(有意水準)에서 $$r^2{\geq_-}0.41$$를 나타낸 인자(因子)는 9월(月) 평균최저기온(平均最低氣溫)으로 이 온도(溫度)가 높을수록 송이 발생량(發生量)이 많았으며, 이런 현상(現象)이 있는 지역(地域)은 영덕(盈德), 울진(蔚珍), 삼척(三陟), 봉화(奉化) 등(等) 9개(個) 지역(地域)이었다. 이들 지역(地域)에서는 식생정리(植生整理)를 통해 임내(林內)에 햇빛이 많이 들어 오도록 하여 균환(菌環) 주변(周邊)의 온도(溫度)를 높이면 송이 증수(增收)에 효과적(效果的)일 것으로 예상(豫想)되었다. 9월(月) 강수량(降水量)이 $$r^2{\geq_-}0.41$$인 지역(地域)은 남원(南原), 문경(聞慶), 상주지역(尙州地域)이었고, 이 지역(地域)에서는 송이 발생시기(發生時期)에 균환주변(菌環周邊)에 관수(灌水)하는 것이 효과(效果)를 거둘 것으로 기대(期待)되었다. 1994년(年)에는 가뭄으로 인(因)하여 1993년(年)에 버금가는 흉년(凶年)을 맞이하였는데, 문경(聞慶)과 거창지역(居昌地域)에서는 관수처리(灌水處理)를 통해 가뭄을 극복(克服)하여 송이를 증산(增産)할 수 있는 가능성(可能性)을 확인(確認)할 수 있었다.

Identification and genetic characterization of bacterial isolates causing brown blotch on cultivated mushrooms in Korea

  • Fluorescent bacteria were isolated from sporocarps that browned into various mushrooms during survey at places of the production in Korea. We examined the pathogenicity, biodiversity, and genetic characteristics of the 19 strains identified as Pseudomonas tolaasii by sequence analysis of 16S rRNA and White Line Assay. The results emphasize the importance of rpoB gene system, fatty acid profiles, specific and sensitive PCR assays, and lipopeptide detection for the identification of P. tolaasii. As a result of these various analyses, 17 strains (CHM03~CHM19) were identified as P. tolaasii. The phylogenetic analysis based on the 16S rRNA gene showed that all strains were clustered closest to P. tolaasii lineage, two strains (CHM01, CHM02) were not identified as P. tolaasii and have completely different genetic characteristics as a result of fatty acids profile, specific and sensitive PCR, lipopetide detection, rpoB sequence and REP-PCR analysis. Pathogenicity tests showed 17 strains produce severe brown discolouration symptoms to button mushrooms and watersoaking of sporophore tissue within three days after inoculation. But two strains did not produce discolouration symptoms. Therefore, these two strains will be further investigated for correct species identification by different biological and molecular characteristics.

  • 버섯 세균갈성색무늬병원균인 Pseudomonas tolaasii에 대한 독소저해균으로 보고된 Pseudomonas sp. HC1 균주의 배양적 특성과 최적 배양을 위한 대량배지를 선발하였다. CH1균주의 생육온도는 $20{\sim}40^{\circ}C$, pH는 5.0~11.0까지 넓은 범위에서 왕성한 생육을 보였다. 대량배양을 위한 효율적인 영양원 선발을 위하여 기본배지에 탄소원 fructose 등 18종, 무기질소원 $NH_4Cl$ 등 6종, 유기질소원 peptone 등 6종 그리고 아미노산 asparagine 등 11종을 각각 1%씩 첨가하였고, 무기염류 13종을 1 mM 농도로 첨가하여 각각에 대한 생육에 미치는 영향을 조사하였다. 또한 선발된 각각의 영양성분들에 대한 최적 농도를 조사하기 위하여 각각의 성분을 0.1%에서 4.0%까지 배지에 첨가하여 배양후 생육정도를 조사하였다. 그 결과, 대량배양을 위한 생육최적조건은 온도 $20^{\circ}C$, pH5, 탄소원 0.9% dextrine, 유기질소원 1.5% yeast extract, 무기질소원 0.5% $(NH_4)_2HPO_4$, 아미노산 3.0% cysteine, 무기염류 4 mM $FeCl_3$에서 왕성한 생육을 보였다.

참나무류(類) 칩을 이용(利用)한 표고버섯 재배(栽培) (On the Mushroom Cultivation of Oak(Quercus spp.) Chip)

  • To increase the production of mushrooms and the economic efficiency of mushroom cultivation, the pinchips(PC) and sawdusts(SD) of three Korean oak species were tested as cultivating media for Lentinus edodes with varing the composition of the media. The results are as follows. 1. Organic acids(tannic acid and citric acid), which were added in the PC medium to enrich the quality of mushrooms, did not increase the growths of the mycelia and the quality. 2. The mushrooms were able to be harvrested from six months after beginning the cultivation. No significant differences were found among the qualities of the mushrooms produced by various treatments. 3. The production rate of mushroom was relatively high with, 28.4~29.3% (700~730gr./2,500gr pack). The net profit estimated was about 1,450 won per pack.

Automation of Solid-state Bioreactor for Oyster Mushroom Composting

  • This study focused on the production of high quality compost for the growth of aero-thermophilic fungi, which has a promoting effect on the growth rate and production of oyster mushrooms. The automated solid-state bioreactor system was designed on the basis of a Three-Phase-One system, which controls the serial steps of prewetting, pasteurization and fermentation processes. High numbers of thermophilic fungi and bacteria were recovered from the mushroom composts prepared by this solid-state bioreactor. The rates of composting process were depended on physical as well as chemical factors. Among these factors, the parameters of moisture content and temperature were found to be particularly important. In our automated system, constant levels of moisture content, temperature and ventilation via mixing were provided by a centralized control apparatus including PLC, water tank and water jacket systems. These features induced higher microbiological activity of aero-thermophiles.

담자균류 목질섬유소 분해효소의 특성과 산업적 이용 (Industrial applications and characteristics of lignocellulolytic enzymes in Basidiomycetous fungi)

  • Basidiomycetous fungi are one of the most potent biodegraders because many of its species grow on dead wood or litter, in environments rich in lignocellulose. For the degradation of lignocellulose, basidiomycetes utilize their lignocellulytic enzymes, which typically include laccase (EC 1.10.3.2), lignin peroxidase (EC 1.11.1.14), xylanase (EC 3.2.1.8), and cellulase (EC 3.2.1.4). In recent years, the practical applications of basidiomycetes have ranged from the textile to the pulp and paper industries, and from food applications to bioremediation processes and industrial enzymatic saccharification of biomass. Recently, spent mushroom substrates of edible mushrooms have been used as sources of bulk enzymes to decolorize synthetic dyes in textile wastewater. In this review, the occurrence, mode of action, general properties, and production of lignocellulytic enzymes from mushroom species will be discussed. We will also discuss the potential applications of these enzymes.

Fungal and mushroom hydrophobins: A review

  • Hydrophobins are surface active proteins that are produced by filamentous fungi including mushrooms. Their ability to self-assemble into an amphipathic membrane at any hydrophilic-hydrophobic interface is most intriguing. These small secreted proteins comprise of eight conserved cysteine residues which form four disulfide bridges and an extraordinary hydrophobic patch. Hydrophobins play critical roles in fungal (and/or mushrooms) growth as structural components and in the interaction of fungi and mushrooms with the environment. The biophysical and biochemical properties of the isolated proteins are remarkable, such as strong adhesion, high surface activity and the formation of various self-assembled structures. With the increasing demands of hydrophobins from fungi and mushroom sources, production and purification in large scale is under challenge. Various applications, ranging from food industries, cosmetics, nanotechnology, biosensors and electrodes, to biomaterials and pharmaceuticals are emerging and a bright future is foreseen.

Comparison of Cultivation, Mushroom Yield, and Fruiting Body Characteristics of Lentinula edodes Strains according to the Inoculation Method

  • The cultivation in sawdust media, mushroom productivity, and fruiting body characteristics of Lentinula edodes strains NIFoS 2778 and NIFoS 3363 were compared according to the inoculation conditions. The cultivation period was 5% shorter when liquid spawn was used. Fruiting bodies were induced after 113 days of incubation on media inoculated with liquid spawn, and the cultivation period was 119 days on media inoculated with solid spawn. Mushroom productivity of NIFoS 2778 was the highest (661.4 g) when 36 mL of liquid spawn was used. For NIFoS 3363, mushroom production was higher under liquid inoculation conditions when the same amount of liquid and solid spawns were used. The mushroom characteristics of the two strains were not significantly different, except for gill width and stipe diameter.
