Optimum Conditions for the Culture of Hericium erinaceum in a Jar Fermenter with the Addition of Ginseng Extract in the Liquid Medium (수삼추출물을 이용한 노루궁뎅이버섯 균사체의 jar fermenter에서의 발효조건 최적화)

  • To obtain functional materials from a submerged culture of Hericium erinaceum, a suitable basal medium for flask culture was screened and the optimal culture conditions in a jar fermenter were investigated with the addition of ginseng extracts (GE) to the basal liquid medium. Of all tested basal liquid media, the mushroom complete medium (MCM) supplemented with 0.5% of GE produced the highest mycelial dry weight (MDW) of 5.91 g/L in the flask, which reached a plateau at $25^{\circ}C$, pH 5.5 after 10 days. The submerged culture conditions for the mass production of mycelia in a 50 L jar fermenter were also optimal at $25^{\circ}C$, pH 5.5, 120 rpm agitation speed and 0.4 vvm aeration rate. Under these conditions, the maximum MDW was produced, which reached a value of 4.28 g/L within 5 days. When we investigated the effects of the amount of GE in the MCM on the production of MDW in the jar fermenter, the addition of 5% GE (HE-GE-5) under the optimal culture conditions produced the maximum MDW (4.93 g/L). In addition, the crude polysaccharide of HE-GE-5 contained mainly neutral sugars (63.2%) with considerable amounts of uronic acid (19.3%) and a small amount of proteins (8.8%) and it had potent immunostimulation properties.

  • 본 연구는 mushroom complete medium(MCM) 액체배지에 수삼 추출물(GE, $65^{\circ}Bx$)을 첨가하여 면역활성이 증진된 노루궁뎅이버섯(Hericium erinaceum) 균사체를 배양하고, 균사체로부터 활성다당성분을 분획하고자 하였다. MCM에 대하여 GE를 5, 10과 15%(v/v) 첨가한 액체배지에서 균사체를 배양하고, 각각의 조다당획분(HE-GE-5-CP, HE-GE-10-CP와 HE-GE-15-CP)으로 분획하여 면역활성을 측정한 결과, HE-GE-10-CP는 HE-GE-5-CP와 HE-GE-15-CP보다 높은 활성을 나타내었으며, GE를 첨가하지 않은 MCM에서 배양된 균사체 조다당획분(HE-CP)보다 유의적으로 증진된 면역활성을 나타내었다. 또한, HE-GE-10-CP의 DEAE-Sepharose CL-6B 분획물 중 가장 높은 활성을 나타낸 HE-GE-10-CP-II획분은 대조군인 HE-CP의 어떠한 획분보다도 유의적으로 높은 면역활성과 암 전이 억제활성을 나타내었다. 한편, 활성획분인 HE-GE-10-CP-II는 arabinose, rhamnose, galactose, glucose와 uronic acid(molar ratio; 0.34:0.26:0.99:1.00:0.39)로 구성되어 있으나, 대조군인 HE-CP의 동일용매 용출획분으로서 HE-GE-10-CP-II보다는 활성이 낮은 HE-CP-II는 fucose, mannose, galactose와 glucose(molar ratio; 0.32:0.55:1.00:0.96)를 함유하여 다른 구성당 분포를 나타내었다. 따라서 노루궁뎅이버섯 균사체 액체배양에서 수삼 추출물 첨가는 균사체의 구성당 변화를 통한 면역활성 증진에 관여하는 것으로 사료되어 기능성 소재 개발에 유용할 것으로 사료된다.

Optimal Production and Characterization of Laccase from Fomitella fraxinea Mycelia (Fomitella fraxinea 균사체로부터 Laccase의 최적생산 및 효소적 특성)

  • The culture conditions were investigated to maximize the production of laccase from Fomitella fraxinea mycelia. Among the tested media, mushroom complete medium (MCM) showed the highest production of the enzyme. The optimum culture medium was 2% dextrose, 0.4% $(NH_4)_{2}HPO_4$, 0.05% $Na_{2}HPO_{4}{\cdot}7H_{2}O$, and 0.05% KCl as carbon, nitrogen, phosphorus, and inorganic salt sources respectively. SDS-PAGE followed by laccase activity staining using 2,6-djmethoxyphenol as the substrate was performed to identify the laccase activity under culture conditions studied. Zymogram analysis of the culture supernatant showed a laccase band with a molecular mass of 50 kDa. The enzyme production from F. fraxinea was reached to the highest level after the cultivation for 10 days at $25^{\circ}C$ and initial pH 8. The enzyme activity of the culture supernatant was most active at $50^{\circ}C$ and pH 5.

Studies on Protoplast Isolation of Pleurotus cornucopiae (노랑느타리버섯의 원형질체(原形質體) 분리(分離)에 관한 연구(硏究))

  • The optimal conditions for high yields of mycelial protoplasts from P. cornucopiae were established. The concentraion of enzyme system containing Novozym 234, ${\beta}-D-glucanase$ and ${\beta}-glucuronidase$ was $5mg\;ml^{-1}$ each. The osmotic stabilizer most effective for protoplast isolation was O.6 M sucrose. The optimal reaction time of mycelium with the lytic mixture was 90 min in a shaking condition at 120 strokes $min^{-1}$. When the myelium of P. cornucopiae was cultured for 4 days on mushroom complete medium at $28^{\circ}C$, the formation of protoplast was effective. When the pH of the digestion mixture with O.6 M sucrose as stabilizer varied between pH 4.0 and 7.0, the production of protoplasts was effective in phosphate buffer (pH 6.2) and Na-maleate buffer (pH 5.0). Generally, phosphate buffer was more effective for protoplast isolation than Na-maleate buffer, but 0.6 M sucrose osmotic stabilizer without adjusting pH was most effective. Using these conditions, protoplasts from P. cornucopiae were obtained at a ratio $1{\times}10^7\;ml^{-1}$.

Grouping of Ganoderma strains based on cultural characteristics and fruitbody morphology (영지버섯 수집균의 배양적 특성 및 자실체 형태에 따른 구분)

  • To establish a genetic relationships of collected Ganoderma strains, mycelium growth according to media and temperature, colony morphology, chlamydospore formation and fruitbody morphology were investigated. For the identification of optimal growth conditions of the strains, five different growth media and four different temperature were tested. GCM (Ganoderma complete medium) at $30^{\circ}C$ was the most effective for mycelial growth of 68 strains with more or less variation. The strains were divided into 28 groups based on their colony shapes, and most of them belong to CM3 or CM8 group. Chlamydospores were observed in the mycelia of 16 strains including ASI 7022 on microscope, but not in most G. lucidum domestic strains, which showed relatively lagging growth on $35^{\circ}C$ in mycelial growth experiment. These results were not similar to those of G. lucidum but those of G. tsugae imported from USA. The strains were cultivated on oak sawdust media to see their fruit body formation. Ninety-seven among 115 strains formed fruitbodies in sawdust cultivation. They showed two forms of fruitbodies, 89.7% of flat type or 10.3% of antler type, although these shapes can be affected by $CO_2$ concentrations. These results suggest that the native strains formerly considered to belong to G. lucidum have to be re-classified with further study.

Immune Enhancement of Polysaccharide from Submerged Culture with Phellinus linteus in the Medium Supplemented with Ginseng Extract (수삼추출물 첨가 혼합배지에서 조제된 상황 균사체 심부배양물 다당획분의 면역활성 증진)

  • Crude polysaccharide (CP) was fractionated from the submerged culture (containing both mycelia and culture broth, SC) with Phellinus linteus (PL) in mushroom complete medium (MCM) supplemented with ginseng extract ($65^{\circ}$Bx, GE) to enhance the immune activity. PL-GE-15-CP from SC cultivated in MCM supplemented with GE-15% (v/v, a ratio of MCM volume to GE) showed significantly higher macrophage stimulation (1.45 fold of the saline control at $100{\mu}g$/mL) than PL-GE-5 and 10-CP with GE-5 and 10%, or PL-CP from SC without GE. The potent intestinal immune system modulating activity through Peyer's patch was also obtained by PL-GE-15-CP (1.46 fold). When PL-GE-15-CP further fractionated on DEAE-Sepharose CL-6B (Cl- form), PL-GE-15-CP-II was the significantly higher than others from PL-GE-15-CP or PL-CP on macrophage stimulation, interleukin (IL)-12 production and intestinal immune system modulation (1.54, 3.96 and 1.56 fold, respectively). PL-GE-15-CP-II also had higher anti-metastatic activity against colon 26-M3.1 carcinoma cell (57.3% inhibition of tumor control, $200{\mu}g$/mouse) rather than PL-CP-II. This active fraction (PL-GE-15-CP-II) mainly contained neutral sugar (82.45%) and uronic acid (12.99%), and component sugar analysis showed that PL-GE-15-CP-II consisted mainly of uronic acid, Ara, Man, Gal and Glc (molar ratio of 0.52:0.97:0.63:1.00:0.54). Furthermore, the activity of GE culture was higher compared with culture without GE, indicating that GE helped to enhance the immune activity of P. linteus; also, it is assumed that the polysaccharide plays an important role in immune enhancement.

  • Positron emission tomography (PET) as the functional component of current hybrid imaging (like PET/CT or PET/MRI) seems to dominate the horizon of medical imaging in coming decades. $^{18}$Flourodeoxyglucose ($^{18}FDG$) is the most commonly used probe in oncology and also in cardiology and neurology around the globe. However, the major capital cost and exorbitant running expenditure of low to medium energy cyclotrons (about 20 MeV) and radiochemistry units are the seminal reasons of low number of cyclotrons but mushroom growth pattern of PET scanners. This fact and longer half-life of $^{18}F$ (110 minutes) have paved the path of a centralized model in which $^{18}FDG$ is produced by commercial PET radiopharmacies and the finished product (multi-dose vial with tungsten shielding) is dispensed to customers having only PET scanners. This indeed reduced the cost but has limitations of dependence upon timely arrival of daily shipments as delay caused by any reason results in cancellation or rescheduling of the PET procedures. In recent years, industry and academia have taken a step forward by producing low energy, table top cyclotrons with compact and automated radiochemistry units (Lab-on-Chip). This decentralized strategy enables the users to produce on-demand doses of PET probe themselves at reasonably low cost using an automated and user-friendly technology. This technological development would indeed provide a real impetus to the availability of complete set up of PET based molecular imaging at an affordable cost to the developing countries.

Some Factors Affecting the Protoplast Release from Pleurotus ostreatus (느타리버섯 균사(菌絲)의 원형질체(原形質體) 나출(裸出)에 관한 연구(硏究))

  • Some factors affecting the protoplast release from mycelia of Plurotus ostreatus using commercial lytic enzymes were investigated. The highest yields of the protoplast were obtained from four days old mycelia grown in mushroom complete medium. The solution of 0.8M $MgSO_4$, or KCI showed good results as the osmotic stabilizer for releasing the protoplast. Novozym 234 was the most effective among commercials tested. The concentration of the enzyme and pH of the enzyme solution were optimal at 15mg/ml and $5.5{\sim}6.0$ for the protoplast release, respectively. Mycelial digestion was optimal at about $28^{\circ}C$ and was better in the reciprocal shaking bath (75 oscillations/min) than the stationary culture.

  • This experiment was undertaken to investigate proper conditions for protoplast isolation from strains of Agaricus bisporus, Flammulina velutipes, Lentinus edodes, Pleurotus florida, Pleurotus ostreatus, Pleurotus sajor-caju and Volvariella volvacea. Combination of ${\beta}-D-glucanase$, novozym 234 and snail enzyme with 0.6M $MgSO_4$ in 0.05M Na-maleate buffer, pH 5.8 was the most effective for isolation of protoplasts. High yields of protoplasts were obtained from $4{\sim}5$ days old mycelia of P. florida and P. ostreatus on mushroom complete agar medium. Protoplasts were also obtained in good yield from other fungi but A. bisporus and V. volvacea. In the latter two cases protoplast release was observed; however, the yield was much lower than those of the other fungi.

Optimun Conditions for Production of Mycelia and Extracellular Polysaccharide from Inonotus obliquus and Their Immunomodulating Acitivities (차가버섯의 균사체 및 세포외다당체의 생산조건과 면역활성)

  • Optium conditions for the production of mycelia and extracellular polysaccharide (EXPS) from submerged mycelial culture of Inonotus obliquus and their immunomodulating activities were investigated. The optmium production of mycelia and EXPS from I. obliquus was observed in mushroom complete medium (MCM). The optimum pH, temperature, and agitation speed for the production of mycelia and EXPS were 5.5, $25^{\circ}C$, and 150 rpm, respectively. The culture period for maximum production of mycelia (10.89 g/l) and EXPS (1.25 g/l) in shake flask cultivation was 11 days. The anticomplementary activity of intracellular polysaccharide (INPS) and EXPS form I. obliquus increased in a dose-dependent manner. Lysosomal enzyme activity of EXPS and INPS increased by 2.0- and 2.2-fold at $100{\mu}g/ml$ concentration, respectively, compared to the control group.
