보툴리눔 A형 독소가 저작효율에 미치는 영향 (The Influence of Botulinum Toxin Type A Masticatory Efficiency)

  • 보툴리눔 독소는 Clostridium botulinum이라는 박테리아가 만들어내는 독소를 의료용으로 희석, 정제한 약제로서 이 독소가 시냅스 전 신경말단에 부착되어 신경근접합부에서 아세틸콜린의 분비를 억제함으로써 근육의 수축을 차단하는 역할을 한다. 보툴리눔 독소 주사 요법은 구강안면 분야에서 저작근의 근경축, 근경련, 운동이상증, 측두하악관절질환, 근막통증, 이갈이, 특발성 교근비대증 또는 만성편두통을 비롯한 두통 등 다양한 질환에서 가역적, 보존적 치료법으로 활발히 사용되고 있다. 사용 범위가 넓어지면서 교근이나 측두근 같은 저작근 부위에 주사를 하는 경우가 많아지고 있으며 그러한 경우 약제자체의 약리학적 작용에 의해 일시적인 근마비 및 근위축에 따라 저작기능에 변화가 생기게 된다. 하지만 지금까지 보툴리눔 독소 주사치료의 효과나 활용에 대한 연구만이 주를 이루었고 부작용 중의 하나인 저작기능 변화에 대한 연구는 거의 이루어지지 않았다. 몇몇 저작기능과 관련한 연구 또한 근전도 등을 이용한 정적인 상태의 저작력 감소에 대한 연구로, 실제로 동적인 저작 환경을 반영하는 저작 효율에 대해서는 현재까지 전혀 연구가 이루어지지 않은 실정이다. 이에 본 연구에서는 총 40명의 건강한 성인환자를 대상으로 보툴리눔 A형 독소 주사 요법을 교근 부위에만 시술 받은 사람 과 교근과 측두근 부위 모두에 시술 받은 사람 각각 20명씩 두 군으로 나누어 주사를 시행하고, 주사 전과 주사 후 4주, 8주, 12주째의 저작 효율의 변화를 주관적 평가방법인 식품섭취능 (Food Intake Ability, 이하 FIA)과 저작 능력에 대한 주관적 인식도 (Visual Analogue Scale, 이하 VAS), 객관적 평가 방법인 Mixing Ability Test를 시행하여 알아보았다. 그 결과 보툴리눔 A형 독소 주사 후 4주 째에 저작효율의 뚜렷한 감소가 나타났으며 이후 서서히 회복되는 경향을 보였다. 주사부위를 달리하여 본 결과 교근과 측두근에 모두 주사를 시행하는 경우, 교근에만 주사를 시행하는 경우에 비해 주관적, 객관적인 값 모두 낮게 나타났다. 주관적인 값에 있어서 4주 째 유의하였으나 이후에는 유의하지 않았으며 객관적인 값은 전 기간에 걸쳐 유의하지 않았다. 실험에서 정한 주사 용량에 대해서는 측두근까지 주사가 이루어지더라도 교근만 주사를 시행한 사람과 비교하여 저작효율의 큰 차이가 없는 것으로 보인다. 또한 식품성상에 따라 저작 효율을 살펴본 결과 보툴리눔 A형 독소 주사가 단단하고 질긴 음식 섭취에 더 큰 영향을 미치는 것으로 보이며 무르고 약한 음식의 섭취 시에도 유의할만한 영향을 받는다는 사실을 확인 할 수 있었다.

Synergistic Effects of Dietary Vitamins C and E on Methylmercury-Induced Toxicity in Juvenile Olive Flounder Paralichthys olivaceus

  • This experiment was conducted to evaluate the synergistic effects of vitamin C and E on methylmercury (MeHg) toxicity in juvenile olive flounder Paralichthys olivaceus. In a $3{\times}3$ factorial design, 9 experimental diets containing three different vitamin C (0, 200 or 400 mg/kg diet in the form of l-ascorbyl-2-monophosphate) and vitamin E (0, 100 or 200 mg/kg diet in the form of dl-${\alpha}$-tocopheryl acetate) levels with the Hg toxicity level (20 mg/kg diet in the form of MeHg) were formulated. Triplicate groups of fish averaging $2.3{\pm}0.05g(mean{\pm}SD)$ were fed one of the 9 diets in a flow through system for 8 weeks. Fish fed 400 mg vitamin C/kg diet with 100 or 200 mg vitamin E/kg diet showed significantly (P < 0.05) higher weight gain (WG) than did fish fed the other diets. Fish fed 400 mg vitamin C/kg diet at all vitamin E levels and those which fed vitamin C and E equally at a rate of 200 mg/kg diet showed significantly (P < 0.05) higher feed efficiency (FE), specific growth rate (SGR) and protein efficiency ratio (PER) than did fish fed the other diets. Fish fed 200 and 400 mg vitamin C/kg diet exhibited significantly (P < 0.05) lower Hg concentration in their muscle as well as kidney than did fish fed the other diets. Therefore, these results clearly indicated that the synergistic effects of these two vitamins on MeHg toxicity by supplementing dietary vitamin C (200 and 400 mg/kg diet) with vitamin E (100 and 200 mg/kg diet) in juvenile olive flounder.

  • Carp, Cypinus carpio, has been esteemed as one of the most tasty fresh water fishes in Korea. But little study on its food has been reported. The composition of fatty acids, amino acids, nucleotides and their related compounds as chemical component of carp muscle were analyzed by gas-liauid chromatography, amino acid autoanalyzer and high speed liauid chromatography. The fatty acids of the carp muscle lipid consisted of the large amount of $C_{18}\;:\;2,$ $C_{18}\;:\;1,$ $C_{18}\;:\;3,$ $C_{16}\;:\;0,$ $C_{16}\;:\;1,$ acids, small amount of $C_{16}\;:\;2,$ $C_{18}\;:\;0,$ $C_{18}\;:\;4,$ $C_{14}\;:\;0,$ $C_{14}\;:\;1,$ $C_{12}\;:\;0,$ acids, and $C_{13}\;:\;1,$ $C_{13}\;:\;0,$ $C_{20}\;:\;1,$ $C_{17}\;:\;0,$ $C_{15}\;:\;1,$ $C_{17}\;:\;1,$ $C_{22}\;:\;1$ acids were smaller. In fresh carp, the results showed that inosine $17.25{\mu}mole/g$, hypoxanthine $10.06{\mu}mole/g$ were dominant and the content of GMP, IMP, AMP, UMP, CMP were 1.56, 1.36, 0.92, 0.49, $0.34\:{\mu}mole/g$ on dry base, respectively. Histidine and lysine were dominant amino acids in carp extract, having 49.4% (214.6mg%), 33.9% (147.1mg%) of total free amino acid contents, respectively, but the content of glycine, serine, alanine and glutamic acid were low, and arginine, aspartic acid, threonine, proline, valine, methionine, isoleucine, phenylalanine were detected in trace amount. The content of amino acid composition showed that glutamic acid, aspartic acid, lysine, leucine, alanine, valine and arginine were the most abundant amino acids, while such amino acids as glycine, isoleucine, phenylalanine, threonine, histidine, methionine, serine and tyrosine were low, and proline was detected in trace amount.

과도한 운동 스트레스에 대한 숙성생강 추출물의 방어효과 (Protective Effects of Aged Ginger Extracts on Excessive Exercise-induced Stress)

  • 생강의 운동 스트레스에 대한 방어효과를 알아보기 위해 동결건조, 열풍건조, 숙성한 생강을 제조하여 추출물을 근육세포에 H2O2와 함께처리한 결과 생강을 숙성한 시료는 숙성하지 않은 시료에 비하여 우수한 근육세포 보호효과를 나타내었다. 또한 숙성생강 추출물을 마우스에 투여시 운동으로 인한 혈액내 LDH, lactate, GOT 증가를 억제시키는 경향을 나타내었다. 따라서 숙성생강을 섭취시 과도한 운동으로 인한 스트레스에 대한 방어효과를 기대할 수 있을 것으로 사료된다. 이러한 결과를 토대로, 향후 생강의 숙성 온도 및 시간에 따른 운동 스트레스 방어효과를 측정하여 가장 높은 활성을 나타내는 최적의 숙성 조건을 확립하는 연구를 진행할 예정이다.

고불포화 지방식이가 흰쥐의 Plasma High Density Lipoprotein Cholesterol 량과 혈청 및 조직 내의 지방성분에 미치는 영향 (Effects of Dietary Polyunsaturated Fat on HDL - Cholesterol, Total Cholesterol and Triglyceride in Plasma and Tissues of Adult Rats)

  • The study was designed to observe the effects of dietary intake of three different levels of polyunsaturated to saturated fatty acid (P/S) ratio on plasma HDL - cholesterol and triglyceride in plasma and tissues of adult rats. Rats were assigned into the three diet groups, composed of protein 24%, carbohydrate 46%, and fat 30% of total caloric intake. However, the P/S ratio of fat was adjusted by using beef tallow, corn oil and perilla oil to give 0.2, 1.1 and 6.0. All groups were fed ad libitum for 4 week. Plasma cholesterol level was decreased but net significantly with increasing P/S ratio of dietary fat, and showed significant negative correlation coefficient. Plasma TG level was also decreased with increasing PUFA level of dietary fat. However, the effect of P/S ratio (6.0) was not greater than that of P/S ratio (1.1) on plasma TG concentration. This implied that high level of PUFA was not required to change plasma TG level as much in the case of HDL-cholesterol. Plasma HDL-cholesterol was significantly increased only in the group of P/S ratio 6.0 which suggested that high level of dietary PUFA was required to increase HDL level. There was a decreasing trend in the levels of total cholesterol and TG per g liver with increasing P/S ratio of dietary fat. There was no effect in the levels of TG per g skeletal muscle by P/S ratio but the level of cholesterol per unit skeletal muscle was rather increased with increased P/S ratio.

소의 혈액 및 근육 중 choline 농도 분석을 위한 효소측정법의 적용기법의 개발 (Application of Enzymatic method to Determine Choline Concentration in Bovine Blood and Muscle)

  • 콜린은 정상적인 세포벽의 기능, 아세틸콜린의 합성, 지질의 운송, 그리고 매칠기의 공급원으로서 중요한 유기화합물이다. 생물학적인 조직과 식품들 중에 있는 콜린의 분석을 위해 많은 기기분석법들 사용되고 있다. 그러나 이러한 기기분석법들은 고가의 장비를 필요로 하며, 조작이 어렵고, 많은 시간을 필요로 한다. 본 연구에서는 우유 중의 콜린함량을 분석하는데 사용되는 효소측정법을 응용하여 신속하게 소의 혈액과 근육내에 함유된 콜린을 측정할 수 있는 적용기법을 확립하였다. 소의 혈청과 근육에 콜린 표준용액을 spike하여 얻은 표준곡선들은 직선성을 나타내었으며, 0.994 이상의 상관계수를 보였다. 소의 혈청과 근육에서의 회수율은 70.6-85.2%를 보였다. 본 연구를 통하여 확립된 콜린분석법은 생체시료와 식품에 함유된 콜린 함량분석에 용이하게 적용할 수 있을 것으로 기대된다.

  • 뱀장어의 동결저장 및 해동후에 일어나는 섬유의 변화를 현미경적으로 관찰한 결과를 요약하면 다음과 같다. 1. 동결 1개월 후의 조직은 근세포내외에 수결정이 많이 생겼으며 $-40^{\circ}C$ 저장한 것은 세포내에 수결정이 생성하고, $-20^{\circ}C$로 저장한 것은 세포외에 수결정이 생겼다. 2. 해동후의 피하지방층은 저장온도에 관련없이 별 변화가 없었으나, 근육조직은 높은 온도($-20^{\circ}C$)로 저장한 것이 근세포의 복원도 불충분하고 탈수변형이 심하였다. 3. 근육파쇄편을 현미경 관찰한 결과, 동결한 것은 투명감이 소실되고, 건조한 것은 변형수축되며, 가열한 것은 수분유리 및 변형이 일어난다는 것을 알 수 있었다.

Short-term protein intake increases fractional synthesis rate of muscle protein in the elderly: meta-analysis

  • The precise effects of protein intake on fractional synthesis rate (FSR) of muscle protein are still under debate. The sample size of these studies was small and the conclusions in young and elderly subjects were inconsistent. To assess the effect of dietary protein intake on the FSR level, we conducted a meta-analysis of controlled protein intake trials. Random-effects models were used to calculate the weighted mean differences (WMDs). Ten studies were included and effects of short-term protein intake were evaluated. In an overall pooled estimate, protein intake significantly increased the FSR (20 trials, 368 participants; WMD: 0.025%/h; 95%CI: 0.019-0.031; P < 0.0001). Meta-regression analysis suggested that the protein dose was positively related to the effect size (regression coefficient = 0.108%/h; 95%CI: 0.035, 0.182; P = 0.009). A subgroup analysis indicated that protein intake significantly increased FSR when the protein dose was ${\leq}$ 0.80 g/kg BW (16 trials, 308 participants; WMD: 0.027%/h; 95%CI: 0.019-0.031; P < 0.0001), but did not affect FSR when the protein dose was > 0.80 g/kg BW (4 trials, 60 participants; WMD: 0.016%/h; 95%CI: 0.004-0.029; P = 0.98). In conclusion, this study is the first integrated results showing that a short-term protein intake is effective at improving the FSR of muscle protein in the healthy elderly as well as young subjects. This beneficial effect seems to be dose-dependent when the dose levels of protein range from 0.08 to 0.80 g/kg BW.

Guava (Psidium guajava L.) Powder as an Antioxidant Dietary Fibre in Sheep Meat Nuggets

  • This study was conducted to explore the antioxidant potential and functional value of guava (Psidium guajava L.) powder in muscle foods. Guava powder was used as a source of antioxidant dietary fibre in sheep meat nuggets at two different levels i.e., 0.5% (Treatment I) and 1.0% (Treatment II) and its effect was evaluated against control. Guava powder is rich in dietary fibre (43.21%), phenolics (44.04 mg GAE/g) and possesses good radical scavenging activity as well as reducing power. Incorporation of guava powder resulted in significant decrease (p<0.05) in pH of emulsion and nuggets, emulsion stability, cooking yield and moisture content of nuggets while ash and moisture content of emulsion were increased. Total phenolics, total dietary fibre (TDF) and ash content significantly increased (p<0.05) in nuggets with added guava powder. Product redness value was significantly improved (p<0.05) due to guava powder. Textural properties did not differ significantly except, springiness and shear force values. Guava powder was found to retard lipid peroxidation of cooked sheep meat nuggets as measured by TBARS number during refrigerated storage. Guava powder did not affect sensory characteristics of the products and can be used as source of antioxidant dietary fibre in meat foods.

인진쑥의 scopoletin함량 분석 및 뇌혈류량에 미치는 영향 (Analysis of Scopoletin Content and Effect on Regional Cerebral Blood Flow)

  • In this study, Artemisia capillaris, which has been used as oriental medicine area and folks remedy, was investigated for effective substance (scopoletin) analysis. As an attempt to develop new functional beverage by using medicinal herb, Artemisia capillaris. Therefore we investigate the effect of scopoletin in Artemisia capillaris on vessel and regional cerebral blood flow (rCBF) of rats. Artemisia capillaris extract was significantly decreased vessel constraction and significantly increased rCBF. Artemisia capillaris has often been cited in medical literature for its medicinal effects. Although the Korean Food and Drug Administration indicated Artemisia capillaris as the In-Jin-Sook which was allowed as a food stuff. From the view point of the subjects who eat In-Jin-Sook products, most of them ingested it for a certain pharmacological efficacy rather than as a beverage or a food. According to this facts, we need develop functional foods using In-Jin-Sook. Also scopoletin, get known to smooth muscle relaxation content was 326.13 ug/g. These results demonstrate that Artemisia capillaris has pharmaceutical effects and balanced nutritional contents. In according with Artemisia capillaris has a sufficiency of the terms use of food stuff in functional foods.
